Use of a tissue clearing technique combined with retrograde trans-synaptic viral tracing to evaluate changes in mouse retinorecipient brain regions following optic nerve crush.

Successful establishment of reconnection between retinal ganglion cells and retinorecipient regions in the brain is critical to optic nerve regeneration. However, morphological assessments of retinorecipient regions are limited by the opacity of brain tissue. In this study, we used an innovative tissue cleaning technique combined with retrograde trans-synaptic viral tracing to observe changes in retinorecipient regions connected to retinal ganglion cells in mice after optic nerve injury. Specifically, we performed light-sheet imaging of whole brain tissue after a clearing process. We found tha

Spatial organization and sex differences of paraventricular hypothalamic oxytocin neurons projecting to distinct brain regions.

Hypothalamic oxytocin (OXT) neurons play crucial roles in modulating various behaviors, including reward, feeding, and affective behaviors, via targeting downstream projections. Although the whole-brain projections of the paraventricular nucleus (PVN) OXT neurons have been dissected, the spatial distribution and sex differences of subpopulations targeting distinct regions remain poorly investigated. Here, utilizing retrograde tracing in mice, we mapped PVN oxytocin neurons projecting to the nucleus accumbens (NAc), lateral septum (LS), central amygdala (CeA), ventral tegmental area (VTA), and

Multidimensional maturation of oxytocin neurons: A principal component analysis of postnatal development.

Oxytocin (Oxt) is a neuropeptide critical for higher brain functions, with its physiological functions dependent on the activity of Oxt-producing neurons in the paraventricular nucleus (PVN). However, the postnatal maturation process of these neurons remains poorly understood, particularly whether it involves a uniform trajectory or a dynamic functional reorganization. Here, we investigated the electrophysiological maturation of PVN Oxt neurons in rats using a multifaceted approach including electrophysiology, principal component analysis (PCA), and f-I curve fitting. Our analysis revealed a h

PVN-mPFC OT projections modulate pup-directed pup care or attacking in virgin mandarin voles.

Many species of animals exhibit caregiving or aggression toward conspecific offspring. The neural mechanisms underlying the infanticide and pup care remain poorly understood. Here, using monogamous mandarin voles (Microtus mandarinus), we found that more oxytocin (OT) neurons in the paraventricular nucleus (PVN) were activated during pup caring than infanticide. Optogenetic activation of OT neurons in the PVN facilitated pup caring in male and female mandarin voles. In infanticide voles, optogenetic activation of PVN OT cells or PVN-medial prefrontal cortex (mPFC) OT projection fibers prolonge

Acute sleep deprivation (ASD) and cardioprotection: Impact of ASD on oxytocin-mediated sympathetic nervous activation preceding myocardial infarction.

INTRODUCTION: This study explored how acute sleep deprivation (ASD) before myocardial ischemia influences oxytocin release from paraventricular (PVN) neurons and its correlation with sympathetic nervous system (SNS) activity post-acute sleep loss, impacting subsequent left ventricular (LV) remodeling following myocardial infarction (MI). METHODS: The study was conducted in two phases: induction of ASD, inducing MI, blood sampling, euthanizing animals and collecting their heart and brain for histological and gene expression evaluations. The animals in first and second phase were euthanized 24

Cardiorenal Syndrome: The Role of Neural Connections Between the Heart and the Kidneys.

The maintenance of cardiovascular homeostasis is highly dependent on tightly controlled interactions between the heart and the kidneys. Therefore, it is not surprising that a dysfunction in one organ affects the other. This interlinking relationship is aptly demonstrated in the cardiorenal syndrome. The characteristics of the cardiorenal syndrome state include alterations in neurohumoral drive, autonomic reflexes, and fluid balance. The evidence suggests that several factors contribute to these alterations. These may include peripheral and central nervous system abnormalities. However, accumul

Beyond the GnRH axis: kisspeptin regulation of the oxytocin system in pregnancy and lactation.

Circulating oxytocin is critical for normal birth and lactation. Oxytocin is synthesised by hypothalamic supraoptic and paraventricular neurons and is released from the posterior pituitary gland into the circulation. Oxytocin secretion depends on action potentials initiated at the cell body, and we have shown that intravenous (IV) administration of kisspeptin-10 transiently increases the firing rate of supraoptic nucleus oxytocin neurons in anaesthetised, non-pregnant, pregnant and lactating rats. This peripheral effect is likely via vagal afferent input, because disruption of vagal afferents

Magnetization transfer changes of grey and white matter in Parkinson's disease.

Since the attempt to evidence structural brain damage in Parkinson's disease (PD) by conventional magnetic resonance imaging (MRI) is usually disappointing, we have investigated whether the magnetization transfer ratio (MTR) can reflect changes in grey and white matter of PD patients. MTR was quantified in 44 regions of interest (ROIs) in both grey and white matter of 11 non-demented PD patients, ranging from 2 to 4 on the Hoehn and Yahr Scale, and eight age-matched healthy subjects. MTR differences between patients and controls were found in the supratentorial white matter and in the brainste

Locus coeruleus noradrenergic lesions attenuate intraoral intake.

I.p. injections of DSP-4 in male rats decreased norepinephrine (NE) levels to varying degrees throughout the brain with 66.7% reductions in the hypothalamic paraventricular nucleus. Intake of intraorally infused sucrose was reduced for 14 days but daily pellet intake recovered within 5 days post-injection. Intraventricular NE restored intraoral sucrose intake in DSP-4-lesioned rats without affecting controls. Intraventricular infusion of neuropeptide Y (NPY) reduced intraoral intake in controls but had no effect in DSP-4-lesioned rats. NPY markedly inhibited intraoral intake in DSP-4-treated r

Suprachiasmatic nucleus in a patient with multiple system atrophy with abnormal circadian rhythm of arginine-vasopressin secretion into plasma.

We performed a quantitative investigation of arginine-vasopressin (AVP) immunopositive neurons in the suprachiasmatic nucleus (SCN), which is the endogenous clock of the brain of a patient with multiple system atrophy (MSA) who exhibited nocturnal polyuria associated with decreased urinary specific gravity and depression of nocturnal AVP secretion. Eleven age- and sex-matched subjects were used as controls. Although, the number of AVP-positive neurons was decreased in neither the supraoptic nucleus nor the paraventricular nucleus, the number of AVP-positive neurons in the SCN was decreased and

Clozapine-induced Fos-protein expression in rat forebrain regions: differential effects of adrenalectomy and corticosterone supplement.

Unlike classical antipsychotic drugs, clozapine activates the hypothalamo-pituitary-adrenal axis and induces a specific regional pattern of Fos-protein expression in the rat forebrain. Whether corticosterone plays a role in the clozapine-induced Fos response is the subject of this study. Some rats were adrenalectomized and in a number, including intact animals, a corticosterone pellet (100 mg s.c.) was implanted; after 1 week, a single dose of clozapine (20 mg kg(-1) i.p.) was administered. The clozapine-induced Fos response was not affected by adrenalectomy, apart from the nucleus accumbens s

Oxytocinergic input from the paraventricular nucleus to the nucleus accumbens core modulates methamphetamine-conditioned place preference.

Methamphetamine (METH) is a highly addictive psychostimulant, yet its addiction mechanisms remain unclear. Oxytocin (OXT), a neuropeptide, shows promise in reducing METH addiction, but how OXT exerts its effects is poorly understood. Using conditioned place preference (CPP), we first found that intranasal OXT other than Arginine Vasopressin (AVP) administration suppressed METH-CPP in mice, which could be reversed by OXT receptors (OXTRs) blockade in the nucleus accumbens (NAc) core. Activating OXTRs in the NAc core similarly reduced METH-CPP. Then, we found repeated METH exposure inhibited oxy

Leptin action on ARC-PVN neural circuit regulates ejaculation behavior by altering sympathetic neuroplasticity.

BACKGROUND: Although some studies have revealed the close relationship between leptin and premature ejaculation in clinical practice, whether and how leptin participates in the regulation of ejaculatory behaviors are still unknown. OBJECTIVE: To explore the role of leptin on ejaculatory behaviors and its underlying mechanism. MATERIALS AND METHODS: Copulation behavior tests were performed after acute and chronic leptin administration at peripheral and central levels. To compare changes in sympathetic nervous system activity, lumbar sympathetic nervous activity, serum noradrenaline levels, an

Stress integration by an ascending adrenergic-melanocortin circuit.

Stress is thought to be an important contributing factor for eating disorders; however, neural substrates underlying the complex relationship between stress and appetite are not fully understood. Using in vivo recordings from awake behaving mice, we show that various acute stressors activate catecholaminergic nucleus tractus solitarius (NTSTH) projections in the paraventricular hypothalamus (PVH). Remarkably, the resulting adrenergic tone inhibits MC4R-expressing neurons (PVHMC4R), which are known for their role in feeding suppression. We found that PVHMC4R silencing encodes negative valence i

Glial regulation of neuronal function: from synapse to systems physiology.

Classically, glia have been regarded as non-excitable cells that provide nourishment and physical scaffolding for neurones. However, it is now generally accepted that glia are active participants in brain function that can modulate neuronal communication via several mechanisms. Investigations of anatomical plasticity in the magnocellular neuroendocrine system of the hypothalamic paraventricular and supraoptic nuclei led the way in the development of much of our understanding of glial regulation of neuronal activity. In this review, we provide an overview of glial regulation of magnocellular ne

Keeping oxytocin neurons under control during stress in pregnancy.

Oxytocin synthesised by magnocellular neurons in the supraoptic and paraventricular nuclei plays an important role in mammalian parturition. Accordingly, in late pregnant rats, oxytocin neurons are restrained from premature activation and stimulated oxytocin secretion is inhibited, preserving the expanded neurohypophysial oxytocin stores for parturition. A wide range of stressors stimulate oxytocin secretion in the rat. Some physical stressors, in particular immune challenge with systemic interleukin-1beta (IL-1beta, a cytokine that mimics infection) signal to magnocellular oxytocin neurons vi

Chronic stress and histological effects on the human central nervous system and other organs.

The effects of stress are usually studied in animal models. The organs of 103 subjects who died instantly or very rapidly from gunshot wound were histologically studied. Everyone, within organized criminality, lives under chronic stress. In the hypothalamus a marked depletion of neurosecretive granules of neurons in the supraopticus nucleus, paraventricular nucleus and median eminence was observed, with frequent spreading of these granules along the axons, towards the neurohypophysis. Glial proliferation and frequent chronic perivascular edema were observed in the white substance, as were typi

Changes in Alzheimer's disease in the magnocellular neurones of the supraoptic and paraventricular nuclei of the hypothalamus and their relationship to the noradrenergic deficit.

When compared with those of age-matched control patients the number of nerve cells in the locus caeruleus in 30 patients with Alzheimer's disease is reduced by 65% while nucleolar volume in surviving cells of the locus caeruleus and in those of the paraventricular and supraoptic nuclei of the hypothalamus is reduced by 25%, 48% and 26% respectively. Furthermore the reduction in cell number and nucleolar volume in these 3 cell types are all interrelated, emphasizing the close functional linkage of these cell groups. Similar changes (for age) were seen in a group of 10 patients with a mixed Alzh

Involvement of central nesfatin-1 neurons on oxytocin-induced feeding suppression in rats.

Peripheral anorectic hormones, such as peptide YY (PYY) and oxytocin (OXT), suppress food intake. A newly identified anorectic neuropeptide, nesfatin-1, is synthesized in both peripheral tissue and the central nervous system, particularly by various nuclei in the hypothalamus and brainstem. Here, we examined the effects of intraperitoneal (ip) administration of PYY3-36, OXT, and OXT analog, on nesfatin-1-immunoreactive (ir) neurons in the rat hypothalamus and brainstem, using Fos double fluorescence-immunohistochemistry. The ip administration of OXT and OXT analog significantly increased the n
