Two different oxygen sensors regulate oxygen-sensitive K+ transport in crucian carp red blood cells.

The O2 dependence of ouabain-independent K+ transport mechanisms has been studied by unidirectional Rb+ flux analysis in crucian carp red blood cells (RBCs). The following observations suggest that O2 activates K+-Cl- cotransport (KCC) and deactivates Na+-K+-2Cl- cotransport (NKCC) in these cells via separate O2 sensors that differ in their O2 affinity. When O2 tension (PO2) at physiological pH 7.9 was increased from 0 to 1, 4, 21 or 100 kPa, K+ (Rb+) influx was increasingly inhibited, and at 100 kPa amounted to about 30% of the value at 0 kPa. This influx was almost completely Cl- dependent a

[Effect of furosemide on GABAA-induced 36Cl transport on ATPase activity in synaptic membranes of carp (Cyprinus carpio L.)].

We studied the effect of furosemide on GABAA-induced 36Cl transport and GABAA-induced Cl- -ATPase activity in synaptic membranes of fish brain. At physiological pH of 7.4, GABA (0.1-100 microM) stimulated 36Cl transport in synaptoneurosomes and Cl- -ATPase activity in synaptic membranes. Furosemide (0.1-0.5 mM) removed the activating effect of the mediator on chlorine transport and enzyme activity (I50 equaled 0.16 and 0.12 mM, respectively). In the absence of the mediator, picrotoxin (50 microM) activated the basal 36Cl influx in synaptoneurosomes and the basal Mg2+ -ATPase activity of synapt

Comprehensive circRNA Profiling Reveals a Testis-Specific circRNA circdmrt1 as a Novel Regulator of Gonadal Development in Crucian Carp.

Fish gonad development is a central process in reproductive regulation, and elucidating its molecular mechanisms is highly important for developing aquatic genetic resources. However, the involvement of circular RNAs (circRNAs) in the gonad development of freshwater fish is unknown. In this study, a dynamic expression profile of circRNA during the development of red crucian carp testes was systematically constructed via whole-transcriptome sequencing. Notably, the circular characteristics and high testis-specific expression of the key circRNA circdmrt1, formed through the reverse splicing of t

Ultra-fast generation of all-female grass carp via transplantation of female germline stem cells into zebrafish.

Surrogate reproduction offers a promising biotechnological approach for accelerating aquaculture breeding. Grass carp (Ctenopharyngodon idellus), a key freshwater species, faces significant constraints in breeding due to its prolonged sexual maturation of 5 years and huge body size. Here, we establish an ultra-fast breeding strategy to generate all-female grass carp within six months via surrogate production in tiny laboratory fish, zebrafish (Danio rerio). We identify and isolate female GSCs from juvenile ovaries of 3-month-old grass carp. Three months after transplantation into zebrafish lar

Modulation of innate immunity, antioxidant capacity, intestinal barrier and disease resistant in common carp by dietary sodium acetate.

This study examined the effects of sodium acetate (SA) on immunity, antioxidant status, intestinal barrier function, and disease resistance in juvenile common carp. A 1-week short-term gavage trial was first conducted using SA concentrations of 0, 0.25%, 0.5%, 1.0%, 1.5%, and 2.0%. SA at various doses enhanced immune and antioxidant responses, with optimal gavage ranges of 0.87%-1.04% and 0.92%-1.20%, respectively. Subsequently, an 8-week feeding trial was performed with five groups: two controls (CK, CK+, 0% SA) and three SA-supplemented groups (0.5%, 1.0%, 1.5%), with three replicates per gr

Thirty Weeks of Microplastics Exposure Alters Muscle Quality in Grass Carp (Ctenopharyngodon idella) Revealed by Transcriptomics and Metabolomics.

Microplastics (MPs) are tiny particles with a diameter of less than 5 mm generated from the breakdown of plastic waste. In recent years, MPs have extensively entered aquatic environments and accumulated in fish tissues, including the intestine, liver, kidney, and muscle. Previous studies have demonstrated that MP exposure can damage fish organs such as the gastrointestinal tract, gills, and liver, and may pose potential health risks to humans through the food chain. However, the toxicological effects of long-term MP exposure on the muscle tissue of grass carp (Ctenopharyngodon idella), an impo

Houttuynia cordata Extract Alleviates SVCV Infection by Modulating the RARα-A/Type I Interferon Axis in Common Carp.

Spring viremia of carp virus (SVCV) poses a major threat to aquaculture industry, triggering severe economic losses and substantial damage to fish health. Effective antiviral therapeutic approaches are currently insufficient in aquaculture, highlighting an urgent demand for viable intervention strategies. This study assessed the antiviral and immunomodulatory potential of Houttuynia cordata against SVCV in common carp. Combined cellular and in vivo fish experiments demonstrated that Houttuynia cordata markedly suppresses SVCV replication, reduces viral titer, and enhances disease resistance of

Development of monoclonal antibodies against IgM of common carp (Cyprinus carpio) and analysis of phagocytosis by mIgM+ B lymphocytes.

Phagocytosis is a specific function of phagocytes, but recent studies have revealed that B cells, such as those of mammals, amphibians, and teleost, also exhibit phagocytic activity. However, the phagocytic capacity of B cells in common carp (Cyprinus carpio), an economically important freshwater fish, remains unexplored. In this paper, we generated a mouse anti-carp IgM monoclonal antibody (mAb) and confirmed its specificity by demonstrating no cross-reactivity with IgMs from three other teleost species by Western blotting and ELISA. Indirect fluorescence assay (IFA) showed that common carp m

Negative Phonotaxis Behavior of Juvenile Grass Carp (Ctenopharyngodon idella) to Different Acoustic Stimuli in Natural Aquatic Environments.

Hydraulic engineering structures can threaten freshwater fish by entraining them into hazardous areas. Acoustic barriers have been proposed as a non-physical method to guide fish away from these zones. In this study, we investigated the behavioral responses of juvenile grass carp to different acoustic stimuli under semi-natural conditions using outdoor net cages. Four sound types were tested: a 1000 Hz pure tone and three broadband sounds, including Alligator sinensis hissing, pile-driving noise, and outboard motor noise. Behavioral responses were quantified using response frequency, total mid

Scallion White and Ginger Extracts Alleviate Stress-Induced Muscle Quality Deterioration in Crucian Carp During Transportation.

This study evaluated the effects of scallion white and ginger extracts on stress indicators, gene expression, tissue structure, and muscle quality in crucian carp during transportation. Compared with the control group, ginger extract effectively alleviated long-term transportation stress (48 h), as evidenced by lower levels of glucose and lactate dehydrogenase activity, along with reduced pathological damage in gill, liver, and muscle tissues. Consequently, muscle quality parameters including shear force, glycogen content, and inosine monophosphate levels were notably improved. These improveme

Identification and differential regulation of proteasome β family genes by viral infection and cytokines in grass carp (Ctenopharyngodon idella).

Proteasome β (PSMB) subunits are essential components of the proteasome complex and play important roles in antigen processing and immune regulation. In this study, we identified 14 Psmb genes in grass carp (Ctenopharyngodon idella), including seven constitutive Psmbs (Psmb1-7), three immunoproteasome genes (Psmb8-10), two thymoproteasome-related paralogs (Psmb11a and Psmb11b), and two telelost-specific members (Psmb12 and Psmb13). Comparative genomic analyses showed that grass carp Psmb genes are highly conserved in genomic organization, gene synteny, and predicted β-subunit-like protein stru

Grass carp reovirus VP35 hijacks DHX15 into phase-separated inclusion bodies to evade host antiviral immunity.

Many viral proteins undergo liquid-liquid phase separation (LLPS) to form biomolecular condensates known as viral inclusion bodies (VIBs), which are utilized for genome replication and virion assembly, thus serving as potential targets for antiviral drugs. However, the role of VIBs in viral immune evasion has rarely been explored. In this study, we demonstrated that VP35 protein of type II grass carp reovirus (GCRV-II) formed VIBs through LLPS in cells and in vitro. Moreover, we identified a host interaction partner of GCRV-II VP35, DEAH (Asp-Glu-Ala-His)-box helicase 15 (DHX15), which promote

Combined toxicity of polystyrene microplastics and triphenyl phosphate on common carp gills: mechanistic insights from physiology and transcriptomics.

The combined toxicity of microplastics (MPs) and co-occurring organic pollutants is a critical concern in environmental pollution risk assessment. Triphenyl phosphate (TPHP), a widely used flame retardant, frequently coexists with microplastics in aquatic environments. However, research on the impact of MPs on the toxicity of TPHP remains limited. This study integrated physiological, biochemical, and transcriptomic approaches to investigate the individual and combined effects of polystyrene (PS) MPs and TPHP on common carp (Cyprinus carpio) gills and the underlying mechanisms following 3 and 7

The dual enhancement of gel properties and storage stability by protamine in low-salt silver carp (Hypophthalmichthys molitrix) surimi.

Reducing salt from 2.0% to 1.0% in silver carp (Hypophthalmichthys molitrix) surimi compromises gel quality and storage stability. This study evaluated the dual effects of protamine on gelation and storage stability in low-salt surimi. Salt reduction decreased gel strength from 2161.66 to 1842.10 g·cm and water-holding capacity from 63.43% to 56.24%, while increasing cooking loss from 9.72% to 19.75%. Addition of protamine at 8 g per 100 g of initial surimi (NP18) reversed these changes, increasing gel strength to 3919.59 g·cm, restoring water-holding capacity to 63.72%, and reducing cooking l

Integrative Multi-Omics Analysis of Gill Responses to Long-Term Salinity Stress in Grass Carp (Ctenopharyngodon idella).

Salinity is an important environmental factor affecting the physiological homeostasis of freshwater fish, yet the underlying mechanisms in grass carp (Ctenopharyngodon idella) gills remain unclear. Therefore, grass carp were exposed to different salinity levels for 60 days, and gill responses were evaluated using histopathological, ion regulatory, antioxidant, transcriptomic, and metabolomic analyses. Histological observations showed that high salinity (8 g/L) caused marked structural damage to the gill lamellae. Specifically, Na+ and Ca2+ concentrations and Na+/K+-ATPase activity significantl

Grass carp DDX3X inhibits SVCV replication by activating IFN through the TRAF6-IRF3/7 signaling axis.

Spring viremia of carp virus (SVCV) is one of the most devastating viral pathogens threatening the aquaculture of grass carp (Ctenopharyngodon idellus), and its outbreaks frequently result in severe economic losses worldwide. DEAD-box RNA helicase DDX3X has been established as a critical regulator of innate immune responses in mammals; however, its functional role and underlying mechanism in fish against SVCV infection remain largely unexplored. In this study, the coding sequence of grass carp DDX3X (gcDDX3X) was cloned and characterized. Sequence analysis revealed that gcDDX3X possesses conse
