Neonatal undernutrition and short term administration of hydrocortisone and thyroxine: effects on rat brain hydrolases.

Brain enzymes activities that are likely to be involved in the catabolism of gangliosides were determined in controls (20% casein diet), postnatally undernourished (6.5% casein diet) and undernourished rats treated with either thyroxine or hydrocortisone, at 21 days of age. Postnatal undernutrition imposed by maternal protein deficiency during lactation resulted in a decrease in body weight and brain wet weight of the pups at 21 days of age. Administration of thyroxine or hydrocortisone to the undernourished pups every day between 16 and 21 days caused a further decrease in the body weight of …

[Effect of hydrocortisone on synaptic transmission in surviving slices of the olfactory cortex from the rat brain].

A study was made of the effect of bath-applied hydrocortisone (10(-4) M) on excitatory transmission of the lateral olfactory tract (LOT) - superficial pyramidal cell synapse of the rat olfactory cortex slice. Population EPSP and IPSP were depressed for 6-12 min., then they were increased (60 min.) by hydrocortisone. Recovery of evoked potentials was long (40-60 min.) at washing of slices hy a normal incubation medium. Habituation of synaptic potentials during LOT repeating stimulation was deteriorated by hydrocortisone application in perfusion solution. The data obtained were considered to be …

[Reproduction and differentiation of cells in the tissues of growing rat teeth in response to parathyroid hormone and hydrocortisone].

Influence of parathyroid hormone (PTH) and hydrocortisone (HC) on growth and development of teeth during prefunctional period has been studied, employing radioactive tracers. Biosynthetic and reproductive activity of cells is estimated in the molar and incisor tissues. It has been established that PTH accelerates the process of local resorption of the bone along the alveolar crest, but does not influence upon the teeth growth, functional and reproductive activity of the pulp cells. HC inhibits protein and glycoprotein synthesis, decreases amount of proliferating cells, accelerates their differ…

Effects of colchicine and hydrocortisone on unimpeded eruption rates of root-resected mandibular incisors of rats.

A single injection of colchicine and daily injections of hydrocortisone have opposite effects on tooth eruption, respectively causing marked reduction and marked increase in unimpeded eruption rates in the root-resected incisors of rats. Unimpeded eruption rates of both root-resected and normal, non-resected, incisors responded identically to these two drugs, providing support for the view that the eruptive process following root resection is physiological. It was not possible to obtain direct evidence relating to either the hydrostatic pressure or the periodontal fibroblast hypotheses of toot…

Modified-release hydrocortisone (Efmody®) in children with congenital adrenal hyperplasia: a retrospective registry study.

BACKGROUND: Replacement therapy for congenital adrenal hyperplasia (CAH) in childhood includes hydrocortisone and, in salt-wasting forms, addition of fludrocortisone. Immediate-release hydrocortisone often fails to adequately suppress early morning 17-hydroxyprogesterone (17OHP) levels. Hydrocortisone modified-release capsules (HMRC, Efmody®) were approved by the EMA in 2021 for the treatment of patients aged ≥12 years with CAH. This study evaluates the efficacy and safety of HMRC in children and adolescents based on real-world data collected from the I-CAH registry. METHODS: A single-center …

Comparative efficacy and prognostic impact of continuous versus intermittent hydrocortisone administration in septic shock patients.

This study investigates the effects of continuous versus intermittent hydrocortisone administration on septic shock patients. Sixty patients were randomized into two groups: one receiving intermittent doses of 50 mg of hydrocortisone every 6 h and the other a continuous infusion of 200 mg/day. After a 7-day treatment period and a 28-day follow-up, we observed no significant differences in the duration of sustained shock, hospital, and ICU stays between the groups. However, those in the continuous infusion group experienced shorter periods of mechanical ventilation and vasopressor use, with sig…

Total hydrocortisone dosage in extremely low birth weight infants and neurodevelopment up to school age.

BACKGROUND: An association between total hydrocortisone (HC) dosage in infants with extremely low birth weight (ELBW) and subsequent neurodevelopmental outcomes up to school age remains unclear. METHOD: We conducted a retrospective longitudinal cohort study across eight centers in Japan, including ELBW infants born between 2015 and 2017. We investigated the association between total HC dosage administered up to 36 weeks postmenstrual age and neurodevelopmental outcomes to school age. RESULTS: Linear mixed model analysis showed a significant association between higher HC dosage and lower deve…

Efficacy of Ascorbic Acid, Thiamine, and Hydrocortisone Combination Therapy: Meta-analysis of Randomized Controlled Trials.

BACKGROUND/AIM: Sepsis is a life-threatening biological condition that induces systemic tissue and organ dysfunction and confers a high mortality risk. Although the use of hydrocortisone in combination with ascorbic acid and thiamine (HAT therapy) significantly reduced mortality from sepsis or septic shock in a previous study, it did not improve mortality in subsequent randomized controlled trials (RCTs). Therefore, no definitive conclusion has been established on the benefits of HAT therapy for sepsis or septic shock. We performed a meta-analysis to assess the treatment outcomes of HAT therap…

Haemodynamic effects of prophylactic post-operative hydrocortisone following cardiopulmonary bypass in neonates undergoing cardiac surgery.

Multiple studies have endeavoured to define the role of steroids in paediatric congenital heart surgery; however, steroid utilisation remains haphazard. In September, 2017, our institution implemented a protocol requiring that all neonates undergoing cardiac surgery with the use of cardiopulmonary bypass receive a five-day post-operative hydrocortisone taper. This single-centre retrospective study was designed to test the hypothesis that routine post-operative hydrocortisone administration reduces the incidence of capillary leak syndrome, leads to favourable postoperative fluid balance, and le…

Twice Daily Compared to Three Times Daily Hydrocortisone in Prepubertal Children with Congenital Adrenal Hyperplasia.

INTRODUCTION: Glucocorticoid therapy in children with congenital adrenal hyperplasia (CAH) must be finely balanced between optimizing adrenal control and minimizing side effects. Twice (BID) rather than three times daily (TID) hydrocortisone may provide similar adrenal control and reduce metabolic risk. We compared BID and TID regimens with respect to adrenal control, growth, and metabolic effects. METHODS: A retrospective chart review (n = 128 visits, 36 individual patients) of prepubertal children with classical CAH was conducted at a tertiary care center between March 2007 and February 202…

Hydrocortisone Improves Oxygenation Index and Systolic Blood Pressure in Term Infants With Persistent Pulmonary Hypertension.

Persistent pulmonary hypertension of the newborn (PPHN) is an essential cause for hypoxic respiratory failure with significant morbidity and mortality in term and near-term neonates. Hydrocortisone has been shown to decrease oxygen dependency and pulmonary hypertension in neonates with meconium aspiration syndrome and animal studies, respectively. We hypothesize that hydrocortisone will improve oxygenation in term and near-term infants with pulmonary hypertension. We performed a retrospective chart review of all infant with PPHN who received intravenous hydrocortisone therapy as a rescue for s…

Hydrocortisone relieves the immediate post-operative scrotal edema after inguinal varicocelectomy: A prospective clinical trial.

This study aimed to assess the effect of a single IM injection of hydrocortisone succinate in relieving the immediate post-varicocelectomy scrotal edema. In all, 117 patients with grades II and III varicocele who developed post-varicocelectomy scrotal edema were randomly classified into group A (n = 59) that received a single IM hydrocortisone sodium succinate injection plus ordinary post-operative treatment and Group B (n = 58), which received the post-operative treatment alone. All patients were followed up to assess; changes in scrotal edema, the day of return to work and emergence of compl…
