African swine fever virus pNP419L inhibits host translation and increases viral replication through the PERK/eIF2α pathway.

African swine fever (ASF) is a highly contagious disease caused by African swine fever virus (ASFV), which poses a significant threat to pig farming worldwide. Although ASFV encodes many self-sustaining proteins, its mRNA translation relies on the host's translation machinery. However, the mechanisms by which ASFV-encoded proteins regulate the host's translation process in infected cells remain largely unexplored. In this study, we investigated how ASFV-encoded proteins affect the translation of host cells. We identified 21 ASFV genes, excluding those from multigene families, that significantl

A Compound Screen Based on Isogenic hESC-Derived β Cell Reveals an Inhibitor Targeting ZnT8-Mediated Zinc Transportation to Protect Pancreatic β Cell from Stress-Induced Cell Death.

Pancreatic β cell loss by cellular stress contributes to diabetes pathogenesis. Nevertheless, the fundamental mechanism of cellular stress regulation remains elusive. Here, it is found that elevated zinc transportation causes excessive cellular stress in pancreatic β cells in diabetes. With gene-edited human embryonic stem cell-derived β cells (SC-β cells) and human primary islets, the results reveal that elevated zinc transportation initiates the integrated stress response (ISR), and ultimately leads to β cell death. By contrary, genetic abolishment of zinc transportation shields β cells from…

The General Control Non-derepressible 2 (GCN2) kinase and the downstream integrated stress response represent the best candidates mediating this process; however, by pharmacological approaches, RNA interference and genomic editing, we demonstrate that they are not implicated. Instead, the response requires MEK/ERK and/or JNK activity and is reprod …

Hydrogen sulfide-mitigated salinity stress impact in sunflower seedlings was associated with improved photosynthesis performance and osmoregulation.

BACKGROUND: Salinity is one major abiotic stress affecting photosynthesis, plant growth, and development, resulting in low-input crops. Although photosynthesis underlies the substantial productivity and biomass storage of crop yield, the response of the sunflower photosynthetic machinery to salinity imposition and how H2S mitigates the salinity-induced photosynthetic injury remains largely unclear. Seed priming with 0.5 mM NaHS, as a donor of H2S, was adopted to analyze this issue under NaCl stress. Primed and nonprime seeds were established in nonsaline soil irrigated with tape water for 14 d

Stress and the "extended" autonomic system.

This review updates three key concepts of autonomic neuroscience-stress, the autonomic nervous system (ANS), and homeostasis. Hans Selye popularized stress as a scientific idea. He defined stress variously as a stereotyped response pattern, a state that evokes this pattern, or a stimulus that evokes the state. According to the "homeostat" theory stress is a condition where a comparator senses a discrepancy between sensed afferent input and a response algorithm, the integrated error signal eliciting specific patterns of altered effector outflows. Scientific advances since Langley's definition o

Activation of the hypothalamic-pituitary adrenal axis in response to a verbal fluency task and associations with task performance.

Speech fluency can be impaired in stressful situations. In this study, it was investigated whether a verbal fluency task by itself, i.e. without the presence of any further stressors, induces responses of the hypothalamic-pituitary adrenal (HPA) axis and of the sympathetic nervous system (SNS). The sample consisted of n = 85 participants (68.2% female; 33.3 ± 15.2 years) who performed two consecutive verbal fluency tasks for two minutes each. The categories were either 'stress' or 'disease' and 'animals' or 'foods' which were presented in a randomized order. Three saliva samples were collected

Diabetes as a disease of endoplasmic reticulum stress.

Endoplasmic reticulum (ER) stress is an integral part of life for all professional secretory cells, but it has been studied to greatest depth in the pancreatic β-cell. This reflects both the crucial role played by ER stress in the pathogenesis of diabetes and also the exquisite vulnerability of these cells to ER dysfunction. The adaptive cellular response to ER stress, the unfolded protein response, comprises mechanisms to both regulate new protein translation and a transcriptional program to allow adaptation to the stress. The core of this response is a triad of stress-sensing proteins: prote

Vaccine-Associated Maintenance of Epithelial Integrity Correlated With Protection Against Virus Entry.

To identify the mechanisms by which human immunodeficiency virus type 1 (HIV-1) might penetrate the epithelial barrier during sexual transmission to women and the mechanisms of vaccine-associated protection against entry, we characterized early epithelial responses to vaginal inoculation of simian immunodeficiency virus strain mac251 (SIVmac251) in naive or SIVmac239Δnef-vaccinated rhesus macaques. Vaginal inoculation induced an early stress response in the cervicovaginal epithelium, which was associated with impaired epithelial integrity, damaged barrier function, and virus and bacterial tran

Allostatic load: A theoretical model for understanding the relationship between maternal posttraumatic stress disorder and adverse birth outcomes.

BACKGROUND: Adverse birth outcomes such as preterm birth and low birth weight are significant public health concerns and contribute to neonatal morbidity and mortality. Studies have increasingly been exploring the predictive effects of maternal posttraumatic stress disorder (PTSD) on adverse birth outcomes. However, the biological mechanisms by which maternal PTSD affects birth outcomes are not well understood. Allostatic load refers to the cumulative dysregulations of the multiple physiological systems as a response to multiple social-ecological levels of chronic stress. Allostatic load has b

Transcriptomic and metabolomic analysis of allelopathic responses in Elymus nutans.

To explore the allelopathic response mechanism of Elymus nutans to the extracts of Ligularia sagitta, seed germination tests combined with integrated transcriptome and metabolome analyses were performed to systematically investigate the effects of the extracts at different concentrations on its germination characteristics and molecular metabolism. The results showed that the concentration range of 0.01-0.10 mg/mL was the sensitive interval for the allelopathic response of Elymus nutans to the extracts of Ligularia sagitta, and germination energy (GE) exhibited higher sensitivity to allelopathi

Heat exposure affects jejunal tight junction remodeling independently of adenosine monophosphate-activated protein kinase in 9-day-old broiler chicks.

Dysfunction of the intestinal epithelial barrier under elevated temperatures is assumed to prompt pathological conditions and to eventually impede chickens' growth, resulting in massive economic losses in broiler industries. The aims of this research were to determine the impact of acute heat stress on the intestinal tight junction network of broiler chicks (Gallus domesticus L.) and to elucidate whether adenosine monophosphate-activated protein kinase (AMPK) was involved in the integrated response of the broiler's gastrointestinal tract to heat stress. A total of 80 9-day-old Arbor Acres chic

AP2/ERF transcription factors in plant stress adaptation: evolutionary expansion, functional diversification and hormone crosstalk.

APETALA2/ethylene response factor (AP2/ERF) transcription factors constitute a plant-specific regulatory family that coordinates plant development, environmental stress responses, and hormone signaling. However, the contribution of AP2/ERF family expansion and functional diversification to environmental adaptation has not been fully integrated across evolutionary and mechanistic scales. This review synthesizes classical findings and representative advances from the past five years in structural classification, duplication-driven expansion, functional diversification, stress adaptation, and hor

Defining subcellular synovial responses in TMJ osteoarthritis onset via mechanical stress and articular disk derangement models.

Temporomandibular joint osteoarthritis (TMJ-OA), the most common degenerative disease of the TMJ, is influenced by various adaptive, inflammatory, and mechanical stressors. In this study, we describe molecular alterations of the synovium of the articular disk in response to mechanical and inflammatory stimuli. Using an integrated transcriptomic approach combining subcellular spatial transcriptomics and single-cell RNA sequencing in murine models of mechanical stress and articular disk derangement, we characterized synovial changes associated with adipogenesis, fibrosis, and macrophage activati

Qingshu Yiqi tea prescription attenuates heat-stressed injury: network pharmacology and experimental validation.

Heat stress (HS) poses a significant threat to health, necessitating effective interventions. This study investigated the therapeutic mechanisms of Qingshu Yiqi Tea Prescription (QSYQTP) against HS-induced organ damage in mice. Utilizing an integrated pharmacology approach combining histopathology, network pharmacology, biochemical assays, and qRT-PCR, we assessed QSYQTP's effects in a heat-stressed murine model. QSYQTP supplementation suppressed serum aminotransferase activities (ALT, AST), reduced MDA concentrations, and enhanced antioxidant enzymes (T-SOD, GSH). It also downregulated heat s

Systems redox biology in health and disease.

Living organisms need to be able to cope with environmental challenges and other stressors and mount adequate responses that are as varied as the spectrum of those challenges. Understanding how the multi-layered biological stress responses become integrated across and between different levels of organization within an organism can provide a different perspective on the nature and inter-relationship of complex systems in health and disease. We here compare two concepts which have been very influential in stress research: Selye's 'General Adaptation Syndrome' and Sies's 'Oxidative Stress' paradi

Integrated assessment of wastewater pollution in a karstic coastal lagoon: linking nutrients, caffeine, stable isotopes, and fish physiological responses.

Coastal water pollution is a global problem, especially in karstic areas, where pollutants from point and non-point sources reach coastal systems. The Chelem Lagoon in the Yucatan Peninsula exemplifies such environments, where the presence and effects of pollutants have been previously reported. This study presents and evaluates an integrated assessment framework to characterize pollution sources and ecotoxicological impacts in complex coastal systems. This approach combines nutrient concentrations, caffeine (as an urban wastewater tracer), carbon and nitrogen stable isotopes, and a suite of p

Chemogenetic activation of CRF neurons as a model of chronic stress produces sex-specific physiological and behavioral effects.

Trauma and chronic stress exposure are the strongest predictors of lifetime neuropsychiatric disease presentation. These disorders often have significant sex biases, with females having higher incidences of affective disorders such as major depression, anxiety, and PTSD. Understanding the mechanisms by which stress exposure heightens disease vulnerability is essential for developing novel interventions. Current rodent stress models consist of a battery of sensory, homeostatic, and psychological stressors that are ultimately integrated by corticotropin-releasing factor (CRF) neurons to trigger

Identification of Cysl-2 as a Compensatory Factor in Vitamin B12-Mediated Protection in a Mitochondrial Aβ Toxicity Model.

Alzheimer's disease (AD) is a highly sophisticated disease associated with mitochondrial deterioration, which potentiates proteotoxic stress and accelerates neuronal decline. Vitamin B12 (B12), an essential cofactor in one-carbon metabolism and cellular homeostasis, plays a pivotal role in modulating neurodegenerative pathology. Here, we established a mitochondrial-associated AD model by using Caenorhabditis elegans (C. elegans) expressing Aβ combining with the mtDNA deletion uaDf5, and evaluated the effect of B12 as a metabolic modulator of neurodegenerative stress. We observed that mitochond

Therapeutic implications of mitochondrial stress-induced proteasome inhibitor resistance in multiple myeloma.

The connections between metabolic state and therapy resistance in multiple myeloma (MM) are poorly understood. We previously reported that electron transport chain (ETC) suppression promotes sensitivity to the BCL-2 antagonist venetoclax. Here, we show that ETC suppression promotes resistance to proteasome inhibitors (PIs). Interrogation of ETC-suppressed MM reveals integrated stress response-dependent suppression of protein translation and ubiquitination, leading to PI resistance. ETC and protein translation gene expression signatures from the CoMMpass trial are down-regulated in patients wit…
