Metal-free glycosylation with glycosyl fluorides in liquid SO2.

Liquid SO2 is a polar solvent that dissolves both covalent and ionic compounds. Sulfur dioxide possesses also Lewis acid properties, including the ability to covalently bind Lewis basic fluoride ions in a relatively stable fluorosulfite anion (FSO2 -). Herein we report the application of liquid SO2 as a promoting solvent for glycosylation with glycosyl fluorides without any external additive. By using various temperature regimes, the method is applied for both armed and disarmed glucose and mannose-derived glycosyl fluorides in moderate to excellent yields. A series of pivaloyl-protected O- an

Decarboxylative Cross-Coupling of Acyl Fluorides with Potassium Perfluorobenzoates.

We report the transition metal-free decarboxylative cross-coupling reactions of acyl fluorides with potassium perfluorobenzoates. Compared with traditional transition metal-catalyzed cross-couplings, this protocol presents an extremely environmentally benign pathway to afford unsymmetrical diaryl ketones. To install perfluorophenyl groups, this method highlights highly selective, inexpensive, and nontoxic conditions. The reaction system tolerates various functional groups in acyl fluorides. Remarkably, all of the starting materials can be prepared from abundant carboxylic acids and the reactio

Stabilization and Extraction of Fluoride Anion Using a Tetralactam Receptor.

A neutral tetralactam macrocycle was prepared in a few minutes in one pot and at high concentration using commercially available starting materials. NMR titration studies in DMSO revealed an anion affinity order of F- > AcO- > Cl- > Br-. The receptor affinity for F- is very high due in part to formation of a self-complementary dimer comprised of two "saddle shaped" complexes. An X-ray crystal structure showed that the two F- ions within the dimer are separated by 3.39 Å. The electrostatic penalty for this close proximity is compensated by attractive interactions provided by the surrounding tet

Stannous chloride and stannous fluoride are inhibitors of matrix metalloproteinases.

OBJECTIVES: Matrix metalloproteinases (MMPs) in dentin and saliva can degrade collagen. Divalent metals are known inhibitors of MMPs, but stannous - such as in the form of stannous chloride (SnCl2) or stannous fluoride (SnF2) - is yet to be tested for a possible inhibitory effect. In this study, we tested the inhibitory effect on the proteolytic activity of MMP-2 and MMP-9. METHODS: Sodium chloride (NaCl), sodium fluoride (NaF), and chlorhexidine (CHX) were used as controls. Gelatin zymography was performed with recombinant human MMP-2 and MMP-9. SnCl2, SnF2, NaF, NaCl, and CHX were included

F-18 fluoride uptake in primary breast cancer.

OBJECTIVE: Bone-specific radiotracers are known to accumulate in breast lesions. Tc-99m diphosphonates have been widely studied in differentiating breast lesions. In this retrospective study, we aimed to assess the uptake of the bone-specific PET radiotracer, F-18 fluoride (NaF), in primary breast cancers to determine its sensitivity and to identify any differences in NaF uptake between calcified and non-calcified tumors, histological subtypes, and patients with or without axillary lymphadenopathy. METHODS: NaF positron emission tomography/computed tomography (PET/CT) images of 69 newly diagn

Evaluation of the staining potential of silver diamine fluoride: in vitro.

BACKGROUND: Silver diamine fluoride (SDF) is a cariostatic agent used to adjunctively manage dental caries in high-risk groups. However, black staining is a frequently reported side effect following SDF application and influences the acceptability of this treatment. AIM: To evaluate the staining potential of SDF and the influence of the potassium Iodide (KI) application on the degree of black staining over time. DESIGN: Extracted carious primary molars were collected and sorted into pairs matched by tooth type, size and location of the carious lesion (n = 35). Teeth were stratified to receiv

Dentin Erosion: Method Validation and Efficacy of Fluoride Protection.

The aging population experiences more gingival recession and root exposure which increases the opportunity for dentin erosion. This study tested the use of transverse microradiography (TMR) methods to assess dentin erosion and the interaction between fluoride and citric acid on the amount of erosion in the dentin samples. In a 4 × 3 interaction experimental design, four fluoride concentrations (0.00, 25.0, 50.0, and 100.0 mg/L) and three citric acid concentrations (0.0, 0.25, and 1.00%) were combined to form 12 experimental solutions. Forty-eight dentin samples were placed in the experimental

Evaluation of exposure to fluoride in child population of North Argentina.

Fluoride is an important element for humans. It inhibits initiation and progression of dental caries and stimulates bone formation. However, excessive intake may lead to the appearance of dental and/or skeletal fluorosis and a decrease in intellectual coefficient in child populations. This study evaluates exposure to fluoride in the child population of Chaco province (Argentina) by analysis of drinking water, food and its bioaccessible fraction (quantity of fluoride solubilised by gastrointestinal digestion and available for intestinal absorption) and urine as a biomarker of internal dose. The

Effects of sodium fluoride on immune response in murine macrophages.

Excessive fluoride intake may be harmful for health, producing dental and skeletal fluorosis, and effects upon neurobehavioral development. Studies in animals have revealed effects upon the gastrointestinal, renal and reproductive systems. Some of the disorders may be a consequence of immune system alterations. In this study, an in vitro evaluation is made of fluoride immunotoxicity using the RAW 264.7 murine macrophage line over a broad range of concentrations (2.5-75mg/L). The results show that the highest fluoride concentrations used (50-75mg/L) reduce the macrophage population in part as a

Does fluoride in drinking water delay tooth eruption?

OBJECTIVES: The objectives of this study are to determine the effect of fluoride exposure on permanent tooth eruption patterns as well as to understand its effect on caries attack rate by accounting for the number of erupted tooth surfaces. METHODS: We analyzed data from the 1986-1987 National Survey of Oral Health of US Schoolchildren to determine the mean number of erupted permanent teeth and permanent first molars according to fluoride level in drinking water. The analysis included 13,348 children aged 5-17 years with a history of single residence. We also estimated the attack rate (decaye

Position of the Academy of Nutrition and Dietetics: the impact of fluoride on health.

It is the position of the Academy of Nutrition and Dietetics to support optimal systemic and topical fluoride as an important public health measure to promote oral health and overall health throughout life. Fluoride is an important element in the mineralization of bone and teeth. The proper use of topical and systemic fluoride has resulted in major reductions in dental caries and its associated disability. Dental caries remains the most prevalent chronic disease in children and affects all age groups of the population. The Centers for Disease Control and Prevention has named fluoridation of wa…

The role of fluoride on the process of titanium corrosion in oral cavity.

Titanium is known to possess excellent biocompatibility as a result of corrosion resistance, lack of allergenicity when compared with many other metals. Fluoride is well known as a specific and effective caries prophylactic agent and its systemic application has been recommended widely over recent decades. Nevertheless, high fluoride concentrations impair the corrosion resistance of titanium. The purpose of this article is to summarize the current data regarding the influence of fluoride on titanium corrosion process in the last 5 years. These data demonstrate noxious effects induced by high f

Fluoride's effects on the formation of teeth and bones, and the influence of genetics.

Fluorides are present in the environment. Excessive systemic exposure to fluorides can lead to disturbances of bone homeostasis (skeletal fluorosis) and enamel development (dental/enamel fluorosis). The severity of dental fluorosis is also dependent upon fluoride dose and the timing and duration of fluoride exposure. Fluoride's actions on bone cells predominate as anabolic effects both in vitro and in vivo. More recently, fluoride has been shown to induce osteoclastogenesis in mice. Fluorides appear to mediate their actions through the MAPK signaling pathway and can lead to changes in gene exp

Sulfonamide-imines as selective fluorescent chemosensors for the fluoride anion.

Two new sulfonamide-type fluorescent chemosensors in organic media are reported. The two receptors, [N,N'-bis(2-tosylaminobenzylidene)-1,2-diaminoethane and N,N'-bis(2-tosylaminobenzylidene)-1,3-diamino-2-propanol], display marked changes in the fluorescence emission intensities as a result of deprotonation by basic anions, and show high selectivity for fluoride over other inorganic anions, such as acetate or dihydrogenphosphate. These results suggest that the presence of the imine group as an intramolecular H-bond acceptor enhances the selectivity of these sensors compared to previous example

How does fluoride affect dentin microhardness and mineralization?

Fluoride (F) has been a useful instrument in caries prevention. However, only limited data exist on the effect of its long-term use on dentin mineralization patterns and microhardness. The objective of this study was to evaluate the influence of tooth F concentration ([F]) and dental fluorosis (DF) severity on dentin microhardness and mineralization. We collected 137 teeth in Montreal and Toronto, Canada, and Fortaleza, Brazil, where optimum or suboptimum levels of water F were 0.2 ppm, 1 ppm, and 0.7 ppm, respectively. Teeth were analyzed for DF severity, dentin [F], enamel [F], dentin microh

[Effects of fluoride in drinking water on health of deciduous teeth].

INTRODUCTION: High incidence of decayed deciduous teeth, as well as lack of adequate therapy, makes tooth decay prevention very important. One of the simplest ways to reduce tooth decay is fluoridation of drinking water. The optimal concentration of fluoride in drinking water is 1 ppm/l, and many waters naturally contain this quantity. Waters in Vojvodina are mainly poor in fluoride, except in a few regions. It has long been postulated that fluoride has a prophyilactic effect during intrauterine life. Today a theory of greater local impact of fluoride has been accepted, as well as its role in …

The biochemistry and physiology of metallic fluoride: action, mechanism, and implications.

Fluoride is a well-known G protein activator. Activation of heterotrimeric GTP-binding proteins by fluoride requires trace amounts of Al3+ or Be2+ ions. AlFx mimics a gamma-phosphate at its transition state in a Galpha protein and is therefore able to inhibit its GTPase activity. AlFx also forms complexes with small GTP-binding proteins in the presence of their GTPase-activating proteins (GAP). As phosphate analogs, AlFx or BeFx affect the activity of a variety of phosphoryl transfer enzymes. Most of these enzymes are fundamentally important in cell signal transduction or energy metabolism.
