Harnessing eDNA metabarcoding for mosquito surveillance: a pilot study in Ethiopia's Great Rift Valley.

BACKGROUND: Vector-borne diseases account for over 17% of all infectious diseases worldwide, with malaria and mosquito-borne arboviruses emerging as a significant and escalating threat. In Ethiopia, malaria and arboviruses such as Dengue and Yellow fever viruses pose substantial public health challenges; however, surveillance efforts are hindered by traditional methods that are costly and labor-intensive, yet unable to identify cryptic species. Environmental DNA (eDNA) metabarcoding presents a promising alternative, facilitating noninvasive and high-throughput detection of vector species direc

Rapid recombinase polymerase amplification-based detection of the bar gene for environmental monitoring of genetically modified soybean and rice.

Rapid and reliable methods are required for environmental monitoring of unintended releases of genetically modified (GM) crops. We developed a real-time recombinase polymerase amplification (RPA) assay targeting the bar gene for the rapid detection of GM soybean and rice, combined with a simplified NaOH-based DNA extraction method for field applications. Primer-probe set 4 provided the most specific and reliable detection, while soybean lectin and rice actin served as internal positive controls. Lectin and actin were consistently detected in GM and non-GM samples, respectively. A 0.1 N NaOH ex

DNA repair as an emerging target for COPD-lung cancer overlap.

Cigarette smoking is the leading cause of lung cancer and chronic obstructive pulmonary disease (COPD). Many of the detrimental effects of cigarette smoke have been attributed to the development of DNA damage, either directly from chemicals contained in cigarette smoke or as a product of cigarette smoke-induced inflammation and oxidative stress. In this review, we discuss the environmental, epidemiological, and physiological links between COPD and lung cancer and the likely role of DNA damage and repair in COPD and lung cancer development. We explore alterations in DNA damage repair by DNA rep

Nutrition and epigenetics: an interplay of dietary methyl donors, one-carbon metabolism and DNA methylation.

DNA methylation is the most extensively studied mechanism of epigenetic gene regulation. Increasing evidence indicates that DNA methylation is labile in response to nutritional and environmental influences. Alterations in DNA methylation profiles can lead to changes in gene expression, resulting in diverse phenotypes with the potential for increased disease risk. The primary methyl donor for DNA methylation is S-adenosylmethionine (SAM), a species generated in the cyclical cellular process called one-carbon metabolism. One-carbon metabolism is catalyzed by several enzymes in the presence of di

Epigenetic Mechanisms Underlying Cognitive Dysfunction in Parkinson's Disease: Current Evidence and Future Prospects.

PURPOSE: Epigenetics studies inheritable characteristics and lasting cellular changes that occur without alterations in the DNA sequence. This field is crucial for understanding how environmental factors interact with genes to influence memory, learning, and cognition. In the context of neurodegenerative disorders, particularly Parkinson's disease (PD), epigenetic mechanisms may help explain the molecular basis of cognitive impairment. The purpose of this review is to explore how epigenetic alterations affect gene expression and their potential role in cognitive dysfunction associated with PD.

Biomonitoring of genotoxicity of industrial fertilizer pollutants in Aiolopus thalassinus (Orthoptera: Acrididae) using alkaline comet assay.

Phosphate fertilizer industry is considered as one of the main sources of environmental pollutants. Besides solid waste products, e.g. phosphates, sulphates, and heavy metals, also atmospheric pollutants, such as hydrofluoric acid fumes (HF), sulphur dioxide (SO2), nitrogen oxides (NO2), and particulate matter with diameter up to 10 μm (PM10) can be dangerous. Genotoxic effect of these pollutants was monitored by assessing the DNA damage using alkaline comet assay on cells from brain, thoracic muscles and gut of Aiolopus thalassinus collected at three sites (A-C) located at 1, 3, and 6 km away

Aromatic DNA adducts in adjacent tissues of breast cancer patients: clues to breast cancer etiology.

The etiology of the majority of human breast cancers is unknown. Environmental factors have long been suspected to play a role, but no specific causative agent has been identified. If the hypothesis that environmental carcinogen exposure contributes to human breast cancer is true, carcinogen-DNA adducts would be expected to be present in human breast tissues. To address this possibility, aromatic DNA adducts were measured in 87 surgical specimens of normal human breast tissues from 87 breast cancer patients undergoing mastectomy using the nuclease P1-enhanced version of the 32P postlabeling as

Structural remodeling and activation of the hdeAB and proVWX operons in response to pH and salt stress.

Escherichia coli is highly sensitive to acid and osmotic stress but adapts by modulating the expression of stress-responsive genes. Nucleoid-associated proteins (NAPs) play key roles in DNA organization and sensing environmental changes. The histone-like nucleoid structuring protein H-NS is an NAP acting as a global regulator of stress genes. H-NS may alter local chromatin structure to modulate the expression of such genes in response to environmental stress. The H-NS homolog suppressor of td phenotype A (StpA) co-regulates several target genes, but its role is poorly defined. To investigate t

Dual Z-scheme In2S3/Bi2S3/ZnS heterojunction with broad-spectrum response as a photoactive material for ultrasensitive detection of environmental Pollutant tetracycline.

Herein, a novel dual Z-scheme heterojunction In2S3/Bi2S3/ZnS (IBZS) with core-shell structure was prepared to establish a photoelectrochemical (PEC) biosensor for ultrasensitive detection of tetracycline (TC) referred to environmental pollution. Compared with the traditional single Z-scheme heterojunction with low PEC response, the dual Z-scheme heterojunction exhibited a strong PEC response due to its broad-spectrum response and highly efficient carrier migration. Furthermore, a redesigned target-triggered entropy-driven DNA reaction (TEDR) was implemented to mitigate spontaneous transient st

Exploitation of genomic sequences in a systematic analysis to access how cyanobacteria sense environmental stress.

The perception and subsequent transduction of environmental signals are primary events in the acclimation of living organisms to changes in their environment. Many of the molecular sensors and transducers of environmental stress cannot be identified by traditional and conventional methods. Therefore, the genomic information has been exploited in a systematic approach to this problem, performing systematic mutagenesis of potential sensors and transducers, namely, histidine kinases and response regulators, respectively, in combination with DNA microarray analysis, to examine the genome-wide expr

Oxidative stress from environmental pollutants.

Recently progress has been made on O2 toxicity and pathology related to numerous environmental contaminants in insects. The pro-oxidants studied included: dioxin, paraquat, and an assorted array of quinones, 8-methoxypsorlen, arsenic, and mercury. The responses to these oxidants are diverse, but they arise from the reactive oxygen species. These pro-oxidants in insects cause lipid peroxidation, protein and enzyme oxidation, and GSH depletion. Potentially, they may also cause DNA oxidation, and form DNA adducts. Oxidative challenge is alleviated by antioxidant compounds, but more importantly by

RAPD- and ERIC-Based Typing of Clinical and Environmental Pseudomonas aeruginosa Isolates.

Pseudomonas aeruginosa is a major cause of nosocomial infection in children and adults, resulting in significant morbidity and mortality due to its ability to acquire drug resistance. The ability of P. aeruginosa in the environment to cause infection in individuals has been reported previously; henceforth, surveillance of the emergence and transmission of P. aeruginosa strains among patients is important for infection control in a clinical setup. Various gene-typing methods have been used for epidemiological typing of P. aeruginosa isolates for the purpose of surveillance. In this work, the su

DNA methylation-independent growth restriction and altered developmental programming in a mouse model of preconception male alcohol exposure.

The preconception environment is a significant modifier of dysgenesis and the development of environmentally-induced disease. To date, fetal alcohol spectrum disorders (FASDs) have been exclusively associated with maternal exposures, yet emerging evidence suggests male-inherited alterations in the developmental program of sperm may be relevant to the growth-restriction phenotypes of this condition. Using a mouse model of voluntary consumption, we find chronic preconception male ethanol exposure associates with fetal growth restriction, decreased placental efficiency, abnormalities in cholester

The role of oxidative stress in carcinogenesis induced by metals and xenobiotics.

In addition to a wide range of adverse effects on human health, toxic metals such as cadmium, arsenic and nickel can also promote carcinogenesis. The toxicological properties of these metals are partly related to generation of reactive oxygen species (ROS) that can induce DNA damage and trigger redox-dependent transcription factors. The precise mechanisms that induce oxidative stress are not fully understood. Further, it is not yet known whether chronic exposures to low doses of arsenic, cadmium or other metals are sufficient to induce mutations in vivo, leading to DNA repair responses and/or

Use of mitochondrial and ribosomal DNA polymorphisms to classify clinical and soil isolates of Histoplasma capsulatum.

We have developed an improved scheme for the classification of environmental and clinical isolates of Histoplasma capsulatum that is based on analysis of mitochondrial DNA (mtDNA) and ribosomal DNA (rDNA). Strains were initially divided into mtDNA groups according to restriction digests of whole-cell DNA and Southern hybridization with cloned mtDNA probes. Strains within a mtDNA class could be further grouped by polymorphisms in rDNA. The majority of soil and clinical isolates from the United States had identical mtDNA patterns; however, rDNA polymorphisms were common in both types of isolates

Wild Boar (Sus scrofa) as Reservoir of Pathogenic and Intermediate Leptospira.

Leptospirosis is a globally widespread zoonosis, with wildlife species playing a key role in the maintenance and environmental dissemination of Leptospira spp. Wild boar (Sus scrofa) are increasingly recognized as potential reservoir; however, molecular data on renal carriage of Leptospira spp. in Europe and particularly in Germany, remain scarce. In this study, 1281 wild boar kidney samples collected in the federal state of Saxony-Anhalt were analyzed by real-time PCR to investigate the presence of pathogenic and intermediate Leptospira DNA. Pathogenic Leptospira DNA was detected in 3.1% (95%

Arid1a Deficiency Drives Aristolochic Acid-Induced Liver Tumorigenesis through Ctnnb1 Mutation and Defective Nucleotide Excision Repair.

ARID1A, which encodes an important subunit of SWI/SNF complex, is frequently mutated in non-malignant tissues and tumors. However, how ARID1A loss enables environmental carcinogens to initiate tumorigenesis remains unknown. Here, liver-specific Arid1a-deficient (Arid1aLKO) mice are exposed to aristolochic acid I (AAI), a potent herbal carcinogen. Notably, AAI dramatically accelerated hepatocarcinogenesis in Arid1a-deficient livers, accompanied by a specific 3' splice-site mutation in Ctnnb1 in most tumors and adjacent non-tumorous tissues. This mutation results in exon 3 skipping and subsequen

Longitudinal viral shedding and antibody response characteristics of men with acute infection of monkeypox virus: a prospective cohort study.

Understanding of infection dynamics is important for public health measures against monkeypox virus (MPXV) infection. Herein, samples from multiple body sites and environmental fomites of 77 acute MPXV infections (HIV co-infection: N = 42) were collected every two to three days and used for detection of MPXV DNA, surface protein specific antibodies and neutralizing titers. Skin lesions show 100% positivity rate of MPXV DNA, followed by rectum (88.16%), saliva (83.78%) and oropharynx (78.95%). Positivity rate of oropharynx decreases rapidly after 7 days post symptom onset (d.p.o), while the rec

Insights into mycobacteriome composition in Mycobacterium bovis-infected African buffalo (Syncerus caffer) tissue samples.

Animal tuberculosis significantly challenges global health, agriculture, and wildlife conservation efforts. Mycobacterial cultures are resource-intensive, time-consuming, and challenged by heterogeneous populations. In this study, we employed a culture-independent approach, using targeted long-read-based next-generation sequencing (tNGS), to investigate the mycobacterial composition in 60 DNA samples extracted from Mycobacterium bovis infected culture-confirmed African buffalo tissue. We detected mycobacterial DNA in 93.3% of the samples and the sensitivity for detecting Mycobacterium tubercul

Assessment of indigenous reductive dechlorinating potential at a TCE-contaminated site using microcosms, polymerase chain reaction analysis, and site data.

A combination of microcosm studies, polymerase chain reaction (PCR) analysis, and site data was used to assess the indigenous reductive dechlorinating potential in a trichloroethene (TCE)-contaminated aquifer at Cape Canaveral Air Station, Florida. Sediment and groundwater were obtained from two distinct locations approximately 10 m apart. Microcosm studies were performed to assess dechlorinating activity under a variety of nutrient and electron donor amendment conditions. Most live microcosms constructed using material from the first location, near well 9 (W09), were negative for dechlorinati
