Purification and Product Characterization of Lipoxygenase from Opium Poppy Cultures (Papaver somniferum L.).

Opium poppy (Papaver somniferum L.) is an ancient medicinal plant producing pharmaceutically important benzylisoquinoline alkaloids. In the present work we focused on the study of enzyme lipoxygenase (LOX, EC 1.13.11.12) from opium poppy cultures. LOX is involved in lipid peroxidation and lipoxygenase oxidation products of polyunsaturated fatty acids have a significant role in regulation of growth, development and plant defense responses to biotic or abiotic stress. The purpose of this study was to isolate and characterize LOX enzyme from opium poppy callus cultures. LOX was purified by ammoni

Fatty acid composition of umbilical arteries and veins: possible implications for the fetal EFA-status.

Fatty acid compositions were determined of phospholipids, isolated from umbilical arteries and veins, obtained from Dutch neonates after vaginal delivery, terminating normal pregnancy. The fatty acid profiles of the cord vessels were characterized by the absence of eicosapentaenoic (timnodonic) acid, a low (2-3%) content of linoleic acid and reasonable amounts of arachidonic acid (10-15%) and docosahexaenoic (cervonic) acid (3-5%). Significant amounts of Mead acid (1-4%) and its direct elongation product (0.5-2%) were also observed. In each cord, the efferent blood vessels contained significan

Formation and cytotoxicity of fatty acid chlorohydrins during sodium hypochlorite bleaching of tree nuts.

Fatty acid chlorohydrins are an emerging class of contaminants formed when hypochlorous acid reacts with unsaturated fatty acids during food processing. While their formation has been reported under high sodium hypochlorite (NaOCl; ≤200 mg/L as Cl2) disinfection of vegetables, much higher levels-≤ 60,000 mg/L of Cl2 or 6% NaOCl-are used to brighten shells. Monochlorohydrins of palmitoleic, oleic, and linoleic acids, along with dichlorohydrins of linoleic acid, were measured in walnuts, pistachios, and hazelnuts after NaOCl bleaching. Formation increased with oxidant dose and kernel accessibili…

Leukoencephalomyelopathy in cats linked to abnormal fatty acid composition of the white matter of the spinal cord and of irradiated dry cat food.

Four outbreaks of leukoencephalomyelopathy in colonies of SPF cats on a long-term diet of irradiated dry cat food were observed in the Netherlands between 1989 and 2001. As a primary defect in myelin formation was suspected to be the cause of the disease and myelin consists mainly of lipids and their fatty acids, we investigated the fatty acid composition of the white matter of the spinal cord of affected and control cats and of irradiated and non-irradiated food. The irradiated food had low levels of alpha-linolenic acid compared to linoleic acid as well as a high total omega-6:omega-3 ratio

Metabolome analyses of mouse uteri two to four days post coitum.

To elucidate the temporal and dynamic changes in uterine metabolites in mice, uterine samples were collected on Day 2 (D2; n = 8), Day 3 (D3; n = 6), and Day 4 (D4; n = 7) postcoitum and subjected to an untargeted metabolomic analysis. The results revealed significant metabolic alterations across the time points. Compared to D2, D3 showed 124 differentially expressed metabolites (95 upregulated and 29 downregulated), whereas D4 exhibited 377 (262 upregulated and 115 downregulated). Between D4 and D3, 370 metabolites were altered (242 upregulated and 128 downregulated). K-means clustering ident

[Essential fatty acids in parenteral nutrition].

Fat is a very important nutrient in that it supplies energy, essential fatty acids and fat soluble vitamins. The importance of n-6 essential fatty acids, i.e., linoleic and arachidonic acid, in total parenteral nutrition was demonstrated in the last 15 years by several cases of essential fatty acid deficiency. In addition, n-3 fatty acids, i.e., alpha-linolenic acid, eicosapentaenoic acid and docosahexaenoic acid, are essential nutrients in forming an independent family of eicosanoids with biological effects different from those of the n-6 fatty acids. The requirement of different essential fa

Prepartum organic selenium supplementation influences postpartum inflammatory response by regulating plasma oxylipin profiles in periparturient dairy cows.

Excessive lipolysis and enhanced inflammation in periparturient dairy cows may lead to metabolic disorders and diseases. Membrane phospholipids and lipid mobilisation-released polyunsaturated fatty acids (PUFAs) can be oxidised into oxylipins that have pro- and/or anti-inflammatory roles. Selenium has immunomodulatory and anti-inflammatory effects, but the mechanisms are not yet fully understood. The objective of this study was to examine the effects of prepartum selenium-yeast (SY) supplementation on the inflammatory response in dairy cows during the periparturient period by regulating plasma

Discovery of potent GPR120 activators with fatty orosensation enhancement activity.

GPR120 is expressed in the tongue and plays an important role in fatty acid taste perception. Natural polyunsaturated fatty acids, including α-linolenic acid and linoleic acid, are found in various edible oils such as soybean, rapeseed, and safflower oils. These fatty acids activate GPR120 but are susceptible to oxidation during storage and heating. To address these limitations while retaining GPR120 activity, a series of conformationally restricted, aliphatic side-chain-conjugated aryl short-chain fatty acid derivatives was synthesized. Several compounds exhibited potent GPR120-mediated activ

The role of oxidized lipid species in insulin resistance and NASH in children.

During the last two decades, nonalcoholic fatty liver disease (NAFLD) has emerged as the most common hepatic disease in pediatrics, mainly owing to the rising prevalence of pediatric obesity. Epidemiological studies have shown that the progressive increase in NAFLD prevalence is associated not only with obesity but also with changes in dietary habits experienced by all age groups, characterized by the increased intake of added sugars and certain fatty acids. In this review article, we focus on the effect of oxidized fatty acids deriving from linoleic acid and arachidonic acid on the pathogenes

Phytochemical profiling and antioxidant potential of Phlomoides rotata essential oils.

The antioxidant properties and characteristic metabolites of Phlomoides rotata (P. rotata) essential oils (EOs) remain largely unexplored. To address this gap, we combined metabolomic profiling with in vitro antioxidant assays to systematically characterize EOs from P. rotata. Cryoprecipitation of EOs yielded two fractions: crystals (Crs) and crystal-free EOs (CEs). GC-MS identified 125 components (84.41-94.86%), including 94 first reports in P. rotata. Dominant constituents were long-chain fatty acids (LCFAs, 42.33-75.73%) and their esters (3.44-15.21%), notably palmitic acid (14.49-63.13%),

Abnormality in membrane fatty acid compositions of cells measured on erythrocyte in alcoholic liver disease.

It has been proven that the fatty acids of esterified phospholipids in the cell membrane play an important role in membrane fluidity. Our previous in vitro experiment indicated that the impairment of erythrocyte membrane fluidity might be largely because of the change in fatty acids. The aim of this study is to clarify changes of cell membrane fatty acids in more detail in relation to various stages and pathology of alcoholic liver disease. For the analysis, erythrocyte membranes were exploited on the assumption that their fatty acid compositions may be similar to those of other organs. In alc

Metabolomic signatures and causal effects of circulating metabolites on age-related cataract.

BACKGROUND: Age-related cataract (ARC) is a leading cause of visual impairment and blindness among older adults, yet effective pharmacological options remain limited. This study aimed to identify candidate circulating metabolic biomarkers and pathways associated with ARC risk. METHODS: Genetic data for blood metabolites and ARC were derived from three metabolite genome-wide association studies (GWASs) and FinnGen. Mendelian randomization (MR) analyses assessed associations between 121 metabolites and ARC. UK Biobank cohort analyses evaluated whether MR-identified associations were observed in

On the effect of brain phospholipase A1 on specifically labelled glycerophospholipids in the course of subacute sclerosing panencephalitis.

The effect of phospholipase A1 of human brain on 1,2-diacyl-sn-glycero-3-phosphorylcholine, -ethanolamine and -serine, specifically labelled with different fatty acids at either the 1 or 2 position, was determined in subacute sclerosing panencephalitis. An increase of approximately 40% in the specific activity of phospholipase A1 could be observed for all substrates investigated during the demyelinating disorder. On investigating the specific activity of the enzyme with various molecular species of phosphatidylcholine and -ethanolamine, labelled at the 1 position with different radioactive fat
