Exosomes from Cyclic Stretched Periodontal Ligament Cells Induced Periodontal Inflammation through miR-9-5p/SIRT1/NF-κB Signaling Pathway.

Abundant evidence demonstrates that mechanical stress could induce an inflammatory response in periodontal tissue, but the precise mechanism … tch loading to reveal the detailed mechanism by which mechanical stimuli initiate immunoreaction in periodontium. In the present study, we found that cyclic stretch could stimulate human PDLCs to secret exosomes and that these exosomes could further induce the increase of phagocytic cells in the periodontium in Sprague-Dawley rats and the M1 polarization of th …

Vital Pulp Therapy as a Conservative Approach for Management of Invasive Cervical Root Resorption: A Case Series.

INTRODUCTION: Clear treatment guidelines with predictable outcomes for invasive cervical root resorption (ICRR) do not exist. Recent studies show that an internal approach for the excavation and repair of resorptive lacunae using bioactive materials might be preferred over external surgical approaches in cases with extensive ICRR. The procedure appears to interrupt and stop the resorptive process and preserves the periodontium. The aim of this case series was to report on the clinical and radiographic outcomes in cases with ICRR treated with vital pulp therapy techniques using calcium-enriched…

IL-35 may maintain homeostasis of the immune microenvironment in periodontitis.

T lymphocyte cells, including regulatory T (Treg) and T helper 17 cells, have important roles in the human periodontium. However, the basis for Treg cytokine expression in various compartments of the periodontium remains unclear. The aim of the present study was to investigate the expression of interleukin (IL)-35 in the peripheral blood mononuclear cells (PBMCs) and periodontal tissues of patients with chronic periodontitis (CP), with a view to understanding its role in this disease, and ultimately providing improved treatments. Peripheral blood, periodontal tissues and gingival crevicular fl…

Hypoxia promotes CEMP1 expression and induces cementoblastic differentiation of human dental stem cells in an HIF-1-dependent manner.

Cementum covering the tooth root provides attachment for the tooth proper to the surrounding alveolar bone via non-mineralized periodontal ligament (PDL). Cementum protein 1 (CEMP1) has been shown to induce a cementoblastic phenotype in cementoblast precursors cells of PDL. Oxygen availability is a critical signal for correct development of many tissues; however, its role in tooth root and periodontium development remains poorly understood. In this study, we demonstrated that reduced oxygen tension increased CEMP1 expression, mineral deposition, and alkaline phosphatase activity in human denta…

Immunohistochemical evaluation of matrix molecules associated with wound healing following regenerative periodontal treatment in monkeys.

The aim of the study was to investigate immunohistochemically the expression of matrix molecules associated with periodontal tissues reformed after regenerative periodontal treatment. Chronic intrabony defects were treated with guided tissue regeneration, enamel matrix proteins, the combination of both, or access flap surgery. Five months after healing, the animals were killed, and the healed periodontal tissues were evaluated immunohistochemically by means of polyclonal antibodies against osteopontin, collagen I, and collagen III. The intact (nontreated) parts of the periodontium served as co…

Rescue therapy with orthodontic traction to manage severely impacted mandibular second molars and to restore an alveolar bone defect.

This case report describes the successful treatment of severely impacted mandibular second molars with severe apical root resorption of the mandibular first molars. The vertically impacted second molars were orthodontically moved (using orthodontic mini-implants) without additional root resorption of the first molars. The orthodontic treatment provided a satisfactory and stable outcome by improving the periodontium surrounding the first and second molars. The treatment also eliminated the need for prosthetic treatment by preserving the first and second molars.

Characterization of dental anatomy and gingival biotype in Asian populations.

Gingival and dental characteristics are risk factors for periodontal problems. With short or fused roots, a decreased periodontium results in some attachment loss, compromising periodontal stability. Similarly, with an increased incidence of thin gingival biotype, inflammatory and traumatic insults may result in gingival recession. Anecdotally, Asian dentitions have been described as having short roots with "thin gingiva". This cross-sectional study will utilize clinical data and radiographic interpretation to ascertain whether this clinical impression is valid.

Management of trauma to supporting dental structures.

Teeth, periodontium, and supporting alveolar bone are frequently involved in trauma and account for approximately 15% of all emergency room visits. The cause of the dentoalveolar trauma varies in different demographics but generally results from falls, playground accidents, domestic violence, bicycle accidents, motor vehicle accidents, assaults, altercations, and sports injuries. Dentoalveolar injuries should be considered an emergency situation because successful management of the injury requires proper diagnosis and treatment within a limited time to achieve better outcomes.

Periodontal immune homeostasis: neutrophils as guardians and betrayers, and strategies for precision modulation.

The health of periodontal tissues fundamentally depends on a delicate, dynamic immune homeostasis; disruption of this balance constitutes the pathological basis driving the initiation and progression of chronic periodontitis. Due to their high heterogeneity and phenotypic plasticity, neutrophils play multifaceted roles in maintaining periodontal homeostasis and driving disease progression. Under homeostatic conditions, through precisely regulated mechanisms including chemotaxis, phagocytosis, and extracellular trap formation, neutrophils actively clear microorganisms and sustain tissue toleran

High-Frequency Ultrasound Characterization of Periodontal Soft Tissues Pre- and Post-Bacterial Inoculation.

OBJECTIVE: Current diagnostic methods of inflammatory periodontal diseases, e.g., visual evaluation, periodontal probing, and radiographs, are either subjective or insensitive. Intra-oral high-frequency ultrasound was investigated to quantify inflammation by detecting tissue dimensional and perfusion changes. METHODS: A cohort of 15-month-old mini-pigs, 4 female/male each, was analyzed. Pre-molars (PM) 3 and 4, as well as first molars (M1), were scanned. In bi-weekly time intervals all 4 quadrants were randomly enrolled and bacterial injection followed each quadrant scan in a weekly fashion.

Potential mechanisms between periodontitis and Alzheimer's disease: a scoping review.

BACKGROUND: Research has suggested 2 potential mechanisms by which the periodontal inflammatory response may communicate to distant organs: … y the introduction of periodontal bacteria and/or proinflammatory cytokines locally produced at the periodontium. CONCLUSION: Inflammation is an important mechanism in the onset and progression of both periodontitis and Alzheimer's disease. Nevertheless, further studies are necessary to better understand the multifactorial pathogenesis of Alzheimer's disease.

Programmed cell death of periodontal ligament cells.

The periodontal ligament is a crucial tissue that provides support to the periodontium. Situated between the alveolar bone and the tooth root, it consists primarily of fibroblasts, cementoblasts, osteoblasts, osteoclasts, periodontal ligament stem cells (PDLSCs), and epithelial cell rests of Malassez. Fibroblasts, cementoblasts, osteoblasts, and osteoclasts are functionally differentiated cells, whereas PDLSCs are undifferentiated mesenchymal stem cells. The dynamic development of these cells is intricately linked to periodontal changes and homeostasis. Notably, the regulation of programmed ce…
