My Voice Library: Protocol for Developing Audio and Visual Datasets to Enable Personalized Real-Time Communication for People With Dysarthria.

BACKGROUND: Communication is a human right; despite this, children with cerebral palsy and dysarthria experience constraints due to their physical disability, affecting their opportunity to reach their full potential and ability to fully participate in play, school, and socializing. For many, their challenge to produce clear speech that can be understood is interpreted by others as an intellectual impairment. Children with moderate-severe speech impairments such as dysarthria often rely on technology solutions, which can translate text or symbols to speech manually or using a single binary swi

Model-Informed Precision Dosing in Pediatric Patients: Current Software Tools and Bedside Guided Application.

BACKGROUND: Personalized dosing is particularly important in pediatric patients, as age-related physiological maturation, developmental changes, and patient-specific factors contribute to substantial pharmacokinetic variability. Standard dosing approaches often fail to adequately account for this heterogeneity and may lead to subtherapeutic or supratherapeutic drug concentrations. Model-informed precision dosing (MIPD) addresses these limitations by integrating individual patient data with population pharmacokinetic/pharmacodynamic models to optimize dosing at the individual level more accurat

Targeting regulatory T cells for cancer immunotherapy: promises and pitfalls.

BACKGROUND: Regulatory T cells (Tregs) form a distinct subset of CD4+ T cells essential for maintaining immune balance and preventing autoimmunity by expressing CD25 and CTLA-4. Tregs, which are defined by the expression of the transcription factor FOXP3 in mice, restrain excessive immune activation through mechanisms such as interleukin 10 (IL-10) expression, transforming growth factor-β expression, metabolic regulation, acting as an IL-2 cytokine sink, and cell-to-cell interaction through co-inhibitory receptors. Although they are indispensable for immune tolerance, Tregs can be co-opted by

Long-read, high-throughput sequencing of 10,000 human mitogenomes using the PacBio Sequel IIe.

Sequence analysis of the human mitochondrial genome (mitogenome) is of interest to the molecular anthropology, medical, and forensic communities. Quality mitogenome data is an essential component of haplotype search databases, serving as an important element of forensic investigations to ensure that weight estimates are reflective of accurate coincidental match probabilities. The European DNA Profiling group (EDNAP) Mitochondrial DNA Population database (EMPOP) is considered the gold standard for this purpose, serving as a reference database and a quality-control tool. The current study report

The Challenge of Somatic Variants in Focal Cortical Dysplasia.

OBJECTIVE: The advent of next-generation sequencing (NGS) enabled the detection of low-level brain somatic variants in postsurgical tissue of focal cortical dysplasia (FCD). The genetic background of FCD Type I remains elusive, while the mammalian target of rapamycin (mTOR) pathway seems to have a relevant role in the pathogenesis of FCD Type II. Our goal was to uncover information on the molecular basis of FCD, performing whole genome sequencing (WGS) in postsurgical tissue to detect candidate brain-specific somatic variants, and evaluate their clinical significance. DESIGN: WGS was performe

Optimization of a Glucagon-Like Peptide 1 Receptor Antagonist Antibody for Treatment of Hyperinsulinism.

UNLABELLED: Congenital hyperinsulinism (HI) is a genetic disorder in which pancreatic β-cell insulin secretion is excessive and results in hypoglycemia that, without treatment, can cause brain damage or death. Most patients with loss-of-function mutations in ABCC8 and KCNJ11, the genes encoding the β-cell ATP-sensitive potassium channel (KATP), are unresponsive to diazoxide, the only U.S. Food and Drug Administration-approved medical therapy and require pancreatectomy. The glucagon-like peptide 1 receptor (GLP-1R) antagonist exendin-(9-39) is an effective therapeutic agent that inhibits insuli

Characterisation and management of dermatologic adverse events to agents targeting the PD-1 receptor.

BackgroundDermatologic adverse events (AEs) are some of the most frequently observed toxicities of immune-checkpoint inhibitor therapy, but they have received little attention. The drugs, pembrolizumab and nivolumab are recently approved inhibitors of the programmed death (PD)-1 receptor that have overlapping AE profiles however, the incidence, relative risk (RR), and clinico-morphological pattern of the associated dermatologic AEs are not known.MethodsWe conducted a systematic review of the literature, and performed a meta-analysis of dermatologic AEs observed with the use of pembrolizumab an

Mapping of modifiable barriers and facilitators of medication adherence in bipolar disorder to the Theoretical Domains Framework: a systematic review protocol.

INTRODUCTION: People with bipolar disorder require long-term treatment but it is estimated that 40% of these people do not adhere to prescribed medication regimens. Non-adherence increases the risk of relapse, hospitalisation and suicide. Some evidence syntheses report barriers to mental health treatment adherence but rarely delineate between modifiable and non-modifiable barriers. They also fail to distinguish between the patients' perspective and that of other stakeholders such as clinicians despite of their different understanding and priorities about adherence. Facilitators of adherence, w
