Regulation of a hair follicle keratin intermediate filament gene promoter.

During hair growth, cortical cells emerging from the proliferative follicle bulb rapidly undergo a differentiation program and synthesise large amounts of hair keratin proteins. To identify some of the controls that specify expression of hair genes we have defined the minimal promoter of the wool keratin intermediate filament gene K2.10. The region of this gene spanning nucleotides -350 to +53 was sufficient to direct expression of the lacZ gene to the follicle cortex of transgenic mice but deletion of nucleotides -350 to -150 led to a complete loss of promoter activity. When a four base subst

Glucocorticoids regulate the expression of the human osteoblastic endothelin A receptor gene.

The endothelial cell-derived peptide endothelin 1 (ET1) stimulates cell proliferation and differentiated functions of human osteoblastic cells (HOC), and HOC constitutively express the endothelin A receptor (ETRA). Therefore, ET1 may play an important role in the regulation of bone cell metabolism. As glucocorticoids (GC) exert a profound influence on bone metabolism and increase the effects of ET1 on bone cell metabolism in vitro, the effects of GC on ETRA expression in HOC were investigated. Dexamethasone (DEX) increased ETRA mRNA levels in a dose- and time-dependent fashion. The effects of

Regulation of hepatic stearoyl-CoA desaturase gene 1 by vitamin A.

The effect of vitamin A supplementation on stearoyl-CoA desaturase gene 1 expression in mouse liver was characterized. Normal BALB/c mice were fed 0.01% and 0.1% retinol palmitate as components of nonpurified diets. This treatment resulted in a 3-fold and a 7-fold induction of SCD1 mRNA levels, respectively, as determined by RNase protection analysis. Vitamin A-deficient animals were also fed diets containing 0.01% and 0.1% retinol palmitate, resulting in a similar pattern of SCD1 mRNA induction. Fatty acid synthase and beta-actin mRNA levels did not respond consistently or significantly to re

The role of nuclear factor-kappa B in cytokine gene regulation.

Transcription factors are DNA-binding proteins that regulate gene expression. Nuclear factor-kappa B (NF-kappa B) is a critical transcription factor for maximal expression of many cytokines that are involved in the pathogenesis of inflammatory diseases, such as adult respiratory distress syndrome (ARDS) and sepsis syndrome. Activation and regulation of NF-kappa B are tightly controlled by a group of inhibitory proteins (I kappa B) that sequester NF-kappa B in the cytoplasm of immune/inflammatory effector cells. NF-kappa B activation involves signaled phosphorylation, ubiquitination, and proteo

Role of the staphylococcal accessory gene regulator (sar) in septic arthritis.

Staphylococcus aureus arthritis is a highly erosive disease in which both host and bacterial factors are of importance for its induction and progression. At the transcriptional level, three known loci act in regulating production of exoproteins and expression of cell wall structures. The aim of our study was to assess the role of the sar locus as a virulence determinant in the pathogenesis of septic arthritis. A recently established murine model of hematogenously spread S. aureus arthritis was employed. S. aureus strains, isogenic for the sar locus, were inoculated intravenously into NMRI mice

Temperature regulation of the streptococcal pyrogenic exotoxin A-encoding gene (speA).

The gene encoding the bacterial superantigen streptococcal pyrogenic exotoxin A is often found in streptococcal strains associated with the recently described streptococcal toxic shock syndrome. Here we demonstrate that this gene is expressed at approximate fourfold higher levels in cells grown at 37 degrees C when compared to cells grown at 26 degrees C. This suggests there is increased production of this toxin when Streptococcus pyogenes is found in infections of the soft tissues and bloodstream, as opposed to S. pyogenes that have not breached the epithelial layers and are living on the sur

Myometrial transcriptional regulation of the gap junction gene, connexin-43.

The mechanisms that enable the myometrium to switch from a state of relative quiescence during pregnancy to a muscle that is spontaneously active, very responsive to endogenous uterotonins and exhibits a high degree of cell-cell coordination are poorly understood. It is hypothesized that this switch or 'activation' of the myometrium results from the coordinated expression of a cassette of 'contraction-associated proteins'. The molecular mechanisms that regulate the expression of one of these, namely the myometrial gap junction protein connexin-43 (Cx-43), have been analysed. Myometrial Cx-43 e

Tumor suppressor genes as negative growth regulators in development and differentiation.

Tumor suppressor genes have received much attention for their roles in the development of human malignancies. However, their gene products (proteins) function as negative growth regulators and are highly expressed in many tissues during embryonic development, suggesting that they might function as critical proteins in differentiation and development. The evidence implicating tumor suppressor proteins in cellular differentiation and embryonic development will be presented with special attention to the neurofibromatosis 1, neurofibromatosis 2, and tuberous sclerosis 2 gene products.

Molecular cloning and analysis of the cDNA for an auxin-regulated calmodulin gene.

An auxin-regulated calmodulin cDNA (arCaM) were isolated by differential screening from a mung bean (Vigna radiata) cDNA library. The expression of the arCaM transcript in the etiolated mung bean hypocotyl was examined by RNA gel blot analysis. The arCaM transcript was induced depending on indole-3-acetic acid (IAA) concentrations. An increase in level of the arCaM transcript upon treatment of hypocotyl segments with 10 microM IAA was detected after 1 h and a maximum level was detected at 2 h. Induction of the arCaM transcript occurred upon treatment with 10 microM 2,4-dichlorophenoxyacetic ac

The PKD1 gene produces a developmentally regulated protein in mesenchyme and vasculature.

Autosomal dominant polycystic kidney disease (ADPKD) is one of the most common human genetic diseases. In addition to polycystic kidneys, the disease can cause cystic changes in liver and other organs, cardiac valvular insufficiency and cerebral arterial aneurysms. Using antibodies raised against the predicted gene product of PKD1, which is mutated in about 85% of ADPKD cases, we show that PKD1 is a 530-kD protein localized to the extracellular matrix of kidney, liver and cerebral blood vessels. We discovered that the PKD1 protein was highly expressed in the mesenchyme of developing kidney and

Syndromal mental retardation due to mutations in a regulator of gene expression.

Mental handicap is a common clinical problem that has been a relatively neglected area of research. Though the causes are varied and complex, molecular biologists are making progress in understanding the mechanisms in some cases, particularly where there are distinguishing phenotypic or genetic markers. The fortuitous association of alpha thalassaemia with a form of mental retardation has allowed us to define a specific X-linked syndrome (ATR-X). Positional cloning was used to define a disease interval and examination of candidate genes demonstrated that mutations in a gene, XH2, showing homol

Vitamin A regulates the expression of apolipoprotein AI and CIII genes in the rat.

The effect of vitamin A on the expression of apoAI and CIII genes in intact rats was studied. In vitamin A-deficient rats, the hepatic level of apoAI mRNA was increased and enhanced by an oral administration of excess retinoic acid(RA). A similar administration to normal rats caused an increase in the apoAI mRNA level in the intestine without affecting that of hepatic mRNA. Though the hepatic level of apo CIII mRNA was not affected by the vitamin A status, the intestinal level was positively regulated by vitamin A. These findings show that vitamin A regulates the expression of apolipoprotein A

Up-regulation of elafin/SKALP gene expression in psoriatic epidermis.

The expression of mRNA for elafin/SKALP, an inhibitor of leukocyte elastase and proteinase 3, in human normal and psoriatic epidermis was examined by in situ hybridization. In normal epidermis, elafin/SKALP mRNA was detected in the granular layer, but not in the spinous or basal layers. In fully developed psoriatic lesions, elafin/SKALP mRNA was found in the suprabasal layers of the ridges, and in the upper two thirds of the stratum malpighii at the elongated rete ridges. Intense staining was noted near the subcorneal microabscess in psoriasis vulgaris and under the subcorneal pustule in local

Estrogen regulation and localization of galanin gene expression in the rat uterus.

To determine whether galanin is a target for estrogenic regulation in the rat uterus, we measured the effects of estrogen on galanin mRNA expression in the uterus of ovariectomized rats and compared the results with the regulation of galanin in the anterior pituitary. Treatment of the animals with a single dose of 17 beta-estradiol resulted in a rapid transient increase of galanin mRNA, similar to that in the pituitary of the same animals, with a peak at 3 h after stimulation and a return to prestimulation levels after 24 h. No galanin mRNA was detected in ovariectomized control animals in eit

Estradiol regulation of proopiomelanocortin gene expression and peptide content in the hypothalamus.

Previous studies have shown that the hypothalamic concentrations of beta-endorphin (beta-EP) and other proopiomelanocortin (POMC)-derived peptides change in the female rat following castration and gonadal steroid replacement. In this study we have measured POMC mRNA by solution hybridization assay in the medial basal hypothalamus (MBH) of ovariectomized rats treated with a regimen of estradiol (E2) that we have previously shown alters brain beta-EP peptide content. In addition the effect of progesterone (P) was also studied. In the first experiment the concentration of beta-EP and alpha-melano

Calcitonin gene-related peptide (CGRP) in the regulation of salivary secretion.

The occurrence of calcitonin gene-related peptide (CGRP) in nerves in the submandibular gland was studied by using indirect immunofluorescence technique. The functional significance of CGRP was further evaluated by using an in vitro method for demonstration of protein and peroxidase secretion. CGRP-immunoreactive nerve fibers were observed mainly around large ducts and around blood vessels in the stromal part of submandibular gland. A moderate amount of CGRP-immunoreactivity were seen around the secretory acini as well. In vitro, CGRP increased significantly total protein and peroxidase secret

Regulation of growth hormone-releasing hormone gene expression and biosynthesis.

Growth hormone-releasing hormone (GRH) was initially isolated, characterized, sequenced, and cloned from human tumors and subsequently from the hypothalamus of humans and other animal species. Extensive structure-function studies have indicated the amino terminus to be most important for its biologic action, and the primary mechanism of its bioinactivation occurs by cleavage of an amino terminal dipeptide. The GRH gene is expressed primarily in the hypothalamic arcuate nucleus but also in the placenta. Expression of the GRH gene is regulated by growth hormone in a classical feedback manner, wi

Thyroid hormone regulation of the rat thyrotropin beta-subunit gene.

In order to determine the molecular mechanisms by which thyroid hormone and other factors regulate the biosynthesis of thyrotropin (TSH), we have isolated and characterized the gene encoding the beta-subunit of rat TSH. The transcriptional unit is 4.9 Kb in size and contains 3 exons and two introns of 3.9 and 0.4 Kb. The gene possesses two promoters and hence two associated transcriptional start sites. Several lines of evidence indicate that there is differential regulation of the two TSH beta transcripts by thyroid hormone. In particular, rat TSH beta mRNA-2, the transcript associated with th

Effects of Schistosoma mansoni on androgen regulated gene expression in the mouse.

Two-dimensional gel electrophoresis of liver mRNA translation products and dot-blot hybridization revealed that the levels of mRNA encoding major urinary proteins were greatly reduced in mice infected with Schistosoma mansoni. Major urinary protein mRNA levels are known to be androgen regulated. Dot-blot hybridization analysis of RNAs from various mouse tissues with a variety of cDNA probes indicated that all androgen-regulated mRNAs tested were reduced in infected mice. Administration of testosterone to infected animals restored urinary major urinary protein levels. Direct measurement of seru
