Trace elements and C and N isotope composition in two mushroom species from a mine-spill contaminated site.

Fungi play a key role in the functioning of soil in terrestrial ecosystems, and in particular in the remediation of degraded soils. The contribution of fungi to carbon and nutrient cycles, along with their capability to mobilise soil trace elements, is well-known. However, the importance of life history strategy for these functions has not yet been thoroughly studied. This study explored the soil-fungi relationship of two wild edible fungi, the ectomycorrhizal Laccaria laccata and the saprotroph Volvopluteus gloiocephalus. Fruiting bodies and surrounding soils in a mine-spill contaminated area

"Mind the gap"-Assessing methods for aligning age determination and growth rate in multi-molar sequences of dietary isotopic data.

OBJECTIVES: Creating multi-tooth sequences of micro-sampled stable isotope (SI) analytical data can help track 20+ years of individual dietary history. Inferences about individual and population level behavioral patterns require cross-calibration of the timing of dietary changes recorded by each tooth. Dentin sections from contemporaneous tissues (eg, in M1 and M2) reflect dietary signature for the time of growth. Contemporary sections should produce similar values, allowing alignment of temporally overlapping portions of teeth into multi-tooth sequences. Published methods for determining the

1H, 15N and 13C resonance assignments of the C-terminal domain of the P protein of the Nishigahara strain of rabies virus.

The C-terminal domain of the P protein of rabies virus is a multifunctional domain that interacts with both viral and host cell proteins. Here we report the 1H, 13C and 15N chemical shift assignments of this domain from P protein of the Nishigahara strain of rabies virus, a pathogenic laboratory strain well established for studies of virulence functions of rabies virus proteins, including P protein. The data and secondary structure analysis are in good agreement with the reported predominantly helical structure of the same domain from the CVS strain of rabies solved by crystallography. These a

Backbone and side-chain ¹H, ¹³C and ¹⁵N assignments of the PPIase domain of macrophage infectivity potentiator (Mip) protein from Coxiella burnetii.

Coxiella burnetii is an obligate intracellular gram-negative bacterium uniquely evolved to thrive in the inhospitable phagolysosome of macrophage. C. burnetii causes Q fever in humans and animals, which is emerging as a global public health concern. It is highly infectious and designated as a category B biowarfare agent because of its ubiquitous nature, abundant natural reservoirs, high resistance to environmental conditions, ease of transmission and low infectious dose. The lack of knowledge and awareness of C. burnetii leads to under-reporting and under-diagnosing of Q fever cases. Therefore

Preparation of stable isotope-labeled peripheral cannabinoid receptor CB2 by bacterial fermentation.

We developed a bacterial fermentation protocol for production of a stable isotope-labeled cannabinoid receptor CB2 for subsequent structural studies of this protein by nuclear magnetic resonance spectroscopy. The human peripheral cannabinoid receptor was expressed in Escherichia coli as a fusion with maltose binding protein and two affinity tags. The fermentation was performed in defined media comprised of mineral salts, glucose and (15)N(2)-L-tryptophan to afford incorporation of the labeled amino acid into the protein. Medium, growth and expression conditions were optimized so that the ferme

Carbon monoxide and cyanide as intrinsic ligands to iron in the active site of [NiFe]-hydrogenases. NiFe(CN)2CO, Biology's way to activate H2.

Infrared-spectroscopic studies on the [NiFe]-hydrogenase of Chromatium vinosum-enriched in 15N or 13C, as well as chemical analyses, show that this enzyme contains three non-exchangeable, intrinsic, diatomic molecules as ligands to the active site, one carbon monoxide molecule and two cyanide groups. The results form an explanation for the three non-protein ligands to iron detected in the crystal structure of the Desulfovibrio gigas hydrogenase (Volbeda, A., Garcin, E., Piras, C., De Lacey, A. Electron spin-lattice relaxation mechanisms of rapidly-tumbling nitroxide radicals.

Electron spin relaxation times at 295 K were measured at frequencies between 250 MHz and 34 GHz for perdeuterated 2,2,6,6-tetramethyl-4-piperidone-1-oxyl (PDT) in five solvents with viscosities that result in tumbling correlation times, τR, between 4 and 50 ps and for three (14)N/(15)N pairs of nitroxides in water with τR between 9 and 19 ps. To test the impact of structure on relaxation three additional nitroxides with τR between 10 and 26 ps were studied. In this fast tumbling regime T2(-1)~T1(-1) at frequencies up to about 9 GHz. At 34 GHz T2(-1)>T1(-1) due to increased contributions to T2(

You are not what you eat during physiological stress: Isotopic evaluation of human hair.

Variation in δ(13) C and δ(15) N values can be assessed to understand not only diet, but also the influence of physiological factors on an individual. The metabolic balance of an individual can impact isotopic signals in tissues that are forming during the periods of metabolic stress. Fluctuating δ(15) N values are associated with physiological stressors that alter an individual's metabolism such as infection, injury, or pregnancy. This study examines variation in δ(13) C and δ(15) N values along sequentially segmented hair in both modern and archaeological individuals. Subjects with an observ

Out of the North Sea: the Zeeland ridges Neandertal.

In 2001, a portion of human frontal bone was discovered in sediments extracted from the bottom of the North Sea, 15km off the coast of the Netherlands. The extraction zone is located in the so-called Zeeland Ridges area located at 51 degrees 40' northern latitude and 3 degrees 20' eastern longitude. The specimen was dredged up from sediments containing Late Pleistocene faunal remains and Middle Palaeolithic artefacts, including well-finished small handaxes and Levallois flakes. The details of the supraorbital morphology, as well as the quantitative assessment of the shape of the external surfa

Preparation of isotopically labelled recombinant beta-defensin for NMR studies.

beta-Defensins are a family of cationic peptides that contain six invariant cysteine residues that form characteristic disulfide bonds between Cys(1)-Cys(5), Cys(2)-Cys(4) and Cys(3)-Cys(6). They have been shown to act as potent antimicrobial agents and chemokines. Human beta-defensin 2 (HBD2) was first isolated from psoriatic skin lesions and the structure of this peptide has been solved by X-ray crystallography and NMR spectroscopy both of which are consistent with a fold that contains an N-terminal alpha-helix and three antiparallel beta-strands. Here, we report the expression and purificat

Regulating Proton Supply for Selective Photocatalytic NO Conversion toward Pollutant Removal or Ammonia Recovery.

Controlling the conversion of nitric oxide (NO) is critical for both air pollution mitigation and nitrogen resource recovery. Proton-coupled electron transfer (PCET) governs photocatalytic NO reduction, where reaction pathways determine whether nitrogen is removed as inert N2 or converted into value-added NH3. However, precise control over this selectivity remains challenging, largely due to the limited understanding and regulation of proton delivery. Alcohols with different α-C-H bond strengths are demonstrated to provide tunable proton fluxes. The energetics of α-C-H bonds in alcohol proton

CO2 agitation combined with magnetized biochar to alleviate "ammonia inhibited steady-state": Exploring the mechanism by combining metagenomics with macroscopic indicators.

The "ammonia inhibited steady-state" phenomenon is frequently observed in the anaerobic digestion (AD) process of nitrogen-rich substrates. Reconfiguring microbial ecosystems has proven to be an effective strategy for mitigating ammonia inhibition. In the current study, biochars were screened and targeted for modification. CO2 agitation combined with magnetized biochar was used to aid the semi-continuous AD systems with "ammonia inhibited steady-state." The results indicated that coconut shell biochar had the best stimulating effect on AD performance. The content of oxygen-containing functiona

Diammonium phosphate supplementation in low-protein diets improves nitrogen utilization and feed efficiency without impairing carcass traits or meat quality in finishing pigs.

Low-protein (LP) diets improve nitrogen utilization efficiency and mitigate environmental pollution, however, excessive reductions in crude protein (CP) result in total nitrogen (TN) deficiency, thereby compromising growth performance. This study investigated the effects of replacing calcium hydrogen phosphate with diammonium phosphate (DP) in an LP diet on growth performance, carcass traits, meat quality, plasma biochemical parameters, hepatic nitrogen-metabolizing enzyme activities, and amino acid (AA) profiles in finishing pigs under TN restriction. Additionally, 15nitroge (15N)-isotope tra

Nitrogen cycling regulates arsenic mobilization in alluvial-lacustrine aquifer systems: DOM molecular signatures and thermodynamic constraints.

High arsenic (As) groundwater commonly co-occurs with elevated ammonium (NH4+) in aquifers worldwide. However, the underlying nitrogen cycling processes regulating As mobilization in groundwater remain elusive. Here, hydrochemistry, stable isotopes, FT-ICR MS, and thermodynamic modelling were integrated to investigate the NH4+ production pathways and their impacts on As mobility within the alluvial-lacustrine aquifer. Hydrochemical and isotopic data indicated that groundwater NH4+ was primarily produced through OM mineralization and dissimilatory nitrate reduction to ammonium (DNRA), and both

Reciprocal transport of carbon and nitrogen between plants and the hyphosphere microbiome in a temperate grassland.

Despite its relevance for plant nutrient acquisition, the role of the hyphosphere in structuring fungal-bacterial networks and mediating carbon-nitrogen (C-N) exchange under field conditions remains unclear. We therefore examined reciprocal C-N fluxes along the plant-fungi-bacteria continuum and their link to microbial community assembly across grassland soil microhabitats. Sterilized compartmentalized ingrowth cores were exposed to soil in the field for 3 months. Plants were pulse-labeled with 13CO2 and 13C was traced into microbial biomarkers over 7 d. 15N-labeled urea was supplied to a hyph

Unveiling the effect of Fe(III) and sulfate on ammonium oxidation under anaerobic condition: interactions and extracellular electron transfer.

Enhanced ammonium (10.6 - 14.7%) and total inorganic nitrogen (TIN, 4.3 - 18.1%) removal are achieved with individual sulfate (500 mg/L) and ferric (5 mg/L) addition. The promotion of ammonium and TIN were even higher than the total promotion with individual sulfate and ferric addition, when sulfate and ferric are both supplemented probably demonstrate the cooperation between ferric and sulfate during nitrogen removal. The Feammox/Sulfammox, Anammox and Denitrification contributes 9% ∼ 39%, 53% ∼ 87% and 4% ∼ 13% of nitrogen loss, respectively. However, decline of both ammonium (10.5%) and TIN

An inundation threshold regulates nitrate reduction pathways in mangrove sediments.

Estuarine hydrological regimes are increasingly altered by dam operations and extreme climate events, reshaping inundation in intertidal wetlands. However, how inundation regulates nitrate (NO3-) fate remains unclear. We conducted an 18-month, elevation-controlled marsh-organ experiment with Avicennia marina-planted and unvegetated sediments across a 0 %-65 % gradient in mean inundation frequency. We combined 15N isotope tracing and metagenomics to quantify NO3- reduction pathways and associated microbial attributes. In unvegetated sediments, increasing inundation reduced the contribution of d
