Tannic acid-crosslinked zein-lipase complex stabilized multichamber Pickering emulsion for efficient diacylglycerol production.

This study presents a food-grade, multi-chamber W/O/W high internal phase Pickering emulsion (HIPPE) system for interfacial catalysis, which stabilized by a zein nanoparticle (ZNP)-lipase complex crosslinked with tannic acid (TA). Experiments and molecular dynamics simulations confirmed that the cross-linking between TA, ZNPs, and lipase was predominantly mediated by hydrogen bonding, hydrophobic interactions, and π-π stacking. TA effectively modulated the zein-lipase interaction to form a stable, dispersed complex and substantially enhanced the thermal stability of the lipase. The multi-chamb

A Bioarmored Enzyme Cascade for Sustained Catalysis in Harsh Microenvironments.

Enzyme therapeutics operating in harsh physiological microenvironments are inherently constrained by rapid unfolding, proteolytic degradation, and acid-induced deactivation, especially in stomachs. These vulnerabilities severely limit catalytic longevity and practical efficacy, particularly for acetaldehyde detoxification in humans. Achieving durable enzymatic function in such hostile conditions requires a protective strategy that can physically armor enzymes while preserving molecular accessibility to substrates-an unresolved challenge in current biomaterial designs. Here, we introduce a bioa

Reshaping the Activity-Stability Landscape of a 1,2-Rhamnosyltransferase via Distal Mutational Engineering.

Rhamnosyltransferases are remarkable biocatalysts for the synthesis of rhamnosylated natural products with valuable physicochemical properties and bioactivities. However, their application is hindered by poor stability and low catalytic efficiency. Here, we achieved simultaneous enhancement of catalytic efficiency and stability of a 1,2-rhamnosyltransferase by a distal mutational engineering strategy. The variant M9 exhibited a 589.43-fold extension in half-life, a 2.5 °C increase in Tm, a 13.6 °C increase in T50, and an 8- to 763-fold increase in activity toward diverse flavonoids compared wi

Semi-Rational Engineering of Formate Dehydrogenase Reveals Loop Dynamics as a Key Determinant of Thermostability.

Formate dehydrogenase (FDH) catalyzes the reversible formate oxidation using NAD+ to yield carbon dioxide and NADH. FDH is widely utilized for cofactor regeneration, enabling the continuous supplying of NADH for chiral chemical biosynthesis. Herein, we employed semi-rational engineering to improve the thermostability of highly catalytic FDH from Bacillus simplex (BsFDH). Through two rounds of mutational screening, BsFDHQ125F variant was identified to exhibit significantly improved thermostability with a 25-fold increase in half-life at 60°C. Additionally, the BsFDHQ125F variant revealed signif

Highly enantioselective reduction of benzophenones by engineered Geotrichum candidum alcohol dehydrogenase.

Biocatalytic approaches have gained increasing attention as sustainable alternatives to metal-catalyzed asymmetric reductions of ketones to obtain enantiopure alcohols, important intermediates for pharmaceutical synthesis. For example, enzyme-catalyzed reduction of substituted benzophenone analogs to produce chiral diaryl methanols has attracted interest, as they are the key intermediates in the synthesis of antihistamines. However, benzophenone analogs are difficult to be reduced by enzymes due to steric hindrance. Moreover, the similarities between the two groups adjacent to the carbonyl gro

Enzyme-Directed Charge Redistribution Controls Substrate Regioselectivity in CO2 Fixation by Salicylic Acid Decarboxylase.

2,4-Dihydroxybenzoic acid is an important intermediate in agricultural and food chemistry. Salicylic acid decarboxylase catalyzes the carboxylation of resorcinol with CO2 to yield 2,4-dihydroxybenzoic acid. However, this process suffers from poor regioselectivity and low carboxylation activity. Here, we propose that the unique microenvironment created by the enzyme's substrate-binding pocket, referred to here as the enzyme field, induces charge redistribution in resorcinol, thus altering the enzymatic regioselectivity. Charge distributions at carbon atoms were calculated under 14 different enz

Mechanistic Insights Into the Formaldehyde Dehydrogenase-Catalyzed Reduction of Formate: A Quantum Chemical Study.

The multienzyme cascade conversion of CO2 to methanol is a promising route for sustainable chemical synthesis, yet the reduction of formate to formaldehyde remains its major bottleneck. Here, we use quantum chemical calculations to elucidate the catalytic mechanism of formaldehyde dehydrogenase from Burkholderia multivorans (BmFaldDH). The lowest-energy pathway initiates with formate activation via a proton-relay network involving His52, Ser49, and the NADH ribose, followed by a concerted yet asynchronous proton-hydride transfer to yield methanediol with a barrier of 16.0 kcal mol-1. l-Asparaginase Immobilized on Nanographene Oxide as an Efficient Nanobiocatalytic Tool for Asparagine Depletion in Leukemia Cells.

l-Asparaginase (l-ASNase) catalyzes the hydrolysis of l-asparagine, leading to its depletion and subsequent effects on the cellular proliferation and survival. In contrast to normal cells, malignant cells that lack asparagine synthase are extremely susceptible to asparagine deficiency. l-ASNase has been successfully employed in treating pediatric leukemias and non-Hodgkin lymphomas; however, its usage in adult patients and other types of cancer is limited due to significant side effects and drug resistance. Recent research has explored alternative formulations and delivery methods to enhance i

Functional Characterization of Residues Affecting the Catalytic Activity of Glucosyltransferase from Streptococcus mutans.

Dental caries is a multifactorial, biofilm-mediated disease primarily caused by Streptococcus mutans, a key etiological agent. This bacterium secretes extracellular enzymes known as glucosyltransferases (Gtfs), also termed glucansucrases, which play a pivotal role in the synthesis of exopolysaccharides through the metabolism of dietary sucrose. These exopolysaccharides provide binding sites for the attachment and colonization of other microorganisms, contributing to the initiation and progression of dental caries. This study investigates the catalytic mechanisms of glucosyltransferases from S.

Design and Fabrication of a Versatile Microreactor to Study Transaminase-Based Heterogeneous Biocatalysts in Flow.

Microfluidics is a very attractive discipline for implementing more versatile high-throughput screening methods to improve enzymes. However, state-of-the-art microfluidics set-ups are mainly devoted to screening enzymes in solution, while screening of enzyme immobilization protocols using microfluidics is scarce. In this work, a microreactor device is designed, fabricated and applied to test different heterogeneous biocatalysts with transaminase activity. This microsystem set-up can be operated in two different modes: packed-bed (PBR) and fluidized (FBR) microreactors without exchanging the sa

Ethyl lactate synthesis in organic media using a magnetic supported CALB.

The growing demand for sustainable chemical processes has spurred interest in enzymatic synthesis, particularly for valuable compounds like ethyl lactate. Traditional chemical methods often suffer from drawbacks, highlighting the potential of enzymatic catalysis using immobilized lipases. This study evaluated the performance of magnetic biocatalyst, prepared by immobilizing Candida antarctica lipase B (CALB) on magnetic nanoparticles, for the batch synthesis of ethyl lactate in hexane. Initial experiments using free CALB and commercial Novozym435 proved problematic due to enzyme denaturation a

Enhancing Alcohol Dehydrogenase Activity for the Efficient Synthesis of (S)-2-Chloro-1-(2,4-dichlorophenyl)ethanol Using Computer-Aided Combinational Mutagenesis.

(S)-2-chloro-1-(2,4-dichlorophenyl)ethanol ((S)-CPEO) is an important chiral precursor of the antifungal drug luriconazole. In this study, a mutant alcohol dehydrogenase, LkADHM0 from Lactobacillus kefir, was redesigned for the efficient synthesis of (S)-CPEO by using virtual saturation mutagenesis to assess beneficial site combinations. Five poorly conserved sites in the active pocket of the enzyme were identified via multiple sequence alignment with enzymes exhibiting high activity toward acetophenone derivatives. To stimulate potential synergies while minimizing the screening effort, the fi

Chanoclavine synthase operates by an NADPH-independent superoxide mechanism.

More than ten ergot alkaloids comprising both natural and semi-synthetic products are used to treat various diseases1,2. The central C ring forms the core pharmacophore for ergot alkaloids, giving them structural similarity to neurotransmitters, thus enabling their modulation of neurotransmitter receptors3. The haem catalase chanoclavine synthase (EasC) catalyses the construction of this ring through complex radical oxidative cyclization4. Unlike canonical catalases, which catalyse H2O2 disproportionation5,6, EasC and its homologues represent a broader class of catalases that catalyse O2-depen

Concentration of docosahexaenoic acid from tuna oil via a two lipase-catalyzed reaction.

Docosahexaenoic acid (DHA) was concentrated successfully in the glyceride fractions from tuna oil via a two-step enzyme reaction involving hydrolysis and ethanolysis. In the first step, Candida rugosa lipase-catalyzed hydrolysis was carried out to concentrate DHA in the glyceride fractions. The DHA content in the glyceride fraction after hydrolysis increased from 30% in the initial tuna oil to 46%. In the second step, Lipozyme RM IM-catalyzed ethanolysis was conducted with the reaction mixture from the first step to further concentrate DHA in the glyceride fraction. In this step, the reaction

Amino-Alkyl Group Dual-Functional Modification Synergistically Regulated Lipase-Carrier Interactions and Enhanced Phytosterol Ester Synthesis Efficiency.

Precisely controlling enzyme conformation to enhance catalytic performance is a highly sought-after yet challenging goal in the immobilization of biocatalysts. Excessively strong enzyme-carrier interactions can restrict enzyme dynamics and reduce catalytic efficiency, while excessively weak interactions may lead to enzyme leakage, thereby reducing reusability. In this study, we developed a novel strategy to finely regulate the interaction between the carrier and the enzyme through the adjustment of the ratio of amino and octadecyl functional groups. The expressed activity of the novel immobili

Understanding the Enzyme (S)-Norcoclaurine Synthase Promiscuity to Aldehydes and Ketones.

The (S)-norcoclaurine synthase from Thalictrum flavum (TfNCS) stereoselectively catalyzes the Pictet-Spengler reaction between dopamine and 4-hydroxyphenylacetaldehyde to give (S)-norcoclaurine. TfNCS can catalyze the Pictet-Spengler reaction with various aldehydes and ketones, leading to diverse tetrahydroisoquinolines. This substrate promiscuity positions TfNCS as a highly promising enzyme for synthesizing fine chemicals. Understanding carbonyl-containing substrates' structural and electronic signatures that influence TfNCS activity can help expand its applications in the synthesis of differ

Nitrogen reduction by the Fe sites of synthetic [Mo3S4Fe] cubes.

Nitrogen (N2) fixation by nature, which is a crucial process for the supply of bio-available forms of nitrogen, is performed by nitrogenase. This enzyme uses a unique transition-metal-sulfur-carbon cluster as its active-site co-factor ([(R-homocitrate)MoFe7S9C], FeMoco)1,2, and the sulfur-surrounded iron (Fe) atoms have been postulated to capture and reduce N2 (refs. 3-6). Although there are a few examples of synthetic counterparts of the FeMoco, metal-sulfur cluster, which have shown binding of N2 (refs. 7-9), the reduction of N2 by any synthetic metal-sulfur cluster or by the extracted form

High Regio- and Stereoselective Multi-enzymatic Synthesis of All Phenylpropanolamine Stereoisomers from β-Methylstyrene.

We present a one-pot cascade for the synthesis of phenylpropanolamines (PPAs) in high optical purities (er and dr up to >99.5 %) and analytical yields (up to 95 %) by using 1-phenylpropane-1,2-diols as key intermediates. This bioamination entails the combination of an alcohol dehydrogenase (ADH), an ω-transaminase (ωTA) and an alanine dehydrogenase to create a redox-neutral network, which harnesses the exquisite and complementary regio- and stereo-selectivities of the selected ADHs and ωTAs. The requisite 1-phenylpropane-1,2-diol intermediates were obtained from trans- or cis-β-methylstyrene b
