Na+/H+ Exchangers (NHEs) in Mammalian Sperm: Essential Contributors to Male Fertility.

Na+/H+ exchangers (NHEs) are known to be important regulators of pH in multiple intracellular compartments of eukaryotic cells. Sperm function is especially dependent on changes in pH and thus it has been postulated that NHEs play important roles in regulating the intracellular pH of these cells. For example, in order to achieve fertilization, mature sperm must maintain a basal pH in the male reproductive tract and then alkalize in response to specific signals in the female reproductive tract during the capacitation process. Eight NHE isoforms are expressed in mammalian testis/sperm: NHE1, NHE

Epididymal SPAM1 and its impact on sperm function.

The most widely conserved mammalian sperm antigen is sperm adhesion molecule 1, SPAM1/PH-20, which is also the major testicular hyaluronidas … icated in ES regulation. Based on regional expression, the protein is likely to play a role in both sperm maturation and storage. A minor secretory glycoprotein, ES is present in the epididymal luminal fluid in both a soluble and insoluble form (epididymosomes), with the latter having an intact lipid anchor. Genetic approaches have provided evidence for sperm upta …

From reservoir to rendezvous: the journey of sperm through the oviduct.

Once viewed simply as a pipeline connecting the ovary and uterus where fertilization and early development occur, the oviduct is becoming more appreciated for its complex functions. The utero-tubal junction (UTJ) restricts the entry of sperm to those meeting specific morphological and molecular requirements. Once sperm passes the UTJ, the isthmus functions as a storage reservoir, retaining sperm before fertilization and prolonging their lifespan, allowing for more asynchrony between sperm deposition and ovulation. Some mammals, such as bats, store sperm for months, but a few days of storage is

In vitro maturation of Nile tilapia (Oreochromis niloticus) testicular spermatozoa in different media considering with motility, oxidative stress, fatty acid profile and fertilization capacity.

Collection and usage of testicular spermatozoa are often required for reproduction practices due to low sperm volume, reduced sperm density, urine contamination, and utilization of spermatozoa of sex-reversed broodstocks. No previous study has investigated in vitro maturation (IVM) of Nile tilapia (Oreochromis niloticus) testicular spermatozoa, despite being a widely cultured species in the world. The main aim of the present study was to establish an IVM protocol for testicular spermatozoa of this species. Accordingly, testicular sperm samples were first diluted with four different media inclu

Is there a specific abnormality of sperm morphology in men with varicoceles?

With the use of a case-control design, we have studied seminal cytology in 53 infertile men with large left varicoceles to determine whether they had a specific morphologic pattern. An equal number of infertile controls without varicoceles were matched for characteristics such as left testicular size, serum follicle-stimulating hormone levels, and sperm concentration to ensure similar degrees of testicular dysfunction. No significant difference in the morphologic patterns was observed, especially in the hallmarks of the "stress pattern": increased percentages of spermatozoa with tapering heads

Role of antioxidants in fertility preservation of sperm - A narrative review.

Male fertility is affected by multiple endogenous stressors, including reactive oxygen species (ROS), which greatly deteriorate the fertility. However, physiological levels of ROS are required by sperm for the proper accomplishment of different cellular functions including proliferation, maturation, capacitation, acrosomal reaction, and fertilization. Excessive ROS production creates an imbalance between ROS production and neutralization resulting in oxidative stress (OS). OS causes male infertility by impairing sperm functions including reduced motility, deoxyribonucleic acid damage, morpholo

Methods for Investigating the Sperm Epigenome.

In recent years, high-throughput technologies for investigating different aspects of sperm epigenome are gaining ground in order to elucidate the complex role of epigenetic regulation in sperm maturation, fertilization, and embryogenesis. From the point of view of transgenerational inheritance, the analysis of sperm epigenome could provide data related to abnormal epigenetic inheritance through the male germline, clarifying the possible effects on offspring health. Overall, these aspects could step into a leading role the sperm epigenome in the field of human reproduction. In this view, it is …

Proteomic identification of sperm from mice exposed to sodium fluoride.

Fluoride is a widespread environmental pollutant which can induce low sperm quality and fertilizing ability. However, effect of fluoride on proteomic changes of sperm is unknown. In this study, two-dimensional electrophoresis (2DE) and mass spectrometry (MS) were used to investigate the differently expressed proteins of sperm from mice exposed to fluoride. 180 male mice were randomly divided into three groups, and were administrated with the distilled water containing 0, 25, and 100 mg L-1 NaF, respectively. After 45, 90 and 180 day's exposure, mice were sacrificed and sperm from the cauda epi

Alteration of transporter activities in the epididymides of infertile initial segment-specific Pten knockout mice.

A fully functional initial segment, the most proximal region of the epididymis, is important for male fertility. Our previous study generated a mouse model to investigate the importance of initial segment function in male fertility. In that model, phosphatase and tensin homolog (Pten) was conditionally removed from the initial segment epithelium, which resulted in epithelial de-differentiation. When spermatozoa progressed through the de-differentiated epithelial duct, they developed angled flagella, suggesting compromised sperm maturation, which eventually resulted in male infertility. To unde…

Fibrinogen in Hamster Cauda Sperm.

Epididymis plays a vital role in promoting sperm maturation and maintaining sperm viability. It has been shown the presence of nonviable sperm in cauda epididymis. We previously identified a secretory protein (260/280KDa oligomers) of hamster cauda epididymal principal cells that binds to nonviable sperm. The 260/280KDa oligomers are composed of 64kDa FGL2 (fibrinogen-like protein-2) and 33kDa FGL1) (fibrinogen-like protein-1). In addition, we have demonstrated that FGL2 is a phospholipid-activated serine protease; the conversion of prothrombin to thrombin by FGL2 followed by the conversion of…

Hydrogen Sulfide as a Mediator of Protein Persulfidation and Motility Regulation in Mammalian Sperm.

AIMS: Hydrogen sulfide (H2S) is a signaling molecule implicated in diverse physiological processes, yet its role in sperm function remains poorly understood. This study characterizes protein persulfidation, a post-translational modification mediated by H2S in boar spermatozoa, tracks its distribution during sperm maturation, and assesses whether H2S influences sperm motility during capacitation. METHODS: Persulfidation was localized using fluorescent labeling and confirmed by Western blotting across epididymal and post-ejaculatory stages. LC-MS/MS identified persulfidated proteins, which were…

Transfer at the blastocyst stage of embryos derived from testicular round spermatid injection.

BACKGROUND: Intracytoplasmic injection of testicular round spermatids has been suggested as a salvage treatment in couples when testicular sperm extraction does not yield any mature sperm. However, the success of the procedure is debatable, and controversy surrounds issues such as the presence and (if present) identification of spermatids in testicular tissue. Progression rate to the blastocyst stage of spermatid-derived embryos appears to be low. METHODS: In this study, we investigated the feasibility and outcome of blastocyst stage embryo transfer after round spermatid injection (ROSI). ROS

[Methylation pattern of imprinted genes H19 and IGF2r in spermatogenic cells cultured in vitro].

OBJECTIVE: To compare the methylation patterns of imprinting control region (ICR) of H19 gene and differentially methylated region (DMR) of IGF2r gene in mature sperms derived from epididymis of Kunming mice and in vitro cultured haploid spermatids. METHODS: The H19 ICR and IGF2r DMR2 were detected by bisulfite sequencing PCR (BSP). The results were compared with standard sequence derived from GenBank using a DNAman software. RESULTS: 96.67% (15 CpG sites) of H19 ICR was found to be methylated, and 94.29% IGF2r DMR2 was found to be unmethylated in mature sperms. By contrast, 69.33% of H19 IC

The role of galectin-3 in spermatozoa-zona pellucida binding and its association with fertilization in vitro.

Human spermatozoa can fertilize an oocyte only after post-testicular maturation and capacitation. These processes involve dynamic modification and reorganization of the sperm plasma membrane, which allow them to bind to the zona pellucida (ZP) of the oocyte. Defective sperm-ZP binding is one of the major causes of male subfertility. Galectin-3 is a secretory lectin in human seminal plasma well known for its action on cell adhesion. The aim of this study was to determine the role of galectin-3 in spermatozoa-ZP interaction and its association with fertilization rate in clinical assisted reprodu

The blood-epididymis barrier and inflammation.

The blood-epididymis barrier (BEB) is a critical structure for male fertility. It enables the development of a specific luminal environment that allows spermatozoa to acquire both the ability to swim and fertilize an ovum. The presence of tight junctions and specific cellular transporters can regulate the composition of the epididymal lumen to favor proper sperm maturation. The BEB is also at the interface between the immune system and sperm. Not only does the BEB protect maturing spermatozoa from the immune system, it is also influenced by cytokines released during inflammation, which can res…
