d-Sulpiride inhibits oral behaviour elicited from the nucleus accumbens of freely moving rats.

The present study analyzed the effect of intra-accumbens administration of the stereoisomers of sulpiride upon (3,4-dihydroxyphenylimino)-2-imidazoline (DPI)-induced changes in oral behaviours and electromyographic patterns of jaw muscles. In line with earlier findings, DPI (5 micrograms) administered into the nucleus accumbens increased chewing and tremor. l-Sulpiride (2-50 ng) had no effect on DPI-induced oro-facial behaviours. d-Sulpiride (10-50 ng) significantly antagonized the DPI-induced increase in chewing and had a biphasic effect on tremor with potentiation (10 ng) followed by attenua

Long-range circuit of the mouse spinal trigeminal nucleus caudalis.

BACKGROUND: The spinal trigeminal nucleus caudalis (SPVC) is a key central relay in the trigeminal circuit that processes orofacial nociceptive and somatosensory information. However, its long-range connectivity patterns remain poorly characterized. METHODS: Using viral tracing and fluorescence micro-optical sectioning tomography (fMOST), we obtained three-dimensional datasets of long-range input-output circuits of the SPVC in adult male C57BL/6J mice. An orofacial pain model was established by injecting formalin into the whisker pad of adult male C57BL/6J mice, followed by pain behavior assa

Craniofacial postherpetic neuralgia managed by stereotactic spinal trigeminal nucleotomy.

Postherpetic craniofacial neuralgias are notoriously difficult to deal with. Nevertheless, stereotactic spinal trigeminal nucleotomy seems to be a rational approach, as both experimental and clinical data strongly suggest the relevance of nucleus caudalis for certain facial neurogenic pain phenomena. From a series of 136 consecutive nucleotomies, 80 were performed for deafferentation pain. The long-term results of 25 such cases, who underwent this procedure for postherpetic neuralgia, are reported. Their pain was referred to the Vth, to the VII, IX and Xth, and to the C2-3 dermatomes. Abolitio

Trigeminal Herpes Zoster With Longitudinally Extensive Spinal Trigeminal Tract Involvement Presenting as an Isolated Facial Palsy Mimicking Ramsay Hunt Syndrome.

Varicella-zoster virus (VZV) is a neurotropic alpha herpesvirus capable of producing a wide spectrum of CNS disease, ranging from aseptic meningitis and vasculopathy to encephalitis and myelitis. Less widely appreciated is its capacity to extend centrally along the trigeminal pathway into the brainstem, producing intra-axial spinal trigeminal tract involvement that may remain clinically silent or manifest only as a cranial neuropathy, particularly when cerebrospinal fluid (CSF) polymerase chain reaction (PCR) is negative. We describe an immunocompetent 41-year-old man who presented with herpes

Descending serotonergic modulation from rostral ventromedial medulla to spinal trigeminal nucleus is involved in experimental occlusal interference-induced chronic orofacial hyperalgesia.

BACKGROUND: Dental treatment associated with unadaptable occlusal alteration can cause chronic primary myofascial orofacial pain. The serotonin (5-HT) pathway from the rostral ventromedial medulla (RVM) exerts descending modulation on nociceptive transmission in the spinal trigeminal nucleus (Sp5) and facilitates chronic pain. The aim of this study was to investigate whether descending 5-HT modulation from the RVM to the Sp5 is involved in the maintenance of primary myofascial orofacial hyperalgesia after persistent experimental occlusal interference (PEOI) or after delayed removal of experime

Exogenous histamine and H 2 receptor activation and H 3 receptor inhibition in nucleus accumbens modulate formalin-induced orofacial nociception through opioid receptors.

It has been demonstrated that the nucleus accumbens (NAc) plays an important role in modulation of nociception due to its extensive connections with different regions of the brain. In addition, this nucleus receives histaminergic projections from tuberomammillary nucleus. Considering the role of the central histaminergic system in nociception, the effect of histamine and its H 2 and H 3 receptors agonist and antagonist microinjections into the NAc on orofacial formalin nociception was investigated. In male Wistar rats, using stereotaxic surgery, two guide cannulas were bilaterally implanted in

Interleukin-6 induces spatially dependent whole-body hypersensitivity in rats: implications for extracephalic hypersensitivity in migraine.

BACKGROUND: Migraine is a complex neurological disorder that is characterized by throbbing head pain, increased sensitivity to light, sound, and touch, as well as nausea and fatigue. It is one of the most common and most disabling disorders globally but mechanisms causing migraine are poorly understood. While head pain is a typical feature of attacks, they also often present with cutaneous hypersensitivity in the rest of the body. In contrast, primary pain conditions in the lower parts of the body are less commonly associated with cephalic hypersensitivity. Previous studies indicate that appli

SCN2B in the Rat Trigeminal Ganglion and Trigeminal Sensory Nuclei.

The beta-2 subunit of the mammalian brain voltage-gated sodium channel (SCN2B) was examined in the rat trigeminal ganglion (TG) and trigeminal sensory nuclei. In the TG, 42.6 % of sensory neurons were immunoreactive (IR) for SCN2B. These neurons had various cell body sizes. In facial skins and oral mucosae, corpuscular nerve endings contained SCN2B-immunoreactivity. SCN2B-IR nerve fibers formed nerve plexuses beneath taste buds in the tongue and incisive papilla. However, SCN2B-IR free nerve endings were rare in cutaneous and mucosal epithelia. Tooth pulps, muscle spindles and major salivary g

B7.2 on activated and phagocytic microglia in the facial axotomy model: regulation by interleukin-1 receptor type 1, tumor necrosis factor receptors 1 and 2 and endotoxin.

Co-stimulatory factors are involved in different forms of brain pathology and play an important role in the activation of T-cells. In the current study, we explored the regulation of B7.2, a prominent member of the B7 family of costimulatory factors, in the facial motor nucleus (FMN) following facial axotomy and systemic application of lipopolysaccharide (LPS, endotoxin) using light and electron immunohistochemistry and cytokine-receptor-deficient mice. Facial axotomy led to a gradual increase of B7.2 immunoreactivity (IR) on microglial cell surface; similar effects were also observed followin

Integrating mind and body: Investigating differential activation of nodes of the default mode network.

BACKGROUND: The default mode network (DMN) is a large-scale brain network tightly correlated with self and self-referential processing, activated by intrinsic tasks and deactivated by externally-directed tasks. OBJECTIVE: In this study, we aim to investigate the novel approach of default mode activation during progressive muscle relaxation and examine whether differential activation patterns result from the movement of different body parts. METHODS: We employed neuroimaging to investigate DMN activity during simple body movements, while performing progressive muscle relaxation. We focused on

Motonuclear changes after cranial nerve injury and regeneration.

Little is known about the mechanisms at play in nerve regeneration after nerve injury. Personal studies are reported regarding motonuclear changes after regeneration of injured cranial nerves, in particular of the facial and oculomotor nerves, as well as the influence that the natural molecule acetyl-L-carnitine (ALC) has on post-axotomy cranial nerve motoneuron degeneration after facial and vagus nerve lesions. Adult and newborn animal models were used. Massive motoneuron response after nerve section and reconstruction was observed in the motonuclei of all nerves studied. ALC showed to have s

An Autopsy Case of Amyotrophic Lateral Sclerosis With Sudden Death Showed Histological Features of Lewy Body Disease.

We present the case of an 81-year-old man diagnosed with probable amyotrophic lateral sclerosis (ALS) based on the Updated Awaji criteria. The patient exhibited progressive motor neuron degeneration with muscle weakness, atrophy, and fasciculations primarily in the right lower limb and later extending to the right upper limb. Three months after being referred to a home care clinic, he collapsed in front of his family members and died. An autopsy revealed phosphorylated TDP-43 pathology consistent with ALS, with involvement of the hypoglossal nucleus, facial nerve nucleus, and medulla oblongata

Functional brain stem circuits for control of nose motion.

Rodents shift their nose from side to side when they actively explore and lateralize odors in the space. This motor action is driven by a pair of muscles, the deflector nasi. We studied the premotor control of this motion. We used replication-competent rabies virus to transsynaptically label inputs to the deflector nasi muscle and find putative premotor labeling throughout the parvocellular, intermediate, and gigantocellular reticular formations, as well as the trigeminal nuclei, pontine reticular formation, midbrain reticular formation, red nucleus, and superior colliculus. Dynamic associations in the cerebellar-motoneuron network during motor learning.

We assessed here true causal directionalities in cerebellar-motoneuron (MN) network associations during the classical conditioning of eyelid responses. For this, the firing activities of identified facial MNs and cerebellar interpositus (IP) nucleus neurons were recorded during the acquisition of this type of associative learning in alert behaving cats. Simultaneously, the eyelid conditioned response (CR) and the EMG activity of the orbicularis oculi (OO) muscle were recorded. Nonlinear association analysis and time-dependent causality method allowed us to determine the asymmetry, time delays,

Osteopontin-immunoreactivity in the rat trigeminal ganglion and trigeminal sensory nuclei.

Osteopontin-immunoreactivity (OPN-ir) was examined in the oro-facial tissues and trigeminal sensory nuclei (principal sensory nucleus and spinal trigeminal nucleus) to ascertain the peripheral ending and central projection of OPN-containing primary sensory neurons in the trigeminal ganglion (TG). No staining was observed using mouse monoclonal anti-OPN antibody preabsorbed with recombinant mature OPN. OPN-immunoreactive (ir) peripheral endings were classified into two types: encapsulated and unencapsulated types. Unencapsulated endings were subdivided into two types: simple and complex types.

Basic FGF-like immunoreactivity in the developing and adult rat brainstem.

Although a variety of in vitro and in vivo actions of basic fibroblast growth factor (bFGF) on neuronal cells have been documented, the physiological role of this protein in the nervous system is still contested. Since the distribution of a molecule in the nervous system may provide cues for an understanding of its possible roles, we have begun to study its cellular localization in the central and peripheral nervous system using immunocytochemistry with an anti-bFGF-specific antibody. Here we provide an account on the distribution of bFGF-like immunoreactivity (bFGF-IR) in the brainstem of the

[The nucleus tractus solitarius: neuroanatomic, neurochemical and functional aspects].

The nucleus tractus solitarii (NTS) has long been considered as the first central relay for gustatory and visceral afferent informations only. However, data obtained during the past ten years, with neuroanatomical, biochemical and electrophysiological techniques, clearly demonstrate that the NTS is a structure with a high degree of complexity, which plays, at the medullary level, a key role in several integrative processes. The NTS, located in the dorsomedial medulla, is a structure of small size containing a limited number of neurons scattered in a more or less dense fibrillar plexus. The dis
