Identification of Differentially Expressed K-Ras Transcript Variants in Patients With Leiomyoma.

PURPOSE: Molecular studies have demonstrated a wide range of gene expression variations in uterine leiomyoma. The rat sarcoma virus/rapidly accelerated fibrosarcoma/mitogen-activated protein kinase (RAS/RAF/MAPK) is the crucial cellular pathway in transmitting external signals into nucleus. Deregulation of this pathway contributes to excessive cell proliferation and tumorigenesis. The present study aims to investigate the expression profile of the K-Ras transcripts in tissue samples from patients with leiomyoma. METHODS: The patients were leiomyoma cases who had no mutation in mediator comple

PRAME Gene Copy Number Variation Is Related to Its Expression in Multiple Myeloma.

Multiple myeloma (MM) patients commonly present abnormal expression of cancer-testis antigens, which may serve as immunotherapeutic targets and prognostic factors. We previously reported that preferentially expressed antigen of melanoma (PRAME) overexpression in bone marrow mononuclear cells is related to progression in MM patients treated with non-bortezomib-containing regimens. The mechanism underlying variations in PRAME expression remains unknown. To investigate the impact of gene copy number variation (CNV) on PRAME expression, plasma cells were sorted from 50 newly diagnosed patients and

Family mapping of previously identified patients with pathogenic or likely pathogenic ALPL variants using predictive genotyping and detailed phenotyping approach: the FAME case-control study.

Hypophosphatasia (HPP) is an inborn error of metabolism caused by loss-of-function variants in the ALPL gene, which encodes the tissue nonspecific isozyme of alkaline phosphatase (ALP). There is no typical phenotype in adults. We used a genotyping first approach to determine whether ALPL pathogenic variants were associated with musculoskeletal symptoms, mineral metabolism abnormalities, and an impact on quality of life. We recruited individuals with a pathogenic (or likely pathogenic) variant in ALPL gene (n = 26) and their relatives (n = 44). We performed genetic tests and compared the relati

Cascade testing in an ovarian cancer traceback genetic testing program: The GRACE study experience.

Approximately 20% of individuals diagnosed with ovarian cancer (OC) have an inherited pathogenic or likely pathogenic variant (P/LP) in a cancer risk gene. Though genetic testing for hereditary cancer risk is currently recommended at OC diagnosis, individuals who have not received risk information and their at-risk relatives (ARR) can benefit from genetic testing at any point. In a single-arm implementation study, the Genetic Risk Assessment in Ovarian CancEr (GRACE) study offered traceback genetic testing using a panel of cancer risk genes to (1) living survivors with a prior OC diagnosis who

Genome-Wide Identification and functional characterization of chitinase genes in the sorghum aphid (Melanaphis sacchari).

Insect chitinases are integral to a spectrum of vital physiological processes, including growth, development, molting, and metamorphosis. Consequently, chitin metabolic enzymes are potential targets for eco-friendly insecticide development. This study employed bioinformatics approaches to analyze the chitinase gene family in the sorghum aphid(Melanaphis sacchari), identifying seven MsCht genes. Phylogenetic distribution analysis revealed that the chitinases of the sorghum aphid are distributed across multiple clades, and the presence of several orthologous relationships with the pea aphid sugg

Rhizopus arrhizus infection in mice causes brain histopathological alterations and exacerbates neuronal apoptosis.

Rhizopus arrhizus is a fungus that can cause central nervous system infections in animals, resulting in high morbidity and mortality, but the mechanism of injury is rarely reported. In this study, we investigated the mechanism of Rhizopus arrhizus damage to the central nervous system of mice by observing the clinical neurological symptoms and resolving the pathological changes in the ultrastructure of brain tissues, combined with the alteration of apoptosis-related genes and immunohistochemistry (IHC). The results showed that all the mice in the treated group died, the brain pyknosis of neuron

Novel REIC/Dkk-3-encoding adenoviral vector as a promising therapeutic agent for pancreatic cancer.

Reduced expression in immortalized cells (REIC)/dickkopf-3 (Dkk-3), a tumor suppressor gene, is downregulated in various cancers. We previously reported the tumor-inhibitory effects of the REIC/Dkk-3 gene, delivered by a conventional adenoviral vector (Ad-CAG-REIC) in pancreatic cancer. Here, we developed an Ad-REIC vector with a novel gene expression system, termed the super gene expression (SGE) system, and assessed its therapeutic effects relative to those of Ad-CAG-REIC in pancreatic cancer cells. Human pancreatic cancer cell lines ASPC1 and MIAPaCa2 were used. REIC/Dkk-3 expression was as

Genetic counselling for affective disease.

Classic genetic studies utilizing twin methods, adoption and cross-fostering data and pedigree analysis, have confirmed the hypothesis that the familial component in affective disease is biological, rather than social or environmental. Only recently, however, has it been possible to clarify the mode of inheritance of affective disorder or to identify specific DNA segments which are physically linked to genes conferring susceptibility to affective illness. At present, two susceptibility genes have been identified in individual families, and it is certain that additional vulnerability genes rema

Soil-plant microbiomes shaped by soil type and wastewater quality as shown by a combination of cultivation-dependent and independent techniques.

The effects of wastewater irrigation on antibiotic resistance genes (ARGs), mobile genetic elements (MGEs), and bacterial communities in plant-associated microbiomes and soils are still poorly understood. We conducted a column experiment using Leptosol and Vertisol monoliths, planted with cilantro (Coriandrum sativum) and irrigated for eight weeks with untreated or treated (TWW) wastewater, with or without spiked antibiotics and disinfectants. Total community-DNA was analyzed by qPCR and 16S rRNA gene sequencing. Third-generation cephalosporin-resistant (3GCR) enterobacteria were isolated. Spi

RANKL is associated with persistent primary teeth and delayed permanent tooth emergence.

BACKGROUND: Tooth eruption is a process that is not fully understood. AIM: To evaluate whether genetic polymorphisms for RANK/RANKL/OPG are associated with delayed tooth emergence. To evaluate whether the relative expression of this genes is associated with persistent primary teeth. DESIGN: To evaluate whether genetic polymorphisms for RANK/RANKL/OPG could be involved in delayed tooth emergence, saliva samples from 160 children, aged 6-13 years old, were analysed. To test if there is correlation between gene expression of RANK/RANKL/OPG in children with delayed tooth emergence and persistent

Susceptibility alleles for testicular germ cell tumour: a review.

Family history is among the strongest and most consistent of the risk factors for testicular germ cell tumour (TGCT). Brothers of affected cases have an 8- to10-fold relative risk and fathers/sons have a risk between four and sixfold. The familial relative risk of TGCT is higher than for most other cancer types, which rarely exceeds four. The high relative risk suggests that inherited susceptibility to TGCT may account for a substantial fraction of TGCT cases. The search for TGCT susceptibility genes has proven difficult and a recent genome-wide linkage study for TGCT susceptibility loci demon

Contribution of the BRCA1 and BRCA2 mutations to breast cancer in Tunisia.

Hereditary breast cancer accounts for 3-8% of all breast cancers, with mutations in the BRCA1 and BRCA2 genes responsible for up to 30% of these. To investigate the prevalence of BRCA1 and BRCA2 gene mutations in breast cancer patients with affected relatives in Tunisia, we studied 36 patients who had at least one first degree relative with breast and/or ovarian cancer Thirty-four 34 patients were suggestive of the BRCA1 mutation and two were suggestive of the BRCA2 mutation, based on the presence of male breast cancer detected in their corresponding pedigrees. Four mutations in BRCA1 were det

Polymorphism in the MSX1 gene in a family with upper lateral incisor agenesis.

OBJECTIVE: The MSX1 gene plays a key role in odontogenesis regulation, particularly during early stages. Since only a few genetic variants have thus far been associated with non-syndromic tooth agenesis, we screened for mutations in this gene, aiming to detect a relationship between genotype and phenotype. DESIGN: The sample consisted of one proband with non-syndromic hypodontia involving upper lateral incisors, three relatives and ten unaffected controls. The proband and two affected relatives showed the same phenotype. DNA was extracted from buccal epithelial cells, and direct sequencing wa

High relative content of lysophospholipids of Helicobacter pylori mediates increased risk for ulcer disease.

Helicobacter pylori phospholipase A (OMPLA) degrades bacterial membrane phospholipids to lysophospholipids. High levels of lysophospholipids are associated with higher hemolytic activity, increased release of urease and vacA and better adherence to epithelial cells in vitro. The phospholipase A gene (pldA) displays phase variation due to a slippage in a homopolymeric tract. The aim of this study was to determine if the relative amount of lysophospholipids in the cell wall is associated with ulcer disease, and to further investigate the significance of pldA phase variation. H. pylori isolates o

Splicing mutations in the COL3 domain of collagen IX cause multiple epiphyseal dysplasia.

We report on a three-generation family with multiple epiphyseal dysplasia (MED). The propositus had typical MED findings of knees, ankles, elbows, and hands in childhood. The 2 other affected relatives were adults. The main clinical findings consisted of osteochondritis dissecans and osteoarthritis of the knees. DNA of the propositus was screened for mutations by conformation sensitive gel electrophoresis in all known candidate genes for MED, cartilage oligomeric matrix protein, and the COL9A1, COL9A2, and COL9A3 genes coding for the alpha1, alpha2, and alpha3 chains of collagen IX. The screen

Theoretical considerations on HLA-associated illness-susceptibility genes in dermatitis herpetiformis and psoriasis.

The frequency of an HLA-associated illness-susceptibility (Is) gene can be estimated by a simple equation: the frequency of the Is gene is equal to the prevalence of the disease in the population divided by the penetrance of the illness in first-degree relatives with the HLA marker antigen, with the quotient further divided by two. This equation is valid if the gene is dominant and its gene frequency is low. An analysis of HLA data of 61 patients with dermatitis herpetiformis (DH) showed that the HLA-associated DH susceptibility gene is obviously dominant. The frequency of this DH susceptibili

Gene Expression of CRAL_TRIO Family Proteins modulated by Vitamin E Deficiency in Zebrafish (Danio Rerio).

An evaluation of the impact of vitamin E deficiency on expression of the alpha-tocopherol transfer protein (α-TTP) and related CRAL_TRIO genes was undertaken using livers from adult zebrafish based on the hypothesis that increased lipid peroxidation would modulate gene expression. Zebrafish were fed either a vitamin E sufficient (E+) or deficient (E-) diet for 9 months, then fish were euthanized, and livers were harvested. Livers from the E+ relative to E- fish contained 40-times more α-tocopherol (P <0.0001) and one fourth the malondialdehyde (P = 0.0153). RNA was extracted from E+ and E- liv

Joubert syndrome: genotyping a Northern European patient cohort.

Joubert syndrome (JBS) is a rare neurodevelopmental disorder belonging to the group of ciliary diseases. JBS is genetically heterogeneous, with >20 causative genes identified to date. A molecular diagnosis of JBS is essential for prediction of disease progression and genetic counseling. We developed a targeted next-generation sequencing (NGS) approach for parallel sequencing of 22 known JBS genes plus 599 additional ciliary genes. This method was used to genotype a cohort of 51 well-phenotyped Northern European JBS cases (in some of the cases, Sanger sequencing of individual JBS genes had been
