Hepatoprotective effects of carvacrol: molecular mechanisms and insights from pre-clinical Studies.

Natural compounds have long been recognized for their therapeutic potential in managing various diseases, including liver disorders. Among these, Carvacrol, a monoterpenoid phenol present in the essential oils of oregano and thyme, has attracted significant attention for its potent hepatoprotective effects. Carvacrol protects the liver by modulating multiple critical molecular pathways. It activates the Nrf2/HO-1 pathway, strengthening the antioxidant defense system and reducing oxidative stress in hepatocytes. It also suppresses NF-κB signaling, resulting in decreased production of pro-inflam

Cuminaldehyde in combination with tetracycline shows promising antibiofilm activity against drug-resistant Pseudomonas aeruginosa.

Pseudomonas aeruginosa, an opportunistic pathogen often causes biofilm-linked infections. A combinatorial approach involving tetracycline (antibiotic) and cuminaldehyde (phytochemical) was explored to combat this infectious pathogen. The results showed that both tetracycline and cuminaldehyde individually demonstrated antibacterial effects. However, when the compounds were applied together, there was a significant increase in their antimicrobial potential. The determined fractional inhibitory concentration index of 0.43 indicated a synergistic interaction between the two compounds. Furthermore

In vitro susceptibility testing of essential oils and their main components against antifungal-resistant Malassezia pachydermatis.

Canine Malassezia dermatitis (CMD) and otitis externa are generally treated by antifungal drugs. However, azole-resistant strains have been isolated from canine skin and ear canals worldwide. Phytochemicals isolated from essential oils are effective alternatives for inhibiting Malassezia pachydermatis. To evaluate the usefulness of phytochemicals against azole-resistant isolates, we performed in vitro susceptibility testing using the phytochemicals carvacrol, citral, and thymol. Eight antifungal-resistant isolates were obtained from 7 cases of dermatitis and 1 case of otitis externa during 202

Preparation, in vitro and in vivo assessment of novel carvacrol@pro-phytomicelles for the treatment of Salmonella enteritidis infection in mice.

In livestock and poultry farming, the use of antibiotics has been abused, which seriously endangers human health. Thus, antibiotic alternatives are urgently needed. The phytochemical carvacrol (CAR) has attracted attention as an antibiotic alternative due to its excellent antibacterial activity and anti-inflammatory activity. However, CAR has high volatility and low water solubility, which seriously affect its antibacterial activity. In this study, two plant-derived small-molecule phytochemicals-glycyrrhizin and rebaudioside A-were selected as nanocarriers for the preparation of a novel solid

The coumarin osthole is a non-electrophilic agonist of TRPA1.

The naturally occurring coumarin osthole has antipruritic properties, and recent reports suggest that this effect is due an inhibition or desensitization of the cation channels TRPV1 and TRPV3. Osthole was also suggested to activate TRPA1, an effect that should rather be pruritic than antipruritic. Here we characterized the effects of osthole on TRPA1 by means of ratiometric calcium imaging and patch clamp electrophysiology. In HEK 293 expressing human (h) TRPA1, osthole induced a concentration-dependent increase in intracellular calcium that was inhibited by the TRPA1-inhibitor A967079. In mo

Plant defences against ants provide a pathway to social parasitism in butterflies.

Understanding the chemical cues and gene expressions that mediate herbivore-host-plant and parasite-host interactions can elucidate the ecological costs and benefits accruing to different partners in tight-knit community modules, and may reveal unexpected complexities. We investigated the exploitation of sequential hosts by the phytophagous-predaceous butterfly Maculinea arion, whose larvae initially feed on Origanum vulgare flowerheads before switching to parasitize Myrmica ant colonies for their main period of growth. Gravid female butterflies were attracted to Origanum plants that emitted h

Regio- and enantioselective oxidation by CYP108N12: substrate scope and specificity.

Growing demand for efficient, safe and economically viable catalytic processes has driven increasing interest in using enzymes for large-scale, biocatalytic transformations. Cytochromes P450 are enantio-, regio- and chemo-selective enzymes renowned for catalysing oxygen insertion into unactivated C-H bonds across a wide range of compounds including steroids, alkanes and terpenes. Despite their impressive catalytic ability and diverse substrate scope, these enzymes are somewhat limited in utility by their requirement for auxiliary redox partner proteins as well as stoichiometric amounts of an e
