In a controlled double blind study the therapeutic efficacy and tolerability of equimolar doses of [1-(p-chlorobenzoyl)-5-methoxy-2-methylindol-3-acetoxy] acetic acid (acemetacin, TV 1322, Rantudil) and indometacin (acemetacin: 180 mg/day, indometacin: 150 mg/day) were studied in 40 patients suffering from rheumatoid arthritis. The period of treatment lasted 42 days. With the exception of the parameter "state of the joint" a statistically confirmed improvement was detectable in the acemetacin group. The parameter "pain" was significantly improved after acemetacin as well after indometacin. Wit

Ulcerative colitis (UC) is a chronic inflammatory disease of the human colon. Dysbiotic gut microbiota play a central role in its pathogenesis, and alterations in microbial composition and function are closely linked to disease activity. Humanized gnotobiotic mice are increasingly used to study how dysbiotic, human-derived microbial communities shape intestinal inflammation. However, the fidelity of microbiota engraftment and its impact on host physiology and metabolism remain incompletely understood. In this study, we performed a multiomics analysis following fecal microbiota transfer (FMT) f

Melatonin integrates multiple biological pathways to enhance rice drought tolerance with cooperation of WRKY, bHLH, ERF, MYB and NAC transcription factors, and crosstalk of salicylic acid, ethylene and auxin. Drought is one of the primary environmental threats, and detrimentally affects plant growth and development, impeding crop yield and food quality worldwide. Melatonin has recently emerged as a multifunctional biomolecule with promising aspects in plant stress tolerance. However, the way in which melatonin improves drought tolerance in rice has not been investigated systematically. Here, w

The development of bladder cancer is a complex, multistep process influenced by both genetic and environmental factors, but its exact etiology remains unclear. Increasing evidence suggests that gut microbiota dysregulation can impact the initiation and progression of cancers, including colorectal cancer, breast cancer, and pancreatic cancer. This study observes that the beneficial gut bacterium Parabacteroides distasonis (P. distasonis) is significantly decreased in bladder cancer patients, and its low abundance is strongly associated with poor prognosis. P. distasonis is a next-generation pro

Olive oil polyphenol hydroxytyrosol (HT) significantly repairs intestinal barrier function, but its absorption in the stomach and small intestine is limited. The metabolites of unabsorbed HT that reach the colon are crucial, yet their effects on colonic microbiota and intestinal barrier repair remain unclear. This study utilized in vitro simulated digestion and colonic fecal fermentation to investigate HT's digestion and fermentation. Results indicated that 79.25% of HT potentially reached the colon intact. Further 16S rDNA, targeted, and untargeted metabolomics analyses showed that HT can be

Salicylic acid (SA) plays a central role in plant disease resistance, and emerging evidence indicates that auxin, an essential plant hormone in regulating plant growth and development, is involved in plant disease susceptibility. GH3.5, a member of the GH3 family of early auxin-responsive genes in Arabidopsis (Arabidopsis thaliana), encodes a protein possessing in vitro adenylation activity on both indole-3-acetic acid (IAA) and SA. Here, we show that GH3.5 acts as a bifunctional modulator in both SA and auxin signaling during pathogen infection. Overexpression of the GH3.5 gene in an activati

The octopine synthase (ocs or ocs-like) element has been previously reported to be responsive to the plant hormones, auxin, salicylic acid, and methyl jasmonate. Using transient assays with carrot protoplasts, we have demonstrated that an ocs element from the soybean auxin-inducible GH2/4 promoter is not only activated by strong auxins (i.e., 2,4-dichlorophenoxyacetic acid, 2,4,5-trichlorophenoxyacetic acid, alpha-naphthalene acetic acid) and salicylic acid, but also by weak auxin analogues (beta-naphthalene acetic acid), inactive auxin analogs (i.e., 2,3-dichlorophenoxyacetic acid, 2,4,6-tric

In silico analysis of different ABCB-type proteins revealed structural similarities within the linker domain, including the presence of [R/K-X-S/T] and [R/K-X-X-S/T] motifs, which are potential phosphorylation sites for kinases of the AGCVIII superfamily. Based on these findings, a phosphorylation-based regulatory mechanism is proposed for ABCB alkaloid transporters. Plants avoid self-toxicity by compartmentalizing secondary metabolites in specialized cells. Even when this mechanism keeps them away from sensitive metabolic processes, it also could restrict them from reaching the different loca

Lead (Pb) contamination presents a widespread environmental plague. Sedum alfredii is widely used for soil phytoremediation owing to its capacity to extract heavy metals, such as Pb. Although efficient Pb extraction is mediated by complex interactions between the roots and rhizospheric bacteria, the mechanism by which S. alfredii recruits microorganisms under Pb stress remains unclear. The Pb-accumulating ecotype (AE) and non-accumulating ecotype (NAE) of S. alfredii recruited different rhizobacterial communities. Under Pb stress, AE rhizosphere-enriched bacteria assembled into stable-connecte

BACKGROUND: Bone mineral density (BMD) is influenced by multiple factors. Recent studies have highlighted a possible relationship between serotonin and BMD. Patients with neuroendocrine tumours (NETs) frequently have elevated urinary 5-hydroxy-indoleacetic acid (5-HIAA) levels, a serotonin metabolite. Evaluation of the relationship between 5-HIAA and BMD in patients with NETs may provide insights into the relationship between serotonin and BMD.
METHODS: One-year audit of consecutive patients with NETs within two institutions. Relationships between urinary 5-HIAA and dual X-ray absorptiometry

Plant growth promoting rhizobacteria (PGPR) are known to influence plant growth by various direct or indirect mechanisms. In search of efficient PGPR strains with multiple activities, a total of 72 bacterial isolates belonging to Azotobacter, fluorescent Pseudomonas, Mesorhizobium and Bacillus were isolated from different rhizospheric soil and plant root nodules in the vicinity of Aligarh. These test isolates were biochemically characterized. These isolates were screened in vitro for their plant growth promoting traits like production of indoleacetic acid (IAA), ammonia (NH(3)), hydrogen cyani

BACKGROUND: Rheumatoid arthritis (RA) is an autoimmune disease that may be associated with gut microbiota via the aryl hydrocarbon receptor (AhR). Human umbilical mesenchymal stem cells (HUMSCs) have therapeutic potential against RA, but the underlying mechanism has not been fully elucidated. The purpose of this study was to explore the mechanism of action of HUMSCs in rats with collagen-induced arthritis (CIA).
METHOD: HUMSCs (1 × 106) were transplanted into each rat with CIA. The tissue localization of HUMSCs and the therapeutic effects in the ankles were assessed. The immune status and exp

A novel bacterial strain, designated JK66T, was isolated from the rhizosphere soil of diseased Cistanche plants collected in Turpan, Xinjiang, China. Strain JK66T grew at 20-42 °C (optimum, 37 °C), pH 5.0-10.0 (optimum, pH 7.0), and with 0-3.0% (w/v) NaCl (optimum, 2.0% (w/v)). The strain was selected for taxonomic characterization because of its antagonistic activity against Fusarium acuminatum and its potential to produce indole-3-acetic acid. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain JK66T belongs to the genus Pseudomonas, showing the highest sequence simi

Weaning stress can severely damage the piglets' intestines. Microbial tryptophan catabolites play a vital role in maintaining the health of the intestinal mucosa. Indole-3-acetic acid (IAA), an indole derivative with known anti-inflammatory properties, has not yet been studied for its impact on piglets' intestinal health. Twenty-four weaned crossbred piglets (Duroc × Yorkshire × Landrace, weighing 6.58 ± 0.07 kg) were randomly allocated to receive diets containing 0, 120, or 240 mg/kg indole-3-acetate sodium (IAA-Na). Although dietary IAA-Na did not significantly impact growth performance or d

Plant immunity relies on dynamic modulation of phytohormone signaling to balance growth and defense, with auxin contributing to resistance against the necrotrophic pathogen Botrytis cinerea (B. cinerea). However, how plants rapidly and precisely regulate auxin biosynthesis during pathogen infection remains unclear. Here, we identified the nitrilases NIT1, NIT2, and NIT3, as key components of auxin biosynthesis during infection. Upon chitin perception, CERK1 associates with and phosphorylates nitrilases to stimulate the conversion of indole-3-acetonitrile (IAN) into indole-3-acetic acid (IAA),

In the root apical meristem, the stem cell niche (SCN) comprises a mitotically inactive quiescent center (QC) and adjacent, mitotically active stem cells that divide to form root tissues. Auxin dynamics are essential for specification and maintenance of the root SCN; however, the underlying mechanisms remain to be explored. Here, we report that Arabidopsis (Arabidopsis thaliana) AUXIN RESPONSE FACTOR3 (ARF3), ARF3-INTERACTING PROTEIN1/2 (AIP1/2), and SIN3-ASSOCIATED POLYPEPTIDE OF 18 KDA (SAP18) form a protein complex that specifies root SCN cell fate in response to auxin level. Melatonin increases Pb tolerance in P. ovata seedlings via the regulation of growth and stress-related phytohormones, ROS scavenging and genes responsible for melatonin synthesis, metal chelation, and stress defense. Lead (Pb) is a highly toxic heavy metal that accumulates in plants through soil and air contamination and impairs its plant growth and development. Because of its pharmaceutical importance, improvements in Plantago ovata yield against abiotic stresses are necessary. Melatonin (MEL) is a stress-alleviating biostimulator and our results showed a reduction in Pb induced phytotoxicity
