The effects of cyanate in vitro on red blood cell metabolism and function in sickle cell anemia.

Cyanate, which is in equilibrium with urea, combines with the alpha-amino group of the aminoterminal valine of hemoglobin in an irreversible … tologous plasma. Glycolysis, ATP and 2,3-diphosphoglyceric acid (DPG) stability, autohemolysis, and osmotic fragility were not affected by carbamylation. Potassium loss in carbamylated cells (2.8 mmol/liter) was less than in control cells (9.0 mmol/liter). Pyruvate kinase activity of carbamylated cells was decreased ( approximately 25%) but the activities of other …

Band 3 Cape Town (E90K) causes severe hereditary spherocytosis in combination with band 3 Prague III.

Hereditary spherocytosis (HS) is an inherited haemolytic anaemia, characterized by spheroidal, osmotically fragile red blood cells. This disorder exhibits heterogeneity in terms of both clinical severity and underlying molecular defect. We have studied a South African Cape Coloured individual with severe HS owing to a band 3 deficiency caused by two mutations, occurring in trans, in the band 3 gene: a novel variant that we have designated band 3 Cape Town and a previously described mutation, band 3 Prague III. Analysis of erythrocyte membrane proteins indicated a deficiency of both band 3 and

Ammonia-induced taurine release from cultured rabbit Müller cells is an osmoresistant process mediated by intracellular accumulation of cyclic AMP.

A previous study demonstrated the release of newly loaded radiolabelled taurine (Tau) from cultured rabbit Müller glia not only following typical cell volume-increasing treatments with high (65 mM) potassium ions or hypotonic media, but also with ammonium chloride (further referred to as ammonia), in a dose-dependent manner, at doses ranging from physiological (0.25 mM) to those accompanying hyperammonemic coma (5 mM) (Faff-Michalak et al., Glia 10:114-120, 1994). Stimulation of Tau release by ammonia, but not by 65 mM potassium, was correlated with a dose-dependent increase of intracellular c

Stabilizing effects of some amino acids on membranes of rabbit erythrocytes perturbed by chlorpromazine.

Hemolysis experiments indicated that glycine (20 mM) remarkably suppressed the chlorpromazine (CPZ)-induced osmotic permeability enhancing effect in rabbit erythrocytes. Lysine and aspartic acid also showed a similar protecting effect at the same concentration. Thus, the acid-base properties of amino acids were not a determining factor in stabilizing the cell membranes perturbed by CPZ. These results suggest that amino acids stabilize the membranes of the erythrocytes by inhibiting the osmotic water transfer from the outside to the inside of the cells. The kinetic studies in isotonic suspensio

A novel ANK1 gene mutation associated with hereditary spherocytosis: a case report.

BACKGROUND: Hereditary spherocytosis (HS) is an inherited form of hemolytic anemia resulting from defects in the red blood cell membrane ske … n revealed hepatosplenomegaly. Laboratory investigations demonstrated normocytic anemia, a positive osmotic fragility test, and the presence of spherocytes on peripheral blood smear, which was subsequently confirmed by electron microscopy. Whole-exome sequencing identified a novel heterozygous pathogenic mutation (c.2388 + 2T > A) in the ANK1 gene (NM_000037.4, Int …

[Diagnosis of Congenital Hemolytic Anemias in Adults].

Hereditary hemolytic anemias (HHAs) are a heterogeneous group of genetic disorders characterized by the premature destruction of red blood c … n form in Europe, are primarily identified using assays such as eosin-5-maleimide (EMA) binding and osmotic fragility testing. Enzymopathies, most notably pyruvate kinase and glucose-6-phosphate dehydrogenase deficiencies, necessitate a combination of enzymatic activity assays and confirmatory genetic testing. Advances in molecular diagnostics, especially next-gene …

Production of Feline Universal Erythrocytes with Methoxy Polyethylene Glycol.

Blood group mismatch in veterinary medicine is a significant problem in blood transfusion, sometimes leading to severe transfusion reactions … ne anti-A alloantibodies. Furthermore, the mPEG-treated RBCs did not cause spontaneous hemolysis or osmotic fragility, compared to control RBCs. According to a monocyte monolayer assay, mPEG treatment significantly reduced feline anti-A antibody-mediated phagocystosis of RBCs. These results confirm the potential of using activated mPEG on feline A-type RBC to creat …

Utility of screening tools to differentiate beta thalassemia trait and iron-deficiency anemia - do they serve a purpose in blood donors?

BACKGROUND: The aim of this study was to estimate the prevalence of the beta thalassemia trait (BTT) and differentiate it from iron-deficiency anemia (IDA) among blood donors. METHODS: A total of 1,000 samples from blood donors were subjected to complete hemogram with red blood cell indices. Further, Mentzer index (MI) was calculated for samples with mean corpuscular volume (MCV) below 80 fL and mean corpuscular hemoglobin (MCH) below 27 pg. Samples with Mentzer index <12 were subjected to naked-eye single-tube red cell osmotic fragility test (NESTROFT) followed by hemoglobin electrophoresis …

[Clinical characteristics and genetic analysis of hereditary spherocytosis caused by mutations of ANK1 and SPTB genes].

This study analyzed the clinical features of 5 children with hereditary spherocytosis (HS) and the characteristics of ANK1 and SPTB gene mutations. All 5 children were confirmed with HS by peripheral blood genetic detection. Anemia, jaundice and splenomegaly were observed in all 5 children. Three children had an increase in erythrocyte osmotic fragility. All 5 children had negative results of the Coombs test, glucose 6 phosphate dehydrogenase test, sucrose hemolysis test, acidified-serum hemolysis test and thalassemia gene test. Peripheral blood smear showed an increase in spherocyte count in …

Novel Gardos channel mutations linked to dehydrated hereditary stomatocytosis (xerocytosis).

Dehydrated hereditary stomatocytosis (DHSt) is an autosomal dominant congenital hemolytic anemia with moderate splenomegaly and often compensated hemolysis. Affected red cells are characterized by a nonspecific cation leak of the red cell membrane, reflected in elevated sodium content, decreased potassium content, elevated MCHC and MCV, and decreased osmotic fragility. The majority of symptomatic DHSt cases reported to date have been associated with gain-of-function mutations in the mechanosensitive cation channel gene, PIEZO1. A recent study has identified two families with DHSt associated wi…

Red blood cell processing for cryopreservation: from fresh blood to deglycerolization.

BACKGROUND: Cryostorage of red blood cells (RBCs) represents a valid alternative to liquid storage, since units can be preserved safely for … ocessing for cryostorage resulted in increased RBC volumes. Shape alterations caused an increase in osmotic fragility and permeability to ions. A significant pH drop was observed which could not to be attributed to a higher metabolic rate, since the levels of lactate did not show substantial fluctuation during the cell processing steps tested in this study. Membran …
