Maternal intake of milk and milk proteins is positively associated with birth weight: A prospective observational cohort study.

BACKGROUND: A striking number of low birth weight (LBW) Indian babies are born annually. Previous studies have confirmed the positive association between milk intake and birth weight. However, the relations between protein and vitamin B12 from milk and birth weight have not been systematically explored. AIMS: We examined the relations between birth weight and maternal intake of milk, protein from milk and vitamin B12 from milk. METHODS: This prospective, observational cohort study was conducted in an urban South Indian hospital. The dietary intakes of milk and milk products were assessed usi…

Breast Milk and Hair Testing to Detect Illegal Drugs, Nicotine, and Caffeine in Donors to a Human Milk Bank.

BACKGROUND: The use of illegal drugs and tobacco is an exclusion criteria for accepting a nursing mother as a milk donor. The detection window for human milk testing is typically a few hours. Hair testing has been considered the gold standard to assess chronic exposure to these toxic substances. OBJECTIVE: The aim of this study was to determine the levels of illegal drugs, nicotine, and caffeine in breast milk and hair samples from donors to assess whether these substances were being used during the donation period and the months leading up to it. METHODS: Thirty-six samples of hair and brea…

Exposure Assessment of Infants to Aflatoxin M₁ through Consumption of Breast Milk and Infant Powdered Milk in Brazil.

Aflatoxin M₁ (AFM₁) is an important biomarker that can be used to evaluate aflatoxin exposure in both humans and animals. The aim of this study was to evaluate the exposure degree of infants to AFM₁ through consumption of breast milk and infant powdered milk in Brazil. For this purpose, the estimated daily intake (EDI) for infants was calculated based on the AFM₁ levels analyzed in 94 breast milk (BM) samples collected in Southern Brazil, and 16 infant powdered milk (IPM) samples commonly commercialized in Brazil. AFM₁ was detected in 5.3% (n = 5) and 43.8% (n = 7) of BM and IPM samples, with …

Determination of diphenhydramine in rat milk and plasma and its effects on milk composition and mammary gland nucleic acids.

To study the excretion of diphenhydramine into rat milk, milk and plasma concentrations of diphenhydramine were determined in lactating rats after single or multiple oral doses. Four hours after a single dose of 40 or 100 mg/kg of diphenhydramine, milk concentrations of the drug averaged 0.30 and 2.2 micrograms/mL, respectively, in two experiments, and the milk:plasma ratios ranged from 4.4 to 7.5. Multiple doses did not significantly affect the plasma or milk concentrations or the milk:plasma ratios, which were similar to the theoretical milk:plasma ratio based on pH partitioning for this com…

Comparative studies on the chemical and immunochemical properties of human milk, human pancreatic juice and bovine milk lactoferrin.

Lactoferrin from human milk, pancreatic juice and bovine milk were purified by heparin-Sepharose affinity chromatography procedure. Urea-sodium dodecylsulfate-polyacrylamide gel electrophoresis of these lactoferrin preparations indicated similar molecular weights (80,000). Metal analyses showed that lactoferrin of bovine milk contained the highest amount of iron while lactoferrin of human milk and human pancreatic juice were similar in content of iron, approximately four-fold lower than bovine milk. All these lactoferrin preparations were also found to contain minor amounts of copper and manga…

Immunogenicity of cows' milk in man. (Influence of age and of disease on serum antibodies to five cows' milk proteins).

Passive haemagglutination has been used to measure serum antibodies to the five proteins of cows' milk--bovine serum albumin, bovine gamma-globulin, alpha-lactalbumin, beta-lactoglobulin and casein. Titres of antibodies to these proteins, and a composite cows' milk antibody (CMA) score, have been measured in 1,087 sera from children and adults. Antibodies to all five proteins were detected in many specimens. Age and disease influenced not only the absolute titres of antibody, but also the relative distribution of antibodies to the five antigens. In infants aged 2 months the typical pattern was…

Detection of bovine milk-, but likely not soy-derived, peptides in human milk after maternal consumption of bovine milk and soy beverage: a randomized, cross-over, dietary intervention trial.

BACKGROUND: Food-borne allergens in human milk (HM) may cause allergic responses in HM-fed infants, but variability of allergen transfer complicates recommendations for individuals nursing food-allergic infants. OBJECTIVE: We aimed to identify bovine- and soy-derived peptides in HM after maternal elimination and reintroduction of bovine milk (BM) and soy beverage (SB). METHODS: In this randomized, cross-over, dietary intervention trial, 38 lactating participants underwent 2 study phases, each including a 5-day diet elimination, 3-day diet intervention, and 2-day washout. Each diet interventi…

Phenotypic and Genomic Profiling of Escherichia coli from Irish Raw Milk and Raw Milk Products: A Baseline Study.

Background/Objectives:Escherichia coli is a ubiquitous commensal organism in humans, animals, and the environment, but certain strains harbour virulence and antimicrobial resistance (AMR) determinants that can cause significant disease. Food-producing animals, including dairy cattle, may act as reservoirs for AMR E. coli, and raw milk and raw milk products can serve as potential exposure pathways to humans. However, data on the prevalence and genomic characteristics of AMR E. coli in raw milk in Ireland are limited. This study aimed to describe the occurrence of commensal and clinically releva…

In vitro Fermentation of Digested Milk Fat Globule Membrane From Ruminant Milk Modulates Piglet Ileal and Caecal Microbiota.

Lipids in milk are secreted as a triacylglycerol core surrounded by a trilayer membrane, the milk fat globule membrane (MFGM). This membrane, known to have important roles in infant brain and intestinal development, is composed of proteins, glycoproteins, and complex lipids. We hypothesized that some of the beneficial properties of MFGM are due to its effects on the gastrointestinal microbiota. This study aimed to determine the effect of a commercial phospholipid concentrate (PC) and enriched bovine, caprine, and ovine MFGM fractions on ileal and hindgut microbiota in vitro. Digestion of PC an…

Fasciola hepatica seroprevalence in Northern German dairy herds and associations with milk production parameters and milk ketone bodies.

Infections with the liver fluke Fasciola hepatica remain a serious problem in dairy herds causing significant production losses. In sheep, a strong relationship between F. hepatica infections and an increase in serum ketone bodies due to reduced feed intake and liver damage was demonstrated. We hypothesized that F. hepatica infections might contribute to an increase in milk ketone bodies in dairy herds. Thus, the objective of the study was to estimate the association between F. hepatica bulk tank milk (BTM) antibodies and milk production parameters (milk yield, milk protein, fat yield), somati…

[Detection of Coxiella burnetii in dairy cattle bulk tank milk and single tank milk samples by confirmatory testing].

Q fever is a worldwide zoonotic disease caused by the pathogen Coxiella (C.) burnetii. A wide range of animal species is susceptible to this intracellular bacterium with great importance in ruminants. Human infections occur mainly by airborne transmission. C burnetii was detected in animal products such as raw milk, raw-milk cheese and butter prepared from raw milk as well as in the meat of infected animals. In cattle milk, the pathogen was detected up to 13 months after calving. The risk of human foodborne C. Burnetii infection is still considered to be low, but cannot be completely ruled out…

Severe anaphylaxis to sheep's milk cheese in a child desensitized to cow's milk through specific oral tolerance induction.

Specific oral tolerance induction to food (SOTI) is a new promising treatmentfor persistent IgE-mediatedfood allergy. Our paper reports a case of a 5-year-old girl with cow's milk allergy, who developed severe anaphylaxis after the ingestion of a croissant containing sheep's milk ricotta cheese, even though she had been previously desensitized to cow's milk through SOTI. The sheep's milk specific allergen causing the severe allergic reaction (a derivative of alpha-casein of 54,1kDa) was identified by SDS-PAGE and immunoblotting. We conclude that SOTI is a species-specific procedure and the ind…

Confirmed low prevalence of Listeria mastitis in she-camel milk delivers a safe, alternative milk for human consumption.

She-camel milk is an alternative solution for people allergic to milk; unfortunately, potential harmful bacteria have not been tested in she-camel milk. Listeria monocytogenes is one harmful bacterium that causes adverse health effects if chronically or acutely ingested by humans. The purpose of this study was to estimate the prevalence, characterize the phenotypic, genetic characterization, virulence factors, and antibiopotential harmful bacteria resistance profile of Listeria isolated from the milk of she-camel. Udder milk samples were collected from 100 she-camels and screened for mastitis …

Eliciting doses of positive challenge test in cow's milk allergy are related to cow's milk specific IgE levels.

BACKGROUND: Ingestion of small amounts of cow's milk (CM) can elicit adverse reactions in patients with IgE-mediated CM allergy. Knowing the dose eliciting allergic reactions and the factors affecting it can be of great help in avoiding these reactions. OBJECTIVE: To analyse the eliciting doses of positive challenge test in patients with CM allergy and to determine its association with the level of CM specific IgE. METHODS: Ninety-eight positive challenge tests in 56 children, median age of 11 months (3-80) with IgE-mediated CM allergy were retrospectively analysed. Open oral challenge tests…

Production of toxic shock syndrome toxin by Staphylococcus aureus isolated from mastitic cow's milk and farm bulk milk.

The production of toxic shock syndrome toxin (TSST) by Staphylococcus aureus isolated from mastitic cow's milk and farm bulk milk was examined by a reverse passive latex agglutination method (RPLA). TSST was detected in 25 (58.1%) of 43 isolates from clinical mastitic cow's milk, in 79 (76.7%) of 103 isolates from subclinical mastitic cow's milk, and in 95 (75.4%) of 126 isolates from farm bulk milk, respectively. When the quantity of TSST in the isolates was determined by RPLA, the titers ranged from 5 to 2560. TSST with RPLA titers of 640 to 2,560 was produced by 83 (30.5%) of 272 isolates t…

[Cow's milk protein antibodies determined by immunofluorescence (RIFT) in cow's milk protein intolerance and in controls (author's transl)].

An immunofluorescent method using antigencoupled red cells was used to determine antibody titres against five main cow's milk proteins in 20 children with CMPI, in 130 normal controls, and in 70 children with various gastrointestinal disorders. Antibody findings were related to age and nutritional conditions. For no antibody group, a pathogenetic significance was found with respect to antigen or immunoglobulin class. Formation of serum cow's milk antibodies was far from indicating a pathological process. Nevertheless, determination of cow's milk antibody titres by RIFT had some use in the diff…
