The fate of the embedded virgin sand flea Tunga penetrans: hypothesis, self-experimentation and photographic sequence.

We describe the morphological development of a single penetrated female sand flea (Tunga penetrans) in a medical expatriate working in Madagascar. The embedded parasite developed abnormally in two aspects. First, it lived twice as long as usually. Second, it did not expel a single egg during a period of two months. We explain these abnormalities by the fact that the female sand flea remained virgin after the penetration into the skin and, therefore, mature eggs did not develop. Our observations are seminal findings in a long dispute between entomologists and suggest that normally the fertiliza…

Ammonium removal pathways and microbial community in GAC-sand dual media filter in drinking water treatment.

A GAC-sand dual media filter (GSF) was devised as an alternative solution for drinking water treatment plant to tackle the raw water polluted by ammonium in place of expensive ozone-GAC processes or bio-pretreatments. The ammonium removal pathways and microbial community in the GSFs were investigated. The concentrations of ammonium, nitrite and nitrate nitrogen were monitored along the filter. Total inorganic nitrogen (TIN) loss occurred during the filtration. For 1 mg ammonium removal, the TIN loss was as high as 0.35 mg, DO consumption was 3.06 mg, and alkalinity consumption was 5.55 mg. It …

Effects of Asian sand dust on mucin gene expression and activation of nasal polyp epithelial cells.

BACKGROUND: Asian sand dust (ASD) contains a variety of chemical and microbiological materials such as lipopolysaccharide (LPS) and beta-glucan that can cause inflammation. The increase in the atmospheric concentration of ASD has been associated with asthma severity and adverse effects on respiratory function. The aim of this study was to evaluate the effect of ASD on the inflammatory process and mucin gene expression in nasal epithelial cells. METHODS: Primary nasal polyp epithelial cells were exposed to ASD for 72 hours, and then the supernatants were collected. To determine the activation …

Expression plasticity of Phlebotomus papatasi salivary gland genes in distinct ecotopes through the sand fly season.

BACKGROUND: Sand fly saliva can drive the outcome of Leishmania infection in animal models, and salivary components have been postulated as vaccine candidates against leishmaniasis. In the sand fly Phlebotomus papatasi, natural sugar-sources modulate the activity of proteins involved in meal digestion, and possibly influence vectorial capacity. However, only a handful of studies have assessed the variability of salivary components in sand flies, focusing on the effects of environmental factors in natural habitats. In order to better understand such interactions, we compared the expression prof…

Indoor protection against mosquito and sand fly bites: a comparison between citronella, linalool, and geraniol candles.

The repellent effect of 3 essential-oil-based candles was evaluated in a high biting pressure environment in Israel. In human landing assays, the repellency rate of 5% citronella candles against mosquitoes was 29.0%, of 5% linalool candles was 71.1%, and of 5% geraniol candles was 85.4%. The candles with geraniol were about twice as effective as those with linalool and were about 5 times as effective as citronella candles in protecting a person from being bitten indoors by mosquitoes. The repellency rate of 5% citronella candles towards sand flies was 24.7%, of 5% linalool candles was 55.2%, a…

Seasonal fluctuations of phlebotomine sand fly populations (Diptera: Psychodidae) in the urban area of Marrakech, Morocco.

Phlebotomine sand flies (Diptera: Psychodidae) were collected continuously, using sticky traps, during 1 year from October 2002 to September 2003, in an urban area of Marrakech city (Morocco). A total of 3277 specimens were collected belonging to five species. Phlebotomus (Phlebotomus) papatasi (54.6%) is the predominant species followed by Sergentomyia (Sergentomyia) minuta (20%), S. (S.) fallax (11.3%), P. (Paraphlebotomus) sergenti (10.3%) and P. (Larroussius) longicuspis (3.8%). Data analyses showed a mono-modal annual pattern for P. sergenti and a bi-modal one for the other species. P. pa…

Antigenic diversity in maxadilan, a salivary protein from the sand fly vector of American visceral leishmaniasis.

The salivary protein maxadilan (MAX) is a vasodilator and immunomodulator from the sand fly vector of the protozoan parasite Leishmania chagasi. Vaccinating BALB/c mice with sand fly salivary gland extracts or with MAX protects the host against L. major infection. Because of the potential use of MAX in an anti-Leishmania vaccine, we characterized the vertebrate host IgG response to MAX in the present study. Our immunochemical analysis indicated that antibodies to MAX were detected in BALB/c mice, as well as in pigs and humans, from a area in Nicaragua endemic for Lutzomyia longipalpis. Previou…

Mobilization and entry of DNAPL pools into finer sand media by cosolvents: two-dimensional chamber studies.

Two-dimensional chamber studies were conducted to determine qualitative and quantitative performance of cosolvents targeted at pooled dense non-aqueous phase liquid (DNAPL) (perchlorethylene, PCE) residing above a fine-grain capillary barrier. Downward mobilization of DNAPL, up gradient along an overriding cosolvent front, was observed. This produced significant pooling above a fine-grain layer that in some cases lead to entry into the capillary barrier beneath. Entry pressure calculations using physical and hydrogeologic parameters provided an excellent prediction of breakthrough of DNAPL int

Delayed-type hypersensitivity to Phlebotomus papatasi sand fly bite: An adaptive response induced by the fly?

The saliva of bloodsucking arthropods contains a large array of pharmacologically active compounds that assist hematophagy. Arthropod saliva is also responsible for causing uncomfortable allergic responses in its vertebrate hosts. In this article, we investigate whether the sand fly Phlebotomus papatasi, known to produce a strong delayed-type hypersensitivity (DTH) in humans, could benefit from, and possibly adaptively induce, this response in their vertebrate hosts. In this study, we show that flies fed on humans to completion nearly twice as fast in DTH sites as compared with normal skin sit…

Effects of permethrin-treated screens on phlebotomine sand flies, with reference to Phlebotomus martini (Diptera: Psychodidae).

Effects of permethrin-treated screens on the sand fly, Phlebotomus martini Parrot, a vector of visceral leishmaniasis in Kenya, were evaluated after the eighth treatment. Screens treated with 0.50 g/m2 of permethrin 20% emulsifiable concentrate were fitted inside houses and re-treated every 6 mo from August 1989 to December 1993. Before application of the treated screens, a geometric mean of 0.83 P. martini were collected per trap night inside houses, whereas after the eighth treatment only 0.05 sand flies were caught. P. martini abundance was reduced by 88.8%. Analysis of the effects of treat…

Effect of cerastobin, a thrombinlike enzyme from Cerastes vipera (Egyptian sand snake) venom, on human platelets.

Cerastobin, a thrombinlike enzyme with arginine esterase activity, was purified from crude Cerastes vipera (Egyptian Sahara snake) venom. Unlike thrombinlike enzymes isolated from other snake venoms, cerastobin had a potent platelet aggregatory effect. The activation of human platelets was not related to adenosine diphosphate release and/or prostaglandin synthesis. Cerastobin showed a proteolytic activity towards protein constituents of the platelets' cytoskeleton. It hydrolyzed actin, actin-binding protein, and P235. This may explain at least a part of the aggregatory mechanism(s) of cerastob

Molecular screening for Wolbachia endosymbionts in phlebotomine sand flies within the North Central Province, Sri Lanka.

Cutaneous leishmaniasis is a growing public health threat in Sri Lanka, transmitted primarily by phlebotomine sand flies. While biological control using Wolbachia has proven effective against other vector-borne diseases, its potential application in phlebotomine sand flies remains largely unexplored. Therefore, investigating the natural prevalence of these endosymbionts in Sri Lankan sand fly populations is well justified. To address this knowledge gap and evaluate the feasibility of symbiont-mediated control, we conducted a six-month entomological survey across four endemic sampling areas in …

Sand Enteropathy-What We Know So Far and Why There Is Still So Much to Learn.

Sand enteropathy describes pathophysiological conditions of the equine gastrointestinal tract due to the uptake and accumulation of sand. It is an increasingly recognized cause of gastrointestinal disease in arid geographic areas. Clinical signs can vary widely from colic, fever and inappetence to poor performance. Diagnosis based solely on clinical signs is nearly impossible. The current literature describes a variety of diagnostic aids, with radiographic imaging considered the gold standard. Therapeutic approaches aim at the evacuation of sand from the abdomen. This can be achieved by the ad…

RNA viruses in sylvatic mosquitoes and phlebotomine sand flies from Alto Pantanal, Mato Grosso, Brazil 2019.

The Pantanal biome harbors exceptional biodiversity but has been increasingly impacted by climate change and human activities. This region is considered a high-risk zone for zoonotic spillover, making viral studies in sylvatic mosquitoes and other invertebrates indispensable, as these vectors are involved in the transmission of pathogens of public health concern. This study aimed to describe viral genomes identified in Aedes spp., Ochlerotatus sp., Mansonia sp., Phlebotomus sp., Psorophora spp., and Anopheles spp. dipterans collected in March and June 2019, in Pirizal and Porto São Luiz, Alto

Integrating molecular tools into leishmaniosis surveillance: evaluation of a commercial qPCR kit in sand fly vectors.

BACKGROUND: Leishmaniosis is a zoonotic disease caused by protozoan parasites of the Leishmania genus, primarily transmitted through the bites of infected female phlebotomine sand flies. Leishmania infantum is the most prevalent Leishmania species in the Mediterranean Region, affecting both humans and animals, mainly dogs. Integrated surveillance strategies, including xenomonitoring, are essential for early detection and cost-effective control in endemic areas. In 2021, an increase in human leishmaniosis cases was reported in the Region of Murcia (Spain), prompting an intensified entomological…

Long-term monitoring of sand fly populations: temporal responses to land-use change and climatic variability.

BACKGROUND: Phlebotomine sand flies are the sole vector of leishmaniasis and other sand fly-borne diseases, and their populations are shaped by climate and human-driven environmental change. While warming is expected to expand sand fly ranges, the joint effects of long-term climatic variability and land-use change on population dynamics remain poorly understood, largely owing to limited long-term monitoring. METHODS: We analyzed an 18-year dataset (2005-2021) from a site in semi-arid Ma'ale Adumim, Israel, using CO2-baited traps. Land-cover change was quantified using remote sensing, focusing…
