Early addition of enzyme-treated soybean in the diet improves amino acid absorption and protein digestibility by promoting digestive enzyme activity in broilers.

Early nutritional regulation has become a research hotspot. The present study was undertaken to assess dietary early addition enzyme-treated soybean meal (ESBM) on nutrient digestibility, amino acid absorption, and intestinal development of broilers. Four hundred and fifty 1-day-old broilers were divided into three groups with 10 replicates of 15 broilers and fed with a basic diet, 2.5 or 5% ESBM during 1-10 days. Then, all groups were fed the same basic diets until 42 d. The obtained results indicated that supplementation of ESBM in early stage resulted in heavier BW and a better feed convers

The Transcription of Flight Energy Metabolism Enzymes Declined with Aging While Enzyme Activity Increased in the Long-Distance Migratory Moth, Spodoptera frugiperda.

Of all the things that can fly, the flight mechanisms of insects are possibly the least understood. By using RNAseq, we studied the aging-associated gene expression changes in the thorax of Spodoptera frugiperda females. Three possible flight energy metabolism pathways were constructed based on 32 key metabolic enzymes found in S. frugiperda. Differential expression analysis revealed up to 2000 DEGs within old females versus young ones. Expression and GO and KEGG enrichment analyses indicated that most genes and pathways related to energy metabolism and other biological processes, such as tran

Increasing angiotensin-converting enzyme concentrations and absent angiotensin-converting enzyme activity are associated with adverse kidney outcomes in pediatric septic shock.

BACKGROUND: Sepsis-induced endothelial dysfunction is proposed to cause angiotensin-converting enzyme (ACE) dysfunction and renin-angiotensin-aldosterone system (RAAS) derangement, exacerbating vasodilatory shock and acute kidney injury (AKI). Few studies test this hypothesis directly, including none in children. We measured serum ACE concentrations and activity, and assessed their association with adverse kidney outcomes in pediatric septic shock. METHODS: A pilot study of 72 subjects aged 1 week-18 years from an existing multicenter, observational study. Serum ACE concentrations and activit

Calcium Fluoride/Manganese Dioxide Nanocomposite with Dual Enzyme-like Activities for Uric Acid Sensing: A Comparative Study of Enzyme and Nonenzyme Methods.

The debate over enzyme methods versus nonenzyme methods in the field of nanosensing has lasted for decades despite hundreds of published studies on this topic. In this study, we first present a comparative analysis of these methods using a reaction based on the CaF2/MnO2 nanocomposite (CM Nc) with dual-enzyme activity, presenting oxidase- and peroxidase-like activities. Uric acid (UA) is a byproduct of purine metabolism in the body, and abnormal levels can cause many diseases; hence, tracking the amount of UA in human serum is crucial. The enzyme method was established using uricase and CM Nc:

Antioxidant Activity of Tetranitrosyl Iron Complex with Thiosulfate Ligands and Its Effect on Catalytic Activity of Mitochondrial Enzymes In vitro.

The antioxidant and antiradical properties of the tetra nitrosyl iron complex with thiosulfate ligands (TNIC) were studied in vitro in mouse brain homogenates. It was found for the first time that TNIC is an effective antioxidant. The effect of TNIC on the catalytic activity of mitochondrial enzymes cytochrome c oxidase and monoamine oxidase A was studied. It was shown for the first time that TNIC is an inhibitor of the catalytic activity of cytochrome c oxidase and monoamine oxidase A in animal brain mitochondria in vitro.

Molecular imprint of enzyme active site by camel nanobodies: rapid and efficient approach to produce abzymes with alliinase activity.

Screening of inhibitory Ab1 antibodies is a critical step for producing catalytic antibodies in the anti-idiotypic approach. However, the incompatible surface of the active site of the enzyme and the antigen-binding site of heterotetrameric conventional antibodies become the limiting step. Because camelid-derived nanobodies possess the potential to preferentially bind to the active site of enzymes due to their small size and long CDR3, we have developed a novel approach to produce antibodies with alliinase activities by exploiting the molecular mimicry of camel nanobodies. By screening the cam

Species differences in induction of hepatic enzymes by BM 17.0744, an activator of peroxisome proliferator-activated receptor alpha (PPARalpha).

BM 17.0744, a new anti-diabetic and lipid-lowering agent, leads also to strong hepatomegaly and carnitine acetyl transferase (CAT) increase in the liver of rats, a phenomenon known from fibrates. For information on the relevance of changes in liver of rats to other species, we investigated the effects of BM 17.0744 on lipids and selected marker enzymes related to beta-oxidation in rats, dogs and guinea-pigs, so-called high and low responders to peroxisome proliferators. To examine selectivity other enzymes were also determined, e.g. esterase, urate oxidase (UOX) and cytochrome c oxidase (CYT.C

Serum angiotensin-converting enzyme activity, concentration, and specific activity in granulomatous interstitial lung disease, tuberculosis, and COPD.

Angiotensin-converting enzyme (ACE) activity in serum is used as an aid to the diagnosis and follow-up of patients with sarcoidosis. A theoretical limitation of measurements of activity is that these may be affected by the presence of pharmacologic or endogenous inhibitors of ACE. Immunoassays of ACE concentration avoid this problem and, when combined with tests of ACE activity, permit calculation of specific activity of ACE. In this study, we set out to develop a sensitive radioimmunoassay for ACE to compare results obtained with this method with results of ACE activity and calculated ACE spe

The interference of leukocytes and platelets with measurement of clucose-6-phosphate dehydrogenase activity of erythrocytes with low activity variants of the enzyme.

Complete removal of leukocytes and platelets from whole blood showed that the glucose-6-phosphage dehydrogenase (G6PD) activity in "pure" erythrocytes from G6PD deficient hemizygous Sardinian subjects is consistently lower than reported in the literature. Thus, although non of 27 hemizygous subjects showed undetectable erythrocyte G6PD activity, their levels ranged between 0.0015 and 0.008 IU/g Hb, as compared with a mean value of 4.5 IU/g Hb in normal subjects. Most of the biochemical peoperties that were formerly ascribed to erythrocyte G6PD appear to be those of the enzyme from contaminatin

Engineering a thermostable MHET-active esterase through active-site compaction for efficient dual-enzyme PET depolymerization.

Efficient enzymatic recycling of polyethylene terephthalate (PET) is frequently hindered by the accumulation of mono-(2-hydroxyethyl) terephthalate (MHET), a soluble intermediate that competitively suppresses PET hydrolase activity. Although MHET-hydrolysing enzymes can overcome this limitation, these enzymes often exhibit poor catalytic performance or thermal compatibility with highly active PETases. In this study, we characterized a previously uncharacterized MHET-active esterase from Saccharothrix carnea and enhanced its catalytic and thermal properties through structure-guided semirational

The Platelet Activating Factor-Platelet Activating Factor Acetylhydrolase Enzyme Axis in Anaphylaxis: Current Evidence and Future Perspectives.

Anaphylaxis is a severe systemic hypersensitivity reaction characterized by rapid onset, unpredictable clinical course, and potentially fatal outcomes. Although its diagnosis remains primarily clinical, the identification of biomarkers capable of improving risk stratification and understanding disease mechanisms remains an important unmet need. Among the mediators implicated in anaphylaxis, platelet-activating factor (PAF) has emerged as a key effector molecule involved in vascular permeability, bronchoconstriction, platelet activation, and cardiovascular dysfunction. The biological activity o

Changes in Disposition of Ezetimibe and Its Active Metabolites Induced by Impaired Hepatic Function: The Influence of Enzyme and Transporter Activities.

Ezetimibe (EZE) is a selective cholesterol absorption inhibitor. Hepatic impairment significantly increases the systemic exposure of EZE and its main active phenolic glucuronide, EZE-Ph. Although changes in efflux transporter activity partly explain the changes in EZE-Ph pharmacokinetics, the causes of the changes to EZE and the effects of the administration route on EZE-Ph remain unclear. A carbon tetrachloride (CCl4)-induced hepatic failure rat model was combined with in vitro experiments to explore altered EZE and EZE-Ph disposition caused by hepatic impairment. The plasma exposure of EZE a

Active Site Hydrophobicity and the Convergent Evolution of Paraoxonase Activity in Structurally Divergent Enzymes: The Case of Serum Paraoxonase 1.

Serum paraoxonase 1 (PON1) is a native lactonase capable of promiscuously hydrolyzing a broad range of substrates, including organophosphates, esters, and carbonates. Structurally, PON1 is a six-bladed β-propeller with a flexible loop (residues 70-81) covering the active site. This loop contains a functionally critical Tyr at position 71. We have performed detailed experimental and computational analyses of the role of selected Y71 variants in the active site stability and catalytic activity in order to probe the role of Y71 in PON1's lactonase and organophosphatase activities. We demonstrate

In vitro acaricidal activity of 1,8-cineole against Sarcoptes scabiei var. cuniculi and regulating effects on enzyme activity.

1,8-Cineole found in many essential oils is a monoterpene and acts as a repellent against Sarcoptes scabiei var. cuniculi. In the present study, the acaricidal activity of 1,8-cineole against S. scabiei var. cuniculi was evaluated and the acaricidal mechanism was also investigated by assaying enzyme activities. The results showed that the lethal concentration of 50% (LC50) value (95% confidence limit (CL)) and the lethal time of 50% (LT50) value (95% CL) of 1,8-cineole were 2.77 mg/mL and 3.606 h, respectively. The pathological changes under transmission electron microscopy showed that the mor

Proteolytic activity at quantum dot-conjugates: kinetic analysis reveals enhanced enzyme activity and localized interfacial "hopping".

Recent studies show that polyvalent, ligand-modified nanoparticles provide significantly enhanced binding characteristics compared to isolated ligands. Here, we assess the ability of substrate-modified nanoparticles to provide enhanced enzymatic activity. Energy transfer assays allowed quantitative, real-time measurement of proteolytic digestion at polyvalent quantum dot-peptide conjugates. Enzymatic progress curves were analyzed using an integrated Michaelis-Menten (MM) formalism, revealing mechanistic details, including deviations from classic MM-behavior. A "hopping" mode of proteolysis at

The role of enzyme activity and charge properties on the presynaptic neurotoxicity and the contracture-inducing activity of snake venom phospholipases A2.

Two toxic phospholipases A2, caudoxin from Bitis caudalis venom and the basic phospholipase A2 from Naja nigricollis venom, were selected for a study of the contribution of enzyme activity and charge properties to the presynaptic neurotoxicity and the contracture-inducing activity of the enzyme. Reducing catalytic activity by lowering the incubation temperature or by substitution of Sr2+ for Ca2+ in the bathing medium causes a significant prolongation of the time to neuromuscular block by caudoxin, but does not change the contracture-inducing activity of Naja nigricollis basic phospholipase A2

New mechanistic frontiers in carbohydrate-active enzymes revealed by QM/MM simulations.

Carbohydrate-active enzymes (CAZymes) catalyze reactions central to glycan degradation, biosynthesis, and remodeling. Although their mechanisms have traditionally been described using a limited set of canonical models, recent studies are revealing a broader and more dynamic mechanistic landscape. We discuss how QM/MM-based enhanced-sampling methods have expanded our understanding of CAZyme catalysis by uncovering substrate-assisted pathways, complex proton-transfer processes, unconventional catalytic residues, and the roles of substrate distortion and enzyme dynamics in shaping reactivity. Counterion-Engineered Redox-Active Lipid Cubic Phases for Stable Enzyme Bioelectrodes.

The fabrication of stable and highly performing enzyme-based electrodes is key for the effective generation of biodevices and bioelectronics, such as electrochemical biosensors. In this context, redox-active lyotropic liquid crystals based on 3D nanomaterials, known as lipid cubic phases (LCP), hold great potential due to the large specific surface area and the possibility of being functionalized. In this study, we functionalized a monoolein (MO) LCP matrix by incorporating an amphiphilic redox shuttle within its matrix with the aim of enhancing the electrochemical performance of a glucose oxi
