Strongyloides venezuelensis has been used in different experimental studies, such as those aimed at the evaluation of diagnostic techniques for human strongyloidiasis, mainly the molecular diagnosis. In this study, three regions (genus, 18S and 28S targets) of Strongyloides ribosomal DNA were evaluated for the molecular diagnosis of experimental strongyloidiasis. Rats were infected subcutaneously with 400 or 4000 S. venezuelensis infective larvae (400iL3 and 4000iL3), and kept for 35 days. Fecal samples were collected daily to count eggs per gram of feces (EPG) and to perform the polymerase ch

Sex is determined in Caenorhabditis elegans by an X-chromosome-counting mechanism that reliably distinguishes the twofold difference in X-chromosome dose between males (1X) and hermaphrodites (2X). This small quantitative difference is translated into the 'on/off' response of the target gene, xol-1, a switch that specifies the male fate when active and the hermaphrodite fate when inactive. Specific regions of X contain counted signal elements whose combined dose sets the activity of xol-1. Here we ascribe the dose effects of one region to a discrete, protein-encoding gene, fox-1. We demonstrat

During a parasitological survey of elasmobranchs of the Atlantic Ocean on the Mexican coast, the digestive tract of some specimens of the southern stingray, Hypanus americanus (Hildebrand & Schroeder, 1928), and the Atlantic sharpnose shark, Rhizoprionodon terraenovae (Richardson, 1836), were analyzed for tapeworms. These elasmobranchs were obtained from commercial fishing in four localities. Five taxa of Trypanorhyncha were recovered: Kotorella pronosoma (Stossich, 1901) Euzet & Radujkovic, 1989, Oncomegas wageneri (Linton, 1890) Dollfus, 1929, Pterobothrium kingstoni Campbell & Beveridge, 19

BACKGROUND: Chitin is an essential structural component of the wall of fungal cells and is present in the exoskeleton of arthropods. It has been generally assumed that mammals lack the ability to produce chitinase proteins, the enzymes responsible for chitin degradation. However, recent studies have indicated that mammals produce chitinases and chitinase-like proteins, and chitinase plays a potential role in human asthma and allergic inflammation. In this study, the effect and brief signaling pathway of chitinase on MUC5B expression were investigated in human airway epithelial cells.

On two occasions in November and December 2009, whilst being captured and handled for banding in Tasman Bay, New Zealand, two variable oystercatchers (Haematopus unicolor Forster, 1844) voided a number of cestode strobilae from their cloaca. Their morphology indicates that they belong to the family Hymenolepididae, confirmed by BLASTn searches of large and small subunits of ribosomal DNA partial sequences (18S and 28S, respectively). However, they cannot currently be assigned to any species reported for any oystercatcher species from New Zealand nor from oystercatchers worldwide. We present a

PURPOSE: Cyathocotylidae Mühling, 1896 is a cosmopolitan family of diplostomoidean digeneans characterized by a broad range of vertebrate definitive hosts, including fishes, birds, reptiles, and mammals. Cyathocotylid diversity in Australasia remains poorly known. In the present study, we describe a new genus and species of cyathocotylid digenean collected from the small intestine of the Australasian gannet, Morus serrator, in Queensland, Australia.
METHODS: Adult cyathocotylid specimens were collected from the small intestine of an Australasian gannet in Queensland. Specimens were stained, m

Bothriocestus claviceps (Goeze, 1782) is a cestode parasite infecting several fish species. One of its principal definitive hosts is the European eel Anguilla anguilla. The parasite is widely distributed across Europe and has been reported from numerous countries, such as Poland, Czechia, and France. In southern France, the European eel co-occurs with the Aquitanian pike, Esox aquitanicus, an endemic species. This pike is threatened by human activities and hybridization with the introduced Northern pike Esox lucius and is therefore classified as "vulnerable" on the French Red List of the Inter

Creptotrema funduli Mueller, 1934, has long presented an uncertain systematic position within the family Allocreadiidae. Previous phylogenetic analyses based on partial 28S rDNA sequences recovered this species as distinct from other members of Creptotrema, suggesting that its current generic placement may not reflect its evolutionary relationships. In the present study, we re-evaluate the taxonomic status of C. funduli through an integrative approach combining morphological observations and phylogenetic analyses based on 28S rDNA sequences. Our results consistently recover C. funduli outside

In the present study, helminths from six Didelphis virginiana and one Philander vossi are reported using morphological techniques (clearing, staining, and scanning electron microscopy). Additionally, the 28S rRNA sequences of individuals from nine helminth taxa are provided. Phylogenetic analyses were performed with the new 28S rRNA sequences to confirm the identification and the genealogical relationships of the parasites. Thirteen helminth taxa were identified, comprising the trematodes Brachylaima sp. and Platynosomum illiciens, the cestode Mathevotaenia sp., the nematodes Cruzia americana,

BACKGROUND: Four species of Strongyloides, Strongyloides felis, Strongyloides planiceps, Strongyloides stercoralis and Strongyloides tumefaciens, have been identified in cats based on morphology and location in the host with limited data on the prevalence and disease potential of these different species. Strongyloides tumefaciens adults are located in colonic nodules while the other three species are in the small intestine. The literature on Strongyloides in cats is scattered and has never been compiled. The aim of this article is to provide a short review of the existing literature on Strongy

Polyopisthocotylean monogenean parasites of fishes are highly host specific and have been used as an appropriate model to study the host-parasite co-evolution. In the present study, eight monogeneans of the order Mazocraeidea were characterized by nuclear 28S rDNA sequences and their phylogenetic relationship with other polyopisthocotylean species was investigated. Neighbour-joining, maximum parsimony, maximum likelihood and Bayesian Inference methods were used for phylogenetic reconstruction. The topology sustained by high bootstrap was: (((Hexabothriidae (Mazocraeidae (Discocotylidae (Diploz

This paper presents the first reported use of 18S rRNA gene sequence to determine the phylogeny of Brugia pahangi. The 18S rRNA nucleotide sequence of a Malaysian B. pahangi isolate was obtained by PCR cloning and sequencing. The sequence was compared with 18S rRNA sequences of other nematodes, including those of some filarial nematodes. Multiple alignment and homology analysis suggest that B. pahangi is closely related to B. malayi and Wuchereria bancrofti. Phylogenetic trees constructed using Neighbour Joining, Minimum Evolution and Maximum Parsimony methods correctly grouped B. pahangi with

Wolbachia are strictly intracellular maternally inherited alpha-proteobacteria, largely widespread among arthropods and filariids (i.e., filarial nematodes). Wolbachia capacities to infect new host species have been greatly evidenced and the transfer of Wolbachia between arthropods and filariids has probably occurred more than once. Interestingly, among nematode species, Wolbachia infection was found in filariids but not in closely related lineages. Their occurrence in filariids has been supposed a consequence of the parasitic lifestyle of worms within Wolbachia-infected arthropods, implying t

In a previous work Ancylostoma caninum sex-specific genes had being identified, however without confirmation, by using an non specific RT-PCR comparing male and female cDNA. In this work more fragments had been identified and a semi-quantitative RT-PCR carried out in order to confirm the sex-specific character of the transcripts obtained. Four fragments were confirmed as sex-specific or as being expressed more in one sex than the other.

Adult Onchocerca volvulus recovered for excised nodules by dissection or treatment with collagenase have been used as a source of RNA for in vitro translation experiments. RNA was purified using either the hot phenol/SDS procedure or the guanidine isothiocyanate protocol. Immunoprecipitation experiments performed on in vitro products demonstrate a marked heterogeneity in responses by individed human infection sera. Further immunoprecipitation experiments demonstrate cross reactivity between O. volvulus and other filarial nematodes.

Myotonic dystrophy disorders are caused by expanded CUG repeats in noncoding regions. Here we used Caenorhabditis elegans expressing CUG repeats to identify genes that modulate the toxicity of such repeats. We identified 15 conserved genes that function as suppressors or enhancers of CUG repeat-induced toxicity and that modulate formation of nuclear foci by CUG-repeat RNA. These genes regulate CUG repeat-induced toxicity through distinct mechanisms including RNA export and clearance, thus suggesting that CUG-repeat toxicity is mediated by multiple pathways. A subset of the genes are also invol

The schistosomal tegument is a dynamic host-interactive layer. Proteins exposed to the host on the tegumental surface are important for completion of the parasitic lifecycle. Dysferlin is a member of the ferlin family and is involved in plasma membrane repair. Based on the results of a proteomics study of tegument surface proteins of Schistosoma japonicum in our laboratory, dysferlin was identified as a tegumental protein of S. japonicum. The gene encoding S. japonicum dysferlin (SjDF), which codes for several Ca(2+) binding sites, was cloned, expressed in Escherichia coli, and characterized.

INTRODUCTION: Hepatic alveolar echinococcosis (HAE), a lethal chronic helminth infection caused by Echinococcus multilocularis, is characterized by pronounced local immune evasion and progressive CD8+ T cell exhaustion at the parasite?host invasive margin. However, the spatial cellular network and core regulatory subsets driving this localizedimmunosuppression remainpoorly understood.
METHODS: We performed high-resolution spatial immune profiling of the lesion invasive margin (collagenous layer tissue, CLT) and paired distal normal liver tissues (DLT) from 12 HAE patients by integrating imagi
