Perfluorooctanoic acid disrupts thyroid hormone biosynthesis by altering glycosylation of Na+/I- symporter in larval zebrafish.

Perfluorooctanoic acid (PFOA) is a well-known thyroid disruptor that has been found to induce hypothyroidism. However, the exact molecular mechanism by which PFOA reduces thyroid hormone levels remains unclear. In this study, we have discovered that PFOA disrupts the glycosylation process of the sodium/iodide symporter (NIS), which inhibits the translocation of NIS onto the plasma membrane of thyroid follicular cells. Our results also demonstrate that PFOA disrupts thyroid stimulating hormone (TSH)-dependent signaling pathways involved in cellular glycosylation, impairing NIS glycosylation and

Maternal exposure to legacy PFAS compounds PFOA and PFOS is associated with disrupted cytokine homeostasis in neonates: The Upstate KIDS study (2008-2010).

There is growing concern that exposure to per/polyfluoroalkyl substances (PFAS), persistent chemicals used widely to make consumer products water- or grease-proof, may alter immune function, leading to reduced vaccine response or greater susceptibility to infections. We investigated associations between two legacy PFAS (PFOA and PFOS) and infant cytokine levels measured in newborn dried bloodspots (NDBS) from a large population-based birth cohort in Upstate New York, to determine whether exposure to legacy PFAS is associated with variability in cytokine profiles in newborns. We performed adjus

Differential impact of per- and polyfluoroalkyl substances exposure on hypertension risk by kidney function and metabolic status.

BACKGROUND: Per- and polyfluoroalkyl substances (PFAS) are persistent environmental pollutants increasingly linked to hypertension. However, the modifying effects of kidney function and metabolic health remain unclear. This study examined the associations between serum PFAS concentrations and hypertension, with a particular focus on kidney function and metabolic status as potential effect modifiers. METHODS: Data were obtained from the Korean National Environmental Health Survey (KoNEHS, 2018-2020), which included adults aged 19 years or older with available serum PFAS, blood pressure, and ki

Bone cell differentiation and mineralization in wild-type and osteogenesis imperfecta zebrafish are compromised by per- and poly-fluoroalkyl substances (PFAS).

Perfluorinated compounds (PFAS) are well recognized toxic pollutants for humans, but if their effect is equally harmful for healthy and fragile people is unknown. Addressing this question represents a need for ensuring global health and wellbeing to all individuals in a world facing the progressive increase of aging and aging related diseases. This study aimed to evaluate the impact of perfluorooctane sulfonate (PFOS), perfluorooctanoic acid (PFOA) and perfluorohexanoic acid (PFHxA) exposure on development and skeletal phenotype using the osteogenesis imperfecta (OI) zebrafish model Chihuahua

Investigating the Mechanism of Neurotoxic Effects of PFAS in Differentiated Neuronal Cells through Transcriptomics and Lipidomics Analysis.

Per- and polyfluorinated alkyl substances (PFAS) are pervasive environmental contaminants that bioaccumulate in tissues and pose risks to human health. Increasing evidence links PFAS to neurodegenerative and behavioral disorders, yet the underlying mechanisms of their effects on neuronal function remain largely unexplored. In this study, we utilized SH-SY5Y neuroblastoma cells, differentiated into neuronal-like cells, to investigate the impact of six PFAS compounds─perfluorooctanoic acid (PFOA), perfluorooctanesulfonic acid (PFOS), perfluorodecanoic acid (PFDA), perfluorodecanesulfonic acid (P

Towards elevated perfluorooctanoic acid (PFOA) enrichment in water: Sequential liquid-liquid extraction pretreatment for ion chromatography detection.

The widespread detection of perfluorooctanoic acid (PFOA) in the environment has raised significant concerns. The standard PFOA analytical method relies on expensive solid-phase extraction (SPE) and liquid chromatography tandem mass spectrometry (LC-MS/MS) instruments, making routine use prohibitive. We herein proposed a cost-effective yet novel enrichment method for determining PFOA at ng L-1 level. This method entailed a two-step sample preparation process: firstly, PFOA was extracted and enriched using a forward-extraction under acidic conditions, followed by a backward-extraction and enric

Specific and potent inhibition of steroid hormone pre-receptor regulator AKR1C2 by perfluorooctanoic acid: Implications for androgen metabolism.

Per- and polyfluoroalkyl substances (PFAS) are ubiquitous environmental pollutants that are highly stable synthetic organofluorine compounds. One congener perfluorooctanoic acid (PFOA) can be detected in nearly all humans and is recognized as an endocrine disrupting chemical (EDC). EDCs disrupt hormone synthesis and metabolism and receptor function. One mechanism of steroid hormone action is the pre-receptor regulation of ligand access to steroid hormone receptors by aldo-keto reductases. Here we report PFOA inhibition of AKR family 1 member C2 (AKR1C2), leading to dysregulation of androgen ac

Perfluoroalkyl substance exposure is associated with asthma and innate immune cell count in US adolescents stratified by sex.

Exposure to perfluoroalkyl substances (PFAS) may be harmful to humans; however, previous studies have been inconsistent regarding the potential for PFAS-induced immunosuppresion. This study explored the relationship between PFAS exposure and risks of asthma, wheezing, and immunosuppression in 12-19 year-olds using the National Health and Nutrition Examination Survey (NHANES) data. Logistic regression models were used to reveal associations between serum PFAS levels and risks of asthma, wheezing, asthma attack, and emergency department visits. Pearson's correlation was used to determine the rel

Glioma is associated with exposure to legacy and alternative per- and polyfluoroalkyl substances.

Data on the occurrences of legacy and alternative per- and polyfluoroalkyl substances (PFASs) in glioma are scarce. It remains unclear if PFASs exposure is related to the prevalence of glioma. A total of 137 glioma and 40 non-glioma brain tissue samples from patients recruited from the Nanfang Hospital, South China were analyzed for 17 PFAS compounds. Perfluorohexanoic acid, perfluorooctanoic acid (PFOA), perfluorooctane sulfonate (PFOS), perfluorooctane sulfonamide (FOSA), and 6:2 chlorinated polyfluorinated ether sulfonate were frequently detected (> 60 %) in glioma. The total concentrations

Autocatalytic degradation of perfluorooctanoic acid in a permanganate-ultrasonic system.

An autocatalytic system, permanganate-ultrasonic (PM-US) system, was applied to degrade perfluorooctanoic acid (PFOA) in aqueous solutions. After a 120-min ultrasonication, a PM dosage of 6 mM increased the pseudo first-order rate constant (k1) for PFOA decomposition from 3.5 × 10-3 to 13.0 × 10-3 min-1 and increased the pseudo zeroth-order rate constant (k2) for PFOA defluorination from 1.5 × 10-3 to 7.9 × 10-3 mM·min-1, respectively. The PFOA degradation rates increased proportionally with the enhanced production rates of MnO2 particles. An initial pH 4 condition was optimal for the PFOA deg

Quantitative bias analysis of a reported association between perfluoroalkyl substances (PFAS) and endometriosis: The influence of oral contraceptive use.

An association between serum levels of perfluoroalkyl substances (PFAS) and endometriosis has recently been reported in an epidemiologic study. Oral contraceptive use to treat dysmenorrhea (pelvic pain associated with endometriosis) could potentially influence this association by reducing menstrual fluid loss, a route of excretion for PFAS. In this study, we aimed to evaluate the influence of differential oral contraceptive use on the association between PFAS and endometriosis. We used a published life-stage physiologically based pharmacokinetic (PBPK) model to simulate plasma levels of perflu

[Persistant perfluorinated compounds cause immunotoxic effects].

Perfluorinated compounds are highly stable and useful industrial chemicals. Both perfluorooctane sulfonic acid and perfluorooctanoic acid cause immunotoxic effects in animal models at serum concentrations similar to human levels. In children who have undergone routine vaccinations, serum concentrations of these substances are inversely associated with concentrations of antibodies against diphtheria and tetanus. Prevention of such effects will require a decrease of exposure limits by at least 100-fold. Immunotoxicity is not included in routine testing of industrial chemicals but urgently needs

A method to quantitatively compare in vivo the effects of gallstone solvents on intestinal mucosal function: a controlled study comparing mono-octanoin with methyl tert-butyl ether in the rat.

During contact dissolution of gallstones, solvents may escape from the gallbladder and damage the intestinal mucosa. In order to compare the extent of this potential injury, we developed a method to objectively quantify the effects of two commonly used cholesterol solvents, methyl tert-butyl ether and mono-octanoin, on mucosal transport function in the rat intestine. Two intestinal segments in each of 184 anesthetized rats were cannulated. Three milliliters of either solvent were instilled in one segment and left for varying periods of time, while saline was instilled in the other as control.

Profiles in altered metabolism. II--(omega -- 1)-hydroxyacid excretion in a case of episodic hypoglycemia.

A patient with recurrent severe hypoglycemia resembling Reye's syndrome was found to have large accumulations of omega -- 1 hydroxy and keto acids in serum and urine that persisted following clinical recovery. A deficiency of mitochondrial medium chain acyl CoA dehydrogenase activity is proposed on the basis of evidence obtained using gas chromatographic mass spectrometric techniques. Analytical data is presented that will allow the recognition of ths variant presenting in other patients.

Poly(o-phenylenediamine) nanosheets-based dual-mode fluorescence probe for ultrasensitive detection of perfluoroalkyls in water environment.

BACKGROUND: Per- and polyfluoroalkyl substances (PFASs) have significant applications in industrial, military and other sectors, particularly perfluorooctanesulfonic acid (PFOS) and perfluorooctanoic acid (PFOA), which are classified as "suspected human carcinogens". However, their release into aquatic environments via both point and non-point sources during production and use poses serious hazards to human health, potentially leading to hepatotoxicity, immunotoxicity, reproductive and developmental toxicity, and carcinogenic effects. PFOS/PFOA levels in tap water from Hong Kong (China) are si

Polystyrene NPs reshape PFOA-driven mitochondrial redox balance, pentose phosphate-nucleotide metabolism, and membrane lipid remodeling in Cipangopaludina cathayensis.

Perfluorooctanoic acid (PFOA) and nanoplastics (NPs) frequently co-occur in freshwater systems, yet their interactive effects on mitochondria-centered toxicity in benthic mollusks remain poorly resolved. We conducted a 28-day exposure of the freshwater snail Cipangopaludina cathayensis at environmentally relevant concentrations, integrating redox biomarkers, mitochondrial functional phenotypes, TUNEL staining, and the IBRv2 index, together with hepatopancreas transcriptomics and mitochondrial metabolomics. PFOA was the dominant driver, eliciting a time-resolved profile of redox imbalance and m
