This is the first longitudinal study of immune profiles and autologous hematopoietic cell transplant (AHCT) survival in B-cell non-Hodgkin lymphoma (B-NHL) patients and the effect of plerixafor mobilization on immune reconstitution in this population. A comprehensive immunophenotyping panel was performed in 104 consecutive adult B-NHL patients (58% diffuse large B cell and 42% mantle cell) who received AHCT (1/2008-11/2014), at a median of 28 days pre-AHCT (N = 104) and Day +100 (N = 83) post-AHCT. Median follow-up post-AHCT was 61 months (range: 8-120 months). Compared to patients mobilized w

Diffuse large B-cell lymphoma (DLBCL) is a frequently encountered aggressive form of non-Hodgkin's lymphoma, characterized by significant clinical and molecular heterogeneity. Owing to inherent limitations of traditional treatments for DLBCL, the advent of circulating tumor DNA (ctDNA) detection, coupled with the integration of multi-omics technologies, has brought about substantial advancements in the diagnosis and treatment of this disease. This review elaborates on the evolution of ctDNA detection and multi-omics integration, its applications in early diagnosis, prognostic model constructio

Diffuse large B-cell lymphoma (DLBCL) is the most common and clinically aggressive subtype of non-Hodgkin lymphoma (NHL). While novel therapies such as rituximab and polatuzumab vedotin have led to improved outcomes, approximately 35% of patients eventually develop relapsed or refractory disease. MicroRNAs (miRNAs), a class of endogenous single-stranded RNAs approximately 22 nucleotides in length, play a pivotal role in the regulation of gene expression at the post-transcriptional level through interactions with complementary target RNAs and contribute significantly to the development, progres

BACKGROUND: Very old or frail patients with newly diagnosed diffuse large B-cell lymphoma (DLBCL) frequently cannot tolerate full-dose R-CHOP, and long-term control with R-miniCHOP is suboptimal. We assessed a sequential, chemotherapy-limiting regimen using zanubrutinib, lenalidomide, and rituximab (ZR2) induction followed by response-adapted ZR2-miniCHOP. METHODS: This single-center, open-label, investigator-initiated phase 2 trial enrolled untreated patients aged ≥ 80 years, or 60–79 years with ECOG performance status ≥ 2. All patients received two 21-day cycles of chemotherapy-free ZR2 (zan

Waldenström's Macroglobulinemia (WM) is a lymphoplasmacytic lymphoma which can rarely transform into diffuse large B cell lymphoma (DLBCL). Usually, the diagnosis of WM precedes DLBCL. A 79-year-old male presented to the hospital for worsening dysphagia and shortness of breath due to a rapidly growing base of tongue mass detected by CT scan. The mass was thought initially to be isolated DLBCL, though incidentally, markedly elevated IgM titers were noted. Biopsy of bone marrow and mass were consistent with histological transformation of WM to DLBCL. Response to treatment was assessed by the siz

CD20 × CD3 bispecific antibodies (BsAbs) have emerged as a meaningful therapeutic option for relapsed or refractory diffuse large B-cell lymphoma (DLBCL), redirecting endogenous T cells against malignant B cells independently of major histocompatibility complex-mediated antigen presentation, and have received regulatory approval after at least two prior lines of therapy. However, a substantial proportion of patients experience primary resistance or early relapse, underscoring the need to characterize the underlying biological mechanisms, which are the focus of this review. Several tumor-intrin

Rosai-Dorfman disease (RDD) can present with lymphadenopathy. However, it is important to note that RDD can also coexist with hematologic malignancies, including non-Hodgkin lymphomas. We present a case of an older male patient with generalized lymphadenopathy who was first diagnosed with RDD. High-dose steroid therapy was rendered without a satisfactory response. Thus, the tissue specimens were re-examined, and the patient was found to have diffuse large B-cell lymphoma (DLBCL) in the background of RDD. The laboratory tests are notable for a slightly elevated serum lactate dehydrogenase (LDH)

Second branchial cleft cyst and lymphoma are two common types of neck masses. The etiology, clinical manifestations, imaging findings, treatment options, and prognosis of these two entities differ. Clinically, the occurrence of both lesions at the same anatomical site is rare. Herein, we report a case of concurrent second branchial cleft cyst and diffuse large B-cell lymphoma in the same anatomical region. The final diagnosis was confirmed by clinical features, ima-ging findings, and ultrasound-guided biopsy. The patient received the R-CHOP (rituximab, cyclophosphamide, doxorubicin, vincristin

Lymphoma is the most frequently diagnosed neoplasm in pigs, predominantly of B-cell origin. Multicentric forms commonly affect the lymph nodes, spleen, liver and kidneys. This report describes a case of a multicentric diffuse large B-cell lymphoma with dural involvement in a 70-day-old nursery piglet presenting with hindlimb paralysis. Necropsy and histological examinations revealed nodular thickening of the meninges of the brain and sacral spinal cord, as well as adjacent paravertebral ganglia, by proliferations of large neoplastic lymphocytes. The same neoplastic cells infiltrated the mesent

Primary vertebral diffuse large B-cell lymphoma (PV-DLBCL) is a rare manifestation of extranodal non-Hodgkin lymphoma that presents with nonspecific symptoms and poses significant diagnostic and treatment challenges. While chemotherapy, radiation, and surgery are frequently employed, their relative impacts on long-term survival remain unclear. Using the SEER database (2000–2021), we identified 584 adult patients with histologically confirmed PV-DLBCL. Demographic, tumor, and treatment characteristics were analyzed. Cox proportional hazards models were used to evaluate predictors of mortality,

BACKGROUND: An increased risk of secondary primary malignancies (SPMs) has been shown in diffuse large B-cell lymphoma (DLBCL) patients after treatment. However, research on SPM outcomes in DLBCL patients is limited.
METHODS: Utilizing the population-based Surveillance, Epidemiology, and End Results Registry (SEER), the authors examined the characteristics, overall survival (OS), and treatments in DLBCL patients who developed secondary cancers of the breast, prostate, lung, colon/rectum, bladder cancers, melanoma, myeloma, and acute leukemia. Each patient was matched by age, sex, race, year o

EZH2 functions as an oncogene in various cancers. However, the role of EZH2 in diffuse large B cell lymphoma (DLBCL) is scarcely reported. This study investigates the potential of EZH2 in DLBCL and the underlying mechanisms. mRNA levels were detected using reverse transcription-quantitative PCR. Protein expression was detected using Western blot. Cell viability was detected using CCK-8 assay. Cell proliferation was detected using methylcellulose colony formation. Cell migration and invasion was detected using transwell. We found that EZH2 is upregulated in DLBCL. EZH2 deficiency effectively en

Background/Objectives: Diffuse Large B-Cell Lymphoma (DLBCL) is the most common subtype of non-Hodgkin lymphoma and demands precise segmentation and classification of nuclei for effective diagnosis and disease severity assessment. This study aims to evaluate the performance of HoVerNet, a deep learning model, for nuclei segmentation and classification in CMYC-stained whole slide images and to assess its integration into a user-friendly diagnostic tool. Methods: A dataset of 122 CMYC-stained whole slide images (WSIs) was used. Pre-processing steps, including stain normalization and patch extrac

We present the case of a 56-year-old male with cecal involvement of non-Hodgkin's lymphoma (NHL), whose tumor was refractory to multiple chemotherapy regimens and ultimately resulted in significant local complications due to tumor erosion, leading to considerable morbidity. NHL is the most common hematologic cancer with many subtypes of disease, with diffuse large B-cell lymphoma (DLBCL) being the most common. Extra-nodular presentation in the gastrointestinal tract is typical in many cases; however, the involvement of the colon is rare. This case details a patient with extra-nodal involvement

Diffuse large B-cell lymphoma (DLBCL) exhibits marked clinical heterogeneity and frequent treatment resistance, particularly in molecularly defined high-risk subtypes such as ABC-DLBCL. While current therapies largely rely on apoptosis induction, non-apoptotic cell death pathways remain underexplored in hematologic malignancies. Here, we identify ferroptosis, an iron-dependent, lipid peroxidation-driven form of regulated necrosis, as an effective baseline in additive therapy with ibrutinib for the treatment of DLBCL. Transcriptomic and lipidomic analyses revealed that DLBCL cells, despite lack

One-third of the patients with diffuse large B-cell lymphoma (DLBCL) experience relapse despite receiving standard R-CHOP chemotherapy. We aimed to elucidate the clinical course and prognosis in older patients with relapsed or refractory (R/R) DLBCL in a single-center experience in Japan. We conducted a retrospective survey of 52 older patients with R/R DLBCL (aged >65 years at diagnosis; 54% men) who received R-CHOP chemotherapy, to assess their clinical course and prognosis. The median progression-free survival was 8.5 months. Seventeen patients had central nervous system (CNS) relapse, with
