Lessons that can be learned from patients with diabetogenic mutations in mitochondrial DNA: implications for common type 2 diabetes.

PURPOSE OF REVIEW: To discuss the role of mitochondria in the development of type 2 diabetes. RECENT FINDINGS: Some mutations in mitochondrial DNA are diabetogenic due to a gradual decline in insulin secretion by the pancreas. These mutations also result in abnormalities in lipid metabolism. A similar situation is seen in patients treated with nucleoside analogues as part of highly active antiretroviral therapy to suppress human immunodeficiency virus infection. These drugs induce a 30-50% reduction in mitochondrial DNA copy number in multiple tissues. Treated individuals develop a redistribu

[Ultrastructural morphometric analysis of normally loaded human myocardial left ventricles from young and old patients (author's transl)].

An ultrastructural and morphometric analysis was performed on human myocardial left ventricle, obtained during surgical heart operations on normally loaded ventricles. The diagnoses of the patients were a) Persistant foramen ovale b) Atrial septal defect stage (I.-II) c) Mitral stenosis (stage III.-IV.). The median values of left ventricular pressure were not pathologically elevated. The patients were divided in 2 groups, the younger one ranging from 5-15 years, the older from 42-78 years. It was shown that the volume density of the interstitial tissue does not differ with the aging process. T

[Ocular myopathy with giant mitochondriae (author's transl)].

A case of progressive external ophthalmoplegia and a slight coincident affection of the scapulohumeral muscle group is recorded. Thorough examination including EMG and biopsy from ocular and skeletal muscle tissue confirmed the clinical diagnosis of myopathy. By means of electron microscopy the affection could be identified as a type of myopathy characterized by giant mitochondriae. Causative factors were not detectable. The function of the thyroid and the carnitine level in the muscle tissue and blood serum were normal.

Targeted overexpression of catalase to mitochondria does not prevent cardioskeletal myopathy in Barth syndrome.

Barth Syndrome (BTHS) is an X-linked recessive disorder characterized by cardiomyopathy and muscle weakness. The underlying cause of BTHS is a mutation in the tafazzin (TAZ) gene, a key enzyme of cardiolipin biosynthesis. The lack of CL arising from loss of TAZ function results in destabilization of the electron transport system, promoting oxidative stress that is thought to contribute to development of cardioskeletal myopathy. Indeed, in vitro studies demonstrate that mitochondria-targeted antioxidants improve contractile capacity in TAZ-deficient cardiomyocytes. The purpose of the present st

Distribution of COX-negative mitochondria in myofibers of rats intoxicated with Senna occidentalis seeds.

We have described that administration of seeds or parts of the seed of Senna occidentalis (coffee senna) for long periods, induces histochemical changes in the skeletal muscles of hens and rats that are characteristic of a mitochondrial myopathy--as decrease of SDH and COX activity, with some COX negative fibers. In this experimental model of mitochondrial myopathy, as in many human mitochondrial diseases, there is a random distribution of COX negative fibers. Some fibers are completely COX negative while others are partially negative and others are completely positive. In the present work we

Abnormal mitochondria on a renal biopsy from a case of mitochondrial myopathy.

Mitochondrial myopathy (MM) is reported in a 5-year-old girl with short stature, hypotonia, ptosis, retinal pigmentation, and Fanconi's syndrome. A muscle biopsy showed the characteristic features of MM, and a renal biopsy revealed abnormal mitochondria in tubular cells that were similar to those seen in the muscle. This is the first report of such findings in association with MM.

Protocol to monitor live-cell, real-time, mitochondrial respiration in mouse muscle cells using the Resipher platform.

Mitochondrial function is typically assessed by measuring oxygen consumption at a given time point. However, this approach cannot monitor respiratory changes that occur over time. Here, we present a protocol to measure mitochondrial respiration in freshly isolated muscle stem cells, primary skeletal muscle, and immortalized C2C12 myoblasts in real time using the Resipher platform. We describe steps for preparing and plating cells, performing media changes, setting up the software and device, and analyzing data. This method can be adapted to other cell types. For complete details on the use and

Fragmented futures: ROCK1 drives mitochondrial fission and muscle loss in CKD.

Patients with chronic kidney disease (CKD) experience skeletal muscle wasting that leads to weakness and negatively impacts quality of life. Decreases in mitochondrial health and function have long been associated with skeletal muscle wasting in CKD, yet mechanisms linking mitochondrial alterations to muscle atrophy are lacking. In this issue, Si et al. present experimental work in mice with CKD linking Rho-associated protein kinase-1 activation to increased mitochondrial fission, oxidative stress, and atrophy. They also investigate therapeutic strategies to alter this mitochondria-muscle atro

[Regressive mitochondrial abnormalities in the muscular biopsy of a bicycling champion].

The onset of severe muscle cramps in the course of road racing led a champion to undergo a muscle biopsy. Fuscinophilic deposits were detected around muscle cell fibers together with a few PAS positive deposits. These findings were related to the accumulation of abnormal mitochondria and glycogen granules. The subsequent study of the mitochondrial respiratory chain did not reveal any modification and so ruled out a genuine mitochondrial myopathy. Such mitochondrial modifications are probably related to the onset of muscle cramps, but no metabolic disturbance could be found to account for these

Multiple defects of the respiratory chain including complex II in a family with myopathy and encephalopathy.

We report severe deficiency of complex II of the mitochondrial respiratory chain and low activities of complex I and II in skeletal muscle mitochondria from a young woman with progressive muscle weakness and encephalopathy. Defects of complex II have only very rarely been described and this is the first report of decreased immunoreactive subunits associated with severe deficiency of this enzyme.

Therapeutic potential of MitoQ, a mitochondria-targeted antioxidant, in age-related physiological dysfunction.

Aging is an inevitable and progressive physiological process marked by a decline in cellular function, accumulation of somatic mutations, decreased ability to maintain homeostasis, and increased tissue and organ dysfunction. These changes contribute to the onset and progression of various age-related diseases. Accumulation of reactive oxygen species (ROS) has been identified as a key mediator of the aging process. Mitochondria, the primary producers of ROS, accumulate defects with aging, leading to increased mitochondrial oxidative stress. These defective mitochondria are both a source and a t

Mutant mitochondrial thymidine kinase in mitochondrial DNA depletion myopathy.

The mitochondrial deoxyribonucleotide (dNTP) pool is separated from the cytosolic pool because the mitochondria inner membrane is impermeable to charged molecules. The mitochondrial pool is maintained by either import of cytosolic dNTPs through dedicated transporters or by salvaging deoxynucleosides within the mitochondria; apparently, enzymes of the de novo dNTP synthesis pathway are not present in the mitochondria. In non-replicating cells, where cytosolic dNTP synthesis is down-regulated, mtDNA synthesis depends solely on the mitochondrial salvage pathway enzymes, the deoxyribonucleosides k

Effect of hydrogen peroxide on rabbit urinary bladder citrate synthase activity in the presence and absence of a grape suspension.

PURPOSE: The etiology of obstructive bladder dysfunction includes free radical damage to mitochondria. Feeding rabbits a standardized grape suspension protects the ability of the bladder to contract and empty in part by preventing mitochondrial damage, thus maintaining smooth muscle and mucosal metabolism. The objective of the current study is to determine the direct effect of this grape suspension on the response of mitochondria to the oxidative effects of hydrogen peroxide. MATERIALS AND METHODS: Six male rabbits were anesthetized with sodium pentobarbital and the bladders excised. Four ful

Augmenter of liver regeneration, a protective factor against ROS-induced oxidative damage in muscle tissue of mitochondrial myopathy affected patients.

Mitochondria-related myopathies (MM) are a group of different diseases defined by a varying degree of dysfunctions of the mitochondrial respiratory chain which leads to reactive oxygen species (ROS) generation followed by oxidative stress and cellular damage. In mitochondrial myopathy muscle tissue an overexpression of antioxidant enzymes has been documented probably as an attempt to counteract the free radical generation. We previously documented, in human non-pathological muscle fibres, the expression of the augmenter of liver regeneration (ALR), a sulfhydryl oxidase enzyme, whose presence i

Cardiomyopathy and short stature associated with mitochondrial and/or lipid storage myopathy of skeletal muscle.

The patient described in this report was an 11-year-old girl with a negligible heart murmur. The length was below 10th percentile. There appeared to be a cardiomyopathy with, at that moment, no signs of obstruction. There were no evident clinical symptoms of myopathy of skeletal muscle. However, electromyography was myopathic. Biochemical studies revealed no abnormalities. In muscle biopsy sections, the Sudan staining reactions revealed the presence of large amounts of sudanophilic droplets, predominantly in type I fibers. Electronmicroscopy demonstrated lipid excess and accumulations of enlar

Differences in the Mitochondrial and Lipid Droplet Morphology in Female Office Workers With Trapezius Myalgia, Compared With Healthy Controls: A Muscle Biopsy Study.

OBJECTIVE: Trapezius myalgia or, more specifically, myofascial dysfunction of the upper trapezius mainly affects women performing jobs requiring prolonged low level activation of the muscle. This continuous low muscle load can be accompanied by a shift to a more anaerobic energy metabolism, causing pain. The aim of the study was to investigate whether morphological signs of an impaired aerobic metabolism are present in female office workers with trapezius myalgia. DESIGN: Muscle biopsy analysis, using electron and light microscopy, was performed to compare mitochondrial and fat droplet morpho

Mitochondrial Ultrastructural Defects in NDUFS3-Related Disorder.

Complex I, the largest multisubunit enzyme complex of the respiratory chain, has a vital role in the energy production of the cell, and the clinical spectrum of complex I deficiency varies from severe lactic acidosis in infants to muscle weakness in adults. Pathogenic variants of NDUFS3 (constitutes the catalytic core of the complex I) have been reported in a small number of patients with variable phenotypes. We describe a girl with a history of infantile-onset nonepileptic myoclonus, who developed myopathy at the age of 2 years. Next-generation sequencing revealed compound heterozygous for tw

Hypermetabolism and hypercatabolism of skeletal muscle accompany mitochondrial stress following severe burn trauma.

Burn trauma results in prolonged hypermetabolism and skeletal muscle wasting. How hypermetabolism contributes to muscle wasting in burn patients remains unknown. We hypothesized that oxidative stress, cytosolic protein degradation, and mitochondrial stress as a result of hypermetabolism contribute to muscle cachexia postburn. Patients (n = 14) with burns covering >30% of their total body surface area were studied. Controls (n = 13) were young healthy adults. We found that burn patients were profoundly hypermetabolic at both the skeletal muscle and systemic levels, indicating increased oxygen c

Clinical and pathological heterogeneity of a congenital disorder of glycosylation manifesting as a myasthenic/myopathic syndrome.

Congenital disorders of glycosylation are often associated with muscle weakness in apparent isolation or as part of a multi-systemic disorder. We report here the clinical and pathological features resulting from a homozygous mutation of ALG2 in an extended family. Phenotypic heterogeneity is observed among the small cohort of patients reported to date and is highlighted by our study. Linkage analysis, homozygozity mapping and whole exome sequencing followed clinical and pathological characterization of patients who presented with a congenital limb girdle pattern of weakness with no ocular or b
