The role of GABAAR phosphorylation in the construction of inhibitory synapses and the efficacy of neuronal inhibition.

GABA(A)Rs [GABA (gamma-aminobutyric acid) type-A receptors] are heteropentameric chloride-selective ligand-gated ion channels that mediate fast inhibition in the brain and are key therapeutic targets for benzodiazepines, barbiturates, neurosteroids and general anaesthetics. In the brain, most of the benzodiazepine-sensitive synaptic receptor subtypes are assembled from alpha(1-3), beta(1-3) and gamma(2) subunits. Although it is evident that the pharmacological manipulation of GABA(A)R function can have profound effects on behaviour, the endogenous mechanisms that neurons use to promote sustain

How does DBS work?

Deep brain stimulation (DBS) produces striking effects in patients with various disorders including Parkinson's disease and dystonia, yet its precise mechanism of action is not clear. Because the clinical benefits of lesioning target structures such as the thalamus, globus pallidus and subthalamic nucleus appear to be similar to those achieved by chronic application of stimulation at these structures, it has been surmized that deep brain stimulation produces a functional inactivation or block of the target. This simplistic proposal has supporting and detracting evidence. In the present work we

Long-lasting depression of motor-evoked potentials to transcranial magnetic stimulation following exercise.

We used transcranial magnetic stimulation to study the modulation of motor cortex excitability after rapid repetitive movements. Eleven healthy subjects aged 24-32 years were evaluated. Serial motor-evoked potential (MEP) recordings were performed from the right thenar eminence every 5 min for a period of 20 min at rest and for a period of 35 min after repetitive abduction-adduction of the thumb at maximal frequency for 1 min. All subjects presented distinct changes in MEP amplitude after exercise with an approximately 55% mean maximal decrease compared with basal conditions and complete recov

Differential modulation of chemical and electrical components of mixed synapses in the lobster stomatogastric ganglion.

1. Two pairs of neurons in the pyloric network of the spiny lobster, Panulirus interruptus, communicate through mixed graded chemical and rectifying electrical synapses. The anterior burster (AB) chemically inhibits and is electrically coupled to the ventricular dilator (VD); the lateral pyloric (LP) and pyloric (PY) neurons show reciprocal chemical inhibition and electrical coupling. We examined the effects of dopamine (DA), serotonin (5HT) and octopamine (Oct) on these mixed synapses to determine the plasticity possible with opposing modes of synaptic interaction. 2. Dopamine increased net i

Actions of 5-hydroxytryptamine on myenteric neurons in guinea pig gastric antrum.

Intracellular recording methods were used to study the actions of 5-hydroxytryptamine (5-HT) on 257 myenteric neurons in the guinea pig gastric antrum. Application of 5-HT caused three types of postsynaptic responses. A fast-activating depolarizing response was accompanied by a decreased input resistance and desensitized quickly to repeated applications. It was mediated by a 5-HT3 receptor. A slowly activating depolarization, accompanied by an increase in the input resistance and enhancement of the excitability, was mainly observed in after hyperpolarizing/type 2 neurons. It was suppressed by

Barbiturate-enhanced paired-pulse depression in neonatal rats.

Paired-pulse depression of the population spike in the hippocampal formation of barbiturate anaesthetized mature rats is increased compared to that in urethane unanaesthetized rats. This barbiturate-induced increase in depression is greatly enhanced in immature animals, which are also known to be behaviorally more sensitive to barbiturates. These results suggest an increased sensitivity of the recurrent inhibitory (possible GABAergic) system to barbiturates in the immature central nervous system.

Opioid antagonists and spinal reflexes in the anaesthetized cat.

In barbiturate-anaesthetized cats, intravenous naloxone (0.025-0.10 mg/kg) increased the amplitude of monosynaptic reflexes produced by electrical stimulation of the nerves to the biceps-semitendinosus and gastrocnemius muscles and the complex reflexes to electrical stimulation of myelinated afferents of the sural and tibial nerves and reflexes to electrical stimulation of unmyelinated primary afferents of the tibial nerve. Increases in reflexes were also produced by the (-)- but not the (+)-isomer of the opiate antagonist N- furylmethylnormetazocine (both isomers being given in the dose range

Inhibition in carotid body chemoreceptors mediated by D-2 dopaminoceptors: antagonism by benzamides.

Inhibition of chemosensory nerve impulses in the cat is evoked by dopamine (DA) applied to carotid body chemoreceptors. Pharmacological characterization of the dopaminoceptors involved in this action was determined through their blockade with benzamides, selective antagonists of D-2 receptors. Both metoclopramide and sulpiride were effective blockers of DA-induced chemosensory inhibition. Furthermore, both drugs induced an immediate increase in the frequency of carotid nerve chemosensory impulses, suggesting the presence of previous tonic inhibition of chemoreceptor discharges by endogenous DA

Inhibitory postsynaptic currents at Aplysia cholinergic synapses: effects of permeant anions and depressant drugs.

Inhibitory postsynaptic potentials (i.p.s.ps) and, under voltage-clamp conditions, inhibitory postsynaptic currents (i.p.s.cs) were recorded in neurons in buccal ganglia of Aplysia juliana. The decay of i.p.s.cs was exponential with a single time constant, tau, which decreased with membrane depolarization. In external solutions containing iodide or bromide ions instead of chloride ions, tau varied according to the sequence tau (I) greater than tau (Br) greater than tau (Cl), and the voltage sensitivity of tau was altered. In iodide solution, the voltage sensitivity of tau was reversed. Further

Primary afferent depolarization in the in vitro frog olfactory bulb.

1. Experiments on the frog olfactory bulb have been performed in vitro in order to determine whether primary afferent transmission is modified by presynaptic inhibition.2. Stimulation of the olfactory nerve resulted in a prolonged depolarization of the olfactory nerve as recorded across a sucrose gap. Unstimulated olfactory nerve fibres adjacent to the stimulated fibres were also depolarized.3. An excitability increase of the olfactory nerve terminals was found that lasted the entire duration of the olfactory nerve depolarization, indicating that the terminals themselves were depolarized. Both

Virus-induced electrotonic coupling: hypothesis on the mechanism of periodic EEG discharges in Creutzfeldt-Jakob disease.

Experimental evidence and computer modeling indicate that periodic synchronous cellular depolarizing bursts (interictal spikes) arise when the balance recurrent inhibition and local excitatory coupling is altered. Such a mechanism may explain the generalized periodic sharp waves that characterize the electroencephalogram of many patients with Creutzfeldt-Jakob disease. In Creutzfeldt-Jakob disease, fusions of neuronal processes, particularly dendrites, may lead to abnormal electrotonic coupling between cells, providing the basis for powerful excitatory interaction whereby large neuronal aggreg

Frequency effects of temporary N1 depression following acoustic overload.

The effect of acoustic overload on the N1 potential at different frequences was determined. The conditions of overstimulation were such as to produce temporary effects. The results of the study were: (1) the N1 potential was substantially depressed by acoustic overloads that did not alter the alternating current (AC) cochlear potential; (2) the maximum N1 depression was at one-half octave above the overstimulation frequency; (3) there was no N1 depression at frequencies below the frequency of overstimulation; and (4) the N1 depression recovered within 24 hours.

Mechanisms of excitation and inhibition in the nigrostriatal system.

The extracellular responses of neurones in the neostriatum following single pulse stimulation of the substantia nigra were investigated in urethane anaesthetized rats. Low intensity stimulation (less than 10 V) evoked single large amplitude spikes while higher intensities (10-20 V) elicit a high frequency burst of small amplitude spikes or waves. When spontaneous or glutamate-induced large spikes are recorded, nigral stimulation causes their inhibition coincidentally with the development of a burst. If the burst is prevented, the inhibitory response disappears. Both the nigral evoked inhibitio

A systematic analysis of the joint effects of ganglion cells, lagged LGN cells, and intercortical inhibition on spatiotemporal processing and direction selectivity.

Simple cells in the visual cortex process spatial as well as temporal information of the visual stream and enable the perception of motion information. Previous work suggests different mechanisms associated with direction selectivity, such as a temporal offset in thalamocortical input stream through lagged and non-lagged cells of the lateral geniculate nucleus (LGN), or solely from intercortical inhibition, or through a baseline selectivity provided by the thalamocortical connection tuned by intercortical inhibition. While there exists a large corpus of models for spatiotemporal receptive fiel

Reciprocal Inhibitory Glomerular Circuits Contribute to Excitation-Inhibition Balance in the Mouse Olfactory Bulb.

The major inhibitory interneurons in olfactory bulb (OB) glomeruli are periglomerular cells (PGCs) and short axon cells (SACs). PGCs and SACs provide feedforward inhibition to all classes of projection neurons, but inhibition between PGCs and SACs is not well understood. We crossed Cre and GFP transgenic mice and used virally-delivered optogenetic constructs to selectively activate either SACs or GAD65cre-ChR2-positive PGCs while recording from identified GAD65cre-ChR2-positive PGCs or SACs, respectively, to investigate inhibitory interactions between these two interneuron types. We show that

Training based on mirror visual feedback influences transcallosal communication.

Mirror visual feedback (MVF) therapy has been demonstrated to be successful in neurorehabilitation, probably inducing neuroplasticity changes in the primary motor cortex (M1). However, it is not known whether MVF training influences the hemispheric balance between the M1s. This topic is of extreme relevance when MVF training is applied to stroke rehabilitation, as the competitive interaction between the two hemispheres induces abnormal interhemispheric inhibition (IHI) that weakens motor function in stroke patients. In the present study, we evaluated, in a group of healthy subjects, the effect

Effects of electrical stimulation of the striatum on bladder activity in cats.

AIMS: Parkinson's disease (PD) affects the nigrostriatal projections leading to micturition disturbance in most cases. Overactive bladder (OAB) symptoms such as urinary urgency or urgent urinary incontinence are common amongst PD patients. Several urodynamic studies have revealed that detrusor overactivity causes OAB symptoms in PD patients. We assert that striatal dysfunction might contribute to the pathogenesis of detrusor overactivity in PD patients. However, the role of the striatum in bladder contraction remains unclear. METHODS: We generated spontaneous isovolumetric bladder contraction

Layer-specific touch-dependent facilitation and depression in the somatosensory cortex during active whisking.

Brains adapt to new situations by retuning their neurons. The most common form of neuronal adaptation, typically observed with repetitive stimulations of passive sensory organs, is depression (responses gradually decrease until stabilized). We studied cortical adaptation when stimuli are acquired by active movements of the sensory organ. In anesthetized rats, artificial whisking was induced at 5 Hz, and activity of individual neurons in layers 2-5 was recorded during whisking in air (Whisking condition) and whisking against an object (Touch condition). Response strengths were assessed by spike
