Labeled cortical mantle distance maps of the cingulate quantify differences between dementia of the Alzheimer type and healthy aging.

The cingulate gyri in 37 subjects with and without early dementia of the Alzheimer type (DAT) were studied by using MRI at 1.0 mm3 isotropic … on changes with normal and abnormal aging. We find no significant change between young controls and healthy aging as measured by the GM volume and cortical mantle distribution as a function of distance in both anterior and posterior regions of the cingulate. Significant progression of GM loss is seen in the very mild DAT and mild DAT groups in all areas of the …

Multi-level mechanistic insights into phytone bioactivity: the healthy aging characteristics of C. elegans, molecular interactions and enzyme inhibition.

Phytone, an important terpene in fermented foods, has anti-aging potential. This study explored its interactions with α-amylase (αAmy), pepsin (PEP), trypsin (TPS), human serum albumin (HSA), and anti-aging effects via Caenorhabditis elegans (C. elegans). In nematodes, it prolongs lifespan, increases pharyngeal pump rate (14.98%), reduces heat stress mortality (10.10%), and preserves reproductive capacity. At the same time, it can significantly enhance the activity of SOD, CAT, and GSH-Px. Multispectral analysis and molecular simulation confirmed phytone forms stable complexes with these prote

Nutritional supplements and cognition in healthy aging and mild cognitive impairment patients: a systematic review and network meta-analysis.

BACKGROUND: Nutritional supplementation is increasingly regarded as a potential strategy to preserve or enhance cognitive function in individuals with healthy aging and mild cognitive impairment (MCI). However, its overall efficacy remains uncertain due to inconsistent findings across clinical trials. METHODS: In accordance with the guidelines outlined by the Preferred Reporting Items for Systematic Reviews and Meta-Analyses of Network Meta-Analyses, we conducted a comprehensive systematic search. Our inclusion criteria focused on randomized controlled trials (RCTs) that examined the impact o…

Krill Oil in Human Nutrition and Healthy Aging: A Narrative Review of Implications for Intrinsic Capacity and Nutrient Bioavailability.

Krill oil (KO) is a marine-derived nutritional ingredient distinguished by its unique structural composition and increasing relevance in human nutrition, particularly in the context of healthy aging and preservation of intrinsic capacity. Approximately 30-65% of its fatty acids are esterified to phospholipids, a molecular configuration associated with enhanced bioavailability and efficient tissue incorporation of omega-3 polyunsaturated fatty acids, even at relatively low doses. Antarctic krill (Euphausia superba) originates from minimally impacted marine ecosystems, contributing to a favorabl…

Investing in equitable healthy aging: Why Africa must reform social pension schemes to improve Alzheimer's disease and dementia outcomes.

The eligibility criteria for social pension schemes in Africa hinder equitable and healthy aging. In 2019, women in 14 sub-Saharan African countries had an average life expectancy of 67 years but a healthy life expectancy of only 57 years, leaving them 5 years in poor health before receiving a pension at age 62. Men had a similar situation-a life expectancy of 62 years and a healthy life expectancy of 53 years, spending 10 years in poor health before becoming eligible for pensions at age 63. Many men do not receive pensions due to early death. Delays and low pension payouts contribute to a 2.5…

Impact of sex and APOE-ε4 genotype on patterns of regional brain atrophy in Alzheimer's disease and healthy aging.

Alzheimer's Disease (AD) is a heterogeneous disease that disproportionately affects women and people with the APOE-ε4 susceptibility gene. We aim to describe the not-well-understood influence of both risk factors on the dynamics of brain atrophy in AD and healthy aging. Regional cortical thinning and brain atrophy were modeled over time using non-linear mixed-effect models and the FreeSurfer software with t1-MRI scans from the Alzheimer's Disease Neuroimaging Initiative (N = 1,502 subjects, 6,728 images in total). Covariance analysis was used to disentangle the effect of sex and APOE genotype …

A joint ventricle and WMH segmentation from MRI for evaluation of healthy and pathological changes in the aging brain.

Age-related changes in brain structure include atrophy of the brain parenchyma and white matter changes of presumed vascular origin. Enlargement of the ventricles may occur due to atrophy or impaired cerebrospinal fluid (CSF) circulation. The co-occurrence of these changes in neurodegenerative diseases and in aging brains often requires investigators to take both into account when studying the brain, however, automated segmentation of enlarged ventricles and white matter hyperintensities (WMHs) can be a challenging task. Here, we present a hybrid multi-atlas segmentation and convolutional auto

Directly Reprogrammed Human Neurons to Understand Age-Related Energy Metabolism Impairment and Mitochondrial Dysfunction in Healthy Aging and Neurodegeneration.

Brain aging is characterized by several molecular and cellular changes grouped as the hallmarks or pillars of aging, including organelle dysfunction, metabolic and nutrition-sensor changes, stem cell attrition, and macromolecular damages. Separately and collectively, these features degrade the most critical neuronal function: transmission of information in the brain. It is widely accepted that aging is the leading risk factor contributing to the onset of the most prevalent pathological conditions that affect brain functions, such as Alzheimer's, Parkinson's, and Huntington's disease. One of th

Psychological Traits, Heart Rate Variability, and Risk of Coronary Heart Disease in Healthy Aging Women-The Women's Health Initiative.

OBJECTIVE: Psychological traits such as optimism and hostility affect coronary heart disease (CHD) risk, but mechanisms for this association are unclear. We hypothesized that optimism and hostility may affect CHD risk via changes in heart rate variability (HRV). METHODS: We conducted a longitudinal analysis using data from the Women's Health Initiative Myocardial Ischemia and Migraine Study. Participants underwent 24-hour ambulatory electrocardiogram monitoring 3 years after enrollment. Optimism (Life Orientation Test-Revised), cynical hostility (Cook-Medley), demographics, and coronary risk

Fitness age outperforms body mass index in differentiating aging patterns and health risk profiles of healthy adults aged 51-80 years.

Physical fitness has been extensively shown to strongly associate with general health status and major health risks. Here we tested the ability of a novel estimate of fitness age (FitAge) to differentiate aging trajectories. This study aimed at (1) testing the ability of FitAge to differentiate aging patterns among decelerated, normal, and accelerated agers in selected health domains, (2) estimating the risk for developing major health issues depending on the aging trajectory, and (3) comparing FitAge to body mass index (BMI) categorization in differentiating healthy from unhealthy aging patter…

Age-Specific Ultrasound Reference Data for the Laterolateral Rectal Diameter in Healthy Children.

Background: Transabdominal ultrasonographic measurement of rectal diameter is increasingly used as a non-invasive adjunct for assessing rectal distension and fecal loading in children. However, interpretation is limited by the lack of age-stratified reference data in pediatric populations without bowel dysfunction. This study aimed to characterize the distribution of the laterolateral (LL) rectal diameter across childhood and adolescence and provide preliminary age-stratified empirical reference percentiles. Methods: This single-center cross-sectional study included pediatric participants aged

Patterns of cognitive and motor decline in Alzheimer's Disease (AD) and ageing in healthy populations.

BACKGROUND: Various patterns may apply to an individual’s health-span, with quality of life deriving from a balance between physical conditions, motor and cognitive abilities (i.e. psychomotor capabilities). METHODS: In this study, the Italian version of the Éxamen Geronto-Psychomoteur was administered to a sample of Alzheimer’s Disease (AD) patients (n = 94) and a group of healthy older adults (n = 333) to compare the patterns of psychomotor decline in pathological and physiological ageing. Three domains were considered to integrate bodily and cognitive dimensions: cognitive functions, motor

Single-cell transcriptomics reveals the impact of sex and age in the healthy human liver.

BACKGROUND & AIMS: The liver is a vital organ composed of parenchymal, non-parenchymal, and immune cell populations. Single-cell sequencing approaches now provide the opportunity to understand how sex and age influence gene expression and cellular function across cell types within the liver. METHODS: We analyzed the cellular composition and intercellular interactions of the human liver through single-nucleus RNA sequencing, incorporating insights from 37 healthy liver samples. The dataset contains cells from female and male donors spanning more than seven decades of life, and analysis was per

Multi-omic profiling reveals age-related immune dynamics in healthy adults.

The generation and maintenance of immunity is a dynamic process that is dependent on age1-3. Here, to better understand its progression, we profiled peripheral immunity in more than 300 healthy adults (25 to 90 years of age) using single-cell RNA sequencing, proteomics and flow cytometry, following 96 adults longitudinally across 2 years with seasonal influenza vaccination. The resulting resource generated a single-cell RNA-sequencing dataset of more than 16 million peripheral blood mononuclear cells with 71 immune cell subsets from our Human Immune Health Atlas and enabled us to interrogate h
