Role of Electroencephalography in the Assessment of Cortical Responses Elicited by Music Therapy in Burn Patients Undergoing Intensive Care.

Music therapy (MT) is increasingly being integrated into intensive care unit (ICU) settings to modulate pain, stress, and emotional dysregulation. Although clinically promising, objective biomarkers for quantifying its neurophysiological effects are still missing. In this context, the electroencephalogram (EEG) represents a valid tool to assess cortical dynamics associated with cognitive-affective engagement elicited by MT. Our study aims to evaluate the role of electroencephalography as an objective tool for monitoring cortical responses to MT in the ICU. EEGs acquired from nine burn patients

Cerebellar Transcranial Alternating Current Stimulation: Frequency-Specific Modulation of Human Gait.

Coordinated locomotion depends on rhythmic neural interactions, with the cerebellum contributing to movement timing and scaling. However, how cerebellar oscillatory dynamics influence human gait remains unclear, despite its relevance for motor learning and rehabilitation. We hypothesized two frequency-dependent effects: (i) gait-matched cerebellar transcranial alternating current stimulation (c-tACS) would produce stronger phase alignment than sham and selectively affect temporal rather than spatial gait parameters; and (ii) higher-frequency stimulation would preferentially modulate spatial ga

Mechanisms of sharp wave initiation and ripple generation.

Replay of neuronal activity during hippocampal sharp wave-ripples (SWRs) is essential in memory formation. To understand the mechanisms underlying the initiation of irregularly occurring SWRs and the generation of periodic ripples, we selectively manipulated different components of the CA3 network in mouse hippocampal slices. We recorded EPSCs and IPSCs to examine the buildup of neuronal activity preceding SWRs and analyzed the distribution of time intervals between subsequent SWR events. Our results suggest that SWRs are initiated through a combined refractory and stochastic mechanism. SWRs i

A Dynamic Mutual Information Measure of Phase-Amplitude Coupling With Uncertainty Quantification.

OBJECTIVE: Phase-amplitude coupling (PAC)-in which the phase of a low-frequency rhythm modulates the amplitude of a higher-frequency oscillation-is widely observed across the brain and linked to cognition and neurological disorders including Parkinson's disease, epilepsy, and depression. Standard PAC metrics aggregate over long windows and return a single summary statistic, obscuring transient structure. We introduce a dynamic PAC framework that captures fast, sample-level fluctuations in coupling strength while supporting multi-trial estimation and uncertainty quantification. METHODS: We dev

SIRT1 in Cardiac Diseases: Molecular Mechanisms, Therapeutic Potential, and Future Directions.

Sirtuin 1 (SIRT1), a nicotinamide adenine dinucleotide (NAD+)-dependent class III histone deacetylase, functions as a central metabolic sensor and stress-responsive regulator in the cardiovascular system. Unlike its well-characterized role in atherosclerosis, SIRT1 exerts multifaceted protective effects directly on cardiac tissue. This review synthesizes recent advances in understanding SIRT1-mediated cardioprotection across a spectrum of heart diseases, including myocardial ischemia/reperfusion (I/R) injury, heart failure (HF), diabetic cardiomyopathy (DCM), cardiac hypertrophy, aging-related

Stephanoside B modulates metabolic gene expression and lengthens the circadian bmal1 oscillation period in differentiated myotubes revealed by real-time bioluminescence.

BACKGROUND: Gymnema (G.) inodorum is a medicinal plant with anti-diabetic, anti-obesity, and anti-inflammatory properties traditionally consumed as tea in Southeast Asia. While bioactive stephanosides and gymnemic acids suppress adipocyte differentiation, their effects on skeletal muscle metabolism and circadian regulation remain unclear. This study investigated the effects of gymnemic acid (GiA-7) and stephanosides B and C in differentiated C2C12 myotubes. METHODS AND RESULTS: Differentiated C2C12 myotubes were treated with GiA-7 and stephanosides B and C, and gene expression was analyzed by

Double-blind, randomized, placebo-controlled pilot clinical trial with gamma-band transcranial alternating current stimulation for the treatment of schizophrenia refractory auditory hallucinations.

Gamma oscillations are essential for brain communication. The 40 Hz neural oscillation deficits in schizophrenia impair left frontotemporal connectivity and information communication, causing auditory hallucinations. Transcranial alternating current stimulation is thought to enhance connectivity between different brain regions by modulating brain oscillations. In this work, we applied a frontal-temporal-parietal 40 Hz-tACS stimulation strategy for treating auditory hallucinations and further explored the effect of tACS on functional connectivity of brain networks. 32 schizophrenia patients wit

Ictal gamma-band interactions localize ictogenic nodes of the epileptic network in focal cortical dysplasia.

OBJECTIVE: Epilepsy surgery fails in > 30% of patients with focal cortical dysplasia (FCD). The seizure persistence after surgery can be attributed to the inability to precisely localize the tissue with an endogenous potential to generate seizures. In this study, we aimed to identify the critical components of the epileptic network that were actively involved in seizure genesis. METHODS: The directed transfer function was applied to intracranial EEG recordings and the effective connectivity was determined with a high temporal and frequency resolution. Pre-ictal network properties were compare

Alterations in High-Frequency Neuronal Oscillations in a Cynomolgus Macaque Test of Sustained Attention Following NMDA Receptor Antagonism.

A growing body of evidence indicates that neuronal oscillations in the gamma frequency range (30-80 Hz) are disturbed in schizophrenic patients during cognitive processes and may represent an endophenotype of the disease. N-methyl-D-aspartate (NMDA) receptor antagonists have been used experimentally to induce schizophrenia-like symptoms including cognitive deficits in animals and humans. Here we characterized neuronal oscillations and event-related potentials (ERPs) in Cynomolgus macaques fully trained to perform a continuous performance test (CPT) in the presence and absence of the NMDA antag

Multi-sensory Gamma Stimulation Ameliorates Alzheimer's-Associated Pathology and Improves Cognition.

We previously reported that inducing gamma oscillations with a non-invasive light flicker (gamma entrainment using sensory stimulus or GENUS) impacted pathology in the visual cortex of Alzheimer's disease mouse models. Here, we designed auditory tone stimulation that drove gamma frequency neural activity in auditory cortex (AC) and hippocampal CA1. Seven days of auditory GENUS improved spatial and recognition memory and reduced amyloid in AC and hippocampus of 5XFAD mice. Changes in activation responses were evident in microglia, astrocytes, and vasculature. Auditory GENUS also reduced phospho

Cortico-Striatal Cross-Frequency Coupling and Gamma Genesis Disruptions in Huntington's Disease Mouse and Computational Models.

Abnormal gamma band power across cortex and striatum is an important phenotype of Huntington's disease (HD) in both patients and animal models, but neither the origin nor the functional relevance of this phenotype is well understood. Here, we analyzed local field potential (LFP) activity in freely behaving, symptomatic R6/2 and Q175 mouse models and corresponding wild-type (WT) controls. We focused on periods of quiet rest, which show strong γ activity in HD mice. Simultaneous recording from motor cortex and its target area in dorsal striatum in the R6/2 model revealed exaggerated functional c

Neurophysiologically-informed markers of individual variability and pharmacological manipulation of human cortical gamma.

The ability to quantify synaptic function at the level of cortical microcircuits from non-invasive data would be enormously useful in the study of neuronal processing in humans and the pathophysiology that attends many neuropsychiatric disorders. Here, we provide proof of principle that one can estimate inter-and intra-laminar interactions among specific neuronal populations using induced gamma responses in the visual cortex of human subjects - using dynamic causal modelling based upon the canonical microcircuit (CMC; a simplistic model of a cortical column). Using variability in induced (spec

Evidence for increased visual gamma responses in photosensitive epilepsy.

BACKGROUND: A sustained gamma (30-70 Hz) oscillation induced in occipital cortex by high-contrast visual stimulation has been well characterised in animal local field potential recordings and in healthy human participants using magnetoencephalography (MEG). The spatial frequency of a static grating stimulus that gives maximal gamma is also that most likely to provoke seizures in photosensitive epilepsy. METHODS: We used MEG to study visual responses induced by grating stimuli of varying contrast and size in twelve patients with photosensitive epilepsy and two matched control groups, one with

An increase in in vivo release of LHRH and precocious puberty by posterior hypothalamic lesions in female rhesus monkeys (Macaca mulatta).

Acetylcholine (ACh) is a neurotransmitter critical for normal cognition. Here we demonstrate heterogeneity of cholinergic signaling in neocortical neurons in the rat prefrontal, somatosensory, and visual cortex. Focal ACh application (100 muM) inhibited layer 5 pyramidal neurons in all cortical areas via activation of an apamin-sensitive SK-type calcium-activated potassium conductance. Cholinergic inhibition was most robust in prefrontal layer 5 neurons, where it relies on the same signal transduction mechanism (M1-like receptors, IP(3)-dependent calcium release, and SK-channels) as exists in

Elevated serum interferon-gamma in atopic asthma correlates with increased airways responsiveness and circadian peak expiratory flow variation.

Interleukin (IL)-4, IL-5 and interferon (IFN)-gamma are thought to play an important role in chronic airway inflammation in asthmatic subjects. Increased airways responsiveness and nocturnal airway obstruction are important clinical manifestations of asthma. The aim of this study was to investigate whether IL-4, IL-5 and IFN-gamma values are elevated in atopic asthma and correlate with its clinical manifestations. Serum IL-4, IL-5 and IFN-gamma levels of 17 atopic asthmatics and eight nonatopic healthy subjects were determined at 16:00 and 04:00 h by a chemiluminescence enzyme-linked immunosor
