Nerve growth factor promotes human hemopoietic colony growth and differentiation.

Nerve growth factor (NGF) is a neurotropic polypeptide necessary for the survival and growth of some central neurons, as well as sensory afferent and sympathetic neurons. Much is now known of the structural and functional characteristics of NGF, whose gene has recently been cloned. Since it is synthesized in largest amounts by the male mouse submandibular gland, its role exclusively in nerve growth is questionable. NGF also causes histamine release from rat peritoneal mast cells in vitro, and we have shown elsewhere that it causes significant, dose-dependent, generalized mast cell proliferatio

Effect of granulocyte colony stimulating factor on neutropenia induced by cytotoxic chemotherapy.

A phase I/II study of granulocyte colony stimulating factor (G-CSF) was undertaken in patients with advanced malignancy receiving melphalan to determine the granulocyte response, side-effects, and pharmacokinetics. Patients received doses of 1-60 micrograms/kg intravenously. There were 3 patients at each dose level. Before chemotherapy the immediate effect of G-CSF was a transient depression in circulating neutrophils followed by a dose-dependent rise. Neutrophil counts up to 80 X 10(9)/l were achieved. G-CSF administration following melphalan reduced the period of neutropenia caused by melpha

In vitro production of colony-stimulating factors by Listeria-immune spleen cells.

In this series of experiments we have shown that there are at least two mechanisms of CSF production by spleen cells in vitro: nonspecific secretion of CSF by nonimmune cells and immune, antigen-driven production by specific T-lymphocytes. M-CSF was mainly produced in the nonspecific reaction. The immune production by T-lymphocytes consisted of increased CSF production and secretion of multiple species of CSFs.

Characterization of the cell surface receptor for granulocyte-macrophage colony-stimulating factor.

125I-labeled recombinant murine granulocyte-macrophage colony-stimulating factor (GM-CSF) was used to characterize receptors specific for this lymphokine on the surface of cells of both myelomonocytic and T-cell origin. GM-CSF binding to these cells was specific and saturable. Equilibrium binding studies revealed that on all cell types examined, GM-CSF bound to a single class of high affinity receptor (1000-5000 receptors/cell) with a Ka of 10(8)-10(9) M-1. More extensive characterization with P388D1 cells showed that binding of GM-CSF was rapid at 37 degrees C with a slow subsequent dissociat

Detection of lymphocytic choriomeningitis virus antibody in colonies of laboratory animals in Japan.

Indirect fluorescent antibody method was applied for a detection of lymphocytic choriomeningitis virus (LCMV) antibody in colonies of laboratory animals in Japan. The results showed that the antibody exist in SPF mice (3/152, 2.2%) and conventional mice (30/539, 5.6%) with the titers ranging from 1:10 to 1:160. The antibody was also detected in 2.2% (2/89) of Syrian hamsters, and 2.9% (2/68) of Apodemus agrarius, 21.4% (3/14) of Japanese harvest mice which have been maintained as laboratory colony for several years. However, the antibody was not demonstrated in Mongolian gerbils, Suncus murinu

[Level of parasitic contact in colonies of the little suslik in the semideserts of western Kazakhstan].

Field experiments were conducted during the settling of young animals of Citellus pygmaeus in four areas 4 to 13.6 hectares in size. From 3 to 6 animals were caught in each area and labeled by administering radioactive substance (0.5 micrograms of glycine 14C or methionine 35S) and colouring hair on the head. In a day or two labeled sousliks were caught again and in 6-7 days a total catching of animals and collection of ectoparasites from their hair and burrows done. In al, 942 sousliks were caught in the experimental areas and 2624 specimens of fleas of different species and 2305 ticks of R.

A human parvovirus-like virus inhibits haematopoietic colony formation in vitro.

Viruses have been shown to cause bone marrow aplasia in animals and have been implicated in bone marrow failure in man; however, until recently, a specific link between human viral infection and bone marrow failure has not been proven. In 1975 Cossart and colleagues found a serum parvovirus-like virus (SPLV, sometimes referred to as B19) in human serum. Antibody to this virus is present in the sera of 30-45% of healthy adults (Y. E. Cossart, P. P. Mortimer, unpublished observations). However, evidence for a direct link came from work by Pattison et al. who found five children with transient ap

Pathogenic and virulence characterization of colonial mutants of Nocardia asteroides GUH-2.

The pathogenicities in mice (comparing LD50 determinations) of two mutant strains and one wild-type strain of Nocardia asteroides GUH-2, each possessing a colonial morphology distinct from the other, were compared at respective stages of growth. Despite the three strains' colinear growth curves and similar physiological properties, unique patterns of pathogenicity emerged for each strain upon analysis. Ultrastructural and fatty acid profiles of cultures at the various growth stages were monitored. The mutant strain SCII-A1 was consistently less virulent than the other strains of N. asteroides

Characteristics of circulating erythroid colony-forming cells in normal and polycythaemic man.

Circulating erythroid progenitors from 10 patients with polycythaemia vera, nine with secondary polycythaemia and 10 normal subjects were studied in culture to determine their frequency, requirements for erythropoietin (ESF) and physical characteristics. Increased numbers of erythroid progenitors were detected in circulation from the patients with polycythaemia vera when compared to patients with secondary polycythaemia or normals, and 19 +/- 3% of these progenitors formed endogenous colonies without added ESF. Velocity sedimentation analysis showed that the majority of ESF-dependent erythroid

In vitro growth of lymphoma colonies from children with non-Hodgkin's lymphoma.

Growth in vitro of malignant lymphoid cells has been previously reported, but those studies involved direct establishment of cell lines from the patients' cells. In the present study, an in vitro system for both primary growth and passage of malignant lymphoblastic colony-forming cells (ML-CFC) was established, and for the first time, colony-forming cells from non-Hodgkin's lymphoma patients were cultured, and cell lines were established from individual colonies. Malignant lymphoid colonies grew from 9 of 16 (56%) bone marrow aspirates, which were histologically involved with lymphoma and from

Bone marrow colony-forming cells in acute drug-induced agranulocytosis.

The capacity to differentiate and form colonies in vitro by bone marrow granulocytic precursor cells and its evolution were studied in eight cases of drug-induced acute agranulocytosis. In three cases colonies and clusters count were normal or high. These cases were probably immunological agranulocytosis. In the five other cases this number was very low and returned to normal from three to thirteen days. These were probably cases of marrow suppression by the drug.

Bacterial and mycoplasma flora of a laboratory colony of the common marmoset (Callithrix jacchus).

A survey was carried out to characterise the bacterial and mycoplasma flora of a closed colony of marmosets, Callithrix jacchus. Bordetella bronchiseptica and Staphylococcus aureus were the only potential bacterial pathogens isolated from the colony; Candida albicans was also identified. S aureus and C albicans were the only organisms which were associated with macroscopic lesions or overt disease. Post mortem examination of the animals proved only reliable method of establishing the presence of some bacterial species in the colony, eg, B bronchiseptica and of obtaining a reliable estimate of

The identification of eosinophil colonies in soft-agar cultures by differential staining for peroxidase.

There has been a need to easily quantitate the incidence of eosinophil colonies within soft agar cultures. This has been realized by layering of the agar with benzidine dihydrochloride that permits detection of peroxidase activity in cells. Eosinophil colonies can be specifically identified by the addition to the substrate of potassium cyanide, an inhibitor of enzyme activity in neutrophils and monocytes. The enumeration of eosinophil colonies can be accomplished by scanning fresh or embedded cultures with low power magnification.

Canine systemic lupus erythematosus. Genetic analysis of an established breeding colony.

Three breeding lines, originating from dogs with SLE, have been established. Two lines were initiated by mating a female with SLE with a normal male. The third line resulted from a mating of two affected dogs. Brother-to-sister matings have reached the third generation in each line. In addition, backcross and outcross matings were carried out. More than one-third of the autopsied dogs had thymic abnormalities. The commonest lesion was a lymphoid follicle; the thymus of one dog contained multiple granulomas, and in one animal a reticulum cell sarcoma of the thymus was found. Multiple serologica
