Proton pump inhibitors and risk of Parkinson's disease: A systematic review and meta-analysis.

Parkinson's disease is a progressive neurodegenerative disorder. We assessed whether proton pump inhibitor (PPI) use is associated with PD risk through a systematic review and meta-analysis of observational studies, exploring heterogeneity and confounding. We searched PubMed, Web of Science, Scopus, and the Cochrane Library to September 2025 for cohort and case-control studies. Study quality was evaluated using the Newcastle-Ottawa Scale, and certainty was assessed using GRADE. Random-effects models pooled ORs; prespecified subgroup analyses examined study design, geography, and adjustment for

Exacerbation Risk by Chronic Proton Pump Inhibitor Use in Obstructive Lung Diseases.

BACKGROUND: Previous studies have shown inconsistent results regarding the use of proton pump inhibitors (PPIs) and the risk on exacerbations in patients with chronic obstructive airway diseases (COADs). RESEARCH QUESTION: Is long-term PPI use associated with exacerbation risk? STUDY DESIGN AND METHODS: Using Belgian nationwide claims-based data of adult patients receiving long-term medication for COADs between 2017 and 2022, we investigated the association between PPI use and exacerbation risk using multivariable Cox models. Based on defined daily doses (DDDs) dispensed during the prior yea

Proton pump inhibitors modulate esophageal epithelial barrier function and crosstalk with eosinophils.

BACKGROUND: Eosinophilic esophagitis (EoE) is a chronic allergic disorder driven by type 2 inflammation, characterized by eosinophilic infiltration and esophageal epithelial abnormalities, including barrier dysfunction, basal cell hyperplasia, epithelial thickening, and loss of differentiation. Although proton pump inhibitor (PPI) therapy is frequently employed in the management of EoE and is known to reduce esophageal eosinophilia, improve barrier function, and exert anti-inflammatory effects, the precise mechanism by which PPIs modulate type 2 inflammation and epithelial integrity remains in

Proton Pump Inhibitor (PPI)-Domperidone Induced Anemia and Galactorrhea: A Case Report.

Gastrointestinal disorders are increasingly common among young adults, particularly medical students exposed to chronic stress and irregular dietary habits. Proton pump inhibitors (PPIs) are widely prescribed for the management of gastroesophageal reflux disease (GERD), gastritis, and related acid-peptic disorders. However, prolonged PPI therapy has been associated with impaired iron absorption and may contribute to iron deficiency anemia (IDA). We report the case of 26-year-old female, unmarried, postgraduate medical student with iron deficiency anaemia and galactorrhea. The patient presented

Proton Pump Inhibitors in Glioblastoma: From Reflexive Prescribing to Evidence-Based Practice.

Glucocorticoids are frequently used in the management of brain tumours to control tumour-associated cerebral oedema and symptoms related to mass effect. Because glucocorticoid exposure may increase the risk of upper gastrointestinal complications in selected patients, proton pump inhibitors (PPIs) are often prescribed prophylactically. Emerging evidence suggests that exposure to PPIs with potent aldehyde dehydrogenase 1A1-activating properties (PA-PPIs) such as omeprazole may be associated with worse progression-free and overall survival in newly diagnosed glioblastoma (GBM). Routine reflexive

Proton pump inhibitors and gut microbiota dysbiosis: insights into the pathogenesis of ulcerative colitis.

BACKGROUND: Ulcerative colitis (UC) is a chronic, relapsing inflammatory bowel disease characterized by continuous mucosal inflammation of the colon and rectum. The global prevalence of UC has been rising steadily, and accumulating evidence suggests a potential association between proton pump inhibitor (PPIs) use and UC development. Nevertheless, the precise role of PPIs in the pathogenesis and clinical course of UC remains unclear. METHODS: The C57BL/6J mice were administered saline, omeprazole (OME) and dextran sulfate sodium to establish control, PPIs-treated and UC models, respectively. T

Site-specific activation of the proton pump inhibitor rabeprazole by tetrathiolate zinc centres.

Proton pump inhibitors have become top-selling drugs worldwide. Serendipitously discovered as prodrugs that are activated by protonation in acidic environments, proton pump inhibitors inhibit stomach acid secretion by covalently modifying the gastric proton pump. Despite their widespread use, alternative activation mechanisms and potential target proteins in non-acidic environments remain poorly understood. Employing a chemoproteomic approach, we found that the proton pump inhibitor rabeprazole selectively forms covalent conjugates with zinc-binding proteins. Focusing on DENR, a protein with a

Class effects of proton pump inhibitors in preventing oxaliplatin-induced peripheral neurotoxicity.

Oxaliplatin-induced peripheral neuropathy (OIPN) is a dose-limiting toxicity with limited countermeasures. Basic research has revealed that omeprazole, a proton pump inhibitor (PPI), exerts preventive effects against OIPN. In this study, we evaluated whether other PPIs exert similar effects via in vitro and in vivo experiments. Notably, esomeprazole, lansoprazole, and rabeprazole, classified as PPIs, prevented oxaliplatin-induced cultured F11 neuronal cell damage, and repeated PPI administration prevented mechanical allodynia in rats. However, vonoprazan, a potassium ion-competitive acid block

Proton Pump Inhibitor Challenge to Confirm Diagnosis of Atrophic Gastritis of the Stomach: A Proposal.

BACKGROUND AND AIMS: Chronic atrophic gastritis (CAG) is a known precancerous condition that can lead to the development of gastric cancer (GC). Low serum pepsinogen (PG) I levels have been proposed as a non-invasive marker for chronic atrophic gastritis of the stomach body, but the adequate upper cut-off for diagnosis remains controversial, as values ranging from 30 to 50 mcg/L are currently considered as a "grey zone". We aimed to identify patients with chronic atrophic gastritis (CAG) of the stomach body amongst subjects with PG-I levels ranging between 30 and 50 mcg/L by means of a proton

In vitro susceptibility testing of Trichomonas gallinae strains to proton pump inhibitors and nitroimidazoles.

Trichomonas gallinae is a unicellular obligate parasite predominantly spread among Columbiformes through the sharing of feeding and drinking stations, as well as during the feeding of squabs with crop milk. It primarily causes lesions in the pharynx and esophagus, impairing the birds' ability to eat, drink, and even breathe, thus it is potentially fatal for infected birds. Currently, five nitroimidazole derivatives are available for treatment, which, however, are prohibited for use in food-producing livestock. The aim of this study is to explore alternative treatment options against Trichomona

Decongestants, proton pump inhibitors, and systemic antibiotics are associated with an increased occurrence of dysbiosis.

BACKGROUND: Dysbiosis (also called dysbacteriosis) is characterized by a disruption of the microbiome, resulting in an imbalance in the microbiota, changes in their functional composition and metabolic activities, and a shift in their local distribution. Dysbiosis is most commonly reported as a condition affecting the gastrointestinal tract, for example with bacterial or fungal overgrowth in the small intestine. Known causes of dysbiosis include antibiotic use, liver disease, and alcohol misuse. AIMS: To determine those variables associated with the diagnosis of dysbiosis using a national dat

F-type proton-pumping ATPase mediates acid tolerance in Streptococcus mutans.

AIMS: Streptococcus mutans is highly sensitive to inhibitors of proton-pumping F-type ATPase (F-ATPase) under acidic conditions. Herein, we investigated the role of S. mutans F-ATPase in acid tolerance using a bacterium expressing the F-ATPase β subunit at lower levels than the wild-type strain. METHODS AND RESULTS: We generated a mutant S. mutans expressing the catalytic β subunit of F-ATPase at lower levels than the wild-type bacterium. The mutant cells exhibited a significantly slower growth rate at pH 5.30, whereas the rate was essentially the same as that of wild-type cells at pH 7.40. I

[Current use of proton pump inhibitors and when to limit, stop or not start treatment].

Proton pump inhibitors (PPIs) are potent inhibitors of gastric acid secretion that have changed treatment practice for gastric acid-related disorders. The major adequate indications for their use are treatment of gastro-esophageal reflux disease, peptic ulcers, eradication of Helicobacter pylori infection in combination with antibiotics and prophylaxis for patients on non-steroidal anti-inflammatory or antiplatelet drugs. Since their introduction, clinical success has been accompanied by widespread use of PPIs, which has steadily increased over the last decades without concomitant increase in

The Association of Proton Pump Inhibitors and QT Interval Prolongation in Critically Ill Patients.

INTRODUCTION: Drug-induced QT interval prolongation has been reported to be related to life-threatening polymorphic ventricular tachycardia (torsade de pointes). Proton pump inhibitors (PPIs) are prescribed widely for hospitalized patients; the QT interval prolongation and torsade de pointes caused by PPIs were reported. We conducted a study to determine the association between PPI treatment and QT interval prolongation in critically ill patients. METHODS: This study included patients with electrocardiography (ECG) reports from the Medical Information Mart for Intensive Care III database (MIM

Proton pump inhibitors induced fungal dysbiosis in patients with gastroesophageal reflux disease.

Gut mycobiota inhabits human gastrointestinal lumen and plays a role in human health and disease. We investigated the influence of proton pump inhibitors (PPIs) on gastric mucosal and fecal mycobiota in patients with gastroesophageal reflux diseases (GERD) by using Internal Transcribed Spacer 1 sequencing. A total of 65 participants were included, consisting of the healthy control (HC) group, GERD patients who did not use PPIs (nt-GERD), and GERD patients who used PPIs, which were further divided into short-term (s-PPI) and long-term PPI user (l-PPI) groups based on the duration of PPI use. Effects of vonoprazan and proton pump inhibitors on the efficacy of bevacizumab: a multicentre retrospective study.

Gastric acid secretion inhibitors such as proton pump inhibitors (PPIs) and vonoprazan may change the duration of treatment with bevacizumab, a vascular endothelial growth factor (VEGF) inhibitor, for cancer. However, there are no data on this prolongation effect. Here, we aimed to determine whether the use of PPIs or vonoprazan in patients with cancer receiving bevacizumab affected the duration of bevacizumab treatment. This observational study was conducted at two national university hospitals in Japan and involved 222 patients using oral PPIs (N = 190) or vonoprazan (N = 32) at the start of
