Plasma melatonin during desmethylimipramine treatment: evidence for changes in noradrenergic transmission.

Plasma melatonin was used to determine overall beta-adrenergic transmission through pineal neuro-effector junctions during desmethylimipramine (DMI) treatment in 10 normal subjects. Changes in plasma melatonin indicated that an initial increase in noradrenaline (NA) transmission produced by DMI was counteracted by adaptive changes which restored transmission to normal by the third week of treatment. A 'rebound' increase in NA transmission was seen on DMI withdrawal. The results suggest that the adaptive changes which occur in NA synapses during DMI treatment do not, as has been proposed, decre

Synaptic vesicle cycling at type-identified diaphragm neuromuscular junctions.

Differences in neuromuscular transmission and neuromuscular junction morphology exist across muscle fiber types. We hypothesized that these fiber-type differences are reflected in the size of the cycling synaptic vesicle pool. Synaptic vesicle cycling at type-identified rat diaphragm neuromuscular junctions was examined by fluorescently labeling presynaptic vesicles with FM4-64. We found that FM4-64 fluorescence uptake was higher at presynaptic terminals of type I/IIa fibers than type IIx/IIb fibers. However, no fiber-type differences in the rate of FM4-64 destaining were found with repetitive

Multiple Mechanisms of Dopamine Modulation of Neuronal Excitability and Neurotransmission in the Striatum: A Personal and Historical Perspective.

3,4-dihydroxyphenethylamine, commonly known as dopamine (DA), is a neuromodulator that fine-tunes neuronal excitability, neurotransmitter release, and the effects of other neurotransmitters on postsynaptic neurons. DA, acting on D1 and D2 receptor families, is involved in myriad functions. In the striatum, it is mainly implicated in motor control, motivation, and reward mechanisms. In the cerebral cortex it participates in attention processes, working memory, long-term memory, etc. DA overproduction or deficits lead to neuronal circuit imbalance that underlies a number of neurological and psyc

Chronic exposure to environmental concentrations of 6PPDQ induces neurotoxicity via perturbing multiple neurotransmission pathways in zebrafish.

Previous research has demonstrated acute toxicity of 6PPD-quinone (6PPDQ), the ozonation product of tire-manufacturing additive 6PPD; however, chronic effects on aquatic organisms under real-world conditions remain unclear. We exposed 2-month-old zebrafish to 6PPDQ at concentrations of 2, 20 and 200 µg/L for 30 days. Whole-body desorption electrospray ionization-mass spectrometry (DESI-MS) revealed accumulation mainly in the muscle, abdomen, and brain of zebrafish exposed to 200 µg/L 6PPDQ, which was verified by MS quantification of muscle and brain tissues. Based on velocity and distance move

Structure and function of the neuromuscular junction in health and myasthenia gravis.

The neuromuscular junction (NMJ) connects a motor neuron to a skeletal muscle cell. Cholinergic synaptic transmission enables muscle contraction, which is crucial for survival. Although synaptic communication at the NMJ is robust, with an inherent safety margin, it becomes hampered in the neuro-immunological disorder myasthenia gravis (MG). The synaptic dysfunction underlies the (fatigable) muscle weakness, which hallmarks this disease. In this chapter, we will review normal NMJ physiology and the pathophysiological consequences of autoimmune attacks in MG, focusing on recent insights and deve…

Ribbon Synapses and Retinal Disease: Review.

Synaptic ribbons are presynaptic protein complexes that are believed to be important for the transmission of sensory information in the visual system. Ribbons are selectively associated with those synapses where graded changes in membrane potential drive continuous neurotransmitter release. Defective synaptic transmission can arise as a result of the mutagenesis of a single ribbon component. Visual diseases that stem from malfunctions in the presynaptic molecular machinery of ribbon synapses in the retina are rare. In this review, we provide an overview of synaptopathies that give rise to reti…

Hetero-pentamerization determines mobility and conductance of Glycine receptor α3 splice variants.

Glycine receptors (GlyRs) are ligand-gated pentameric chloride channels in the central nervous system. GlyR-α3 is a possible target for chronic pain treatment and temporal lobe epilepsy. Alternative splicing into K or L variants determines the subcellular fate and function of GlyR-α3, yet it remains to be shown whether its different splice variants can functionally co-assemble, and what the properties of such heteropentamers would be. Here, we subjected GlyR-α3 to a combined fluorescence microscopy and electrophysiology analysis. We employ masked Pearson's and dual-color spatiotemporal correla

Introduction to the special issue "Pharmacotherapies for the treatment of alcohol abuse and dependence" and a summary of patents targeting other neurotransmitter systems.

This paper introduces the Special Section: Pharmacotherapies for the Treatment of Alcohol Abuse and Dependence and provides a summary of patents targeting neurotransmitter systems not covered in the other four chapters. The World Health Organization notes that alcoholic-type drinking results in 2.5 million deaths per year, and these deaths occur to a disproportionately greater extent among adolescents and young adults. Developing a pharmacological treatment targeting alcohol abuse and dependence is complicated by (a) the heterogeneous nature of the disease(s), (b) alcohol affecting multiple ne

[Use of snake venom proteins in medicine].

Snakes feed exclusively on freshly killed prey animals which, following their immobilization, have to be swallowed whole. Venomous snakes effect prey immobilization by injection of their venom. Snake venoms are highly concentrated, complex mixtures of individual proteins which, either as enzymes, enzyme effectors or blocking ligands, acting as single agents or in synergistic conjunction with other venom components, modify vital structures of the prey organism to destroy their biological function. Predominantly neurotoxic venoms paralyze respiratory activity by pre- or postsynaptic blockade of

[Relationship between the hippocampal formation and the cortical and subcortical regions].

A summary of afferent and efferent connections of the hippocampal formation has been reviewed, especially outputs from the subicular complex. The hippocampal formation receives much information from widely spread cortical and subcortical regions, and in turn projects principally to many of the cortical regions. Particularly, the entorhinal area of primates receives a large number of outputs from cortical regions containing the prefrontal, insular, temporal, parietal, cingular, and retrosplenial regions. On the other hand, the cortex of both rodents and primates projects to almost all of the co

Disorders of neuromuscular transmission due to natural environmental toxins.

A variety of natural toxins of animal, plant, and bacterial origin are capable of causing disorders of neuromuscular transmission. Animal toxins include venomous snakes and arthropods, venoms of certain marine creatures, skin secretions of dart-poison frogs, and poisonous fish, shellfish, and crabs. There are plant poisons such as curare, and bacterial poisons such as botulinum toxin. These act at single or multiple sites of the neuromuscular apparatus interfering with voltage-gated ion channels, acetylcholine release, depolarization of the postsynaptic membrane, or generation and spread of th

AAEE Minimonograph #33: electrodiagnostic approach to defects of neuromuscular transmission.

Clinical testing for neuromuscular dysfunction is supported by an extensive amount of excellent basic information about normal and abnormal subcellular physiology and ultrastructure. This information provides an essential frame of reference for describing the rationale of single-fiber electromyography (SFEMG). SFEMG in turn helps to explain the more conventional clinical testing of neuromuscular function by repetitive nerve stimulation (RNS). Electrical findings in myasthenia gravis, Lambert-Eaton myasthenic syndrome, and botulinum intoxication are discussed from the subcellular level via the

Transmission in squid giant synapses: the importance of oxygen supply and the effects of drugs.

Synaptic transmission was studied in giant synapses of the stellate ganglion of the squid. When bathed in air-saturated sea water, the synapses deteriorate in 10 to 20 min.; if the sea water is saturated with 100 per cent oxygen, they function steadily for up to 12 hours. Optimal results probably require a medium with lower magnesium and higher calcium than the sea water used. Of eighteen compounds known to affect other synapses (Table I), none had stimulatory effects when applied to the preparation, but ten produced synaptic depression in concentrations of 10(-3) gm. per ml. or higher. The on…
