Mechanism of tissue damage through free oxygen radicals during hepatic amoebiasis in guinea pigs.

The capacity of Kupffer cells and blood monocytes to release free oxygen radicals was studied by chemiluminescence (CL) response during hepatic amebiasis in guinea pigs. Guinea pigs infected with Entamoeba histolytica intramesenterically were sacrificed on days 0, 2, 5 and 8 post-infection. Hepatic lesions were graded I-IV. A significant increase in the CL response was observed from day 2 post-infection and it increased with the progress of infection. Maximum increase was observed on the 8th post infection day. Animals with grade III or IV hepatic lesions had significantly elevated CL response…

Increased natural resistance to Listeria monocytogenes in senescent mice correlates with enhanced macrophage bactericidal activity.

Natural resistance to Listeria monocytogenes is increased in aging (18-22-month-old) AB6F1 mice when compared to their younger counterparts. We have investigated several parameters of macrophage function of old AB6F1 mice to establish what macrophage characteristics determine their enhanced resistance to Listeria. Senescent mice had an increased number of macrophages present in unstimulated peritoneal cavity. These macrophages exhibited enhanced bactericidal activity and H2O2 production. Old mice showed a diminished subcutaneous inflammatory response. Kupffer cells obtained from normal uninfec…

Hepatic Ischemia-reperfusion Injury: Protective Approaches and Treatment.

Ischemia and reperfusion damage to the liver is one of the major causes of hepatic dysfunction and liver failure after a liver transplant. The start of hepatic ischemia-reperfusion damage is linked to metabolic acidosis, Kupffer cells, neutrophils, excessive calcium, and changes in the permeability of the mitochondrial membrane. Hypoxia activates Kupffer cells, resulting in the production of reactive oxygen species (ROS). These ROS when accumulated, causes apoptosis and necrosis, as well as activate immune and inflammatory responses that involve many cells and signalling molecules. Numerous an…

Nitric oxide production by hepatocytes contributes to the inhibitory effect of endotoxin on insulin-like growth factor I gene expression.

We tested whether endotoxin (lipopolysaccharide, LPS) inhibits IGF-I gene expression in hepatocytes and the possible role of Kupffer cells and nitric oxide (NO) in this effect. LPS decreased IGF-I mRNA in hepatocyte cultures and increased the nitrite + nitrate levels in the culture medium. Furthermore, there was a negative correlation between the IGF-I mRNA and the nitrite+nitrate levels. When hepatocytes were cocultured with Kupffer cells, the inhibitory effect of LPS on IGF-I mRNA was higher than in hepatocyte cultures, but the stimulatory effect on nitrite+nitrate was similar in both condit…

Experimental African trypanosomiasis: IFN-gamma mediates early mortality.

In this study, we demonstrate that Kupffer cells in the livers of highly susceptible BALB/c mice infected with Trypanosoma congolense were loaded with trypanosomal antigen and appeared highly activated. This was associated with an enlarged capillary bed in the livers and decreased blood pressure of these mice towards the terminal stage. Blocking of murine IL-10 receptor (IL-10R)in vivo shortened the survival time of highly susceptible T. congolense-infected BALB/c mice. Anti-IL-10R treatment decreased the survival of relatively resistant T. congolense-infected C57BL/6 mice dramatically. Blocki…

Clinical Applications of Cell-Specific Carriers Based on Glycosylated Albumin.

Over-activated macrophages and/or fibroblasts have shown involvement in the onset and progression of various diseases, including hepatitis and cancer. Therefore, the development of techniques to inhibit the activation of these cells has become an urgent challenge. Prior to this study, a mannosylated-human serum albumin (Man-HSA) was created to target mannose receptors expressed on Kupffer cells. Here, applications of Man-HSA for medical treatments are reported. To target the reactive oxygen species or inflammation derived from Kupffer cells, a nano-antioxidant, i.e., SH-Man-HSA, produced by in…

ALYREF drives NLRP3 inflammasome-dependent macrophage pyroptosis to promote sepsis-associated acute liver injury by enhancing GADD45A mRNA stability.

Sepsis-associated acute liver injury (SALI) is an independent risk factor that significantly worsens sepsis prognosis, underscoring the need for mechanistic insights to guide targeted therapy. Accordingly, we established a sepsis mouse model via cecum ligation and puncture (CLP) and performed in vitro experiments using murine hepatic Kupffer cells stimulated with lipopolysaccharide (LPS) and adenosine triphosphate (ATP). Here, we found that growth arrest and DNA damage 45A (GADD45A) was highly expressed in liver tissues of CLP-induced septic mice, and its knockdown ameliorated liver injury and…

Bioengineered Multicellular Liver Microtissues for Modeling Advanced Hepatic Fibrosis Driven Through Non-Alcoholic Fatty Liver Disease.

Despite considerable efforts in modeling liver disease in vitro, it remains difficult to recapitulate the pathogenesis of the advanced phases of non-alcoholic fatty liver disease (NAFLD) with inflammation and fibrosis. Here, a liver-on-a-chip platform with bioengineered multicellular liver microtissues is developed, composed of four major types of liver cells (hepatocytes, endothelial cells, Kupffer cells, and stellate cells) to implement a human hepatic fibrosis model driven by NAFLD: i) lipid accumulation in hepatocytes (steatosis), ii) neovascularization by endothelial cells, iii) inflammat…

Overactivation of the immune system by translocated bacteria and bacterial products.

Gut-derived bacteria and their constituents, namely endotoxins, contribute to the pathogenesis of septic multi-organ failure. Ischemia, trauma, chronic inflammation, immunosuppression or reduced blood flow in the gut are conditions that enhance bacterial translocation, which in turn activates the non-specific immune system, i.e. macrophages and granulocytes to release mediators of inflammation such as cytokines, eicosanoids and degranulation products. Besides blood leukocytes, the liver macrophage population (Kupffer cells), the majority of macrophages in the organism, is a central part of the…

Toll-like receptor signaling and liver fibrosis.

Liver fibrosis occurs as a wound-healing scar response following acute and chronic liver inflammation including alcoholic liver disease, non-alcoholic steatohepatitis, hepatitis B and C, and autoimmune hepatitis. Myofibroblasts, mainly transdifferentiated from hepatic stellate cells, are pivotal cell types that produce fibrillar collagen. The activation of inflammatory cells, including Kupffer cells, is a crucial step for activating hepatic stellate cells. Toll-like receptors (TLRs) are pattern recognition receptors that sense pathogen-associated molecular patterns (PAMPs), which discriminate …

Methyl palmitate prevents CCl(4)-induced liver fibrosis.

Liver fibrosis is characterized by an excess of collagen fiber deposition, and it is known that Kupffer cells play an important role by immunomodulation of the toxic response. Methyl palmitate (MP) is an effective Kupffer cell inhibitor. The aim of this work was to evaluate the effect of MP on experimental liver fibrosis. Four groups were formed: the control group, which received the vehicles only; CCl(4) group (0.4 g kg(-1), i.p., three times a week, for eight weeks); CCl(4) plus MP (300 mg kg(-1), i.p., daily); and MP alone. Alanine aminotransferase was increased by CCl(4), and MP did not pr…

Silymarin inhibits the progression of fibrosis in the early stages of liver injury in CCl₄-treated rats.

Liver fibrosis, a common condition occurring during the evolution of almost all chronic liver diseases, is the consequence of hepatocyte injury that leads to the activation of Kupffer cells and hepatic stellate cells (HSC). Silymarin (Si) is a herbal product widely used for its hepatoprotective potential. Our study aims to investigate the effects of two different doses of Silymarin on a CCl4-induced model of liver fibrosis with a focus on the early stages of liver injury. Fifty Wistar rats were randomly divided into five groups (n=10): control group (sunflower oil twice a week); CMC group (car…
