Revealing ROC-derived optimal thresholds of specific seminal plasma proteins as markers of bull semen freezability.

This study examined differences in the concentrations of selected bovine seminal plasma proteins-osteopontin (OPN), clusterin (CLU), lactoferrin (LTF), and angiotensin-converting enzyme (ACE)-between ejaculates with good (GFE) and poor freezability (PFE). In addition, the potential of these proteins as indicators of semen freezability was assessed using receiver operating characteristic (ROC) curve analyses. A total of 72 ejaculates collected from six bulls were included in the study to enhance the biological robustness of the analysis, thereby reducing ejaculate-related variability. Using clu

Histological and proteomic characterization of hyaline material in two dogs with hyalinizing pancreatic adenocarcinoma.

Hyalinizing pancreatic adenocarcinoma is a rare subtype of exocrine pancreatic carcinoma in dogs, characterized by abundant extracellular hyaline material within the tumor. In this study, we investigated two canine cases exhibiting this distinctive hyalinizing morphology using histological, proteomic, immunohistochemical, ultrastructural, and molecular approaches. Histological examination revealed amyloid-like eosinophilic hyaline deposits; however, these deposits lacked diagnostic features of amyloid, as confirmed by Congo red and thioflavin S staining and transmission electron microscopy. How schistosomes alter the human serum proteome.

Schistosomes are intravascular parasitic worms that cause the debilitating disease schistosomiasis. To better understand how these long-lived parasites may subvert host immune and hemostatic capabilities, we examine here the impact of adult Schistosoma mansoni worms on the human serum proteome. Normal human serum (150μl) was incubated at 37°C for one hour either in the presence or absence of adult worms (∼50 pairs). Thereafter parasites were removed, serum samples were labeled and their proteins resolved for comparative analysis by 2D-Differential in-Gel Electrophoresis (2D-DIGE). Several diff

Purification and characterization of astrocyte-secreted apolipoprotein E and J-containing lipoproteins from wild-type and human apoE transgenic mice.

The varepsilon4 allele of apolipoprotein E (apoE) is a genetic risk factor for Alzheimer's disease (AD). In order to gain a better understanding of the molecular mechanisms by which apoE and possibly other apolipoproteins produced in the central nervous system (CNS) influence AD pathogenesis, we have purified and characterized the two most abundant apolipoproteins produced in the CNS, apoE and apoJ. We purified apoE and apoJ from primary cultures of mouse astrocytes, which were derived from transgenic mice expressing human apoE isoforms in the absence of mouse apoE. Utilizing antibody affinity

Increased expression of sulfated glycoprotein-2 and DNA fragmentation in the pancreas of copper-deficient rats.

Recent morphological studies from our laboratory have shown that copper deficiency-induced pancreatic involution in rats is secondary to apoptosis (M. S. Rao, A. V. Yeldandi, V. Subbarao, and J. K. Reddy, 1993, Am. J. Pathol. 142, 1952-1957). To corroborate the morphological findings, we have examined pancreases from copper-deficient rats for expression of sulfated glycoprotein-2 (SGP-2) mRNA and DNA fragmentation, which are considered as specific markers of apoptosis. Agarose gel electrophoresis of the DNA extracted from pancreases of rats maintained on copper deficient diet (CUDD) for 5 and

Epidermal growth factor ameliorates autosomal recessive polycystic kidney disease in mice.

C57BL/6J mice homozygous for the cpk gene exhibit an autosomal recessive (AR) form of polycystic kidney disease (PKD), similar to human ARPKD, with massive collecting duct cysts. These cysts are lined by epithelial with an immature phenotype. Since renal expression of epidermal growth factor (EGF) is also significantly decreased in affected mice, we hypothesized that renal EGF is necessary for normal developmental maturation of the collecting duct. To determine if the lack of EGF may be a decisive factor in the initiation and/or growth of collecting duct cysts, we administered exogenous EGF (1

Regulation of levels of specific Sertoli cell mRNAs by vitamin A.

The regulation of Sertoli cell transferrin and sulfated glycoprotein-2 (SGP-2) mRNA levels by vitamin A was studied in vitamin A deficient rats. Vitamin A deficiency differentially affected the levels of these mRNAs. Sertoli cells from vitamin A deficient rats contained 3-fold less transferrin mRNA and 1.8-fold more SGP-2 mRNA than Sertoli cells from normal rats. Vitamin A deficiency did not result in changes in the amount of transferrin mRNA per liver cell. When vitamin A deficient rats were fed a retinol supplemented diet, Sertoli cell transferrin and SGP-2 mRNA levels returned to normal. In

Proteomics in myelodysplastic syndromes: towards novel prognostic and diagnostic biomarkers.

Myelodysplastic neoplasms (MDS) are clonal hematopoietic disorders defined by ineffective hematopoiesis, cytopenias, and variable risk of progression to acute myeloid leukemia. Although genomic and epigenomic studies have provided insight into disease pathogenesis, reliable biomarkers for diagnosis and prognosis remain limited. Proteomics offers an important advantage because it reflects the functional protein state and captures post-translational modifications, making it highly relevant for risk assessment and therapy guidance. Recent studies have identified several groups of candidate biomar

Seminal ubiquitin as an independent predictor of severe sperm damage and ICSI outcomes: evidence for a redox-proteostasis axis in male infertility†.

Oxidative stress plays a central role in male infertility by inducing lipid peroxidation, DNA damage, and activation of sperm quality control mechanisms. However, the integrated relationship between oxidative stress, genomic damage, cellular stress response, and ubiquitin-mediated sperm elimination remains unclear. This study evaluated seminal plasma levels of malondialdehyde (MDA), 8-hydroxy-2'-deoxyguanosine (8-OHdG), clusterin, and ubiquitin across distinct infertility phenotypes and assessed their predictive value. A total of 132 men were stratified into four groups based on sperm morpholo

Lymphocyte alterations and elevated complement signaling are key features of refractory myasthenia gravis.

BACKGROUND: A significant proportion of patients with myasthenia gravis (MG) remain refractory to standard immunosuppressive therapy, and biomarkers to help guide treatment decisions are lacking. We investigated whether refractory disease is associated with a distinct circulating immune profile. METHODS: We performed comprehensive immune phenotyping of peripheral blood from patients with acetylcholine-receptor-antibody-positive MG with differing treatment requirements and compared them with healthy controls. In a subset of refractory patients treated with anti-CD20 therapy, B cell reconstitut

COPS7B Drives Malignant Progression of Glioblastoma Through Translational Upregulation of the Downstream Functional Effector CLU.

Glioblastoma (GBM) is a highly aggressive primary malignant brain tumor, featuring diffuse infiltrative growth and poor clinical outcomes, which underscores the need to delineate the molecular mechanisms driving its malignant progression. COP9 signalosome subunit 7B (COPS7B), a core component of the conserved COP9 signalosome complex, is significantly upregulated in GBM tissues; however, its biological function and regulatory mechanism in GBM remain largely elusive. Here, we found that elevated COPS7B expression was positively correlated with glioma pathological grade and adverse prognosis in

Snake bite associated with acute kidney injury.

Acute kidney injury (AKI) is a well-known life-threatening systemic effect of snake envenomation which commonly happens secondary to snake bites from families of Viperidae and Elapidae. Enzymatic toxins in snake venom result in injuries to all kidney cell types including glomerular, tubulo-interstitial and kidney vasculature. Pathogenesis of kidney injury due to snake envenomation includes ischaemia secondary to decreased kidney blood flow caused by systemic bleeding and vascular leakage, proteolytic degradation of the glomerular basement membrane by snake venom metalloproteinases (SVMPs), dep

A non-invasive model for diagnosis of primary Sjogren's disease based on salivary biomarkers, serum autoantibodies, and Schirmer's test.

BACKGROUND: Minor salivary gland (MSG) biopsy is a critical but invasive method for the classification of primary Sjögren's disease (pSjD). Here we aimed to identify salivary proteins as potential biomarkers and to establish a non-invasive prediction model for pSjD. METHODS: Liquid chromatography-tandem mass spectrometry was conducted on whole saliva samples from patients with pSjD and non-Sjögren control subjects (non-pSjD). Proteins involved in immune processes were upregulated in the pSjD group, such as complement C3 (C3), complement factor B (CFB), clusterin (CLU), calreticulin (CALR), an

Updated Meta-Analysis of BIN1, CR1, MS4A6A, CLU, and ABCA7 Variants in Alzheimer's Disease.

Genome-wide association studies (GWAS) have associated several genetic variants with late-onset Alzheimer's disease (LOAD), a neurodegenerative disease. Among those, rs3764650 ABCA7, rs6656401 CR1, and rs744373 BIN1 were associated as risk factors for LOAD, while rs11136000 CLU and rs610932 MS4A6A were protective. Recently, several case-control studies have investigated the association of these polymorphisms with AD. However, not all meta-analyses analyzed these variants across different ethnic groups. Therefore, we performed an updated meta-analysis of rs3764650 ABCA7, rs6656401 CR1, rs744373

Discovery of novel biomarker candidates for liver fibrosis in hepatitis C patients: a preliminary study.

BACKGROUND: Liver biopsy is the reference standard for assessing liver fibrosis and no reliable non-invasive diagnostic approach is available to discriminate between the intermediate stages of fibrosis. Therefore suitable serological biomarkers of liver fibrosis are urgently needed. We used proteomics to identify novel fibrosis biomarkers in hepatitis C patients with different degrees of liver fibrosis. METHODOLOGY/PRINCIPAL FINDINGS: Proteins in plasma samples from healthy control individuals and patients with hepatitis C virus (HCV) induced cirrhosis were analysed using a proteomics techniq

Apolipoprotein J expression at fluid-tissue interfaces: potential role in barrier cytoprotection.

Apolipoprotein J (apoJ) is a sulfated secreted glycoprotein that exhibits ubiquitous expression, evolutionary conservation, and diverse tissue inducibility. It has been proposed to have roles in programmed cell death, sperm maturation, complement regulation, and lipid transport. To identify cell types that synthesize apoJ and to aid evaluation of its function, we screened mouse and human tissues by in situ hybridization. ApoJ was expressed at high levels in an array of specialized cell types of adult and fetal mouse tissues and in similar cell types of human tissues. Most of these cell types a

An exploratory analysis of plasma biomarkers associated with cerebral amyloid angiopathy.

Cerebral amyloid angiopathy (CAA) remains diagnostically challenging, particularly in asymptomatic individuals. While CAA often co-exists with Alzheimer's disease (AD), it may even have a direct impact on AD pathophysiology and the cognitive decline within the clinical course of AD. While fluid biomarkers are well established for AD pathology, reliable markers to characterize CAA are lacking. We analyzed two subsets of participants from the Alzheimer's Disease Neuroimaging Initiative with available plasma biomarker measurements from a 145-analyte multiplex immunoassay panel: one with T2*-weigh
