Identification of Rice m6A Methyltransferase Complex Subunits MTA/MTB and Their Effects on Heading Date and Grain Weight.

N6-methyladenosine (m6A) is a vital epitranscriptomic modification that regulates plant development and stress responses. In rice (Oryza sativa L.), the identity of the catalytic subunit (MTA) of the core m6A writer complex has remained controversial because of inconsistent annotations based primarily on sequence homology. Here, phylogenetic analysis identified LOC_Os02g45110 as the putative catalytic subunit OsMTA, whereas LOC_Os01g16180 and LOC_Os03g05420 were classified as the structural subunits OsMTB1 and OsMTB2, respectively. All three proteins showed nuclear enrichment. Yeast two-hybrid

Mitochondrial dysfunction in neurodegenerative disorders: mechanisms and therapeutic advances.

Mitochondrial dysfunction is a core pathogenic mechanism underlying a broad spectrum of neurodegenerative disorders, from Alzheimer's and Parkinson's diseases to inherited optic neuropathies and mitochondrial ataxias. This review provides a comprehensive analysis of how defects in mitochondrial and nuclear DNA converge to disrupt oxidative phosphorylation, mitochondrial dynamics, calcium homeostasis, and quality control pathways, leading to energy depletion, oxidative stress, and neuronal degeneration across multiple disease contexts. Building on this mechanistic foundation, we examine how the

Nutrigenomic regulation of one-carbon metabolism and the circadian clock in health and disease.

Methylation of DNA, histones and RNA is central to the regulation of circadian rhythms, yet the biochemical origin of the methyl groups driving these modifications has received comparatively little attention in circadian biology. This review explores the bidirectional crosstalk between the methyl cycle and the mammalian circadian clock. We describe how S-adenosylmethionine-dependent epigenetic and epitranscriptomic modifications constitute essential layers of circadian gene regulation, and how the clock orchestrates the rhythmic expression of one-carbon metabolism enzymes and oscillations in S

Targeting METTL3 Attenuates Thyroid Inflammatory Injury by Restoring Th17/Treg Balance through a YTHDC2-m6A-Dependent KDR/VEGFA Loop.

Autoimmune thyroiditis (AIT) is characterized by extensive lymphocytic infiltration and progressive destruction of thyroid follicular cells (TFCs), yet the molecular mechanisms underlying persistent thyroid injury remain poorly defined. Here, methyltransferase-like 3 (METTL3) mediated N6-methyladenosine (m6A) modification is identified as a central driver of thyroid inflammation. METTL3 expression and m6A levels were markedly increased in TFCs from AIT patients and EAT mice, and are positively associated with immune inflammation scores. Genetic or pharmacological inhibition of METTL3 suppresse

Epigenetic mechanisms of retroviral regulation: a comparative review.

Major pharmacologic advances the past few decades have transformed infection with Human Immunodeficiency Virus (HIV) from a fatal disease into a chronic, manageable condition for people with access to antiretroviral therapy (ART). However, a cure remains elusive because HIV persists in a latent state throughout the body, evading immune clearance after ART suppression. Our understanding of HIV latency has made tremendous strides the past few decades, but the specific epigenetic mechanisms underlying latency are still being elucidated. Insights might be gained from simpler retroviruses capable o

Targeting IGF2BP3 in Cancer: From Molecular Structure and Biology to Early Drug Discovery.

RNA-binding proteins (RBPs) remain underexplored as small-molecule targets, although their dysregulation contributes to numerous human diseases, including cancer. RBPs are key regulators of post-transcriptional gene expression, controlling multiple stages of RNA metabolism. Among them, insulin-like growth factor 2 mRNA-binding protein 3 (IGF2BP3) is an oncofetal RBP that is highly expressed during embryonic development, largely absent in adult tissues, and re-expressed in multiple malignancies. A growing body of evidence supports IGF2BP3 as a diagnostic and prognostic biomarker and a potent on

Epigenetic modulation in cancer drug discovery: promising targets and clinical applications.

Epigenetic modulation has emerged as a central strategy that can change the fate of tumour cells to offer more rational and precise approaches by modulating reversible changes in chromatin structure, regulating gene expression without altering DNA sequence. Many reports have indicated the contributions of abnormal epigenetic alterations, particularly DNA methylation and histone modification patterns, as well as their association with non-coding RNA interactions during cancer emergence, development or resistance to standard therapies. Ongoing studies on various inhibitors also demonstrate encou

Pathomechanistic Networks of Motor System Injury in Amyotrophic Lateral Sclerosis.

Amyotrophic Lateral Sclerosis (ALS) is the most common, adult-onset, progressive motor neurodegenerative disorder that results in death within 3 years of the clinical diagnosis. Due to the clinicopathological heterogeneity, any reliable biomarkers for diagnosis or prognosis of ALS have not been identified till date. Moreover, the only three clinically approved treatments are not uniformly effective in slowing the disease progression. Over the last 15 years, there has been a rapid advancement in research on the complex pathomechanistic landscape of ALS that has opened up new avenues for success

Exploring the Roles of m6A-Modified circRNAs in Myasthenia Gravis Based on Multi-Omics Analysis.

Myasthenia gravis (MG) is an autoimmune disease mediated by autoantibodies. The important roles of circRNAs modified by m6A methylation have been reported in the pathogenesis of other autoimmune diseases, but remain unclear in MG. To address this point, we collected peripheral blood mononuclear cells from six MG patients and six healthy controls and performed m6A‑circRNA epitranscriptomic microarray and RNA sequencing. Differentially m6A-modified circRNAs and differentially expressed genes (DEGs) were analyzed. A network was constructed containing 17 circRNAs, 30 miRNAs, and 34 DEGs. The GSE85

Restoration of mitochondrial function through activation of hypomodified tRNAs with pathogenic mutations associated with mitochondrial diseases.

Mutations in mitochondrial (mt-)tRNAs frequently cause mitochondrial dysfunction. Mitochondrial myopathy, encephalopathy, lactic acidosis, and stroke-like episodes (MELAS), and myoclonus epilepsy associated with ragged red fibers (MERRF) are major clinical subgroups of mitochondrial diseases caused by pathogenic point mutations in tRNA genes encoded in mtDNA. We previously reported a severe reduction in the frequency of 5-taurinomethyluridine (τm5U) and its 2-thiouridine derivative (τm5s2U) in the anticodons of mutant mt-tRNAs isolated from the cells of patients with MELAS and MERRF, respectiv

5-methylcytosine modification by Plasmodium NSUN2 stabilizes mRNA and mediates the development of gametocytes.

5-methylcytosine (m5C) is an important epitranscriptomic modification involved in messenger RNA (mRNA) stability and translation efficiency in various biological processes. However, it remains unclear if m5C modification contributes to the dynamic regulation of the transcriptome during the developmental cycles of Plasmodium parasites. Here, we characterize the landscape of m5C mRNA modifications at single nucleotide resolution in the asexual replication stages and gametocyte sexual stages of rodent (Plasmodium yoelii) and human (Plasmodium falciparum) malaria parasites. While different represe

RNA-binding protein YTHDF3 suppresses interferon-dependent antiviral responses by promoting FOXO3 translation.

IFN-stimulated genes (ISGs) are essential effectors of the IFN-dependent antiviral immune response. Dysregulation of ISG expression can cause dysfunctional antiviral responses and autoimmune disorders. Epitranscriptomic regulation, such as N6-methyladenosine (m6A) modification of mRNAs, plays key roles in diverse biological processes. Here, we found that the m6A "reader" YT521-B homology domain-containing family 3 (YTHDF3) suppresses ISG expression under basal conditions by promoting translation of the transcription corepressor forkhead box protein O3 (FOXO3). YTHDF3 cooperates with two cofact

Hepatic Mettl14 Knockout Disrupts Bile Acid Homeostasis and Alters Fecal Microbiota Composition.

Bile acids are multifunctional signaling molecules that play significant roles in maintaining microbial homeostasis. N6-methyladenine (m6A), the most abundant epitranscriptomic modification, mediates various biological processes by modulating RNA metabolism. However, the precise regulatory mechanisms of m6A methylation in bile acid metabolism, and its downstream effects on microbiota remain unclear. In this study, liver-specific Mettl14 knockout (Mettl14-LKO) reshaped bile acid profile and expression levels of proteins related to bile acid metabolism, namely CYP7A1, FXR, and BSEP. M6A-seq data

METTL14 promotes trophoblast dysfunction by elevating SZRD1 expression in an m6A-dependent manner.

Preeclampsia, a common hypertensive pregnancy disorder associated with shallow trophoblast invasion, is a leading cause of perinatal and maternal mortality worldwide. Many studies have indicated that genes are differentially expressed between normal and diseased states. This study found that SUZ RNA binding domain containing 1 (SZRD1) expression was increased in placenta from preeclamptic pregnancies. However, the role and mechanism of SZRD1 in patients with preeclampsia are unclear. Gene expression was determined by RT-qPCR and Western blot. Cell proliferation, apoptosis, migration, invasion,

METTL3 regulates exocytosis independently of m6A.

RNA modification pathways are often misregulated in various cancers, with N6-methyladenosine (m6A) having a pivotal role in cancer progression and metastasis. Methyltransferase-like 3 (METTL3), a core component of the m6A methyltransferase complex, not only functions as an m6A writer but also promotes tumorigenesis through m6A-independent mechanisms. Here, we show that METTL3 is mislocalized to the cytoplasm in breast cancer tumors from patients, contributing to the oncogenic phenotype. Cytoplasmic METTL3 interacts with EXOC7, a key regulator of exocytosis, promoting its stabilization. In addi

B cell-specific METTL3 depletion exacerbates experimental autoimmune encephalomyelitis.

N6-methyladenosine (m6A), the most prevalent RNA modification, plays a pivotal role in regulating mRNA metabolism and cellular processes such as immune responses. Although the m6A methyltransferase METTL3 is known to regulate T-cell homeostasis and influence experimental autoimmune encephalomyelitis (EAE, a model for multiple sclerosis (MS)), its function within B cells remains poorly defined. Crucially, we observed that METTL3 expression is significantly downregulated in peripheral blood mononuclear cells (PBMCs) from MS patients and within B cells isolated from EAE mice. To directly investig

METTL16 Modulates GPX4 Expression to Regulate Chondrocyte Ferroptosis.

BACKGROUND: Achondroplasia (ACH), the predominant inherited form of disproportionate short stature, results from specific genetic alterations in fibroblast growth factor receptor 3 (FGFR3). N6-methyladenosine (m6A) modification is reported to modulate mRNA stability and translation. The present investigation systematically explored the epigenetic regulatory function of METTL16, an m6A RNA methyltransferase, within the pathophysiological framework of ACH. METHODS: We generated an ACH mouse model via Fgfr3380R (Fgfr3ach) gene mutation. Primary chondrocytes were isolated from newborn mice and st

Decoding osteosarcoma pathogenesis: the pivotal influence of m6A modification.

Osteosarcoma (OS) is the predominant primary malignant bone tumor in children and adolescents. Current treatments mainly include surgical resection combined with chemotherapy, but they still cannot effectively control tumor metastasis and recurrence, and may pose a risk of long-term complications. N6-methyladenosine (m6A) is the most widespread internal modification. It orchestrates various biological and disease-related events via the reversible m6A machinery comprising writers, erasers and readers. As a key epigenetic regulatory mechanism, m6A modification opens new avenues for deeper explor
