Integration of an electron beam ion trap source into an electrostatic accelerator for pre-clinical cancer research.

A commercial electron beam ion trap (EBIT) source has been integrated into the 5.0 MV Singletron accelerator at the Radiological Research Accelerator Facility (RARAF). Generally used as an injector for particle accelerators, it is the first integration of an EBIT source into a single-ended electrostatic accelerator, for cancer heavy-ion radiotherapy (HIRT) research. The EBIT source generates highly charged ions (B5+, C6+, etc.) in addition to light ions (protons, deuterons, and helium ions) produced by the standard High Voltage Engineering Europe (HVEE) configuration of the RARAF accelerator,

A sandwiched photoelectrochemical biosensing platform for detecting Cytokeratin-19 fragments based on Ag2S-sensitized BiOI/Bi2S3 heterostructure amplified by sulfur and nitrogen co-doped carbon quantum dots.

A sandwiched photoelectrochemical (PEC) immunosensor based on BiOI/Bi2S3/Ag2S was designed for the quantitative detection of cytokeratin-19 fragments (CYFRA21-1) in serum. In this work, due to the intervention of the narrow band gap Bi2S3, the absorption of the light source by the BiOI/Bi2S3 heterostructure has been significantly enhanced. Meanwhile, the matched band structure of BiOI, Bi2S3 and Ag2S promoted the rapid transfer of electrons between the conduction bands and effectively inhibited the recombination of electron-hole pairs, thus enhanced the photoelectric signals. Sulfur and nitrog

Electrochemiluminescent aptasensor based on resonance energy transfer system between CdTe quantum dots and cyanine dyes for the sensitive detection of Ochratoxin A.

In this work, an innovative aptasensor based on electrochemiluminescence resonance energy transfer (ECL-RET) from CdTe quantum dots (QDs) to a cyanine dye (Cy5) fluorophore for the determination of Ochratoxin A (OTA) was fabricated. A strong cathodic ECL emission was obtained by the CdTe QDs modified glassy carbon electrode (GCE). After the immobilization with the capture DNA (cDNA) and the sequential hybridization with the probe DNA-modified Cy5 (pDNA, the aptamer of OTA), the ECL signal enhanced obviously through the ECL-RET. Meanwhile, the spectrum- and distance-related ECL enhancement effe

Development of near-infrared ratiometric fluorescent probe based on cationic conjugated polymer and CdTe/CdS QDs for label-free determination of glucose in human body fluids.

Quantum dots (QDs) have attracted extensive attention in biomedical applications, because of their broad excitation spectra, narrow and symmetric emission peaks etc. Furthermore, near-infrared (NIR) QDs have further advantages including low autofluorescence, good tissue penetration and low phototoxicity. In this work, the electrostatic interaction and fluorescence resonance energy transfer (FRET) between NIR CdTe/CdS QDs and cationic conjugated polymer (CCP) was studied for the first time. Based on the newly discovered phenomena and the result that hydrogen peroxide (H2O2) can efficiently quen

Investigation of trypsin-CdSe quantum dot interactions via spectroscopic methods and effects on enzymatic activity.

The paper presents the interactions between trypsin and water soluble cadmium selenide (CdSe) quantum dots investigated by spectrophotometric methods. CdSe quantum dots have strong ability to quench the intrinsic fluorescence of trypsin by a static quenching mechanism. The quenching has been studied at three different temperatures where the results revealed that electrostatic interactions exist between CdSe quantum dots and trypsin and are responsible to stabilize the complex. The Scatchard plot from quenching revealed 1 binding site for quantum dots by trypsin, the same has been confirmed by

Fast neutron radiotherapy for primary mucosal melanomas of the head and neck.

BACKGROUND: Primary head and neck mucosal melanomas (HNMMs) are rare tumors managed with surgery and/or radiotherapy and associated with poor outcomes. Given their radioresistance, high linear energy transfer radiotherapy with neutrons may improve local control. METHODS: We conducted a retrospective review of 14 patients with HNMM treated with neutrons at the University of Washington from 1990 to 2012. Five patients had T3 disease, 9 had T4 disease, 3 had regional nodal disease, and 4 had distant metastases at diagnosis. Primary sites were sinonasal (n=10), lip (n=2), and palate (n=2). Ten pa…

Adaptive response to gamma radiation in mammalian cells proficient and deficient in components of nucleotide excision repair.

Cells preconditioned with low doses of low-linear energy transfer (LET) ionizing radiation become more resistant to later challenges of radiation. The mechanism(s) by which cells adaptively respond to radiation remains unclear, although it has been suggested that DNA repair induced by low doses of radiation increases cellular radioresistance. Recent gene expression profiles have consistently indicated that proteins involved in the nucleotide excision repair pathway are up-regulated after exposure to ionizing radiation. Here we test the role of the nucleotide excision repair pathway for adaptiv…

Dual-readout lateral flow immunoassay based on upconversion nanoparticle-gold nanoparticle luminescence resonance energy transfer for rapid determination of 17β-estradiol.

A luminescence resonance energy transfer (LRET)-regulated dual-readout lateral flow immunoassay (LFIA) is reported that integrates naked-eye colorimetric screening and near-infrared (NIR)-excited quantitative luminescence analysis on a single test strip. Core-shell upconversion nanoparticles (UCNPs) functionalized with E2-bovine serum albumin conjugates were immobilized on the test line as luminescent donors, while gold nanoparticle (AuNP)-labeled anti-E2 monoclonal antibodies served dually as colorimetric labels and efficient LRET acceptors. Time-resolved luminescence measurements confirmed a

Two-dimensional otolith size trends in myctophid fishes.

Lanternfishes are a key food source for marine and terrestrial predators, and play an important role in energy transfer between surface waters and deeper layers. This study investigates otolith size variation in 105 myctophid species included in the AFORO database. The study covers the five Myctophidae subfamilies: Diaphinae, Gymnoscopelinae, Lampanyctinae, Myctophinae and Notolychninae. Several measurements were taken from the otoliths, and relative shape and sizes indices were calculated for the different fishes. Morphometric analyses revealed that Diaphinae and Gymnoscopelinae possess the l

Actinium-225 Targeted Agents: Where Are We Now?

α-particle targeted radionuclide therapy has shown promise for optimal cancer management, an exciting new era for brachytherapy. Alpha-emitting nuclides can have significant advantages over gamma- and beta-emitters due to their high linear energy transfer (LET). While their limited path length results in more specific tumor 0kill with less damage to surrounding normal tissues, their high LET can produce substantially more lethal double strand DNA breaks per radiation track than beta particles. Over the last decade, the physical and chemical attributes of Actinium-225 (225Ac) including its half…

A far-red FRET fluorescent probe for ratiometric detection of l-cysteine based on carbon dots and N-acetyl-l-cysteine-capped gold nanoparticles.

A novel far-red fluorescence resonance energy transfer (FRET) fluorescent probe for ratiometric detection of l-cysteine (l-Cys) has been designed. The system was established a FRET assembly by positively charged carbon dots (CDs) and negatively charged N-acetyl-l-cysteine capped gold nanoparticles (NAC-AuNPs). The fluorescence of CDs at 539 nm could be effectively quenched in the presence of NAC-AuNPs owing to FRET process, while the emission of NAC-AuNPs at 630 nm was appeared. Subsequently, the interactions between l-Cys and NAC-AuNPs resulted in the decreased emission intensity of NAC-AuNPs

Development of Targeted Alpha Particle Therapy for Solid Tumors.

Targeted alpha-particle therapy (TAT) aims to selectively deliver radionuclides emitting α-particles (cytotoxic payload) to tumors by chelation to monoclonal antibodies, peptides or small molecules that recognize tumor-associated antigens or cell-surface receptors. Because of the high linear energy transfer (LET) and short range of alpha (α) particles in tissue, cancer cells can be significantly damaged while causing minimal toxicity to surrounding healthy cells. Recent clinical studies have demonstrated the remarkable efficacy of TAT in the treatment of metastatic, castration-resistant prosta…

Europium-Functionalized Flexible Luminescent Zeolite-like Supramolecular Assembly for Ratiometric Anthrax Biomarker Determination.

Anthrax spores have been a determining risk to human beings and animals across the world. As such, the qualitative monitoring toward dipicolinic acid (DPA), the unique biomarker of bacillus anthracis, is highly desired and being moved forward in health management. Herein, a breathing zeolite-like supramolecular assembly (ZSA), with the sra topology and 1D hexagon channel, is designed by controlling the bridging angle of the ditopic ligand after systematic analysis in topology. The solvent-dependent dynamic behavior is illustrated by powder X-ray diffraction and reflected in tunable luminescent

Radio-adaptive response to environmental exposures at Chernobyl.

The genetic consequences resulting from environmental exposure to ionizing radiation have a significant impact on both radiation regulatory policies and the comprehension of the human health risks associated with radiation exposure. The primary objectives of the study were to assess 1) genotoxicity of exposure to radiation as a function of absorbed dose and dose rate, and 2) induction of a radio-adaptive response following a priming dose at varying dose rates. Results demonstrated that sub-acute environmental exposures of 10cGy gamma radiation resulted in indistinguishable levels of chromosoma

Bayesian analysis of overdispersed chromosome aberration data with the negative binomial model.

The usual assumption of a Poisson model for the number of chromosome aberrations in controlled calibration experiments implies variance equal to the mean. However, it is known that chromosome aberration data from experiments involving high linear energy transfer radiations can be overdispersed, i.e. the variance is greater than the mean. Present methods for dealing with overdispersed chromosome data rely on frequentist statistical techniques. In this paper. the problem of overdispersion is considered from a Bayesian standpoint. The Bayes Factor is used to compare Poisson and negative binomial …

Ab initio electronic absorption spectra of para-nitroaniline in different solvents: Intramolecular charge transfer effects.

Intramolecular charge transfer (ICT) effects of para-nitroaniline (pNA) in eight solvents (cyclohexane, toluene, acetic acid, dichloroethane, acetone, acetonitrile, dimethylsulfoxide, and water) are investigated extensively. The second-order algebraic diagrammatic construction, ADC(2), ab initio wave function is employed with the COSMO implicit and discrete multiscale solvation methods. We found a decreasing amine group torsion angle with increased solvent polarity and a linear correlation between the polarity and ADC(2) transition energies. The first absorption band involves π → π* transition

Molecularly imprinted polymer based hybrid structure SiO2@MPS-CdTe/CdS: a novel fluorescence probe for hepatitis A virus.

A novel designed fluorescence molecularly imprinted polymer (MIP) probe made from CdTe/CdS quantum dot (QD)-based silica nanoparticles (SiO2@MPS-CdTe/CdS) was successfully created via a sol-gel process. The target virus - hepatitis A virus (HAV) was selectively captured by imprinted polymer layer, resulting in the fluorescence quenching of the QDs within 20 min, which could be explained by the energy transfer mechanism. Under optimized conditions, the limit of detection for the SiO2@MPS-CdTe/CdS MIP was as low as 88 pmol · L-1, and excellent linearity was obtained from 0.2 to 1.4 nM. Additiona

[Adsorptive voltammetric behavior of omeprazole and its application].

AIM: To study the electrochemical behavior of omeprazole (OPZ) and a new method for the determination of the compound was established by adsorptive stripping voltammetry. METHODS: Linear-sweep polarography and cyclic voltammetry were used. RESULTS: In a supporting electrolyte containing 0.1 mol.L-1 NH3-NH4Cl, a reduction peak of OPZ was observed by linear-sweep voltammetry at Hg electrode. The peak showed a potential of -1.04 V (vs Ag/AgCl at 100 mV.s-1) and adsorptive characteristics. The adsorbed species is most probably the neutral molecular of OPZ. The saturated adsorption of OPZ at Hg e
