Archaeometabolomics characterizes phenotypic differences in human cortical bone at a molecular level relating to tobacco use.

Tobacco consumption affects human health, but no studies have investigated its effect on the bone metabolome, or if any changes are traceable after long postmortem intervals. Human osteoarchaeological remains preserve small molecules, making them valuable for studies that aim to examine past conditions. We test if there are molecular differences in the metabolome of cortical bone between archaeological individuals who used tobacco and those who did not, and if these differences are distinct enough to assign tobacco use status to individuals with unknown tobacco use. Cortical bone of 323 known …

Characterization of damage mechanisms in cortical bone: Quantification of fracture resistance, critical strains, and crack tortuosity.

One step towards understanding bone fragility and degenerative diseases is to unravel the links between fracture resistance and the compositional and structural characteristics of cortical bone. In this study, we explore an optical method for automatic crack detection to generate full fracture resistance curves of cortical bone. We quantify fracture toughness, critical failure strains at the crack tip, and crack tortuosity in three directions and analyze how they relate to cortical bone microstructure. A three-point bending fracture test of single-edge notched beam specimens in three direction…

A 3D-CT Study of the Cortical Bone Trajectory Screw Placement Parameters Based on Lumbar CT.

OBJECTIVE: The cortical bone trajectory (CBT) technology is an effective substitute for traditional pedicle screw (PS) technology. However, there is still controversy about the CBT screw technology placement strategy. The objective of this study was to simulate cortical screw placement with the help of three-dimensional (3D) software, to discuss the differences in screws between genders and vertebral segments, and to explore a safer and more efficient strategy for cortical screw placement. METHODS: Mimics Medical software was used to construct a 3D model of the lumbar spine, and the placement…

In situ synchrotron radiation µCT indentation of cortical bone: Anisotropic crack propagation, local deformation, and fracture.

The development of treatment strategies for skeletal diseases relies on the understanding of bone mechanical properties in relation to its structure at different length scales. At the microscale, indention techniques can be used to evaluate the elastic, plastic, and fracture behaviour of bone tissue. Here, we combined in situ high-resolution SRµCT indentation testing and digital volume correlation to elucidate the anisotropic crack propagation, deformation, and fracture of ovine cortical bone under Berkovich and spherical tips. Independently of the indenter type we observed significant depende…

Thermomechanical damage in cortical bone caused by margins of surgical drill bit: A finite element analysis.

BACKGROUND AND OBJECTIVE: Conventional surgical drill bits suffer from several drawbacks, including extreme heat generation, breakage, jam, … t of 0.05-0.13 mm and a margin width of 0.22-0.26 mm can produce minimum thermomechanical damage in cortical bone drilling. The insights regarding the suitable ranges for margin height and width from this study could be adopted in future research devoted to optimizing the margin of the existing surgical drill bit.

A scanning electron microscopy study of projectile entry fractures in cortical bone; genesis and microarchitectural features.

The present paper presents a scanning electron microscope (SEM) analysis of the genesis and microarchitecture of experimentally induced cort … ack propagation. Despite impulsive loading, two forms of plastic deformation were identified in the cortical bone surrounding the entry fracture up to a maximum velocity of 871 m/s. Microscopic radial and concentric cracks were associated with projectile impact, and the role of pores and pits as stress concentrators was captured. Possible underlying mechanisms …

Cortical bone stem cells modify cardiac inflammation after myocardial infarction by inducing a novel macrophage phenotype.

Acute damage to the heart, as in the case of myocardial infarction (MI), triggers a robust inflammatory response to the sterile injury that is part of a complex and highly organized wound-healing process. Cortical bone stem cell (CBSC) therapy after MI has been shown to reduce adverse structural and functional remodeling of the heart after MI in both mouse and swine models. The basis for these CBSC treatment effects on wound healing are unknown. The present experiments show that CBSCs secrete paracrine factors known to have immunomodulatory properties, most notably macrophage colony-stimulatin…

Sfrp4 repression of the Ror2/Jnk cascade in osteoclasts protects cortical bone from excessive endosteal resorption.

Loss-of-function mutations in the Wnt inhibitor secreted frizzled receptor protein 4 (SFRP4) cause Pyle's disease (OMIM 265900), a rare skeletal disorder characterized by wide metaphyses, significant thinning of cortical bone, and fragility fractures. In mice, we have shown that the cortical thinning seen in the absence of Sfrp4 is associated with decreased periosteal and endosteal bone formation and increased endocortical resorption. While the increase in Rankl/Opg in cortical bone of mice lacking Sfrp4 suggests an osteoblast-dependent effect on endocortical osteoclast (OC) activity, whether …

Comparison of cortical bone trajectory versus pedicle screw techniques in lumbar fusion surgery: A meta-analysis.

BACKGROUND: Biomechanical studies have demonstrated that cortical bone trajectory (CBT) screw can provide a 30% increase in uniaxial yield pullout load than pedicle screw (PS). In addition, the insertion torque of CBT screw is 1.71 times higher than that of PS. A meta-analysis was conducted to evaluate clinical results between CBT screw technique and PS technique in lumbar fusion surgery. METHODS: An extensive search of literature was performed in PubMed, Embase, the Cochrane library. The following outcomes were extracted: visual analog scale (VAS), Oswestry disabilities index (ODI), Japanese…

Mitochondrial Function Is Compromised in Cortical Bone Osteocytes of Long-Lived Growth Hormone Receptor Null Mice.

Despite increased longevity and resistance to multiple stressors, growth hormone receptor null (GHRKO) mice exhibit severe skeletal impairment. The role of GHR in maintaining osteocyte mitochondrial function is unknown. We found that GHR ablation was detrimental to osteocyte mitochondrial function. In vivo multiphoton microscopy revealed significant reductions of >10% in mitochondrial membrane potential (MMP) in GHRKO osteocytes and reduced mitochondrial volumetric density. Reductions in MMP were accompanied by reductions in glucose transporter-1 levels, steady state ATP, NADH redox index, oxy

Does Removal of Subchondral Cortical Bone Provide Sufficient Resection Depth for Treatment of Cam Femoroacetabular Impingement?

BACKGROUND: Residual impingement resulting from insufficient resection of bone during the index femoroplasty is the most-common reason for revision surgery in patients with cam-type femoroacetabular impingement (FAI). Development of surgical resection guidelines therefore could reduce the number of patients with persistent pain and reduced ROM after femoroplasty. QUESTIONS/PURPOSES: We asked whether removal of subchondral cortical bone in the region of the lesion in patients with cam FAI could restore femoral anatomy to that of screened control subjects. To evaluate this, we analyzed shape mo…

Microstructural changes associated with osteoporosis negatively affect loading-induced fluid flow around osteocytes in cortical bone.

Loading-induced interstitial fluid flow in the microporosities of bone is critical for osteocyte mechanotransduction and for the maintenance … an negatively affect interstitial fluid flow around osteocytes in the lacunar-canalicular system of cortical bone. This fluid flow reduction could impair mechanosensation of the osteocytic network, possibly playing a role in the initiation and progression of age-related bone loss and postmenopausal osteoporosis.

Transcriptional profiling of cortical versus cancellous bone from mechanically-loaded murine tibiae reveals differential gene expression.

Mechanical loading is an anabolic stimulus that increases bone mass, and thus a promising method to counteract osteoporosis-related bone los … s downregulated at 24h than at 3h in both tissues. Enhanced Wnt signaling dominated the response in cortical bone at 3 and 24h, but in cancellous bone only at 3h. In cancellous bone at 24h many muscle-related genes were downregulated. These findings reveal key differences between cortical and cancellous genetic regulation in response to mechanical loading. Futu …

31P NMR relaxation of cortical bone mineral at multiple magnetic field strengths and levels of demineralization.

Recent work has shown that solid-state (1) H and (31) P MRI can provide detailed insight into bone matrix and mineral properties, thereby potentially enabling differentiation of osteoporosis from osteomalacia. However, (31) P MRI of bone mineral is hampered by unfavorable relaxation properties. Hence, accurate knowledge of these properties is critical to optimizing MRI of bone phosphorus. In this work, (31) P MRI signal-to-noise ratio (SNR) was predicted on the basis of T1 and T2 * (effective transverse relaxation time) measured in lamb bone at six field strengths (1.5-11.7 T) and subsequently

Risedronate does not reduce mechanical loading-related increases in cortical and trabecular bone mass in mice.

To establish whether the combination of anti-resorptive therapy with mechanical loading has a negative, additive or synergistic effect on bone structure, we assessed the separate and combined effects of risedronate and non-invasive dynamic loading on trabecular and cortical bone. Seventeen-week-old female C57BL/6 mice were given daily subcutaneous injections of vehicle (n=20) or risedronate at a dose of 0.15, 1.5, 15 or 150 μg/kg/day (n=10 in each) for 17 days. From the fourth day of treatment, the right tibiae were subjected to a single period of axial loading (40 cycles/day) for three altern…
