[Inflammatory myofibroblastic tumour of the tonsil: case report and literature review].

Inflammatory myofibroblastic tumour (inflammatory pseudotumor) is an idiopathic lesion, rare in the head and neck, of unknown aetiology. It is primarily a soft tissue, lung and orbital condition. In the world literature, only two cases with tonsillar disease have been found. We report a case of a 10-year-old girl admitted to our hospital with clinical complaints of pain in the neck region, cough with vomiting, dyspnoea and dyslexia. Clinical examination revealed halitosis and a neoformation dependent on the left tonsil. Bilateral tonsillectomy was performed.

[Inflammatory myofibroblastic tumor of the prostate: a case report and review of the literature].

OBJECTIVE: To discuss the diagnosis and treatment of inflammatory myofibroblastic tumor (IMT) of the prostate. METHODS: We retrospectively analyzed the clinical data of a case of IMT of the prostate and reviewed relevant literature. The patient was a 62-year-old man, who was twice referred to the local hospital for recurrence of dysuria. He was diagnosed as with benign prostatic hyperplasia (BPH) and twice underwent transurethral resection of the prostate (TURP). But frequent recurrence of dysuria followed postoperatively, for which he came to our hospital and received another TURP and transu

Inflammatory myofibroblastic tumor of the epididymis: case report and review of the literature.

The inflammatory myofibroblastic tumor (IMT) is a rare benign tumor, known by several names and found in several locations throughout the body, including the lungs and genitourinary system (mostly the bladder). We report a case of a paratesticular IMT in a 17-year-old boy. Although the initial clinical diagnosis was rhabdomyosarcoma, the histopathologic examination revealed it to be an IMT. The preoperative diagnosis of a scrotal mass should differentiate between benign and malignant tumors. A definitive diagnosis of a scrotal mass should be established by surgical exploration and resection. I

Retroperitoneal Inflammatory Myofibroblastic Tumor: Case Report and Immunohistochemistry Study.

Inflammatory myofibroblastic tumors (IMT) are a rare clinicopathological entity of yet unknown etiology and those located retroperitoneally are even rarer. Clinical outcome is unpredictable and complete surgical resection of the tumor remains the principal treatment. We report the case of a 41-year old man presented with abdominal pain. An abdominal magnetic resonance imaging scan revealed a retroperitoneal tumor located between the pancreas, stomach small curvature and big vessels. A laparotomy with biopsy was performed because the tumor was not amenable to surgical resection. Histopathologic

MeCP2 controls an epigenetic pathway that promotes myofibroblast transdifferentiation and fibrosis.

BACKGROUND & AIMS: Myofibroblast transdifferentiation generates hepatic myofibroblasts, which promote liver fibrogenesis. The peroxisome proliferator-activated receptor gamma (PPARgamma) is a negative regulator of this process. We investigated epigenetic regulation of PPARgamma and myofibroblast transdifferentiation. METHODS: Chromatin immunoprecipitation (ChIP) assays assessed the binding of methyl-CpG binding protein 2 (MeCP2) to PPARgamma and chromatin modifications that silence this gene. MeCP2(-/y) mice and an inhibitor (DZNep) of the epigenetic regulatory protein EZH2 were used in the c…

Inflammatory myofibroblastic tumor of epididymis: a case report and review of literature.

BACKGROUND: Epididymal inflammatory myofibroblastic tumor, also known by various other synonyms is a rare benign disease. Only eight cases have been reported to date. The most common presentation is a scrotal mass of variable duration. For a scrotal mass it is difficult to distinguish a benign or malignant etiology, in addition to the origin whether from testis or epididymis. As a result the definitive diagnosis can only be established by surgical exploration. CASE PRESENTATION: We report the ninth case of epididymal IMT who based on clinical and radiological findings underwent radical orchid

Homocysteine enhances cell proliferation in hepatic myofibroblastic stellate cells.

Homocysteine is an intermediate in sulfur amino acid metabolism, which takes place mainly in the liver. Recent studies have shown that hyperhomocysteinemia in patients and murine models develop hepatic fibrosis. To define mechanisms underlying homocysteine-induced hepatic fibrosis, the effect of homocysteine on hepatic stellate cell (HSC) proliferation was examined. In the present study, homocysteine promoted proliferation in myofibroblastic HSCs. Homocysteine elicited a transient formation of reactive oxygen species (ROS). The initial ROS activated extracellular signal-regulated kinase and p3

Acute abdomen caused by a perforated inflammatory myofibroblastic tumor of the jejunum.

Inflammatory myofibroblastic tumors are rare pseudosarcomatous tumors found in virtually all anatomic sites. Our case report describes an elderly female patient with an inflammatory myofibroblastic tumor in the proximal jejunum, accidentally discovered at laparotomy for an acute abdomen. The localization in the jejunum is a very rare finding, and perforation has not been described before.

Pseudosarcomatous myofibroblastic proliferations of the bladder: a clinicopathologic study of 42 cases.

Inflammatory pseudotumor or pseudosarcomatous fibromyxoid tumor and postoperative spindle cell nodule of the bladder are unusual lesions of uncertain pathogenesis which share overlapping, if not identical, histologic features. We present our experience with 42 cases, the largest series to date, to study the etio-pathogenesis, histologic features, biologic behavior and relationship to "inflammatory myofibroblastic tumor" of childhood. Patients ranged in age from 7 to 77 years (mean 47 y) and males predominated (3.2:1). Most patients presented with hematuria (31/42). Common associations were smo

[Intraabdominal sclerosing panniculitis--myofibroblast proliferation that can mimic malignancy].

Intraabdominal sclerosing panniculitis is a fibroinflammatory lesion of the intraabdominal fat tissue of unknown origin. The authors report 4 secondary cases, that were associated with other kind of intestinal pathology. The cases had different clinical manifestation (mesenterial sclerosis leading to bowel obstruction, lesion simulating transmural spread or peritoneal metastasis of colorectal carcinoma, and chance finding associated with ulcerative colitis). They review the literature and summarize the features of the reactive process characterized by a spindle cell proliferation, fibrosis (sc

Lysyl oxidase cDNA of myofibroblast from mouse fibrotic liver.

In order to study the regulation of lysyl oxydase (LO) in fibrosis, mRNAs were extracted from an enriched population of myofibroblasts (MF) isolated from liver of schistosomiasis infected mouse. Four mRNAs (5.5kb, 4.5kb, 2.4kb and 2.0kb) hybridizing with a LO cDNA probe were transcribed in fibrotic liver, but only the two largest mRNAs were found in MF. A cDNA library was constructed, allowing the cloning of twenty four cDNAs. The largest clone of 4689bp should correspond to the 5.5kb mRNA. Its sequence was essentially similar to the NIH-3T3 fibroblasts LO-ras recision gene (rrg4) cDNA, with t…

A model for Crohn's disease post-ileal resection fibrosis development using human ileal enteroids and myofibroblasts.

BACKGROUND: Fibrosis and inflammation frequently emerge soon after ileal resection for strictures in Crohn's disease (CD). OBJECTIVE: This study examined the profibrotic and proinflammatory effects of conditioned media from TNF and IFN-γ (T+I)-exposed healthy and inactive CD ileal enteroids (CDiE) on subepithelial myofibroblasts (SEMF), modeling early post-ileal resection conditions in CD. METHODS: Ileal enteroids from healthy subjects (HE) and inactive CD patients (CDiE) were exposed to T+I for 6 hours, followed by an 18-hour conditioning period. Conditioned media were collected and applied…

Antibody-conjugated SIS3-loaded PLGA@polydopamine nanoparticles alleviate cardiac fibrosis by modulating Smad3 signaling in myofibroblasts.

BACKGROUND: Pathological cardiac fibrosis arising from acute/chronic myocardial injury drives ventricular remodeling, functional impairment, and elevated mortality through mechanisms lacking effective therapies. Central to fibrogenesis, myofibroblast activation via TGFβ1-Smad3 signaling necessitates targeted therapeutic strategies. METHODS: Anti-fibroblast activation protein (FAP)-functionalized nanoparticles (NPs-SIS3-Ab) were fabricated using PLGA@PDA cores to deliver a Smad3 inhibitor (SIS3). In vitro targeting was assessed through confocal microscopy in TGFβ1-stimulated myofibroblasts. Th…

Branched-chain amino acid transaminase 1-mediated pathway promotes proline-dependent collagen production in cardiac myofibroblasts.

Myofibroblasts are the cells responsible for collagen production, leading to tissue fibrosis. Because 20.5% of the total amino acids in collagen are proline, myofibroblasts must acquire a well-developed proline-producing mechanism during their differentiation. However, the detailed mechanism for myofibroblasts to acquire and keep the developed proline biosynthesis machinery remains obscure. Here, we show that branched-chain amino acid (BCAA) transaminase 1 (Bcat1) was upregulated in a substantial subset of Periostin (Postn)-expressing protomyofibroblast-like fibroblasts, which are transitional…

Emerging Alternaria Toxins Drive LX-2 Cells Transdifferentiation into Myofibroblasts for Liver Fibrosis and CotA Detoxification.

Alternariol (AOH), alternariol monomethyl ether (AME), and tenuazonic acid (TeA) are three major emerging Alternaria toxins in food. In this lncRNA-mRNA omics-based study, we first decrypt the blueprint: how emerging Alternaria toxins induce hepatic stellate cell LX-2 transdifferentiation for liver fibrosis. AOH and AME caused the fibrotic marker α-smooth muscle actin, extracellular matrix collagen expression, and cell contraction, while TeA had no significant effect. Mechanistically, AOH, AME, and AAT (their combination) activated the NF-κB pathway and induced ferroptosis and AMPK/AKT/m-TOR-r

The role of NLRP3-PGE2 axis in mediating interactions between bovine endometrial myofibroblasts and M1 macrophages.

The bovine endometrium undergoes dynamic structural and functional changes during the estrous cycle, driven by coordinated interactions between stromal and immune cells. Among the endometrial stromal populations, (myo)fibroblasts and macrophages play key roles in tissue remodeling, inflammation, and repair. However, their direct cellular communication and underlying molecular mechanisms are not fully understood. In this study, we investigated the bidirectional crosstalk between bovine endometrial myofibroblasts and M1 macrophages. Treatment of endometrial myofibroblasts with M1 macrophage-cond…

A novel senolytic drug for pulmonary fibrosis: BTSA1 targets apoptosis of senescent myofibroblasts by activating BAX.

Idiopathic pulmonary fibrosis is a progressive and age-related disease that results from impaired lung repair following injury. Targeting senescent myofibroblasts with senolytic drugs attenuates pulmonary fibrosis, revealing a detrimental role of these cells in pulmonary fibrosis. The mechanisms underlying the occurrence and persistence of senescent myofibroblasts in fibrotic lung tissue require further clarification. In this study, we demonstrated that senescent myofibroblasts are resistant to apoptosis by upregulating the proapoptotic protein BAX and antiapoptotic protein BCL-2 and BCL-XL, l…
