Activation of Lysosomal Retrograde Transport Triggers TPC1-IP3R1 Ca2+ Crosstalk at Lysosome-ER MCSs Leading to Lethal Depleting of ER Calcium.

Inter-organellar signaling linkages in oncology are increasingly elucidated. However, the impact of lysosome-endoplasmic reticulum (ER) interaction on tumor cell fate remains relatively unexplored. A novel interaction between lysosomes and the ER, mediated by the flavonoid LW-213 through targeting LIMP2 (lysosomal integral membrane protein type 2)to activate a lysosomal repair pathway, is identified in acute myeloid leukemia (AML). This leads to activated RAB7A activity, enhancing lysosomal retrograde transport to the perinuclear region and increasing contact at lysosome-ER membrane contact si

Zika Virus Exploits Lipid Rafts to Infect Host Cells.

Several flaviviruses such as Hepatitis C virus, West Nile virus, Dengue virus and Japanese Encephalitis virus exploit the raft platform to enter host cells whereas the involvement of lipid rafts in Zika virus-host cell interaction has not yet been demonstrated. Zika virus disease is caused by a flavivirus transmitted by Aedes spp. Mosquitoes, although other mechanisms such as blood transfusion, sexual and maternal-fetal transmission have been demonstrated. Symptoms are generally mild, such as fever, rash, joint pain and conjunctivitis, but neurological complications, including Guillain-Barré s

Lipocalin 2 as a membrane-reorganizing agent.

Lipocalins are a class of proteins that scavenge hydrophobic molecules in diverse contexts, including the immune system, the nervous system, and cancer. A recent study by Watanbe et al. identifies lipocalin 2 produced by the female mouse reproductive tract as a sperm-capacitating agent that alters the membrane properties of sperm in preparation for fertilization. The potential for lipocalins to act as general modulators of plasma membrane bioactivity is discussed.

Antibody binding geometry and affinity control inhibitory hFcγRIIB receptor signaling.

The inhibitory human Fc gamma receptor, hFcγRIIB, is a key mediator of humoral immunity and regulator of antibody-mediated effector function. hFcγRIIB function can be modulated by anti-hFcγRIIB antibodies. We demonstrate that agonistic, but not antagonistic, antibodies reduce hFcγRIIB mobility in the plasma membrane, associated with receptor clustering and redistribution into lipid rafts. Agonists display lower affinity binding with higher off rates compared with antagonists. Using crystallographic structure determination and alanine-scanning mutagenesis, we show that epitopes targeted by agon

Nesfatin-1 as a Cardiovascular-Metabolic-Immune Integrator: Insights from Cellular Cross-talk to Precision Medicine.

Nesfatin-1 is a multifunctional peptide that integrates cardiovascular, metabolic, and immune regulation, particularly along the "Heart-Brain-Gut" axis. Accumulating evidence highlights its pivotal role in inter-organ communication under both physiological and pathological conditions. However, its underlying molecular mechanisms remain incompletely understood, and the prevailing orphan G protein-coupled receptor (GPCR) hypothesis is currently undergoing critical scrutiny due to accumulating data that contest its exclusivity and sufficiency in accounting for nesfatin-1's pleiotropic effects. MYB Proto-Oncogene Like 2 Promotes the Growth of Renal Carcinoma by Attenuating Endoplasmic Reticulum Stress and Activating Lipid Rafts.

OBJECTIVE: Renal cell carcinoma (RCC) is a typical malignant lesion of kidney with high recurrence rate and high mortality. MYB proto-oncogene-like 2 (MYBL2) is significantly expressed in cancer cells and closely linked to cancer development and poor prognosis. This study aimed to investigate the role and possible mechanisms of MYBL2 in renal cancer. METHOD: MYBL2 was silenced and overexpressed in A498 kidney cancer cells. The ability of A498 cells to migrate and proliferate was analysed by 5-ethynyl-2'-deoxyuridine staining, colony formation assay, Transwell assay and cell counting kit-8 ass

Association of lipid rafts cholesterol with clinical profile in fragile X syndrome.

Fragile X syndrome (FXS) is the most prevalent monogenic cause of intellectual disability and autism spectrum disorder (ASD). Affected individuals have a high prevalence of hypocholesterolemia, however, the underlying mechanisms and the clinical significance remains unknown. We hypothesized that decrease in the plasma cholesterol levels is associated with an alteration of cholesterol content within the lipid rafts (LRs) which ultimately affects the clinical profile of FXS individuals. The platelets LRs were isolated by ultracentrifugation on sucrose gradient from 27 FXS and 25 healthy controls

Structural diversity and biological significance of glycosphingolipids in pathogenic and opportunistic fungi.

Glycosphingolipids (GSLs) are ubiquitous membrane components and have key roles in biological systems, acting as second messengers or modula … of fungal infections. Recently, it was shown that GlcCer and GIPCs are preferentially localized in membrane microdomains and monoclonal antibodies directed to these GSLs interfere in several fungal biological processes such as growth and morphological transition. This review focuses on glycan structures carried on sphingolipids of pathogenic/opportunistic fungi, and a …

Emerging pharmacotherapies for Creutzfeldt-Jakob disease.

Only a few years ago, pharmacotherapy of Creutzfeldt-Jakob disease was inconceivable. The enigmatic prion agent causing Creutzfeldt-Jakob disease, consisting solely of a misfolded conformational isoform, the scrapie prion protein, of the normal cellular prion protein was considered hard to treat by routine drug development. However, huge progress has been achieved in recent years, demonstrating principal reversibility of the neuropathological features and protection from clinical symptoms in animal models and introducing potential pharmaceutical agents. Among the most promising ones, antibodie

Role of cholesterol and the ganglioside GM(1) in entry and short-term survival of Brucella suis in murine macrophages.

Brucella species are gram-negative, facultative intracellular bacteria that infect humans and animals. These organisms can survive and replicate within a membrane-bound compartment inside professional and nonprofessional phagocytic cells. Inhibition of phagosome-lysosome fusion has been proposed as a mechanism for intracellular survival in both types of cells. We have previously shown that the maturation inhibition of the Brucella-containing phagosome appears to be restricted at the phagosomal membrane, but the precise molecular mechanisms and factors involved in this inhibition have yet to be

Alterations of membrane curvature during influenza virus budding.

Influenza A virus belongs to the Orthomyxoviridae family. It is an enveloped virus that contains a segmented and negative-sense RNA genome. Influenza A viruses cause annual epidemics and occasional major pandemics, are a major cause of morbidity and mortality worldwide, and have a significant financial impact on society. Assembly and budding of new viral particles are a complex and multi-step process involving several host and viral factors. Influenza viruses use lipid raft domains in the apical plasma membrane of polarized epithelial cells as sites of budding. Two viral glycoproteins, haemagg

Loss of expression of neutrophil proteinase-3: a factor contributing to thrombotic risk in paroxysmal nocturnal hemoglobinuria.

BACKGROUND: A deficiency of specific glycosylphosphatidyl inositol-anchored proteins in paroxysmal nocturnal hemoglobinuria may be responsible for most of the clinical features of this disease, but some functional consequences may be indirect. For example, the absence of certain glycosylphosphatidyl inositol-anchored proteins in paroxysmal nocturnal hemoglobinuria cells may influence expression of other membrane proteins. Membrane-bound proteinase 3 co-localizes with glycosylphosphatidyl inositol-linked neutrophil antigen 2a, which is absent in patients with paroxysmal nocturnal hemoglobinuria

Crystal structure of dihydroorotate dehydrogenase from Helicobacter pylori with bound flavin mononucleotide.

Helicobacter pylori is the primary causative agent of peptic ulcer disease, among other gastrointestinal ailments, and currently affects over half of the global population. Although some treatments exist, growing resistance to these drugs has prompted efforts to develop novel approaches to fighting this pathogen. To generate many of the nucleotides essential to biochemical processes, H. pylori relies exclusively on the de novo biosynthesis of these molecules. Recent drug-discovery efforts have targeted the first committed step of this pathway, catalysed by a class 2 dihydroorotate dehydrogenas

Deactivation of ceramide de novo synthesis induces cerebral angiogenesis and microvascular remodeling post-ischemia/reperfusion in mice via mechanisms not predominated by extracellular vesicles.

Ceramide is a major constituent of membrane microdomains and controls cell signaling. Previous studies showed that ceramide exacerbates cerebral ischemia/reperfusion injury and that inhibition of acid sphingomyelinase (ASM), which converts sphingomyelin to ceramide, promotes microvascular remodeling via induction of extracellular vesicles (EVs) release. ASM inhibitors have so far not translated into clinical stroke therapies. Of note, ceramide levels can also be lowered by inhibition of serine palmitoyltransferase (SPT), which is the rate-limiting enzyme of ceramide de novo synthesis. To evalu…

Blocking glycosphingolipid production alters autophagy in osteoclasts and improves myeloma bone disease.

Glycosphingolipids (GSLs) are key constituents of membrane bilayers playing a role in structural integrity, cell signalling in microdomains, endosomes and lysosomes, and cell death pathways. Conversion of ceramide into GSLs is controlled by GCS (glucosylceramide synthase) and inhibitors of this enzyme for the treatment of lipid storage disorders and specific cancers. With a diverse range of functions attributed to GSLs, the ability of the GSC inhibitor, eliglustat, to reduce myeloma bone disease was investigated. In pre-clinical models of multiple myeloma, osteoclast-driven bone loss was reduc

Simvastatin Inhibits Brucella abortus Invasion into RAW 264.7 Cells through Suppression of the Mevalonate Pathway and Promotes Host Immunity during Infection in a Mouse Model.

Simvastatin is an inhibitor of 3-hydroxy-3-methylglutaryl CoA reductase and has been found to have protective effects against several bacterial infections. In this study, we investigate the effects of simvastatin treatment on RAW 264.7 macrophage cells and ICR mice against Brucella (B.) abortus infections. The invasion assay revealed that simvastatin inhibited the Brucella invasion into macrophage cells by blocking the mevalonic pathway. The treatment of simvastatin enhanced the trafficking of Toll-like receptor 4 in membrane lipid raft microdomains, accompanied by the increased phosphorylatio

Increased plasmin-mediated proteolysis of L1CAM in a mouse model of idiopathic normal pressure hydrocephalus.

Idiopathic normal pressure hydrocephalus (iNPH) is a common neurological disorder that is characterized by enlarged cerebral ventricles, gait difficulty, incontinence, and dementia. iNPH usually develops after the sixth decade of life in previously asymptomatic individuals. We recently reported that loss-of-function deletions in CWH43 lead to the development of iNPH in a subgroup of patients, but how this occurs is poorly understood. Here, we show that deletions in CWH43 decrease expression of the cell adhesion molecule, L1CAM, in the brains of CWH43 mutant mice and in human HeLa cells harbori

CD14 targets complement receptor 3 to lipid rafts during phagocytosis of Borrelia burgdorferi.

Phagocytosis of Borrelia burgdorferi, the causative agent of Lyme disease, is mediated partly by the interaction of the spirochete with Complement Receptor (CR) 3. CR3 requires the GPI-anchored protein, CD14, in order to efficiently internalize CR3-B. burgdorferi complexes. GPI-anchored proteins reside in cholesterol-rich membrane microdomains, and through its interaction with partner proteins, help initiate signaling cascades. Here, we investigated the role of CD14 on the internalization of B. burgdorferi mediated by CR3. We show that CR3 partly colocalizes with CD14 in lipid rafts. The use o…
