BACKGROUND: There is evidence to suggest that childhood leukemia and non-Hodgkin lymphoma have an infective etiology. We investigated the risk of childhood leukemia and non-Hodgkin lymphoma in relation to paternal occupational contact with other individuals, a surrogate for potential exposure to infection.
METHODS: We carried out a case-control study using 792 cases from the Northern Region Young Persons' Malignant Disease Registry, United Kingdom, 1968-1997, and 2 large population-based control groups. Paternal occupations at birth were classified as having standard, high, or very high level

Preconceptional paternal irradiation has been implicated as a causal factor in childhood cancer and it has been suggested that this exposure to radiation may play a role in the occurrence of childhood leukaemia clusters in the vicinity of nuclear installations. Using a transgenic mouse system employing a lambda shuttle vector allowing mutations (in the lacI gene) to be analysed in vitro, we have investigated the possibility that preconceptional paternal irradiation can lead to such transgenerational transmission of genomic instability. We have examined the mutation frequencies in vector recove

BACKGROUND: Maternal immune-mediated conditions during pregnancy have been linked with increased risk of attention-deficit/hyperactivity disorder (ADHD) in offspring. However, we do not know the extent to which these associations are influenced by shared genetic predispositions, as opposed to maternal inflammatory/immune responses during pregnancy. This study contributes by using paternal immune-mediated conditions as a negative control to explore these underlying factors, as we investigate associations between maternal immune-mediated conditions during pregnancy and offspring ADHD.

BACKGROUND: Family violence is a serious and increasingly significant public health issue, both in New Zealand and internationally. While Pacific families in New Zealand experience disproportionately higher rates of violence compared to their Palagi counterparts, little epidemiological information exists about the effect of childhood abuse on Pacific fathers and whether it increases their proclivity on perpetrating violence.
AIMS: To determine the prevalence of physical discipline administered to young Pacific children by their fathers and physical intimate partner violence (IPV) perpetrated

Paternal experiences and exposures before conception can influence fetal development and offspring phenotype. The composition of seminal plasma contributes to paternal programming effects through modulating the female reproductive tract immune response after mating. To investigate whether paternal obesity affects seminal plasma immune-regulatory activity, C57Bl/6 male mice were fed an obesogenic high-fat diet (HFD) or control diet (CD) for 14 weeks. Although HFD consumption caused only minor changes to parameters of sperm quality, the volume of seminal vesicle fluid secretions was increased by

Increased fat and carbohydrate intakes based on the Western diet are important lifestyle modifications that lead to hypercaloric inputs, obesity, and male fertility negative effects. Epigenetic transmission may also predispose descended generations to chronic diseases, such as obesity, type 2 diabetes, behavioral, and reproductive disorders. The present study sought to evaluate the influence of a high-fat-high-sugar (HFHS) diet supplied to Wistar rats from 25 to 90 days of life on reproductive and metabolic parameters in male generations F0, F1, and F2. The standard group received the normocal

Alcohol exposure during fetal and early postnatal development can lead to an increased incidence of later life adult-onset diseases. Examples include central nervous system dysfunction, depression, anxiety, hyperactivity, and an inability to deal with stressful situations, increased infection and cancer. Direct effects of alcohol leading to developmental abnormalities often involve epigenetic modifications of genes that regulate cellular functions. Epigenetic marks carried over from the parents are known to undergo molecular programming events that happen early in embryonic development by a wa

There is no convincing evidence regarding radiation-induced heritable risks of adult-onset multifactorial diseases in humans, although it is important from the standpoint of protection and management of populations exposed to radiation. The objective of the present study was to examine whether parental exposure to atomic-bomb (A-bomb) radiation led to an increased risk of common polygenic, multifactorial diseases-hypertension, hypercholesterolaemia, diabetes mellitus, angina pectoris, myocardial infarction or stroke-in the first-generation (F1) offspring of A-bomb survivors. A total of 11,951

The search for risk factors for development of germ cell tumors (GCT) in children who lived in Mexico City (MC). A protective, observational, case-control study was conducted in children under 15 years of age resident in MC, insurer by the Mexican Institute of Social Security. The study population was selected between January 1st, 1990 and December 31st, 1994. Parents of the children were interviewed with a 230-items precoded questionnaire, validated previously with a pilot study. For analysis were obtained simple frequencies and odds ratios (OR) and 95% confidence interval (95%CI). There were

Di-(2-ethylhexyl) phthalate (DEHP) is an endocrine disruptor (EDC) of significant public health concern due to its widespread use and potential adverse effects. This study investigated DEHP's multigenerational and dual-parent neurotoxic effects on male rats. The dams of F0 and F1 were exposed to DEHP during the gestation and lactation periods (GD0 to PND21). Meanwhile, F1 fathers were exposed prenatally and postnatally during puberty, maturity, and adulthood to evaluate the impact of paternal DEHP (300 mg/kg) exposure on their offspring. A comprehensive battery of tests was employed, including

Parental exposure to ambient fine particulate matter (PM2.5) has been associated with some of adverse health outcomes in offspring. The association between parental PM2.5 exposure and the development of metabolic syndrome (MetS) in offspring, and the effects of parental PM2.5 exposure on the susceptibility of offspring mice to PM2.5, has not been evaluated. The C57BL/6 parental mice (male and female mice) were exposed to filtered air (FA) or concentrated PM2.5 (PM) using Shanghai-METAS for a total of 16 weeks. At week 12 during the exposure, we allowed the parental male and female mice to bree

The oyster's immune system is capable of adapting upon exposure to a pathogen-associated molecular pattern (PAMP) to have an enhanced secondary response against the same type of pathogen. This has been demonstrated using poly(I:C) to elicit an antiviral response in the Pacific oyster (Crassostrea gigas) against Ostreid herpesvirus (OsHV-1). Improved survival following exposure to poly(I:C) has been found in later life stages (within-generational immune priming) and in the next generation (transgenerational immune priming). The mechanism that the oyster uses to transfer immunity to the next gen

WHO estimates that annually 15 million disability-adjusted life-years are lost and 250,000 asthma deaths are reported worldwide. Approximately 500,000 annual hospitalizations are due to asthma. To study the Relationship between environmental factors and childhood asthma has been studied. Maternal stress during pregnancy might affect fetal lung development and subsequently predispose to childhood asthma. The aim of the study was to assess the associations of maternal psychological stress during pregnancy and iden

Nutritional deficiencies are preventable etiological and epigenetic factors causing congenital abnormalities, first cause of infant mortality. Folate deficiency has a well-established teratogenic effect, leading to an increasing risk of neural tube defects. This paper highlights the most recent medical literature about folate deficiency, be it maternal or paternal. It then focuses on associated deficiencies as nutritional deficiencies are multiple and interrelated. Observational and interventional studies have all been consistent with a 50-70% protective effect of adequate women consumption of

OBJECTIVES: Fertility problems are an increasing public health issue in industrialised countries. Exposure to exogenous agents with endocrine disrupting properties, such as some pesticides, are potential risk factors for subfertility. The aim of this study was to determine whether time-to-pregnancy (TTP) is prolonged in male greenhouse workers exposed to pesticides in comparison with a non-exposed reference group.
METHODS: Data were collected through self-administrated questionnaires with detailed questions on TTP, as well as on lifestyle (for example, smoking habits, coffee and alcohol consu

INTRODUCTION: Fetal macrosomia is associated with short- and long-term perinatal complications. Although the role of maternal obesity is well established, the father's independent contribution has been less studied, particularly in middle-income countries such as Peru, where the prevalence of parental obesity has increased steadily over the past decade.
OBJECTIVE: To evaluate the association between obesity in both parents and fetal macrosomia in father-mother-newborn triads nationwide in Peru.
METHODS: Analytical cross-sectional study using data from the Peruvian Demographic and Family Heal

Given that the primordial ovarian follicular pool is established in utero, it may be influenced by parental characteristics and the intrauterine environment. Anti-Müllerian hormone (AMH) levels are increasingly recognized as a biomarker of ovarian reserve in females in adulthood and adolescence. We examined and compared associations of maternal and paternal prenatal exposures with AMH levels in adolescent (mean age, 15.4 years) female offspring (n = 1,399) using data from the Avon Longitudinal Study of Parents and Children, a United Kingdom birth cohort study that originated in 1991 and is sti
