Mitochondrial myopathy simulating spinal muscular atrophy.

A patient with a severe progressive neuromuscular disorder resembling spinal muscular atrophy is reported. The initial muscle biopsy was consistent with a denervating process. DNA analysis did not reveal deletions in exons 7 and 8 of the survival motor neuron gene. Histology, histochemistry, and biochemistry of a second muscle biopsy suggested mitochondrial myopathy accompanying the denervating features. Immunohistochemistry using anti-DNA antibodies revealed only nuclear staining in skeletal muscle, suggesting mitochondrial DNA depletion. In patients with clinical features of spinal muscular

Modification of the atrophic effects of tenotomy on mouse soleus muscles by various hind limb nerve lesions and different levels of voluntary motor activity.

The response of mouse soleus muscles to transection of the proximal tendon was examined in a variety of circumstances which might be expected to modify the amount of activation of soleus motoneurons. (i) Denervation of ankle flexors antagonistic to the soleus produced slight atrophy of intact muscles but protected soleus to some extent from the effects of concurrent tenotomy. The onset of changes in tenotomized fibers was delayed several days. (ii) Tenotomy of the soleus soon after reinnervation of the muscle fibers (following tibial nerve transection) resulted in little loss of mass and no de

Pharmacological reduction of lipid hydroperoxides as a potential modulator of sarcopenia.

We previously reported that elevated expression of phospholipid hydroperoxide glutathione peroxidase 4, an enzyme that regulates membrane lipid hydroperoxides, can mitigate sarcopenia in mice. However, it is still unknown whether a pharmacological intervention designed to modulate lipid hydroperoxides might be an effective strategy to reduce sarcopenia in aged mice. Here we asked whether a newly developed compound, CMD-35647 (CMD), can reduce muscle atrophy induced by sciatic nerve transection. We treated mice daily with vehicle or CMD (15 mg/kg, i.p. injection) starting 1 day prior to denerva

Myopathological findings in interstitial myositis in type II polyendocrine autoimmune syndrome (Schmidt's syndrome).

We report on a patient with an interstitial myositis in type II polyendocrine autoimmune disease (Schmidt's syndrome) showing an immunohistochemical demonstrated perifascicular expression of Leu 19 indicating perifascicular denervation, which could not be detected with classical histological methods. In neurogenic muscular atrophy, idiopathic polymyositis and normal muscle we could not find such an exclusive perifascicular expression of the Leu 19 antigen. We conclude that perifascicular denervation is a characteristic feature in IM and thus might have diagnostic value. Furthermore the interst

[Experimental study on delayed reinnervation of vocal cord adductors].

The adductor branch was anastomosed with the main branch of ansa cervicalis to reinnervate the vocal cord adductors in six dogs at the time of denervation and after varying intervals of chronic denervation. Some recovery of adduction was noted within ten-month interval. Laryngeal electromyography, tension tests and histologic studies of the adductor muscles were correlated with the recovery of adduction. Although adduction didn't recover after twelve-month interval, striations of the adductor muscles were normal, only slight fibrosis was observed.

Denervation and beta-adrenoreceptors of the cat nictitating membrane.

1. Dose-response curves to the relaxant effects of isoprenaline, salbutamol, adrenaline and noradrenaline were determined on the partially contracted smooth muscles of the cat nictitating membrane following alpha-adrenoreceptor blockade in order to test the hypothesis of a causal relationship between the inhibition of neuronal uptake and denervation supersensitivity. 2. Denervation did not produce supersensitivity of the nictitating membrane to the beta-adrenoreceptor-mediated relaxant effects of any of the four agents studied. 3. The sensitivity of the inferior muscle was greater than that of

Aging Accelerates Degradation of Human Neuromuscular Junction Following Peripheral Nerve Injury.

INTRODUCTION/AIMS: Clinical observations indicate differences between young versus older adults in outcomes of peripheral nerve injury (PNI). Here, we present an analysis of motor endplates (MEPs) from normal and denervated deltoid muscle biopsies from young adult (≤ 60 years) and older (> 60 years) patients. METHODS: Under an IRB-approved prospective cohort study protocol, 24 patients who underwent open surgery for either shoulder pathology or deltoid dysfunction secondary to axillary nerve injury, confirmed with electrodiagnostic studies, were evaluated. Deltoid muscle biopsies were procure

XLOC_015548 Mitigates Skeletal Muscle Atrophy via the Gadd45g/MEK/ERK Pathway and Redox Regulation.

BACKGROUND: Skeletal muscle atrophy is a common musculoskeletal disorder that significantly reduces patient quality of life. Long non-coding RNA (lncRNA) XLOC_015548 has been identified as a pivotal regulator of C2C12 myoblast proliferation and differentiation. However, its role in mitigating denervation-induced muscle atrophy and the underlying mechanisms remain unclear. METHODS: We employed lentiviral-mediated stable expression of XLOC_015548 in C2C12 myoblasts and skeletal muscle-specific XLOC_015548-edited mouse models to investigate the function of this lncRNA. Muscle atrophy models were

[Replacement continent bladder in dogs by opening an antro-pyloric graft onto the abdominal wall (seven cases between 1980 and 1983)].

To determine the usefulness of antral bladder controlled by the pyloric sphincter denervated for urological applications, bladder replacement was performed in 17 dogs using the antral segment with its own sphincter, initially with the pylorus supplied by the left gastro-epiploic artery, and later by the right gastro-epiploic artery. The smooth pyloric muscle was denervated to obtain permanent spasm and pulled through the striated anterior abdominal muscle. Both ureters were anastomosed by a Cohen's procedure. The best results were obtained with a gastric pouch supplied by the right gastro-epip

A neurogenic element to urinary genuine stress incontinence.

The motor unit fibre density in the external anal sphincter has been measured in 35 women with genuine stress incontinence and in 14 continent women. The mean (SD) fibre density in the normal women was 1.5 (0.15) and in the incontinent women it was 1.96 (0.34) (P less than 0.002). These data provide evidence of peripheral denervation of the external anal sphincter and imply synchronous denervation of the pelvic floor muscles. It is suggested that denervation is an aetiological factor in genuine stress incontinence.

Oxidative stress in skeletal muscle stimulates early expression of Rad in a mouse model of amyotrophic lateral sclerosis.

Motor neuron degeneration and progressive muscle atrophy characterize amyotrophic lateral sclerosis (ALS) in humans and related mutant superoxide dismutase-1 (SOD1) transgenic mice. Our previous microarray studies on ALS muscle revealed strong up-regulation of Ras-related associated with diabetes (Rad), an inhibitor of voltage-gated calcium channels. The mechanisms controlling Rad expression in disease are unknown. We analyzed Rad expression in skeletal muscle from ALS patients and animal models and investigated whether it is regulated by oxidative stress. In mutant SOD1 mice, Rad up-regulatio

"Popeye muscle" morphology in OBPI elbow flexion contracture.

BACKGROUND: The pathophysiology of elbow flexion contracture (EFC) in obstetrical brachial plexus injury (OBPI) is not established. In basic science models, neonatal denervation leads to impaired muscle growth. In clinical studies, diminished growth is correlated with extent of denervation, and improved with surgical repair. In EFC, the biceps are clinically short and round vs the contralateral size, termed the "Popeye muscle". The objective of this study was to determine if the biceps morphology (muscle belly and tendon length) in arms with EFC secondary to OBPI is different vs the contralate

Central activation deficits contribute to post stroke lingual weakness in a rat model.

Lingual weakness frequently occurs after stroke and is associated with deficits in speaking and swallowing. Chronic weakness after stroke has been attributed to both impaired central activation of target muscles and reduced force-generating capacity within muscles. How these factors contribute to lingual weakness is not known. We hypothesized that lingual weakness due to middle cerebral artery occlusion (MCAO) would manifest as reduced muscle force capacity and reduced muscle activation. Rats were randomized into MCAO or sham surgery groups. Maximum volitional tongue forces were quantified 8 w

Pathological features of reinnervated skeletal muscles after crush injury of the sciatic nerve in ob/ob mice.

INTRODUCTION/AIMS: Obesity is a factor contributing to suboptimal improvement of motor function in peripheral nerve disorders. In this study we aimed to evaluate the skeletal muscles during denervation and reinnervation after nerve crush injury in leptin-deficient (ob/ob) mice. METHODS: Experiments were performed on the skeletal muscles of the hindlimbs in 20 male ob/ob mice and controls. Characteristics of the gastrocnemius muscles were evaluated by histological analysis, immunohistological analysis, and Sircol-collagen assay after measurement of body weight and wet weight of the skeletal mu

Rabbit supraspinatus motor endplates are unaffected by a rotator cuff tear.

Rotator cuff tears are a major cause of morbidity. Following rotator cuff tears, muscle atrophy and fatty infiltration begin in the tissue, limiting repair potential and leading to a higher re-tear rate and a worse functional outcome. We evaluated whether fatty degeneration resulting from a complete supraspinatus tear with retraction is associated with an injury to the suprascapular nerve. Four skeletally mature New Zealand white rabbits were randomized to receive an index procedure on either their right or left shoulder with the opposite shoulder serving as a control. At the index procedure,

Up-regulation of PKA RI alpha subunit mRNA in rat skeletal muscle after nerve injury.

Localization of all subunits of cAMP dependent protein kinase (PKA) mRNAs and their changes of expression after denervation were examined in rat tongue skeletal muscle by in situ hybridization histochemistry. Among all PKA subunits only RI alpha subunit mRNA was detected in the skeletal muscle, whereas positive signal of all subunits mRNA were observed in some haematocytes or lymphocytes within the tongue tissue. The RI alpha mRNA was distributed in a restricted area near the endplate. The mRNA level was substantially induced by the hypoglossal nerve transection, suggesting that the up-regulat

Respiratory chain deficiency in aged spinal motor neurons.

Sarcopenia, muscle wasting, and strength decline with age, is an important cause of loss of mobility in the elderly individuals. The underlying mechanisms are uncertain but likely to involve defects of motor nerve, neuromuscular junction, and muscle. Loss of motor neurons with age and subsequent denervation of skeletal muscle has been recognized as one of the contributing factors. This study investigated aspects of mitochondrial biology in spinal motor neurons from elderly subjects. We found that protein components of complex I of mitochondrial respiratory chain were reduced or absent in a pro

Vagal afferents essential for abdominal muscle activity during lung inflation in cats.

Maintained inflation of the lung evokes abdominal muscle activity in anesthetized cats only if the vagus nerves are intact, indicating the importance of vagal receptors. The location of these receptors was determined in 14 anesthetized cats by comparing prevagotomy inflation responses of the abdominal muscles and diaphragm to the responses obtained after section of the thoracic vagi at one of three different levels. The abdominal muscle and diaphragm responses to maintained lung inflation persisted following vagotomy below the roots of the lung or denervation of the heart and great vessels. De

BACKGROUND: Synkinetic dysfunction of the Depressor Anguli Oris (DAO) and Mentalis muscle (MM) in conjunction with a weak Depressor Labii Inferiors (DLI) produces a characteristic deformity in the perioral region. Chemodenervation, myectomy and facial nerve neurectomy have been utilized to treat this imbalance with varying degrees of success. This study explores the value of highly selective Mentalis denervation combined with transfer of the hypertonic DAO to the hypotonic DLI for improving facial symmetry. METHODS: A retrospective review of ten patients treated with a DAO-DLI transfer and me
