Histochemical analysis of glycoconjugate secretion in the alimentary canal of Anguilla anguilla L.

Conventional histochemical methods as well as lectin-binding techniques were used to study glycoconjugates that are present in the alimentary canal of the European eel (Anguilla anguilla). Specimens from pharynx, oesophagus, stomach and intestine were collected from adult ("silver eel" stage) females. Alcian Blue pH 2.5/PAS and High Iron Diamine/Alcian Blue pH 2.5 reactions were performed to stain neutral and acidic glycoconjugates. In addition, lectin histochemistry was applied to identify acidic glycoconjugates containing O-acylated sialic acids. Finally, the presence of sugar residues in th

Postharvest Immersion in Slightly Acidic Electrolyzed Water Improves Guava Storability by Regulating Phenylpropane Metabolism.

Postharvest guava fruit are at high risk of decay and spoilage, which extremely restrains the further advancement of guava industry in China. Currently, slightly acidic electrolyzed water (SAEW) has been shown to be potent in maintaining the storability of fruits and vegetables. Accordingly, this work was designed to figure out the effectiveness of SAEW on storability maintenance in postharvest guavas via regulating the phenylpropane metabolism. On the harvest day, fresh guavas were immersed in distilled water or SAEW (available chlorine concentration: 30 mg L-1) for 10 min, followed by storag

Gas chromatography-mass spectrometric method-based urine metabolomic profile of rats with pelvic inflammatory disease.

Pelvic inflammatory disease (PID) can lead to a poor outcome of severe sequelae, and the current methods of clinical diagnosis are not satisfactory. Metabolomics is an effective method for the identification of disease-related metabolite biomarkers to facilitate disease diagnosis. However, to the best of our knowledge, no PID-associated metabolomic study has yet been carried out. The metabolomic changes of rats with PID were investigated in the present study. A PID model was constructed by the multi-pathogenic infection of the upper genital tract in rats. Infiltration of inflammatory cells and

Comparison of optoacoustic and photothermal-lens determination of lipopolysaccharides.

As a primary aim, several approaches to photothermal (thermal-lens) and optoacoustic determination of exogenous pyrogens (lipopolysaccharides) based on photometric procedures for their determination at the microgram level and below are compared. The limits of detection of lipopolysaccharides by thermal-lens spectrometry and optoacoustic spectroscopy are at a level of 2-100 ng/mL, and the conditions of optoacoustic and photothermal procedures are the same. Optoacoustic spectroscopy is advantageous in determining suspensions, while thermal lensing is superior in determining lipopolysaccharides f

Antipyrine pharmacokinetics and D-glucaric excretion in kwashiorkor.

Hepatic microsomal oxidation and glucuronidation were studied in 15 children with kwashiorkor on admission to the hospital and again after 3 weeks of nutritional rehabilitation. Microsomal oxidation as measured by antipyrine half-life and clearance was shown to be depressed in the acute phase of malnutrition (T 1/2 = 7.9 +/- 5.0 hr, clearance = 8.4 +/- 5.1 ml/min) improving with nutritional rehabilitation (T 1/2 = 4.3 +/- 2.3 hr, clearance = 15.5 +/- 8.7 ml/min). Urinary D-glucaric acid excretion increased from 60.6 +/- 42.2 mumoles/24 hr to 121.8 +/- 105.0 mumoles/24 hr over the same time per

Effect of sweeteners and sweetness enhancers on weight management and gut microbiota composition in individuals with overweight or obesity: the SWEET study.

Consumption of sweeteners and sweetness enhancers (S&SEs) is a popular strategy to reduce sugar intake, but the role of S&SEs in body weight regulation and gut microbiota composition remains debated. Here, we show that S&SEs in a healthy diet support weight loss maintenance and beneficial gut microbiota shifts in adults with overweight or obesity. In this multi-centre, randomized, controlled trial, we included 341 adults and 38 children with overweight or obesity. Adults followed a 2-month low-energy diet for ≥5% weight loss, followed by a 10-month healthy ad libitum diet with <10% energy from

How Does Lagenaria siceraria (Bottle Gourd) Metabolome Compare to Cucumis sativus (Cucumber) F. Cucurbitaceae? A Multiplex Approach of HR-UPLC/MS/MS and GC/MS Using Molecular Networking and Chemometrics.

Cucurbitaceae comprises 800 species, the majority of which are known for their nutritive, economic, and health-promoting effects. This study aims at the metabolome profiling of cucumber (Cucumis sativus) and bottle gourd (Lagenaria siceraria) fruits in a comparative manner for the first time, considering that both species are reported to exhibit several in-common phytochemical classes and bioactivities. Nevertheless, bottle gourd is far less known and/or consumed than cucumber, which is famous worldwide. A multiplex approach, including HR-UPLC/MS/MS, GNPS networking, SPME, and GC/MS, was emplo

Mixed-phase weak anion-exchange/reversed-phase LC-MS/MS for analysis of nucleotide sugars in human fibroblasts.

Nucleotide sugars (NS) fulfil important roles in all living organisms and in humans, related defects result in severe clinical syndromes. NS can be seen as the "activated" sugars used for biosynthesis of a wide range of glycoconjugates and serve as substrates themselves for the synthesis of other nucleotide sugars. NS analysis is complicated by the presence of multiple stereoisomers without diagnostic transition ions, therefore requiring separation by liquid chromatography. In this paper, we explored weak anion-exchange/reversed-phase chromatography on a hybrid column for the separation of 17

Glycaemic sugar metabolism and the gut microbiota: past, present and future.

Non-communicable diseases (NCDs), such as type 2 diabetes (T2D) and metabolic dysfunction-associated fatty liver disease, have reached epidemic proportions worldwide. The global increase in dietary sugar consumption, which is largely attributed to the production and widespread use of cheap alternatives such as high-fructose corn syrup, is a major driving factor of NCDs. Therefore, a comprehensive understanding of sugar metabolism and its impact on host health is imperative to rise to the challenge of reducing NCDs. Notably, fructose appears to exert more pronounced deleterious effects than glu

Structure, pathology and function of the N-linked sugar chains of human chorionic gonadotropin.

Human chorionic gonadotropin (hCG) contains five acidic N-linked sugar chains, which are derived from three neutral oligosaccharides by sialylation. Each of the two subunits (hCGalpha and hCGbeta) of hCG contain two glycosylated Asn residues. Glycopeptides, each containing a single glycosylated Asn, were obtained by digestion of hCGalpha with trypsin, and of hCGbeta with chymotrypsin and lysyl endopeptidase. Comparative study of the sugar chains of the four glycopeptides revealed the occurrence of site-directed glycosylation. Studies of the sugar chains of hCGs, purified from urine of patients

Food processing and storage as a determinant of protein and amino acid availability.

Protein is perhaps the most reactive of the major food components. During food processing, the essential amino acids, lysine, tryptophan, methionine and cyst(e)ine, may react with other food components causing a loss in amino acid bioavailability and sometimes a reduction in the digestibility of the whole protein molecule. This review first discusses some recent developments concerning protein-polyphenol reactions, racemization and lysinoalanine formation, and then describes the reactions of proteins and reducing sugars (the Maillard reaction) in greater detail. We report on the chemistry of t

Integrated metabolite, volatile, and genome annotation profiling reveals distinct functional phenotypes in cocoa-derived yeasts.

Cocoa fermentation shapes the precursor chemistry underlying chocolate flavour, but the isolate-level contributions of cocoa-associated yeasts remain incompletely resolved. This study compared six cocoa-derived yeasts with a reference Saccharomyces cerevisiae strain by profiling sugar utilisation, organic-acid turnover, volatile formation, and targeted genome annotation under controlled cocoa-relevant conditions. In cocoa pulp extract-based medium, Kluyveromyces marxianus Y1A, Meyerozyma guilliermondii Y1, and Ogataea polymorpha Y3A showed the strongest sugar depletion and organic-acid transfo

The possible relevance of autoxidative glycosylation in glucose mediated alterations of proteins: an in vitro study on myofibrillar proteins.

The present work was carried out to examine the role of glycation and transition metal catalysed autoxidation of sugars in glucose-mediated alterations of myofibrillar proteins. Myofibrils were prepared from rat skeletal muscle and incubated with 1) sugar alone 2) sugar and micromolar concentrations of transition metals (Cu2+ or Fe3+) 3) transition metals alone and the control remained without sugar or transition metals. A significant increase in extent of glycation and decrease in ATPase activity of myofibrils incubated under autoxidative conditions were observed over the other three incubati

Energy metabolism of a unique acetic acid bacterium, Asaia bogorensis, that lacks ethanol oxidation activity.

Acetic acid bacteria (AAB) are known as a vinegar producer on account of their ability to accumulate a high concentration of acetic acid due to oxidative fermentation linking the ethanol oxidation respiratory chain. Reactions in oxidative fermentation cause poor growth because a large amount of the carbon source is oxidized incompletely and the harmful oxidized products are accumulated almost stoichiometrically in the culture medium during growth, but a newly identified AAB, Asaia, has shown unusual properties, including scanty acetic acid production and rapid growth, as compared with known AA

Burkholderia kururiensis sp. nov., a trichloroethylene (TCE)-degrading bacterium isolated from an aquifer polluted with TCE.

A trichloroethylene (TCE)-degrading bacterium was isolated from an aquifer sample collected at a TCE-polluted site in Japan by enriching with phenol as sole carbon source. The isolate, designated strain KP23T, was a Gram-negative, oval-shaped micro-organism. A phylogenetic study based on 16S rRNA gene sequences indicated that strain KP23T should be placed in the genus Burkholderia. Cellular fatty acids of the strain were mainly composed of C16:0, cyclopropanic acid C17:0 and cyclopropanic acid C19:0. Strain KP23T also contained notable amounts of C13:1 and C17:1. The G + C content of total DNA

New antifungal proteins from sugar beet (Beta vulgaris L.) showing homology to non-specific lipid transfer proteins.

Two novel, nearly identical antifungal proteins, IWF1 and IWF2, were isolated from the intercellular washing fluid (IWF) of sugar beet leaves. The proteins were purified to homogeneity and their amino acid sequences were determined. They are basic, monomeric proteins of 91 amino acid residues, 89 of which are identical. Both proteins show strong in vitro antifungal activity against Cercospora beticola, the casual agent of leaf spot disease in sugar beet. Based on primary sequence homology, including the presence of 8 conserved cysteine residues, IWF1 and IWF2 are related to the family of plant

Elucidating the mechanism of action of copper heptagluconate on the plant immune system against Pseudomonas syringae in tomato (Solanum lycopersicum L).

BACKGROUND: Phytopathogenic problems caused by the bacterial pathogen Pseudomonas syringae in tomato are becoming more serious due to the emergence of strains resistant to classical pesticides. This has led to research into new formulations with lower environmental problems. One of the most promising alternatives to the use of classical pesticides is the induction of natural plant defences. New formulations based on Cu complexed with heptagluconic acid induce plant innate defences and could be an alternative to classical treatments based on inorganic Cu against bacterial speck. To study the ef

Microbial D-xylonate production.

D-Xylonic acid is a versatile platform chemical with reported applications as complexing agent or chelator, in dispersal of concrete, and as a precursor for compounds such as co-polyamides, polyesters, hydrogels and 1,2,4-butanetriol. With increasing glucose prices, D-xylonic acid may provide a cheap, non-food derived alternative for gluconic acid, which is widely used (about 80 kton/year) in pharmaceuticals, food products, solvents, adhesives, dyes, paints and polishes. Large-scale production has not been developed, reflecting the current limited market for D-xylonate. D-Xylonic acid occurs n
