Design of new imidazole derivatives with anti-HCMV activity: QSAR modeling, synthesis and biological testing.

The problem of designing new antiviral drugs against Human Cytomegalovirus (HCMV) was addressed using the Online Chemical Modeling Environment (OCHEM). Data on compound antiviral activity to human organisms were collected from the literature and uploaded in the OCHEM database. The predictive ability of the regression models was tested through cross-validation, giving coefficient of determination q2 = 0.71-0.76. The validation of the models using an external test set proved that the models can be used to predict the activity of newly designed compounds with reasonable accuracy within the applic

Development of a human physiologically based pharmacokinetic (PBPK) model for dermal permeability for lindane.

Lindane is a neurotoxicant used for the treatment of lice and scabies present on human skin. Due to its pharmaceutical application, an extensive pharmacokinetic database exists in humans. Mathematical diffusion models allow for calculation of lindane skin permeability coefficients using human kinetic data obtained from in vitro and in vivo experimentation as well as a default compound-specific calculation based on physicochemical characteristics used in the absence of kinetic data. A dermal model was developed to describe lindane diffusion into the skin, where the skin compartment consisted of

Discovery of new inhibitors of resistant Streptococcus pneumoniae penicillin binding protein (PBP) 2x by structure-based virtual screening.

Penicillin binding proteins (PBPs) are involved in the biosynthesis of the peptidoglycan layer constitutive of the bacterial envelope. They have been targeted for more than half a century by extensively derived molecular scaffolds of penicillins and cephalosporins. Streptococcus pneumoniae resists the antibiotic pressure by inducing highly mutated PBPs that can no longer bind the beta-lactam containing agents. To find inhibitors of PBP2x from Streptococcus pneumoniae (spPBP2x) with novel chemical scaffold so as to circumvent the resistance problems, a hierarchical virtual screening procedure w

Antidiabetics: Structural Diversity of Molecules with a Common Aim.

BACKGROUND: Diabetes mellitus type 2 (DMT2) is an endocrine disease of global proportions which is currently affecting 1 in 12 adults in the world, with still increasing prevalence. World Health Organization (WHO) declared this worldwide health problem, as an epidemic disease, to be the only non-infectious disease with such categorization. People with DMT2 are at increased risk of various complications and have shorter life expectancy. The main classes of oral antidiabetic drugs accessible today for DMT2 vary in their chemical composition, modes of action, safety profiles and tolerability. ME

Safe use of selected cephalosporins in penicillin-allergic patients: a meta-analysis.

BACKGROUND: Recent analysis of clinical data and a clearer understanding of the role of chemical structure in the development of cross-reactivity indicate that the increased risk of an allergic reaction to a cephalosporin in penicillin-allergic patients is smaller than previously postulated. METHOD: Medline and EMBASE databases were searched with the keywords: cephalosporin, penicillin, allergy, and cross-sensitivity for the years 1960 through 2005. Among 219 articles retrieved, 9 served as source material for this evidence-based meta-analysis. RESULTS: A significant increase in allergic rea

Association of exposures to environmental chemicals estimated through primary teeth biomatrix and health outcomes in children and adolescents - A systematic review.

Children's heightened susceptibility to environmental exposure arises from their underdeveloped detoxification mechanisms and augmented per-unit body-weight absorption capacity for chemical compounds. Primary teeth are an emerging biomatrix, which aid in storing crucial data on early exposure to harmful substances and developmental illnesses. This systematic review aimed to evaluate the association between environmental chemical exposure and health outcomes in children and adolescents using primary teeth as a matrix. The study protocol was registered with PROSPERO (CRD42023428013). The review

A Comprehensive Review on Pharmacologically Active Phyto-Constituents from Hedychium species.

In this review, we describe and discuss the phytoconstituents present in Hedychium species and emphasize their potential as drug candidates. Though they are widely validated in vitro and in vivo models, to date, no efforts have been made to compile in a single review all the pharmacologically active phytoconstituents from Hedychium species, and their pharmacological and toxicity profile. In this study, we present a reinvestigation of the chemical constituents present in Hedychium species obtained from the essential oil and solvent extraction of the flowers, leaves and rhizomes under considerat

Targeting EZH2 and EGFR therapeutic vulnerabilities of TNBC with a single chemical entity confers preclinical treatment advantage.

Globally, triple-negative breast cancer (TNBC) has a huge unmet medical need, as it has limited targeted therapeutic options. However, it has well-known therapeutic vulnerabilities for selective inhibition of pathways such as EGFR, mTOR, PARP, enhancer of zeste homolog 2 (EZH2), PD1-PDL1, etc. Analysis of publicly available databases clearly suggests that concomitant EZH2 and EGFR overexpression results in poor survival of patients with TNBC compared to their individual overexpression, underscoring the strong rationale for dual-targeted therapeutic intervention. Herein, potent, tumor cell-sele

Thyme, Oregano, and Cinnamon Essential Oils: Investigating Their Molecular Mechanism of Action for the Treatment of Bacteria-Induced Cystitis.

Pathogen infections, exacerbated by emerging drug resistance, remain among the most challenging health issues, for which multitargeting approaches may offer effective solutions. In this context, medicinal plants, including essential oils, provide complex mixtures of diverse molecules that can exert therapeutic effects, either alone or synergistically with established antibiotics. Although several databases comprehensively collect information on the antibacterial properties of medicinal plants, including chemical composition, bioactivity data, and ethnobotanical uses, there is a notable lack of

Origanum Essential Oil and Antifungal Activity: A Systematic Review.

Fungi are pathogens that affect humans, animals, food, and plants. Many strains are resistant to currently available antifungals, which are also associated with high toxicity and can cause environmental pollution. Treatments are lengthy with low adherence rates. Given the historical use of plants in disease treatment, natural products, such as essential oils (EOs), have been studied and developed for fungal infection treatment. Therefore, the aim of this study was to conduct a systematic literature review on Origanum species commercialized in Brazil: O. compactum, O. minutiflorum, O. syriacum,

A comprehensive characterization strategy for natural products using dual-mode hybrid acquisition on high-resolution MS with intelligent subdivision polygonal mass defect filtering: a case study on Typhae Pollen.

The polygonal mass defect filtering (PMDF) technique is widely used for initial filtering of characteristic constituents in natural products. This method still has issues with false positives in blank areas, a complex construction process, and cumbersome target components filtering. The intelligent subdivision polygonal mass defect filtering (Sub-PMDF) technique was developed to solve these issues. Python was introduced to automate both the construction of polygonal regions and the filtering of target components. The concept of subdivision was incorporated to reduce the blank areas within poly

Surface Treatment of Dental Mini-Sized Implants and Screws: A Systematic Review with Meta-Analysis.

Miniscrews are devices that allow for absolute skeletal anchorage. However, their use has a higher failure rate (10-30%) than dental implants (10%). To overcome these flaws, chemical and/or mechanical treatment of the surface of miniscrews has been suggested. There is no consensus in the current literature about which of these methods is the gold standard; thus, our objective was to carry out a systematic review and meta-analysis of the literature on surface treatments of miniscrews. The review protocol was registered (PROSPERO CRD42023408011) and is in accordance with the PRISMA guidelines. A
