Linear alkyl carbonate solvents such as dimethyl carbonate (DMC) are considered to be environmentally safe with a low toxicology risk. The modest viscosity of 0.625 mPa·s and the UV cutoff wavelength of about 215 nm makes DMC a potential alternative aprotic HPLC modifier solvent to acetonitrile. We have developed a generally applicable methodology for cysteine mutagenesis of nanobody (Nb) framework region serine residues. This strategy allows for subsequent labeling with thiol-reactive compounds without disrupting Nb antigen binding. We provide a protocol for production, labeling, and affinity determination of cysteine-engineered Nbs (cys-Nbs) with Alexa Fluor 488-maleimide and the mercury compound para-chloromercuribenzoic acid (PCMB). Alexa Fluor 488- and PCMB-labeled cys-Nbs can be used for immunofluorescence microscopy and experimental phasing in crystallography, respectively.

In order to monitor loco-regional anaesthesia in the upper limbs with a combination of two anaesthetics, the authors participated in a clinical pharmacology study. They describe a method of simultaneous assay of bupivacaine and lignocaine by direct phase HPLC. In particular, the use of heptane sulfonic acid (counter-ion) in the mobile phase allows the elution of the local anaesthetics to be controlled. The criteria of reliability studied enable this technique to be considered for routine use.

Skin diseases due to Nocardia are rare in Europe and usually occur as the secondary localization of a pulmonary lesion in immunocompromised patients. We describe a cutaneous pyoderma and cold abscess due to Nocardia asteroides infection in a cardiac transplant recipient. An insect bite represents the wound of this soil saprophyte belonging to the actinomycetes. Treatment by sulfone and ampicillin-clavulanic acid allowed the regression of lesions without extracutaneous dissemination.

Human scalp hair samples were oxidized to determine the sulfonic acid absorption peak. This peak was monitored at 1044 cm-1 by Fourier transform infrared spectroscopy (FTIR) in hair samples from 135 whites and found to provide a degree of discrimination in treated and untreated hairs. The effects of moisture, laboratory storage, natural hair color, and variation over time were also studied.
