Risk is in the air: an intranasal MPTP (1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) rat model of Parkinson's disease.

The presence of smell loss and the early pathological involvement of the olfactory pathways in the early stages of some neurodegenerative disorders are in accord with the tenants of the olfactory vector hypothesis. This hypothesis postulates that some such diseases may be caused or catalyzed by agents that enter the brain via the olfactory mucosa. In this study, rats infused intranasally (i.n.) with a low concentration of the proneurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) subsequently suffered olfactory, cognitive, and motor function impairments conceivably analogous to thos…

Devices for the electrical stimulation of the olfactory system: A review.

The loss of olfactory function has a profound impact on quality of life, affecting not only sensory perception but also memory, emotion, and overall well-being. Despite this, advancements in olfactory prostheses have lagged significantly behind those made for vision and hearing restoration. This review offers a comprehensive analysis of the current state of devices for electrical stimulation of the olfactory system. We begin by providing an overview of the olfactory system's structure and function, emphasizing the neural pathways involved in smell perception. Following this, we explore the key

Immunohistochemical localization of interleukin-2 and its receptor subunits alpha, beta and gamma in the main olfactory bulb of the rat.

Endogenous interleukin-2 (IL-2) was found in the adult rat brain, however, it has not been reported whether this cytokine is present in the olfactory bulb. Immunohistochemical techniques were used to examine the cellular localization of IL-2 and its receptor subunits in the main olfactory bulb of the rat. Strong IL-2 immunoreactivity was localized in glial cells, specifically in the olfactory nerve layer, glomerular layer and external plexiform layer. IL-2 mRNA was detected in the olfactory bulb by RT-PCR. All three IL-2 receptor subunits also showed distinct laminar distributions. The IL-2Ral

α-Synuclein Spread from Olfactory Bulb Causes Hyposmia, Anxiety, and Memory Loss in BAC-SNCA Mice.

BACKGROUND: Patients with Parkinson's disease (PD) show motor symptoms as well as various non-motor symptoms. Postmortem studies of PD have suggested that initial alpha-synuclein (α-Syn) pathology develops independently in the olfactory bulb and lower brainstem, spreading from there stereotypically. However, it remains unclear how these two pathological pathways contribute to the clinicopathological progression of PD. OBJECTIVE: The objective of this study was to examine the clinicopathological contribution of α-Syn spread from the olfactory bulb. METHODS: We conducted pathological and behav

Pyrfume: A window to the world's olfactory data.

Advances in theoretical understanding are frequently unlocked by access to large, diverse experimental datasets. Our understanding of olfactory neuroscience and psychophysics remain years behind the other senses, in part because rich datasets linking olfactory stimuli with their corresponding percepts, behaviors, and neural pathways remain scarce. Here we present a concerted effort to unlock and unify dozens of stimulus-linked olfactory datasets across species and modalities under a unified framework called Pyrfume. We present examples of how researchers might use Pyrfume to conduct novel anal

Primary olfactory axons form ectopic glomeruli in mice lacking p75NTR.

The restricted expression of the low affinity nerve growth factor receptor p75NTR by olfactory ensheathing cells suggests that this molecule is involved in the development of the olfactory nerve pathway. To begin to understand the role of p75NTR, we examined the development of the primary olfactory system in p75NTR(-/-) and wild-type mice. Our results demonstrate that, although p75NTR is not essential for the initial assembly of the olfactory nerve, it plays an important role in the postnatal maturation of the olfactory bulb. In the absence of p75NTR, there is exuberant growth of some primary

Development of olfactory glomeruli: temporal and spatial interactions between olfactory receptor axons and mitral cells in opossums and rats.

Mitral cells are the primary output neurons of the vertebrate olfactory bulb and are major recipients of sensory input from the periphery. The morphogenesis of mitral cell dendrites was followed to elucidate their early spatial and temporal interactions with olfactory receptor neurons and glia during the construction of olfactory glomeruli. Monodelphis domestica, a marsupial born at an extremely immature stage, and rats were examined. Mitral cells were retrogradely labeled by application of the lipophilic dye 1,1' dihexadecyl-3,3,3'3'-tetramethylin-docarbocyanine perchlorate (DiI) to the later

Dopaminergic periglomerular cells in the turtle olfactory bulb.

With the indirect immunofluorescence technique using antisera to three catecholamine synthesizing enzymes, labeled periglomerular cells as well as their intraglomerular processes were observed in the turtle olfactory bulb. These cells could also be recognized in the EPL and the glomerular layer. Unlabeled periglomerular cells were also seen. Thick labeled processes (presumably dendrites) entered the glomerular neuropil, and there formed a dense network, with numerous terminal varicosities. These results support the existence of a unique, homologous dopaminergic subdivision of the periglomerula

Catecholamine innervation of the basal forebrain. III. Olfactory bulb, anterior olfactory nuclei, olfactory tubercle and piriform cortex.

The catecholamine innervation of the olfactory bulb, anterior olfactory nuclei, olfactory tubercle and piriform cortex was studied in the rat using biochemical analysis and fluorescence histochemistry. Biochemical studies demonstrate a moderate norepinephrine (NE) content in all olfactory structures, a high dopamine (DA) content in the olfactory tubercle and a low DA content in the olfactory bulb, anterior olfactory nucleus and piriform cortex. Following locus coeruleus lesions NE content decreases 71% in the olfactory bulb, 82% in the anterior olfactory nucleus, 62% in olfactory tubercle and

Glutamatergic Neurons in the Piriform Cortex Influence the Activity of D1- and D2-Type Receptor-Expressing Olfactory Tubercle Neurons.

Sensory cortices process stimuli in manners essential for perception. Very little is known regarding interactions between olfactory cortices. The piriform "primary" olfactory cortex, especially its anterior division (aPCX), extends dense association fibers into the ventral striatum's olfactory tubercle (OT), yet whether this corticostriatal pathway is capable of shaping OT activity, including odor-evoked activity, is unknown. Further unresolved is the synaptic circuitry and the spatial localization of OT-innervating PCX neurons. Here we build upon standing literature to provide some answers to

Localization of nerve growth factor-like immunoreactivity in rat nervous tissue.

The use of immunofluorescence with affinity-purified antibodies enabled cytological localization of nerve growth factor-like material in the rat. Immunoreactivity was observed along various nerve tracts of the foetal rat brain and spinal cord at day 15 of gestation. Longitudinal pathways in ventral and dorsal spinal cord, ventral lower brain stem, posterior commissure, retroflex fascicle and in the olfactory bulb were all positive. A weaker and more widely spread immunostaining was visible in many areas in the central nervous system. Cranial nerves were strongly immunoreactive. Neuronal perika

TALE-class homeodomain transcription factors, homothorax and extradenticle, control dendritic and axonal targeting of olfactory projection neurons in the Drosophila brain.

Precise neuronal connectivity in the nervous system depends on specific axonal and dendritic targeting of individual neurons. In the Drosophila brain, olfactory projection neurons convey odor information from the antennal lobe to higher order brain centers such as the mushroom body and the lateral horn. Here, we show that Homothorax (Hth), a TALE-class homeodomain transcription factor, is expressed in many of the antennal lobe neurons including projection neurons and local interneurons. In addition, HTH is expressed in the progenitors of the olfactory projection neurons, and the activity of ht

A subpopulation of nervus terminalis neurons projects to the olfactory mucosa in Xenopus laevis.

Biocytin application to the normal or zinc sulfate-treated nasal cavity of Xenopus laevis was used to trace retrogradely neurons associated with the terminal nerve (TN). Immunocytochemistry was conducted to identify the relationship of gonadotropin-releasing hormone-immunoreactive (GnRH-ir) TN neurons with biocytin-labeled neurons. Neurons that accumulated biocytin were located in olfactory nerve fascicles close to the olfactory mucosa lining the caudal, medial, and rostral walls of the principal cavity. GnRH-ir fibers were observed only in the olfactory nerve fascicle projecting to the rostra

Maternal aggression in rats: effects of olfactory bulbectomy, ZnSO4-induced anosmia, and vomeronasal organ removal.

Previous studies from our laboratory indicate that somatosensory inputs to the snout and ventral trunk, but not visual or auditory stimuli, play critical roles in the elicitation and maintenance of maternal aggression by lactating Norway rats toward a strange male intruder. There are conflicting reports on the influence of olfaction on maternal aggression. We explored the possible roles of central or peripheral anosmia on maternal aggression in Long-Evans rats during early lactation. In Experiment 1, responsiveness to both volatile and non-volatile odors was eliminated by bilateral olfactory b

Chronic recording of vomeronasal pump activation in awake behaving hamsters.

The vomeronasal organs, the receptor organs of the accessory olfactory system, are important in chemical communication. Each organ contains receptor neurons sequestered inside a blind-ending tube with a narrow access duct. Large blood vessels surrounding the vomeronasal lumen act as a pump to draw substances into the lumen, under the control of vasomotor fibers in the nasopalatine nerve. Stimulation of the superior cervical sympathetic ganglion or nasopalatine nerve operates the pump (24) but its schedule of activation in awake behaving animals is unknown. Electrodes, implanted inside the vome

Effect of putative pheromones on the electrical activity of the human vomeronasal organ and olfactory epithelium.

The summated receptor potential was recorded from the vomeronasal organ (VNO) and olfactory epithelium (OE) of 49 human subjects of both sexes (18 to 55 years old) using surface non-polarizable silver-silver chloride electrodes. 15-25 pg of human putative pheromones, clove oil and a diluent were administered to the VNO or the OE in 0.3-1 s pulses from a 0.05 mm dia cannula connected to a multichannel delivery system. Local stimulation of the VNO produces negative potentials of 1.8-11.6 mV showing adaptation. Responses are not obtained when the recording electrode is placed in the nasal respira

Selective deficits in the sense of smell caused by chemical modification of the olfactory epithelium.

A chemically selective procedure for covalent modification of Schiff base-forming binding sites in proteins is demonstrated in vitro. In vivo studies show that the same procedure produces a selective anosmia ("odor blindness") when applied to the olfactory epithelia of experimental animals. Surgical experiments confirm that the sense of smell is specifically affected.

Evaluation of post-traumatic anosmia with MRI and chemosensory ERPs.

Magnetic resonance imaging (MRI) and chemosensory event-related potentials (ERPs) are important methods to evaluate olfactory function, but there is lack of study to explore the application of MRI and chemosensory ERPs in the patients with traumatic anosmia. The data of 26 post-traumatic anosmic patients and 21 healthy controls were retrospectively surveyed; olfaction and olfactory pathway of all participants were measured clinically using the T&T olfactometer, the Sniffin' Sticks, chemosensory ERPs and MRI. All patients were anosmic based on complaints and clinical examinations. In five patie
