Etiology of implant orthopedic infections: a survey on 1027 clinical isolates.

In spite of the recent achievements derived from modern protocols of prophylaxis, orthopedic surgical infections still remain unacceptably frequent, especially in light of the often devastating outcomes of septic complications. The spectrum and the prevalence of the bacteria most frequently involved in orthopedic infections are here explored, with particular reference to those infections associated to implant biomaterials, which were grouped based on device typology. During a 30 months period (from September 2000 to April 2003), 1027 microbial strains were consecutively isolated from 699 patie

In vitro evaluation of E1077, a new cephalosporin with a broad antibacterial spectrum.

E1077 is a novel parenteral cephalosporin with a wide spectrum of potent antibacterial activity against aerobic and anaerobic gram-positive and gram-negative bacteria. Against methicillin-susceptible Staphylococcus aureus, E1077 was twice as active as cefpirome, with an MIC for 90% of strains tested (MIC90) of 0.78 micrograms/ml. Methicillin-resistant S. aureus was moderately to highly resistant to E1077, but E1077 was at least twice as active as other beta-lactams tested. Against Enterococcus faecalis, E1077 was the most active of the cephalosporins tested (MIC90, 12.5 micrograms/ml) and was

[Study of the horizontal transfer of mercury resistance genes in natural populations of bacteria using antibodies to mercury reductases].

Mercury resistant soil and intestinal bacteria were isolated from different mercury deposit areas of the USSR. Mercury reductases from all gram negative bacteria studied (Pseudomonas, Acinetobacter and Enterobacterial species) with a single exception (Flavobacterium sp.) were immunologically cross reactive. Two immunological types of mercury reductases were found among gram positive bacteria (Bacillus, Staphylococcus and Coryneform species). Further subdivisions were done by "spur" formation tests. Despite considerable diversity of mercury reductases revealed in this study, we found several st

Accuracy of the KOH and vancomycin tests in determining the Gram reaction of non-enterobacterial rods.

Two nonstaining tests for the Gram reaction, the KOH and vancomycin disk (5 micrograms) tests, were employed on 488 strains of aerobic gram-negative and gram-positive rods. Since each test may yield false results with certain species, the use of a combination is suggested.

Exploring the Activity of Bacteriocins Against Bacterial Biofilms Using a Simple and Inexpensive Assay.

There is an urgent need for novel antimicrobials that can combat antibiotic resistant bacteria. Biofilms constitute a particular niche where bacteria display an intrinsic higher resistance to antibiotics and that favors resistance appearance and spread among species. However, the susceptibility tests for sessile bacteria are not well standardized. This work provides a high-throughput, affordable and simple methodology to grow biofilms on a three-dimensional surface represented by glass beads placed in 96-well microtiter plates. Two Gram-positive and two Gram-negative bacterial species were tes

Genetic covariance in immune measures and pathogen resistance in decorated crickets is sex and pathogen specific.

Insects are important models for studying immunity in an ecological and evolutionary context. Yet, most empirical work on the insect immune system has come from phenotypic studies meaning we have a limited understanding of the genetic architecture of immune function in the sexes. We use nine highly inbred lines to thoroughly examine the genetic relationships between a suite of commonly used immune assays (haemocyte count, implant encapsulation, total phenoloxidase activity, antibacterial zone of inhibition and pathogen clearance) and resistance to infection by three generalist insect pathogens

Late-Stage Modification of Aminoglycoside Antibiotics Overcomes Bacterial Resistance Mediated by APH(3') Kinases.

The continuous emergence of antimicrobial resistance is causing a threat to patients infected by multidrug-resistant pathogens. In particular, the clinical use of aminoglycoside antibiotics, broad-spectrum antibacterials of last resort, is limited due to rising bacterial resistance. One of the major resistance mechanisms in Gram-positive and Gram-negative bacteria is phosphorylation of these amino sugars at the 3'-position by O-phosphotransferases [APH(3')s]. Structural alteration of these antibiotics at the 3'-position would be an obvious strategy to tackle this resistance mechanism. However,

Clostridioides difficile Modifies its Aromatic Compound Metabolism in Response to Amidochelocardin-Induced Membrane Stress.

Amidochelocardin is a broad-spectrum antibiotic with activity against many Gram-positive and Gram-negative bacteria. According to recent data, the antibiotic effect of this atypical tetracycline is directed against the cytoplasmic membrane, which is associated with the dissipation of the membrane potential. Here, we investigated the effect of amidochelocardin on the proteome of Clostridioides difficile to gain insight into the membrane stress physiology of this important anaerobic pathogen. For the first time, the membrane-directed action of amidochelocardin was confirmed in an anaerobic patho

Gut microbiota-derived metabolites in CRC progression and causation.

BACKGROUND: Based on recent research reports, dysbiosis and improper concentrations of microbial metabolites in the gut may result into the carcinogenesis of colorectal cancer. Recent advancement also highlights the involvement of bacteria and their secreted metabolites in the cancer causation. Gut microbial metabolites are functional output of the host-microbiota interactions and produced by anaerobic fermentation of food components in the diet. They contribute to influence variety of biological mechanisms including inflammation, cell signaling, cell-cycle disruption which are majorly disrupt

Fluorogenic 7-azidocoumarin and 3/4-azidophthalimide derivatives as indicators of reductase activity in microorganisms.

A series of fluorogenic heterocyclic azides were prepared and assessed as reductase substrates across a selection of Gram-negative and Gram-positive microorganisms. The majority of these azides showed similar activity profiles to nitroreductase substrates. Microorganisms that do not produce hydrogen sulfide reduced the azides, indicating reductase activity was not linked to hydrogen sulfide production.
