Role of intracranial bone marrow mesenchymal stem cells in stroke recovery: A focus on post-stroke inflammation and mitochondrial transfer.

Stem cell therapy has become a hot research topic in the medical field in recent years, with enormous potential for treating a variety of diseases. In particular, bone marrow mesenchymal stem cells (BMSCs) have wide-ranging applications in the treatment of ischemic stroke, autoimmune diseases, tissue repair, and difficult-to-treat diseases. BMSCs can differentiate into multiple cell types and exhibit strong immunomodulatory properties. Although BMSCs can regulate the inflammatory response activated after stroke, the mechanism by which BMSCs regulate inflammation remains unclear and requires fu

Fine structure of progenitor cells in early ectopic human embryos.

This study has documented the major types of lineage progenitor cells at the second level of cell differentiation after the establishment of … enitor cells, including those for amniotic, yolk sac and chorionic tissues that are used in current stem cell research. Neural induction by the notochord has been imaged. This study should give valuable clues to understand the pattern of cell differentiation of embryonic stem cells (ESC) in vitro, which are more or less mimicked in ESC colonies, embryoid bodies and …

The potential of alternate sources of cells for neural grafting in Parkinson's and Huntington's disease.

Cell-based therapies for Parkinson's and Huntington's disease have provided mixed clinical outcomes and one of the reasons underlying this is the use of primary fetal tissue as the source of grafted cells. An alternate source of cells, such as stem cells, could overcome many of the issues associated with primary fetal tissue and would help bring forward cell replacement therapy as a reliable and effective treatment for these two neurodegenerative disorders. This review will discuss which stem cells are likely to go to clinic in the next generation of cells, based on trials for Parkinson's and

Bone marrow-derived mesenchymal stem cells for ischemic stroke: mechanisms, delivery strategies, clinical evidence, and translational challenges.

Ischemic stroke remains a major global health burden because of its high incidence, disability, and mortality. Current reperfusion therapies are constrained by narrow therapeutic windows and have limited capacity to reverse established neuronal injury or promote long-term functional recovery, highlighting the need for strategies that support neural repair and neurovascular remodeling. Bone marrow-derived mesenchymal stem cells (BMSCs) have been widely investigated as candidate therapeutic products for ischemic stroke because of their paracrine, immunomodulatory, trophic, angiogenic, and microe

Cell Therapy for Ischemic Stroke: How to Turn a Promising Preclinical Research into a Successful Clinical Story.

Stroke is a major public health issue with limited treatment. The pharmacologically or mechanically removing of the clot is accessible to less than 10% of the patients. Stem cell therapy is a promising alternative strategy since it increases the therapeutic time window but many issues remain unsolved. To avoid a new dramatic failure when translating experimental data on the bedside, this review aims to highlight the indispensable checkpoints to make a successful clinical trial based on the current preclinical literature. The large panel of progenitors/ stem cells at the researcher's disposal i

Adult stem cell therapy in stroke.

PURPOSE OF REVIEW: Acute cerebral infarction causes irreversible locally restricted loss of the neuronal circuitry and supporting glial cells with consecutive functional deficits and disabilities. The currently available and effective therapy targets fast vessel recanalization accompanied by symptomatic measures. Research activities focusing on stem cells, which represent a promising source for organotypic cell replacement and functional recovery after stroke, have gained momentum in recent years, making regenerative cell-based therapies a much more feasible realistic approach. New perspective of skeletal stem cells.

Tissue-resident stem cells are a group of stem cells distinguished by their capacity for self-renewal and multilineage differentiation capability with tissue specificity. Among these tissue-resident stem cells, skeletal stem cells (SSCs) were discovered in the growth plate region through a combination of cell surface markers and lineage tracing series. With the process of unravelling the anatomical variation of SSCs, researchers were also keen to investigate the developmental diversity outside the long bones, including in the sutures, craniofacial sites, and spinal regions. Recently, fluoresce

Engrafted stem cell therapy for Alzheimer's disease: A promising treatment strategy with clinical outcome.

To date, although the microscopic alterations present in Alzheimer's disease (AD) have been well known for over a century only a handful of symptomatic treatments have been developed which are a far cry from a full cure providing volatile benefits. In this context, the intervention of stem cell therapy (SCT) has been proposed as an auxiliary treatment for AD as suggested by the rising number of pre-clinical studies that stem cell engraftment could provide an exciting future treatment regimen against neurodegeneration. Although, most of the primary enthusiasm about this approach was based on re

[Stem cell therapy for non-obstructive azoospermia].

Non-obstructive azoospermia (NOA) is an important factor that causes male infertility. Stem cells are a group of cells capable of self-renewal and multi-directional differentiation, and embryonic stem cells and induced pluripotent stem cells can generate spermatozoa through differentiation, which, however, is confronted with ethical constraints and the risk of tumorigenesis. Spermatogonial stem cells can produce haploid gametes by differentiation but human spermatogonial stem cells are difficult to be cultured in vitro. Mesenchymal stem cells promote spermatogenesis through paracrine activity,

Dental application potential of mesenchymal stromal cells and embryonic stem cells.

In the past decade, research on the potential applications of stem cells in dentistry has made great progress. There are at least five different types of multipotent mesenchymal stromal cells (MSCs) originating from exfoliated primary teeth, including dental pulp, apica papilla, periodontal ligament, and dental follicle. It has been reported that dental tissue-derived MSCs are able to generate dentine-pulp-like complexes as well as differentiate into periodontal and craniofacial progenitor cells. Similar to these dental tissue-derived MSCs, bone marrowderived MSCs are also capable of developin

Skeletal Changes After Hematopoietic Stem Cell Transplantation in Osteopetrosis.

Osteopetrosis is a rare skeletal dysplasia resulting from an osteoclast defect leading to increased bone mass and density. Hematopoietic stem cell transplantation can rescue the disease phenotype and prevent complications. However, little is known about the skeletal changes hematopoietic stem cell transplantation induces in patients with this disease. The purpose of this study was to describe the skeletal changes after hematopoietic stem cell transplantation in a retrospective cohort of patients diagnosed with osteopetrosis in one medical center over 13 years. For this purpose, all available e

Comparative analysis of dental-derived stem cells and alternative stem cell sources: properties and regenerative functions.

Due to their accessibility, wide range of sources and unique biological characteristics, dental stem cells have broad application prospects in regenerative medicine. This cell population mainly includes dental pulp stem cells, periodontal ligament stem cells, stem cells from deciduous teeth, and dental follicle stem cells. In addition, dental stem cells have good microenvironment-specific immunomodulatory functions, including inhibiting T cell activation, promoting the polarization of regulatory T cells and regulating the phenotype of macrophages, thereby promoting tissue repair and reducing i

Development of Stem Cell Therapies for Retinal Degeneration.

Degenerative retinal disease is the major cause of sight loss in the developed world and currently there is a lack of effective treatments. As the loss of vision is directly the result of the loss of retinal cells, effective cell replacement through stem-cell-based therapies may have the potential to treat a great number of retinal diseases whatever their underlying etiology. The eye is an ideal organ to develop cell therapies as it is immune privileged, and modern surgical techniques enable precise delivery of cells to the retina. Furthermore, a range of noninvasive diagnostic tests and high-

Generation of insulin-producing beta cells from stem cells--perspectives for cell therapy in type 1 diabetes.

Cell based therapy for the treatment of type 1 diabetes is limited by the overall shortage of donor organs for transplantation. This is the rationale for the research on the generation of insulin-producing beta cells from an inexhaustible source of cells such as the stem cells. Stem cells are progenitor cells which possess the capacity of self-renewing and differentiation in fully mature cells depending on the culture conditions. The fundamental question is how to make terminally matured pancreatic beta cells. During the last years different approaches for the neogenesis of beta cells have bee

Heart regeneration using reprogramming technology.

Loss of terminally differentiated cardiomyocytes due to heart disease is irreversible and current therapeutic regimes are limited. Cell therapy using stem cell-derived cardiomyocytes is an attractive option to repair injured hearts. The discovery of direct reprogramming of fibroblasts into induced pluripotent stem cells (iPSCs) and successful differentiation of iPSCs into cardiomyocytes provided a revolutionary paradigm in heart regenerative research. During the past decades, significant advances in stem cell culture, differentiation and purification protocols, as well as in cell transplantati

The potential role of adult stem cells in the management of the rheumatic diseases.

Adult stem cells are considered as appealing therapeutic candidates for inflammatory and degenerative musculoskeletal diseases. A large body of preclinical research has contributed to describing their immune-modulating properties and regenerative potential. Additionally, increasing evidence suggests that stem cell differentiation and function are disrupted in the pathogenesis of rheumatic diseases. Clinical studies have been limited, for the most part, to the application of adult stem cell-based treatments on small numbers of patients or as a 'salvage' therapy in life-threatening disease cases
