Identification and characterization of autoantibodies against the nuclear envelope lamin B receptor from patients with primary biliary cirrhosis.

We have identified autoantibodies from two patients with primary biliary cirrhosis (PBC) that recognize the nuclear envelope of mammalian cells on indirect immunofluorescence microscopy. These antibodies bind to a 58-kD integral membrane protein (p58) of the turkey erythrocyte nuclear envelope, which has been previously identified as a membrane receptor for lamin B (Worman, H. J., J. Yuan, G. Blobel, and S. D. Georgatos. 1988. Proc. Natl. Acad. Sci. USA. 85:8531). The antibodies also bind to a 61-kD integral membrane protein (p61) of the rat liver nuclear envelope. Affinity-purified antibodies

Annexin A2 and lamin B join membrane recycling compartments for the assembly of biomolecular condensates operating in mitotic partitioning.

Localization of the actin-, lipid- and mRNA-binding protein Annexin A2 (AnxA2) in dividing cells revealed its presence in large spherical structures which are confined to the cell periphery and frequently co-align with astral microtubules. These structures appear during prometaphase and disappear at telophase, coinciding with the mitotic breakdown and subsequent reformation of the nuclear lamina and envelope. Their size increases as cells progress to anaphase, while their number decreases, suggesting that they are capable of fusion. Treatment of cells with the aliphatic alcohol propylene glyco

Effect of Thickness and Stitch Density on Low-Velocity Impact and Compression After Impact Properties of Stitched Composite Laminates.

In this work, experimental studies were conducted on the damage failure of laminated composite laminates under low-velocity impact and compressive failure behavior under compression after impact. The study primarily investigated the effects of stitch density, impact energy, and laminate thickness on the damage behavior of composite laminates. The experimental results indicate that at impact energies of 10 J, 15 J, and 20 J, the stitched specimens demonstrated higher impact resistance. When the stitch density was 10 × 10 mm, the average maximum impact force of the stitched specimens increased b

Epilepsy-associated FOXJ3 variants link a transcriptional program of the PTEN-mTOR pathway to neuronal specification and cortical lamination.

Focal cortical dysplasia (FCD), a major cause of drug-resistant epilepsy, involves abnormal neuronal migration and cortical architecture, yet its molecular basis remains poorly defined. Here, we identify FOXJ3 pathogenic variants in patients with autosomal dominant focal epilepsy and FCD. In the developing mouse cortex, FOXJ3 declines sharply in neural progenitors after embryonic day 15.5. In utero electroporation-mediated Foxj3 knockdown in mouse brains impairs neuronal migration, disrupts cortical lamination, and alters neuronal specification, promoting upper-layer neuron production at the e

Gluten protein network formation induced by a novel laminated sheeting process: Effect on the textural stability of cooked noodles.

To address the progressive textural deterioration of takeaway noodles during immersion, this study employed a novel laminated sheeting process for noodle preparation by integrating dough sheet with dough crumbs. The effects of the ratio of dough sheet to crumbs (R-DSTC) (1:0, 1:0.25, 1:0.75, and 1:1.5) and sheeting ratio (R-S) (20 %, 30 %, and 40 %) on the textural stability of cooked noodles were investigated. Both cooking quality and textural characteristics of noodles improved with increasing R-DSTC and R-S values. The highest hardness (4894.62 ± 92.39 g) and tensile distance (72.91 ± 3.24

Novel lamin B receptor mutation (c.561C > G) in a patient with Pelger-Huët anomaly: a case report.

Pelger-Huët anomaly (PHA), an autosomal dominant disorder characterized by abnormal granulocyte morphology, was first described in 1928. Mutations in the lamin B receptor (LBR) gene cause a phenotypic spectrum ranging from isolated PHA, PHA with mild skeletal abnormalities, to the embryonic-lethal Greenberg skeletal dysplasia. We report a Chinese boy presenting peripheral blood granulocyte abnormalities associated with a novel LBR gene mutation. Whole-exome sequencing uncovered the LBR gene heterozygous mutation, NM_194442.2: c.561C > G (p.Tyr187*). Notably, the patient exhibited scoliosis sec

Lamin A/C promotes HBV transcription by modulating histone modification associated with cccDNA minichromosome in an HBx-dependent manner.

BACKGROUND: Chronic hepatitis B virus (HBV) infection remains a serious public health challenge, and primary treatment strategies are unable to cure HBV infection due to the persistence of HBV covalently closed circular DNA (cccDNA) in the nuclei of cells. The HBV X protein (HBx) plays a crucial role in regulating cccDNA transcription. Therefore, targeting HBx to identify host proteins that regulate cccDNA transcription could represent a curative approach. METHODS: Here, we used co-immunoprecipitation (co-IP) and mass spectrometry (MS) to identify proteins that interact with HBx. The candidat

Generation of a lamin A/C knockout human induced pluripotent stem cell line (ZJULLi007-A) via CRISPR/Cas9.

Lamin A/C is a protein encoded by the LMNA gene and belongs to the nuclear lamina protein family. Mutations in the LMNA gene lead to several diseases: Emery-Dreifuss muscular dystrophy, familial partial lipodystrophy, limb girdle muscular dystrophy, dilated cardiomyopathy, Charcot-Marie-Tooth disease, and Hutchinson-Gilford progeria syndrome. In this study, a lamin A/C knockout human induced pluripotent stem cell line was successfully generated using the CRISPR/Cas9 genome-editing technology, which was confirmed with normal pluripotency and karyotype.

Study of CFRP Laminate Gradually Modified throughout the Thickness Using Thin Ply under Transvers Tensile Loading.

The use of thin-ply composite materials has rapidly increased due to their tailorable mechanical properties and design flexibility. Considering an adhesively bonded composite joint, peel stress stands out as a key contributor leading to failure among other primary stress factors. Therefore, the reinforcement of carbon fiber-reinforced polymer (CFRP) laminates throughout the thickness could be considered as an approach to improve the joint strength. Using thin plies locally between the conventional CFRP layers in a laminate can enhance the strength, as the sudden change in stiffness means that

Investigation of bionic composite laminates inspired by the natural impact-resistant helicoidal structure in the mandibles of trap-jaw ants.

Many living organisms exhibit exceptional capabilities and have evolved effective strategies to synthesize impact-resistant and damage-tolerant structures. One such example can be observed in the rapid mandible strikes ofOdontomachus monticola, a species of trap-jaw ants from the ponerine subfamily. During trap-jaw strikes, the mandibles can achieve peak speeds of 35.42 m s-1, and the maximum acceleration can reach 71 729 g within an average duration of 0.18 ms. The extreme acceleration results in instantaneous mandible strike forces that can exceed 330 times the ant's body weight, withstandin

Bio-Membrane SELEX as a New Approach for Selecting ss-DNA Aptamers that Bind to the Hydatid Cyst Laminated Layer.

BACKGROUND: Hydatid cyst (HC) is the larval stage of the canine intestinal tapeworm (cestode), Echinococcus granulosus. In addition to the high global economic cost of livestock farming, the infection can lead to dangerous problems for human health. Therefore, research into new diagnosis and treatment approaches is valuable. This study is set out to explore aptamers that bind to HC antigens. METHODS: The similarity between HC genotype in sheep and humans was that sheep HCs were collected and used as a biological membrane for aptamer selection. Four Bio- Membrane SELEX rounds were conducted, a

Dexamethasone induces p21cip1/waf1 expression via FoxO3a independently of the Lamin A/C-HDAC2 interaction in Ataxia Telangiectasia.

Ataxia-Telangiectasia (A-T) is a very rare autosomal recessive multisystemic disorder which to date is still uncurable. The use of glucocorticoid analogs, such as dexamethasone (dex), can improve neurological symptoms in patients, but the molecular mechanism of action of these analogs remains unclear. Here, we report the effects of dex in regulating the interaction between Lamin A/C and HDAC2 in WT and A-T cells. Upon administration of dex to A-T cells, we first observed that the accumulation of HDAC2 on the CDKN1A promoter did not exert a repressive role on p21cip1/waf1 expression, and second

Altered p16Ink4a, IL-1β, and Lamin b1 Protein Expression Suggest Cellular Senescence in Deep Endometriotic Lesions.

Endometriosis causes immunological and cellular alterations. Endometriosis lesions have lower levels of lamin b1 than the endometrium. Moreover, high levels of pro-inflammatory markers are observed in the peritoneal fluid, follicular fluid, and serum in endometriosis lesions. Thus, we hypothesized that the accumulation of senescent cells in endometriosis tissues would facilitate endometriosis maintenance in an inflammatory microenvironment. To study senescent cell markers and the senescence-associated secretory phenotype (SASP) in endometriosis lesions, we conducted a cross-sectional study wit

Colocalization Analysis of Peripheral Myelin Protein-22 and Lamin-B1 in the Schwann Cell Nuclei of Wt and TrJ Mice.

Myelination of the peripheral nervous system requires Schwann cells (SC) differentiation into the myelinating phenotype. The peripheral myelin protein-22 (PMP22) is an integral membrane glycoprotein, expressed in SC. It was initially described as a growth arrest-specific (gas3) gene product, up-regulated by serum starvation. PMP22 mutations were pathognomonic for human hereditary peripheral neuropathies, including the Charcot-Marie-Tooth disease (CMT). Trembler-J (TrJ) is a heterozygous mouse model carrying the same pmp22 point mutation as a CMT1E variant. Mutations in lamina genes have been r
