Identification and characterization of a DR4-restricted T cell epitope within chlamydia heat shock protein 60.

An epitope within the 60 kD Chlamydia trachomatis heat shock protein (hsp) 60, recognized by a HLA-DRB1*0401-restricted T cell clone from a reactive arthritis patient, has been characterized. Stimulatory peptides contained a nine amino acid sequence (residues 38-46) predicted by algorithm to confer strong binding to DRB1*0401, with valine in the P1 position. The overall length of the peptide was critical for efficient recognition; peptides with at least one residue N-terminal to the putative P1 position were markedly more stimulatory than a peptide whose N-terminal is the P1 valine. Optimal re

Effect of heat on antigenicity and immunogenicity of the antigenic determinant shared by Haemophilus influenzae type b and Escherichia coli K100.

Escherichia coli K100 produces an antigenic determinant similar to, or identical with, the capsular antigen of Haemophilus influenzae type b. Studies of the effects of heat on the immunogenicity, erythrocyte-modifying capacity, and antigenicity of this cross-reacting antigen (CRA) revealed the following findings. Immunization of rabbits with viable or formaldehyde-killed suspensions of E. coli K100, producing CRA, engendered CRA antibodies in significant titers, as demonstrated by hemagglutination of erythrocytes modified by H. influenzae type b antigen. Heating of the suspensions for 1 h at 5

Evaluating Memory B Cell Cross-Reactivity Between Ancestral and Future SARS-CoV-2 Variants-Evidence for Original Antigenic Sin.

Background: Despite the circulation of evolutionarily related cold-causing coronaviruses (CCCs) in the pre-COVID era, most individuals lacked pre-existing serum IgG and/or class-switched memory B cell (Bmem) reactivity for the SARS-CoV-2 Spike (S) glycoprotein expressed by the ancestral Wuhan-Hu-1 (WH1) strain. Subsequent priming of the immune system through natural infection or prophylactic COVID-19 mRNA vaccination successfully generated robust Bmem responses against the WH1-S antigen, along with eliciting cross-reactivity for the future Omicron (BA.1) variant responsible for breakthrough in

Dermatology for the elderly: an Indian perspective.

Geriatric dermatology in India is gaining steady momentum, because the aging population is dramatically increasing. India crossed the United Nations definition of an aging country when the population of persons aged older than 60 years exceeded 7%. The dermatologic issues of this aging population are strongly influenced by many social, economic, and cultural factors. Pigmentary disorders are the prime example of cultural factors affecting dermatoses and their treatment. Photoaging differs from what one sees in Western populations due to the inherently dark skin of Indians. Cosmetic dermatology

The 19-kD antigen and protective immunity in a murine model of tuberculosis.

The 19-kD antigen is a cell wall-associated lipoprotein present in Mycobacterium tuberculosis and in bacille Calmette-Guérin (BCG) vaccine strains. Expression of the 19-kD antigen as a recombinant protein in two saprophytic mycobacteria-M. vaccae and M. smegmatis-resulted in abrogation of their ability to confer protection against M. tuberculosis in a murine challenge model, and in their ability to prime a DTH response to cross-reactive mycobacterial antigens. Induction of an immune response to the 19-kD antigen by an alternative approach of DNA vaccination had no effect on subsequent M. tuber

Effective cancer immunotherapy by natural mouse conventional type-1 dendritic cells bearing dead tumor antigen.

BACKGROUND: The manipulation of dendritic cells (DCs) for cancer vaccination has not reached its full potential, despite the revolution in cancer immunotherapy. DCs are fundamental for CD8+ T cell activation, which relies on cross-presentation of exogenous antigen on MHC-I and can be fostered by immunogenic cancer cell death. Translational and clinical research has focused on in vitro-generated monocyte-derived DCs, while the vaccination efficacy of natural conventional type 1 DCs (cDC1s), which are associated with improved anti-tumor immunity and specialize on antigen cross-presentation, rema

Malaria-specific and nonspecific activation of CD8+ T cells during blood stage of Plasmodium berghei infection.

Cerebral malaria is one of the severe complications of Plasmodium falciparum infection. Studies using a rodent model of Plasmodium berghei ANKA infection established that CD8(+) T cells are involved in the pathogenesis of cerebral malaria. However, it is unclear whether and how Plasmodium-specific CD8(+) T cells can be activated during the erythrocyte stage of malaria infection. We generated recombinant Plasmodium berghei ANKA expressing OVA (OVA-PbA) to investigate the parasite-specific T cell responses during malaria infection. Using this model system, we demonstrate two types of CD8(+) T ce

Human Germinal Center Reaction: Assessing T Follicular Helper-B Cells Interaction In Vitro.

Antibody responses deeply rely on the interaction of antigen-primed B cells and CD4 helper T cells in the context of germinal center reactions, through signals provided by costimulatory molecules and cytokines. B-cell proliferation and differentiation in antibody-secreting plasma cells are processes that critically depend on the helper function of a specific CD4 T-cell subset, known as follicular helper T cells (Tfh). Here, we describe a method that mimics in vitro the cross talk between Tfh and B cells occurring in the germinal center. The procedure is based on setting up a coculture system w

Induction of broadly neutralizing H1N1 influenza antibodies by vaccination.

The rapid dissemination of the 2009 pandemic influenza virus underscores the need for universal influenza vaccines that elicit protective immunity to diverse viral strains. Here, we show that vaccination with plasmid DNA encoding H1N1 influenza hemagglutinin (HA) and boosting with seasonal vaccine or replication-defective adenovirus 5 vector encoding HA stimulated the production of broadly neutralizing influenza antibodies. This prime/boost combination increased the neutralization of diverse H1N1 strains dating from 1934 to 2007 as compared to either component alone and conferred protection ag

Stimulation of resistance to Fasciola hepatica infection in sheep by a regime involving the use of the immunomodulatory compound L tetramisole (levamisole).

The effect of prior nematode and cestode infection in sheep on the course of experimental Fasciola hepatica was studied. The cross immunising properties of these parasites per se and the modification of any protective effect conferred by the immunomodulatory compound L tetramisole (levamisole) were investigated. It was recorded that no protection was conferred on sheep against F hepatica infection by virtue of prior infection with any helminth species per se, including Cysticercus tenuicollis. However, a degree of immunity to F hepatica was recorded, for only the second time, in sheep primed w

A HEMODILUTION TECHNIQUE FOR OPEN-HEART SURGERY: A PRELIMINARY CLINICAL REPORT.

With the extension of open-heart procedures and the increasing success of open-heart surgery, a great strain has been placed on many hospital laboratories and on the Red Cross for the maintenance of equipment and the supply of blood. In order to decrease both of these needs, hemodilution techniques combined with disposable oxygenators have been introduced. The authors have used a disposable oxygenator with low molecular weight dextran prime, along with moderate hypothermia, in a consecutive series of 15 patients operated upon for congenital and acquired heart lesions. In this consecutive serie

Prevention of acute graft-versus-host disease by blocking T-cell entry to secondary lymphoid organs.

In acute graft-versus-host disease (aGVHD), donor T cells attack the recipient's gastrointestinal tract, liver, and skin. We hypothesized that blocking access to distinct lymphoid priming sites may alter the specific organ tropism and prevent aGVHD development. In support of this initial hypothesis, we found that different secondary lymphoid organs (SLOs) imprint distinct homing receptor phenotypes on evolving alloreactive effector T cells in vivo. Yet preventing T-cell entry to specific SLOs through blocking monoclonal antibodies, or SLO ablation, did not alter aGVHD pathophysiology. Moreover

Molecular and biophysical foundations of nano-enabled seed priming and smart nanocarriers for crop stress alleviation.

Agricultural productivity is increasingly constrained by interacting abiotic stresses, yet translation of nano-enabled seed treatments from controlled experiments to field agriculture remains limited. Seed nanopriming can initiate physiological and molecular priming before radicle emergence, but outcomes depend strongly on nanomaterial identity, particle size and morphology, surface charge, colloidal stability, cargo chemistry, exposure regime, and crop genotype. This review critically examines mesoporous silica nanoparticles, chitosan-based systems, metal-organic frameworks, hybrid/core-shell

Agonists for cytosolic bacterial receptor ALPK1 induce antitumour immunity.

Targeting innate immunity holds promise in cancer immunotherapy, particularly in improving checkpoint inhibitors. However, the use of agonis … s differentiated from STING agonists in stimulating tumour-cell antigen presentation, macrophage-DC cross-priming and protective memory T cell differentiation, and it does not induce T cell apoptosis. Our study elucidates the antitumour effect of ALPK1 agonism and suggests the potential of ALPK1 agonists in cancer immunotherapy.

Imprinting of serum neutralizing antibodies by Wuhan-1 mRNA vaccines.

Immune imprinting is a phenomenon in which prior antigenic experiences influence responses to subsequent infection or vaccination1,2. The effects of immune imprinting on serum antibody responses after boosting with variant-matched SARS-CoV-2 vaccines remain uncertain. Here we characterized the serum antibody responses after mRNA vaccine boosting of mice and human clinical trial participants. In mice, a single dose of a preclinical version of mRNA-1273 vaccine encoding Wuhan-1 spike protein minimally imprinted serum responses elicited by Omicron boosters, enabling generation of type-specific an

Induction of broadly cross-reactive antibody responses to the influenza HA stem region following H5N1 vaccination in humans.

The emergence of pandemic influenza viruses poses a major public health threat. Therefore, there is a need for a vaccine that can induce broadly cross-reactive antibodies that protect against seasonal as well as pandemic influenza strains. Human broadly neutralizing antibodies directed against highly conserved epitopes in the stem region of influenza virus HA have been recently characterized. However, it remains unknown what the baseline levels are of antibodies and memory B cells that are directed against these conserved epitopes. More importantly, it is also not known to what extent anti-HA

A Set of Rapid Diagnostic Tool for Babesia microti Infection.

BACKGROUND: Human babesiosis caused by Babesia microti is an emerging tick-borne zoonosis, with a global pooled prevalence of 2.23% and regional peaks in Europe (4.17%) and North America (1.54%). Traditional diagnostics like microscopy and polymerase chain reaction (PCR) suffer from low sensitivity in low-parasitemia cases or high costs ($230/test), necessitating accessible, rapid assays for resource-limited regions. METHODS: A cross-priming amplification combined with vertical flow visualization (CPA-VF) assay, a straightforward molecular method targeting the 18S rRNA gene of B. microti, req…

Antibodies to Mycobacterium tuberculosis-specific epitopes in lepromatous leprosy.

Sera from patients with leprosy or tuberculosis and healthy subjects have been analysed for the presence of antibodies to four species-specific mycobacterial epitopes, four different viruses and five autoantigens. Antibodies to the Mycobacterium leprae-specific 35-kD protein and phenolic glycolipid I epitopes were not present in patients with active pulmonary tuberculosis. In contrast, antibody levels to species-specific epitopes of the 38-kD and 14-kD antigens M. tuberculosis were significantly elevated in patients with lepromatous leprosy. Neither of the two antigens is cross-reactive with M

Hydrogel activation of Mincle receptors for tumor cell processing: A novel approach in cancer immunotherapy.

An obstacle in current tumor immunotherapies lies in the challenge of achieving sustained and tumor-targeting T cell immunity, impeded by the limited antigen processing and cross-presentation of tumor antigens. Here, we propose a hydrogel-based multicellular immune factory within the body that autonomously converts tumor cells into an antitumor vaccine. Within the body, the scaffold, formed by a calcium-containing chitosan hydrogel complex (ChitoCa) entraps tumor cells and attracts immune cells to establish a durable and multicellular microenvironment. Within this context, tumor cells are comp

Narrowed TCR repertoire and viral escape as a consequence of heterologous immunity.

Why some virus-specific CD8 TCR repertoires are diverse and others restricted or "oligoclonal" has been unknown. We show here that oligoclonality and extreme clonal dominance can be a consequence of T cell cross-reactivity. Lymphocytic choriomeningitis virus (LCMV) and Pichinde virus (PV) encode NP(205-212) epitopes that induce different but highly cross-reactive diverse TCR repertoires. Homologous viral challenge of immune mice only slightly skewed the repertoire and enriched for predictable TCR motifs. However, heterologous viral challenge resulted in a narrow oligoclonal repertoire with dom
