The upregulation of α2δ-1 subunit modulates activity-dependent Ca2+ signals in sensory neurons.

As auxiliary subunits of voltage-gated Ca(2+) channels, the α2δ proteins modulate membrane trafficking of the channels and their localization to specific presynaptic sites. Following nerve injury, upregulation of the α2δ-1 subunit in sensory dorsal root ganglion neurons contributes to the generation of chronic pain states; however, very little is known about the underlying molecular mechanisms. Here we show that the increased expression of α2δ-1 in rat sensory neurons leads to prolonged Ca(2+) responses evoked by membrane depolarization. This mechanism is coupled to CaV2.2 channel-mediated res

Neural Control of Mastication: Ion-Channel Mechanisms in the Brainstem Central Pattern Generator.

Mastication is a fundamental rhythmic motor behavior controlled by a brainstem central pattern generator (CPG) located within the pontine and medullary reticular formations. Coordinated activation of jaw-opening and jaw-closing muscles is generated by this network and continuously refined through sensory feedback from periodontal mechanoreceptors and muscle spindles, together with descending inputs from the cortical masticatory area (CMA), basal ganglia, and cerebellum. Thus, mastication is regulated by distributed neural circuits rather than a single central locus. At the cellular level, the

Computational Modeling of Dorsal Root Ganglion Stimulation: Understanding Pain Suppression Mechanisms.

This study aims to advance our mechanistic understanding of electrical stimulation of dorsal root ganglia (DRG) for treating chronic pain. While DRG stimulation has shown moderate clinical success in managing certain types of chronic pain, the underlying neural mechanism remains inconclusive, hindering the further development of the technology to treat a broader range of chronic pain symptoms and benefit a larger patient population. In this study, we conducted computational simulations in the NEURON simulation environment to assess the neuromodulatory effect of DRG stimulation on action potent

Nerve Injury-Induced γH2AX Reduction in Primary Sensory Neurons Is Involved in Neuropathic Pain Processing.

Phosphorylation of the serine 139 of the histone variant H2AX (γH2AX) is a DNA damage marker that regulates DNA damage response and various diseases. However, whether γH2AX is involved in neuropathic pain is still unclear. We found the expression of γH2AX and H2AX decreased in mice dorsal root ganglion (DRG) after spared nerve injury (SNI). Ataxia telangiectasia mutated (ATM), which promotes γH2AX, was also down-regulated in DRG after peripheral nerve injury. ATM inhibitor KU55933 decreased the level of γH2AX in ND7/23 cells. The intrathecal injection of KU55933 down-regulated DRG γH2AX expres

Transduction without tip links in cochlear hair cells is mediated by ion channels with permeation properties distinct from those of the mechano-electrical transducer channel.

Tip links between adjacent stereocilia are believed to gate mechano-electrical transducer (MET) channels and mediate the electrical responses of sensory hair cells. We found that mouse auditory hair cells that lack tip links due to genetic mutations or exposure to the Ca(2+) chelator BAPTA can, however, still respond to mechanical stimuli. These MET currents have unusual properties and are predominantly of the opposite polarity relative to those measured when tip links are present. There are other striking differences, for example, the channels are usually all closed when the hair cell is not

Nicotine and brain development.

Preclinical studies, using primarily rodent models, have shown acetylcholine to have a critical role in brain maturation via activation of nicotinic acetylcholine receptors (nAChRs), a structurally diverse family of ligand-gated ion channels. nAChRs are widely expressed in fetal central nervous system, with transient upregulation in numerous brain regions during critical developmental periods. Activation of nAChRs can have varied developmental influences that are dependent on the pharmacologic properties and localization of the receptor. These include regulation of transmitter release, gene ex

Childhood trauma predicts attenuated N170 responses to happy child faces: ERP Evidence of early perceptual gating deficits.

Maternal sensitivity to children's emotional cues is a fundamental determinant of healthy child development, yet childhood trauma can disrup … and RF-T showed no neural associations. These findings suggest that early adversity may recalibrate sensory gating to deprioritize social safety cues. Yet, preserved later evaluative processes indicate neurocognitive resilience. This dissociation identifies a bottleneck in maternal attunement and a targeted pathway for interventions to disrupt intergenerational …

Analysis of Gene, Environment, and Sex Interaction in the Development of Autistic-like Phenotype in Mice.

Autism Spectrum Disorder (ASD) is a developmental disorder that manifests a broad variability of phenotypes. The underlying factors contribu … ve effects on repetitive/stereotyped and social behavior in the offspring, while working memory and sensory gating were not affected by hits. These results indicate that genetics, sex, and environment interact to influence autistic-like phenotypes in a behavior-specific manner.

A population of primary afferent sensory neurons mediates pain relief through nocifensive coping behavior in mice.

When exposed to noxious cutaneous stimuli, animals exhibit nocifensive behaviors, including rapid defensive reflexes that limit tissue damage and subsequent coping behaviors-most commonly licking of the affected area-that alleviate pain. Despite its ubiquity, the neural mechanism underlying pain relief through licking remains poorly understood. Here, we demonstrate that Npy2r-Cre+ primary sensory neurons in the dorsal root ganglion (DRG), a population of Aβ fibers that includes touch-sensitive, rapidly adapting low-threshold mechanoreceptors, are critical for this process. Mice lacking Npy2r-C

Channelrhodopsin fluorescent tag replacement for clinical translation of optogenetic hearing restoration.

Sensory restoration by optogenetic neurostimulation provides a promising future alternative to current electrical stimulation approaches. So far, channelrhodopsins (ChRs) typically contain a C-terminal fluorescent protein (FP) tag for visualization that potentially poses an additional risk for clinical translation. Previous work indicated a reduction of optogenetic stimulation efficacy upon FP removal. Here, we further optimized the fast-gating, red-light-activated ChR f-Chrimson to achieve efficient optogenetic stimulation in the absence of the C-terminal FP. Upon FP removal, we observed a ma

Modulation of medial geniculate nucleus neuronal activity by electrical stimulation of the nucleus accumbens.

Dysfunctional sensory gating has been proposed to result in the generation of phantom perceptions. In agreement, it has been recently suggested that tinnitus, a phantom perception of sound commonly associated with hearing loss, is the result of a breakdown of circuitry involving the limbic system and the medial geniculate nucleus (MGN) of the thalamus. In humans with tinnitus, structural changes and abnormal activity have been found to occur in the auditory pathway as well as parts of the limbic system such as the nucleus accumbens (NAc). However, at present, no studies have been conducted on …

Auditory and Lower Limb Tactile Prepulse Inhibition in Primary Restless Legs Syndrome: Clues to Its Pathophysiology.

The resting sensory discomfort transiently relieved upon movement of the affected area in restless legs syndrome suggests that sensorimotor integration mechanisms, specifically gating, may be altered in the disease. The authors sought to determine the effects of prepulse auditory and tactile stimulation applied to lower limbs on the blink reflex of patients with restless legs syndrome and healthy subjects. Seventeen patients with restless legs syndrome and 17 age- and sex-matched healthy controls were investigated. Auditory stimuli and tactile lower limb stimulation were applied as prepulses.

Current and Developing Therapeutics for Dry Eye Disease: Targeting Ion Channels.

Dry eye disease (DED) is an ocular surface disorder characterized by tear film instability, inflammation, epithelial damage, and neurosensory abnormalities. Due to its multifactorial etiology and pathophysiology, conventional therapies that focus on lubrication and immunosuppression often fall short in addressing the neuropathic component of ocular pain experienced by a growing subset of patients. Recent developments in sensory neuroscience have highlighted the pivotal role of ion channels in mediating ocular surface homeostasis, pain signaling, and inflammation. This review examines the role

Partners in plasticity: serotonergic glial interactions in brain circuit remodeling.

Experience-dependent brain circuit optimization choreographed by environmental sensory input activity involves synapse formation, pruning, and remodeling to sculpt appropriate connectivity. The serotonin (5-HT) neuromodulator acts as a core regulator of this circuit plasticity. Classically, serotonergic control has been understood solely through neuronal mechanisms, however new evidence reveals glial 5-HT signaling roles. This review focuses on recent studies in Drosophila with reference to foundational mammalian work to discuss 5-HT functions in both neurons and glia, particularly experience-

The skeletal connectome: From molecular circuitry to systemic network medicine.

Although classically viewed as a mechanical scaffold, the skeleton is increasingly recognized as a dynamic endocrine and sensory hub that integrates systemic biophysical and biochemical cues. In this review, we rethink skeletal physiology by proposing the 'skeletal connectome,' a network medicine framework that links circuit-level remodeling programs to multiorgan homeostasis. Structurally, we first distill the cellular logic of bone remodeling, emphasizing how osteoblasts, osteoclasts, and osteocytes act as plastic network nodes whose state transitions are constrained by topology, compartment

Evaluation of neurotoxicity and long-term function and behavior following intrathecal 1 % 2-chloroprocaine in juvenile rats.

Spinally-administered local anesthetics provide effective perioperative anesthesia and/or analgesia for children of all ages. New preparations and drugs require preclinical safety testing in developmental models. We evaluated age-dependent efficacy and safety following 1 % preservative-free 2-chloroprocaine (2-CP) in juvenile Sprague-Dawley rats. Percutaneous lumbar intrathecal 2-CP was administered at postnatal day (P)7, 14 or 21. Mechanical withdrawal threshold pre- and post-injection evaluated the degree and duration of sensory block, compared to intrathecal saline and naive controls. Tissu

Prediction of acoustic tinnitus suppression using resting-state EEG via explainable AI approach.

Tinnitus is defined as the perception of sound without an external source. Its perceptual suppression or on/off states remain poorly underst … zed in individuals with BATS. Additionally, hyperconnected auditory-limbic networks in BATS suggest sensory gating may aid suppression. These findings demonstrate robust classification of BATS status, revealing distinct neural traits between tinnitus subpopulations. Our work emphasizes the role of neural mechanisms in predicting and managing tinnitus suppression …

Increased basal ganglia volume in older adults with tinnitus.

Tinnitus is the perception of sounds without external stimuli, affecting 10%-15% of the general population and up to 25% of individuals over 70 years of age. While traditionally viewed as an auditory phenomenon, growing evidence highlights the role of the central nervous system in its pathophysiology. One of the proposed mechanisms, the “gating hypothesis” of tinnitus, suggests an alteration in the modulation of sensory activity by the frontostriatal network. Although structural changes in frontal areas support this idea, gray matter differences in subcortical regions—such as the auditory path

In addition, the sensory gating by alpha oscillations applies not only to the visual but also to the somatosensory domain. We conclude that albeit more refined perceptual paradigms and individualized stimulation practices remain to be systematically adopted, tACS is a promising to …
