New test for urinary glucose (BM33071) evaluated.

Results for urinary glucose by the Boehringer Mannheim BM33071 test pad and a hexokinase-based method agree well. The new test, which involves the glucose oxidase/peroxidase reaction, measures as little as 260 mg of glucose per liter. Acetoacetate, beta-hydroxybutyrate, and human hemoglobin do not interfere.

Use of Clinitest urine test for indirect estimation of blood glucose levels in the critically ill.

The use of the Clinitest urine glucose test as an indirect way to monitor and control blood glucose levels in critically ill patients was studied. Part 1 of the study was a review of 16 patients records to determine the correlation between 348 paired Clinitest readings and blood glucose levels. Part 2 was a study of six patients from whom a total of 83 arterial blood samples were obtained, each followed 15 minutes later by a urine sample that was tested quantitatively by autoanalyzer and qualitatively by Clinitest. In Part 1, only 43% of the paired readings were within clinically acceptable le

Synthesis and Use of the Bifunctional Sulfenic Acid Probe BCN-E-BCN for In Vitro and Cell-Based Assays of Protein Oxidation.

The reversible oxidation of cysteine thiol groups to sulfenic acid by reactive oxygen species (ROS) such as hydrogen peroxide can impact protein function, activity, and localization. As a consequence, ROS have profound effects on cell functions including proliferation, differentiation, and survival. Furthermore, there are clear associations between the effects of ROS on cells and the etiology of several diseases including cancer and neurodegeneration. In spite of the importance of cysteine sulfenylation as a validated post-translational modification, its labile nature impedes efficient and rep

[Specific features of studies of glucose in biological fluids (review)].

The review is devoted to the main questions of investigations of the main index of carbohydrate metabolism--glucose in biological liquids of an organism. Detailed analysis of technological principles and interpretation of biochemical results with due regard to the influence of the complex of inner and external physico-chemical, biological and climatic factors is presented.

Effects of chemical and biological warfare agent decontaminants on trace survival: Impact on fingermarks deposited on paper.

Chemical and biological events present a challenging environment for forensic scientists to perform their work. This research is the follow-up of the previous article that investigated the impact of decontaminants on fingermarks deposited on glass. In the present study, the focus shifted to paper, a porous and sensitive substrate where fingermark residues are absorbed and cannot be optically seen, making their detection more difficult. Paper is also highly relevant in forensic casework, as demonstrated by the anthrax letter events. Here the impact of 16 decontaminants on fingermarks deposited

Rapid and Bifunctional Chemoselective Metabolome Analysis of Liver Disease Plasma Using the Reagent 4-Nitrophenyl-2H-azirine.

Primary sclerosing cholangitis (PSC) is a chronic inflammatory disease of the bile ducts that has been associated with diverse metabolic carboxylic acids. Mass spectrometric techniques are the method of choice for their analysis. However, the broad investigation of this metabolite class remains challenging. Derivatization of carboxylic acids represents a strategy to overcome these limitations but available methods suffer from diverse analytical challenges. Herein, we have designed a novel strategy introducing 4-nitrophenyl-2H-azirine as a new chemoselective moiety for the first time for carbox

Simple colorimetric assay using pectin hydrogel reagent coupled with camera-based photometry for trace arsenic determination.

Humans mainly ingest arsenic through contaminated drinking water, causing serious health effects. The World Health Organization (WHO) has set the permissible limit of arsenic in drinking water at 0.01 mg/L and concentrations should be regularly determined to ensure a safe supply. In this study, a leucomalachite green (LMG) pectin-based hydrogel reagent was prepared that selectively reacted with arsenic over other metals including manganese, copper, lead, iron, and cadmium. Pectin, optimized at 0.2% (w/v), was used to form the hydrogel matrix. Arsenic reacts with potassium iodate in sodium acet

An improved method for rapid identification of hook effect samples in HBsAg quantitative assay.

Quantitative hepatitis B surface antigen assay is widely used for diagnosis of hepatitis B virus infection; however, specimens with high levels of the antigen can cause false-negative results (Hook effect), which needs to be resolved. The hook effect samples and non-hook effect samples were detected on the LiCA® 500 instrument using three methods, viz., 1, 2, and 3. Method 1, the currently used procedure, was performed in two steps with a total reaction time of 25 min in a final volume of 250 µL: first incubation was with two reagents for 15 min and then with one other reagent for 10 min. In M

Rapid determination of hydrogen sulfide-related metabolites in human urine by UHPLC-ESI-MS/MS with fluorinated ion-pairing reagents.

Although primarily considered as a toxic gas, hydrogen sulfide is naturally produced in humans from the amino acid cysteine in low quantities and is commonly referred to as the "third gaseous signaling molecule". There is a need to establish reliable biomarkers for exposure to hydrogen sulfide in human matrices not only at toxic levels but also at trace levels. Herein, we report optimized analytical methods for the direct determination of three compounds related to hydrogen sulfide metabolism in human urine without sample preparation; namely, trimethylsulfonium, thiosulfate, and cystine. The m
